$\gamma$-Oryzanol의 급여가 KK 당뇨 마우스의 지질대사에 미치는 영향 (Supplementary Effect of$\gamma$-Oryzanol on Lipid Metabolism in Diabetic KK Mice)

  • This study was carried out to investigate the supplementary effects of ${\gamma}$-oryzanol extracted from rice bran on lipid metabolism in diabetic mice. We supplied 2 kinds of experimental diets (CO without and GO with 0.2% ${\gamma}$-oryzanol) to diabetic mice for 8 weeks. Diet intake, body weight, organ weight, contents of serum and hepatic lipid profiles, and fecal lipid levels were measured. Though there was no significant difference in diet intake, body weight change and organ weight between experimental groups, the concentration of serum total cholesterol and hepatic total lipid, total cholesterol and HMG-CoA reductase activity was significantly lower in GO group treated with 0.2% ${\gamma}$-oryzanol of diet than CO group after supplementary period of experimental diets. And total lipid, triglyceride, total cholesterol, and bile acid levels excreted to feces were significantly higher in GO group than CO group. These results suggest that ${\gamma}$-oryzanol decrease the serum and hepatic lipid levels by lowing HMG-CoA reductase activity or increasing the contents of fecal lipid in diabetic KK mice.

Screening of Indigenous Strains of Lactic Acid Bacteria for Development of a Probiotic for Poultry

  • In an attempt to develop a probiotic formulation for poultry feed, a number of lactic acid bacteria (LAB) were isolated from chicken intestinal specimens and a series of in vitro experiments were performed to evaluate their efficacy as a potential probiotic candidate. A total of 650 LAB strains were isolated and screened for their antagonistic potential against each other. Among all the isolates only three isolates (TMU121, 094 and 457) demonstrated a wide spectrum of inhibition and were thus selected for detailed investigations. All three selected isolates were able to inhibit the growth of E. coli and Salmonella species, although to variable extent. The nature of the inhibitory substance produced by the isolates TMU121 and 094 appeared to be associated with bacteriocin, as their activity was completely lost after treatment with proteolytic enzymes, while pH neutralization and catalase enzyme had no effect on the residual activity. In contrast, isolate TMU457 was able to resist the effect of proteolytic enzymes while pH neutralization completely destroyed its activity. Attempts were made to study the acid, bile tolerance and cell surface hydrophobicity of these isolates. TMU121 showed high bile salt tolerance (0.3%) and high cell surface hydrophobicity compared to the other two strains studied, while TMU094 appeared the most pH resistant strain. Based on these results, the three selected LAB isolates were considered as potential ingredients for a chicken probiotic feed formulation and were identified to species level based on their carbohydrate fermentation pattern by using API 50CH test kits. The three strains were identified as Lactobacillus fermentum TMU121, Lactobacillus rhamnosus TMU094, and Pediococcus pentosaceous TMU457.

Cholesterol-Lowering Lactic Acid Bacteria from Kimchi

  • Lactic acid bacteria are dominant microflora in fermented kimchi. In this study, lactic acid bacteria were isolated from 5 kinds of fermented kimchi and determined their physiologic property. Total 64 of LAB were isolated from kimch samples. In this study, 8 strains of them were selected by pH and bile salt tolerance test. HFI 58, HFI 40, and Yeulmu E strain had significant cholesterol lowering effect in vitro test. These were made of special feed of chicken by WooJin B&G. A Diet was tested for 5 weeks. The feed of special material supplement HFI58 groups had significant lower cholesterol concentration in egg yolk.

돼지감자 및 치커리 섭취가 흰쥐의 지질대사에 미치는 영향 (Effects of Jerusalem Artichoke and Chicory on Lipid Metabolism in Rats)

  • This study was carried out to determine the effects of Jerusalem artichoke (JA) powder , JA extract and chicory extract on lipid metabolism in SD rats. The experimental groups were divided into 4 groups ; control, JA powder JA extract and chicory extract. The animals were fed ad libitum each of the experimental diets for 3 weeks. After 3 weeks, the wet weights of cecum were significantly increased in rats fed JA powder and chicory extract. Cecal contents were slightly increased in all experimental groups. Serum HDL-cholesterol, HDL-cholesterol/total cholesterol ratio and atherogenic index were significantly increased in the chicory extract group. Serum triglyceride, total cholesterol and LDL-cholesterol levels were not different among the diet groups. Although the feeding of chicory extract significantly lowered total lipid of liver, there was no difference in levels of triglyceride and total cholesterol. The content of fecal lipiid and cholesterol were significantly higher in the Ja extract and chicory extract group than other groups. Fecal bile acid was significantly increased in the chicory extract group. These results indicate that chicory extract is an effective regimen for improvement of lipid metabolism in SD rats.

Effect of Onion Powder Supplementation on Lipid Metabolism in High Fat-cholesterol Fed SD Rats

  • This study was performed to examine the effects of onion powder supplementation on lipid metabolism in male SD rats fed a high fat and high cholesterol diet. Experimental groups were control (C), high fat fed group (HF), high fat+onion powder intake (OP), high fat+quercetin intake (Q). The HF diet contained 1% cholesterol, 4% lard, 0.3% Na-taurocholate, and quercetin supplementation level was 0.1 g/kg diet. The OP group showed lower body weight gains compared to the control, while there was no significant difference in food efficiency ratio efficiency. When the proportion of fecal bile acids per total lipids was calculated, there was a significant decrease in the HF group compared to the control group, while the levels of the control group was same as that of the OP group. There was no significant difference between the HF and Q groups in bile acid/total lipid in feces. The amount of total cholesterol in liver increased significantly in HF group compared to the control group, while total cholesterol decreased significantly in the OP group compared to the HF group. There was a significant decrease in GOT (glutamic oxaloacetic transaminase) activity in OP and Q groups compared to the HF group. In conclusion, feeding onion powder to hyperlipidemic rates appeared to control weight gain, significantly lower the level of total cholesterol in the liver, and recover GOT activity. We also demonstrated that onion powder intake was more effective than quercetin intake.

Fermentation Characteristics of Exopolysaccharide-Producing Lactic Acid Bacteria from Sourdough and Assessment of the Isolates for Industrial Potential

  • Lactic acid bacteria (LAB) with antimicrobial activity and high exopolysaccharide (EPS) production ability isolated from sourdough were studied for their fermentation characteristics as potential new starter cultures. The values of pH, titratable acidity, and viable cell counts were $4.06{\pm}0.009-4.50{\pm}0.015,\;0.787{\pm}0.020%-1.172{\pm}0.018%,\;and\;8.78{\pm}0.08-8.98{\pm}0.06$ log CFU/ml, respectively. In order to select probiotics with a high survival rate in the gut, isolates were tested to assess resistance against the artificial gastric acid and bile juice. Viable LAB counts were significantly (p<0.05) affected by the acidity. At pH 2.0, the total declines in the initial bacterial counts were 4.52$\pm$0.07 log for S. thermophilus St-Body-1, >7.98$\pm$0.03 log for E. flavescens DU-10, >7.95$\pm$0.05 log for E. faecium DU-12, and 3.15$\pm$0.06 log for L. amylovorus DU-21. Among the strains, L. amylovorus DU-21 was the only strain that had bile tolerance under simulated gastrointestinal conditions. In order to improve EPS production by L. amylovorus DU-21, the influence of carbon source was studied. When glucose was used as a carbon source, EPS production dramatically increased to 17.19$\pm$0.28 g/l (p<0.05). The maximum cell growth (10.012$\pm$>0.012 log CFU/ml) and EPS production (18.71$\pm$0.19 g/l) were achieved when 15 g/l of glucose was employed as the carbon source.

  • For a probiotic yoghurt it is desirable to utilize lactic acid bacteria with a high survival rate and beneficial function to human beings. We have examined a variety of lactic acid bacteria to assess the acid and bile tolerance and antibiotic resistance. In addition, an in vitro culture experiment was performed to evaluate their ability to reduce cholesterol levels in the growth medium. Thirteen strains were selected from in vitro cholesterol assays and fed to Sprague-Dawley rats with a high-cholesterol diet. Among the 13 strains tested, 8 strains were shown to reduce serum cholesterol levels significantly after 24 days of administration in vivo. Rats were fed lyophilized yoghurt powder fermented with a combination of 3 selected strains: Bifidobacterium infantis AM-220, Lactobacillus AM-245, and Styeptococcus MA-1. The levels of total cholesterol and low density lipoprotein were significantly lower (p$\leq$0.05) in rats fed the yoghurt powder compared with control group. These studies suggest that yoghurt fermented with appropriately selected lactic acid bacteria may have a anticholesterolemic effect.

Screening of Bacteriocin-producing Bacillus Strains Isolated from Domestic Animal Feces for Antagonistic Activities against Clostridium perfringens

  • The purpose of this study was to isolate and characterize bacteriocin-producing bacteria against Clostridium perfringens from domestic animals to determine their usefulness as probiotics. The feces of cattle and chicken were used as sources to isolate bacteriocin-producing bacteria using the spot-on-lawn method. In total, 900 bacterial stains were isolated from domestic animal feces, and 19 strains were finally selected after determining the inhibitory activity against the pathogenic indicator C. perfringens KCTC 3269. Eighteen strains of Bacillus subtilis and one strain of Brevibacillus parabrevis were identified by 16s rRNA sequencing. Most of the bacterial strains isolated were resistant to 0.5% bile salts and remained viable after 2 h at pH 3.0. Additionally, some B. subtilis strains showed strong inhibitory activity against Listeria monocytogenes. We isolated and screened B. subtilis strains CB 153 and CB 189 from cattle and B. subtilis MSC 156 and B. parabrevis MSC 164 from chickens using probiotic selection criteria such as inhibition activity against C. perfringens and tolerance to acid and bile salts. The isolated bacteriocin-producing bacteria and/or bacteriocin have the potential to be used as probiotics in the livestock industry.

인삼사포인의 흡수 및 배설에 관한 연구 (Studies on the Absorption and Excretion of Ginsenosldes)

  • The metabolic fate of ginsenosides including gastrointestinat absorption, organ distribution, excretion and metabolism in liver was investigated by tracer studies using the radio-labeled ginsenosides. 3H-ginsenosides were shown to be absorbed from the mouse digestive tract and then to be excreted rapidly into urine and/or bile. Bile juice was concluded to play a significant role in absorption of ginsenosides. The total concentration of radioactivity persisted in tissues 24 hrs after oral administration was less than 1.3% of the administered dose and Rbl showed the highest value. The concentrations of radioactivity were relatively high in the liver and kidney. After administration of Rbl radioactivity was detected in the brain. After oral administration of 8H-ginsenosides, major component excreted into urine was found to be the intact ginsenosides and decomposed and/or metabolized products were found in GIT in the case of Rbl. 3H-ginsenoside Rbl was shown to be metabolized in the liver and the metabolite was suggested to be an acylated compound of Rbl by a certain organic acid.

게 껍질분말 첨가식이가 고지혈증 흰쥐의 지진대사에 미치는 영향 (Effect of Crab Shell Powder on Lipid Metabolism in Diet-Induced Hyperlipidemic Rats)

  • This study was performed to investigate the effect of dietary crab shell powder on lipid metabolism in diet induced hyperlippidemic rats. Sixteen male Sprague-Dawley rats were divided into two groups and fed 20% (w/w) carb shell powder supplemented diet for 4 weeks. Energy intake and weight gain were lower in experimental group than in control group. Concentrations of plasma triglyceride, total cholesterol, LDL-cholesterol, and atherogenic index (AI) were decreased and concentration of HDL-cholesterol was increased by crab shell powder supplementation. Concentrations of liver total cholesterol and triglyceride were significantly lower in experimental group. The crab shell decreased triglyceride and cholesterol concentration in plasma and liver, and increased bile acid excretion in feces. Therefore, the crab shell powder is assumed to exhibit function of chitin or chitosan that effects on lipid metabolism and cholesterol reabsorption.
