Studies on Oleoresin Product from Spices 2. Quality Stability of Red Pepper Oleoresin (향신재료를 이용한 Oleoresin제조에 관한 연구 2. 고추 Oleoresin의 품질안정성)

  • Thermal stability and quality change during storage of red pepper oleoresin were studied. Upon heat treatment, carotenoids in oleoresin from red pepper were more stable under nitrogen than air. In the thermal stabilities, the presence of air at $100^{\circ}C$ for 3 hours and 10 hours were 69.1% and 42.3%, respectively ; whereas, in the presence of nitrogen, 95.4% and 92.3%, respectively. In contrast, capsaicin was comparatively stable upon heat treatment in the presence of air. The retentions of capsaicin at $100^{\circ}C$ for 3 hours and 10 hours were 84.7% and 81.3% with air, those were 90.7% and 87.5% with nitrogen, respectively. Color appearance showed close relation to the stability of total carotenoids during 60 days storage at varying temperatures ; $5^{\circ}C,\;25^{\circ}C\;and\;40^{\circ}C$ were 69.4%, 48.0% and 35.1%, respectively. The degradation of total carotenoids during storage could be interpreted as a first order reaction. The activation energy obtained from the Arrhenius plot was 5.03 kcal/mole. Conversely, capsaicin was very stable under storage temperatures. More than 90% of capsaicin was remained upon completion of storage. In the mean time, pH of oleoresin was increased slightly as increasing storage temperatures.

Biochemical Changes Induced due to Staphylococcal Infection in Spongy Alphonso Mango(Mangifera indica L.) Fruits

  • Spongy Alphonso mangoes were found to be infected with Staphylococcus bacteria. A Gram positive Staphylococcus strain was isolated from spongy pulp and identified from CABI Bioscience, UK, by partial 16S rDNA sequence analysis and by morphological and biochemical characterization through IMTECH, Chandigarh, India. Although identification by both of these methods indicated the organism belonged to same genus, different species names were given. Changes in total phenolics, reducing, and non-reducing sugars, respiration rate, total carotenoids, peroxidase(POX), and catalase activities were monitored during ripening of these fruits. The climacteric rise in spongy fruits was marked by an increase in respiration rate and a decrease in sugar content. Total phenolics content increased in spongy fruits as compared to ripe non-spongy fruits. Development of corky white tissue in spongy fruits was associated with about a 2.5-fold reduction in total carotenoids and a concomitant increase in lipoxygenase-mediated, $\beta$-carotene co-oxidation. A marked decrease in soluble protein content and about a 1.5-fold increase in POX activity was observed. Maximum POX activity was confined to 50-70%$(NH_4)_2SO_4$ fraction. The intense dark bands visible after POX specific substrate staining of the Native gel indicated a high expression of isoenzymes of POX in spongy fruits. Similarly, changes in levels of catalase activity were also observed in spongy fruits. The results suggest that infection of Alphonso mangoes with Staphylococcus bacteria affects the normal ripening processes of the fruit interfering with the carbohydrate and carotenoid metabolism. Also, the studies indicate the expression of POX and catalase enzymes as a plant defense response to microbial invasion.

Comparison of Autoxidative Stability and Minor Compounds in Oils Extracted from Bran and Germ of Keumkang Wheat and Dark Northern Spring Wheat (금강밀과 dark northern spring밀의 기울과 배아에서 추출한 기름의 자동산화 안정성 및 미량성분 비교)

  • Autoxidative stability of wheat bran and germ oil extracted from Keumkang wheat (WBG-K) or Dark Northern Spring wheat (WBG-DNS) at $50^{\circ}C$ in the dark was compared by peroxide values (POV) and fatty acid composition by gas chromatography. Changes of minor compounds were monitored by HPLC. WBG-K showed significantly higher linoleic but lower oleic acid content than WBG-DNS. WBG-DNS contained more phospholipids but less tocopherols and carotenoids than WBG-K. POV of oils was increased during storage, with no significant difference in the oxidation rates between two oils. Tocopherols, carotenoids, and phospholipids in both oils were degraded during the autoxidation. Total phospholipids content showed the highest correlation with the degree of oxidation in WBG oils. The results clearly showed that both fatty acid composition and contents of tocopherols, carotenoids, and phospholipids co-affected the autoxidation of WBG oil.

  • Seasonal and regional variation of carotenoid contents in the muscles and tunic of the cultured ascidian(Halocynthia roretzi) was investigated. Carotenoid contents of tunic(47.87 mg/100g wet base) was much higher than that of muscles(2.35mg/100g wet base). The carotenoid contents was increased from April to August and then decreased in September. A total of 13 components were separated from the carotenoids extracted from ascidian tunic. The carotenoids of ascidian tunic accounted for followed by those of alloxanthin($31.3\%$), halocynthiaxanthin($15.5\%$), diatoxanthin($11.9\%$), diadinochrome($11.6\%$), mytilo-xanthin($10.8\%$), and astaxanthin($7.8\%$).

The Study of Lipid-peroxidation, Antioxidant Enzymes, and the Antioxidant Vitamins in NIDDM Patients with Microvascular-diabetic Complications (한국인 제2형 합병증동반 당뇨병 환자에 있어 과산화지질, 항산화 효소, 및 항산화비타민에 관한 연구)

  • The purpose of this study was to determine the extent of oxidative stress in NIDDM patients with diabetic complications and to determine the relationship between oxidative stress and diabetic complications. For this study, 139 NIDDM patients were recruited, 85 with diabetic complications and 54 without complications were recruited. The concentration of malondialdehyde(MDA) and the activities of antioxidant enzymes including catalase, superoxide dismutase(SOD), gluthatione peroxidase(GSH-Px)were determined. The daily intakes and plasma concentrations of beta-carotene, lycopene, lutein nd alpha-tocopherol were determined by food frequency questionnaire and by high performance liquid chromatography(HPLC), respectively. Among the antioxidant enzymes studied, only GSH-Px activity was lower in NIDDM patient, with diabetic complications than in those without complications(2.91$\pm$0.80 vs 3.54$\pm$0.44 U/mgHb, p<0.05). Those NIDDM patients with diabetic complications had higher MDA concentrations than those without diabetic complications(1.40$\pm$0.25 vs 1.25$\pm$0.11 nmol/ml, p<0.05). There were no significant differences in the dietary intakes of total carotenoids(2854 vs 2824ug/day)or vitamin E (9.5$\pm$3.2 vs 9.5$\pm$2.0mg/day)between NIDDA patients with and without complications. However, the plasma concentrations of beta-carotene and lycopene were significantly lower in NIDDM patients with complications than in NIDDM patients without complications (Beta-carotene : 24.2$\pm$12.5 vs 33.1$\pm$16.2(ug/dl), lycopene : 2.8$\pm$2.1 vs 4.3$\pm$2.8(ug/dl)). This study showed that in NIDDM patients with complications, the lipid peroxidation of erythrocytes was higher increased and the antioxidant reserves were significantly dipleted, compared with NIDDM patients without complications. The lower plasma concentrations of beta-carotene and lycopene in NIDDM patients may be due to the presence of diabetic complication, not due to the lower dietary intakes of antioxidant vitamins. To define the role of carotenoids in diabetes, more experimental and clinical studies are needed.

Compositional Analysis of Lentil (Lens culinaris) Cultivars Related to Colors and Their Antioxidative Activity

  • Metabolite profile is a powerful analytical technique to identify the functional characterization of plants. In this study, the phytochemicals and secondary metabolites of lentils (Lens culinaris) were analyzed to compare the anti-oxidative activities according to the different colors. The polar metabolites, fatty acids, carotenoids, flavonoids, anthocyanins, total phenolic acids, DPPH activity were analyzed. Three kind of lentils, French green whole lentil (FG), red whole lentil (LR), and green whole lentil (LG) (ASIA SEED Co., LTD), were used for this study. Fatty acids, phytochemicals, and antioxidative components from each lentil varieties were analyzed by official methods. The contents of lutein in carotenoids were 6-9 times higher than zeaxanthin in all lentils, but were not significantly different among three varieties. The content of carotenoids in FG was lower significantly than those in the LR and LG. Myricetin and luteolin were detected in the only FG. Kaempferol and delphinidin were significantly highest in the FG. Most of the phenolic acids except coumarate were higher in FG and LG than in LR. Also antioxidant effects ($EC_{50}$) were higher in FG and LG than in LR. The analyzed metabolites obtained from lentils showed distinct separation in the PCA results according to the varieties. Also, lentils showed different anti-oxidant profiles according to the colors. FG and LG showing higher contents of phytochemicals showed higher antioxidative activity than LG containing relative low contents of phytochemicals.

Analysis of Carotenoids in Commonly Consumed Agricultural Products in Korea (국내에서 상용되는 농산물의 카로티노이드 함량 분석)

  • A total of 51 vegetables and fruits, commonly consumed agriculture products in Korea, were analyzed for their α-carotene, β-carotene, and β-cryptoxanthin contents as provitamin A. The beta-carotene content (㎍/100 g) was high in a few green leaf vegetables such as coriander (5,924.07), gegeol radish leaf (5.855.72), and curried mallow (5,138.01), while α-carotene and β-cryptoxanthin contents were not detected. The β-carotene in 8 kinds of 20 general vegetables was detected in the range of 214.06~1,437.67 ㎍/100 g, while α-carotene was detected at 460.17 ㎍/100 g in only old pumpkin. The β-cryptoxanthin was detected in the range of 106.55~315.49 ㎍/100 g in Japanese elm, watermelon, white cucumber, and lettuce. However, carotenoids were not detected in 10 kinds of agricultural products including oriental melon, potato, etc. In fruits, the beta-carotene contents ranged from 165.72~3,997.39 ㎍/100 g, showing maximum value in apple mango and minimum value in persimmon. The β-cryptoxanthin was detected at 232.22 ㎍/100 g in only passion fruit, while the α-carotene was detected at 77.25 ㎍/100 g in only darae. Thus, based on the analyzed results of carotenoids of agriculture products consumed or cultivated in Korea, and it was found that green leaf vegetables comprise high beta-carotene overall.

Detection and Measurement of Retinoic Acid in Human Liver Samples (간 조직내의 Retinoic Acid 검출 및 측정 방법)

  • Retinoic acid. the active metabolite of vitamin A. was detected in the human liver for the first time using a new method. A rapid and sensitive technique has been developed using gradient-elution. reverse-phase high performance liquid chromatography. This assay. with simultaneous multiwavelength detection at 294nm, 325nm and 450nm after saponifcation of liver samples. allows us seperation and quantitation of vitamin E, retinoic acid, total retinoids and various carotenoids in one small sample. The proportion of retinoic acid to total retinoids in human liver appears to be quite $consistent(2.4\pm0.2%$ ). With low vitamin A storage in liver, detection at another wavelenth 354nm would increase the sensitivity for retinoic acid of small quantity This method of analysis could be used for other tissues like red blood cells, plasma or serum, also. Hepatic retinoic acid level with total retinoids and carotenoids would serve a better indicator of functional vitamin A nutriture especially for those with disease requiring needle biopsy of liver.

  • Variations of carotenoid and chlorophyll during the course of after-ripening in hotpepper fruits were studied: 1. Carotenoid pigments were fractionized into six fragments by means of column chromatography and each of six were applied to the TLC. By these procedures 30 kinds of carotenoids were identified among 54 kinds of different separations. 2. Total carotenoids increased very rapidly in cli. stage. And when compared group by group, Diol showed highest in amounts as well as in increasing index. 3. Decrease of phytoene in post-cli. stage and notable increase of capsanthin, capsorubin and violaxanthin amounts in carotenoid, as major pigments, were remarkable one. 4. Total amounts of chlorophyll a and b showed decreasing tendency during after-ripening and, finally, disappeard at the post-cli. stage.

Contents of Isoflavones and Antioxidative Related Compounds in Soybean Leaf, Soybean Leaf Jangachi, and Soybean Leaf Kimchi (콩잎 밑 콩잎 요리의 이소플라본 함량 및 항산화 관련 성분들의 비교)

  • Soybean is an important plant as it is the source of protein and oil as well as various phytochemicals that are related with biological activity. Over the past decades, scientists have conducted considerable research on the physiological properties of soybeans, especially isoflavones, which are the characteristic components in soybeans. However, there is no research on the properties or the bio-functionality of soybean leaf. Jangachi and kimchi are two of the traditional special dishes of Gyungsang Province in Korea which we made from soybean leaves. Depending on the recipe, green or yellow soybean leaves are used for the preparation of these two side dishes. We compared the antioxidative activity and measured the contents of isoflavones, total phenol, chlorophylls, carotenoids, and vitamin C in the ingredients (green and yellow soybean leaf) and the final side dishes (jangachi and kimchi). We Int report that isoflavones were contained in soybean leaf and that jangachi had the highest isoflavone contents among the samples. Yellow soybean leaf contained higher isoflavones than green soybean lear and kimchi. From the TEAC assay results, the sequence or antioxidative activities was yellow soybean leaf > soybean leaf jangachi > green soybean leaf > soybean leaf kimchi. The sequence was the same with total phenol contents, indicating that antioxidative activity is highly related with total phenol level. Chlorophylls, carotenoids and vitamin C existed abundantly in green soybean leaf. In conclusion, soybean leaf could be a good material for health due to the presence of isoflavones and the other useful antioxidants mentioned above.
