Processing of medicinal plants is one of the processing methods for reducing of toxicity or improving of effect on medicinal plants. In this study, we studied the changes in the constituents and antioxidant activity in accordance with the processing conditions by the salt water of Citrus unshiu Markovich. Changes in antioxidant activity was measured by the DPPH method and changes in the components were analyzed by high performance liquid chromatography. As a result, the content of the main constituents (narirutin, hesperidin) and total polyphenol were increased by increasing the concentration of salt water. Moreover, antioxidant activity was increased gradually in proportion to the increase of the total polyphenol content.

Polyphenol profiles, physicochemical properties, antioxidant activities, and inhibitory effect of adipocyte differentiation of Houttuynia cordata fermented with Lactobacillus brevis B84 were evaluated. Six polyphenols were characterized for this plant by using high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS), and the results were compared with total phenolic content by a spectrophotometric method. The total amount of the identified polyphenols was lower than that determined by the spectrophotometric method. However, the fermentation process influenced polyphenol composition such as content of vanillic acid and caffeic acid. The phytochemical profiles were evaluated by high-performance liquid chromatography with UV and electrospray ionization mass spectrometry detection ($HPLC-DAD-ESI-MS^n$). Total sugar and reducing sugar contents decreased after fermentation. Antioxidant activities such as DPPH, ABTS, and superoxide anion radical scavenging and reducing power were evaluated to compare the beneficial effect after fermentation. Fermented H. cordata increased the lipolytic effect in 3T3-L1 adipocytes. Overall, the results indicate that the fermentation of H. cordata with L. brevis B84 produces changes of phenolic compounds, antioxidant activity, and lipolytic effect.
Triphala churna is a widely used herbal formulation that contains equal proportion of dried fruit powder of Emblica officinalis, Terminalia chebula and Terminalia belirica. In the Indian system of medicine, it is used in cleaning wounds, urinary disorders, diabetes mellitus, leprosy, constipation, eyesight promotion, piles, and as a rejuvenator. In the present study, the methanolic extract of 5 commercial Triphala was evaluated for antioxidant activity by 1,1-diphenyl-2-picryl-hydrazyl free radical scavenging method, total phenolic content by Folin-Ciocalteu method and gallic acid equivalents (GAE) by high performance thin layer chromatographic (HPTLC) method. All extracts exhibited antioxidant activity significantly. The $IC_{50}$ of the extracts ranged between $7.16\;to\;12.96\;{\mu}g/ml$. The total phenolic content of the extracts was found to be 195.3-296.4 mg of GAE/gm of GAE/gm dw. The HPTLC chromatographic data reveal that the content of GAE present in the extract was found to be $7.17-4.11\;{\mu}g/ml$. The study reveals that out of the churnas analysed, C was found to exhibit the most potent antioxidant activity. A clear correlation between $IC_{50}$ and content of GAE nor the total phenolic content could be observed. The study reveals that the consumption of Triphala would exert several beneficial effects by virtue of its antioxidant activity.

Objective: The effect of garlic powder (GP) and phenyl acetic (PA) acid throughout the fattening period of broiler chickens on performance, blood parameters, immune, and antioxidant parameters as well as carcass traits was evaluated. Methods: A total of 210 day-old Cobb broiler chicks were randomly distributed into seven dietary treatments having five replications with six chicks per replicate. The first group (control) fed a basal diet without supplements, whereas the 2nd, 3rd, and 4th group were fed basal diet plus 0.25, 0.50, and 0.75 g GP/kg diet, respectively and the group 5th, 6th, and 7th were fed on the basal diet plus 0.25, 0.50, and 0.75 g PA/kg diet. Results: Broiler body weight and gain at 21 and 42 days were increased (p<0.05) with diets supplemented with GP and PA. Red blood cells and hemoglobin were improved in chickens fed diets enriched with GP. Broiler chickens received diets containing either GP or PA recorded the higher values (p<0.05) of total protein, globulin, high-density lipoprotein, immunoglobulin M (IgM), and IgG, superoxide dismutase and total antioxidant capacity; while, blood total cholesterol, low-density lipoprotein, aspartate-aminotransferase, and malondialdehyde were lowered (p<0.05) compared to control-diet. Liver and immune-related organs weight were improved (p<0.05) in broilers fed diet supplemented with GP and PA. Conclusion: Feeding of GP or PA in diet had positive effects on performance traits and immunological, antioxidant and physiological status of broilers. Thus, the use of tested feed additives as an eco-friendly alternative to antibiotics produced a positive effect on animal health.
BACKGROUND/OBJECTIVES: Angelica keiskei is a green leafy vegetable rich in plant pigment phytochemicals such as flavonoids and carotenoids. This study examined bioavailability of flavonoids and carotenoids in Angelica keiskei and the alteration of the antioxidant performance in vivo. SUBJECTS AND MATERIALS: Absorption kinetics of phytochemicals in Angelica keiskei were determined in healthy older adults (> 60 y, n = 5) and subjects with metabolic syndrome (n = 5). Subjects consumed 5 g dry Angelica keiskei powder encapsulated in gelatin capsules with a low flavonoid and carotenoid liquid meal. Plasma samples were collected at baseline, 0.5, 1, 2, 3, 4, 5, 6, 7, and 8 h. Samples were analyzed for flavonoids and carotenoids using HPLC systems with electrochemical and UV detection, respectively, and for total antioxidant performance by fluorometry. RESULTS: After ingestion of Angelica keiskei increases in plasma quercetin concentrations were observed at 1-3 and 6-8 hr in the healthy group and at all time points in the metabolic syndrome group compared to baseline (P < 0.05). Plasma lutein concentrations were significantly elevated in both the healthy and metabolic syndrome groups at 8 hr (P < 0.05). Significant increases in total antioxidant performance were also observed in both the healthy and the metabolic syndrome groups compared to baseline (P < 0.05). CONCLUSIONS: Findings of this study clearly demonstrate the bioavailability of phytonutrients of Angelica keiskei and their ability to increase antioxidant status in humans.

The aim of this study was to investigate the antioxidant effects of O. robusta stem extract (ORE) and to determine the total polyphenol and flavonoid contents. Free radical scavenging properties were evaluated using DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid)), and ·OH (hydroxyl radical) scavenging assays. Total polyphenol and flavonoid contents were measured using Folin-Ciocalteu reagent and aluminum chloride colorimetric methods, respectively. Active compounds of ORE were determined using high-performance liquid chromatography (HPLC). The results of the study showed that ORE exhibited DPPH, ABTS, and ·OH radical scavenging activities in a dose-dependent manner. Especially, 1,000 ㎍/mL of ORE showed the strongest radical scavenging properties against DPPH, ABTS, and ·OH. ORE contained total polyphenol content of 57.4 mg GAE/g and total flavonoid content of 5.4 mg QE/g, which may contribute to their antioxidant effects. As a result of HPLC, the contents of active compounds in ORE, dihydrokaempferol (0.65 ㎍/mL), and 3-O-methylquercetin (1.10 ㎍/mL) were confirmed. In conclusion, ORE may be useful as a functional material with antioxidant properties.
In this study, we examined the antioxidant activity of the naturaceutical extract in vitro using total phenolic contents, total flavonoids contents, DPPH radical scavenging activity, reducing power assay, and phenolic acid contents. The total phenolic and total flavonoids contents of naturaceutical extract were 5.46 mg/g, 2.21 mg/g, respectively. The DPPH radical scavenging activity of natraceutical extract varied from 18.77 ($200{\mu}g/mL$) to 3.44 ($1,000{\mu}g/mL$). The reducing power of the natraceutical extract absorbance varied from 1.07 (0.78 mg/mL) to 3.44 (12.5 mg/mL), and reducing power of extract presented a concentration-dependent activity increase. The highest amounts of trans-ferulic acid, p-hydroxybenzoic acid, p-coumaric acid, vanillin, vanillic acid, p-hydroxybenzaldehyde, and trans-cinnamic acid were observed in the naturaceutical extract at the levels of 750.79, 619.75, 531.34, 222.04, 219.28, 107.40, and $89.56{\mu}g/g$, respectively. The results imply that this antioxidant effect of the naturaceuticals extract could be harnessed in the management and prevention of degenerative diseases associated with oxidative stress.
Objective: This study was performed to investigate the effects of supplementing sesame seed meal (SSM) with phytase and lysine on performance, egg quality, blood biochemical and antioxidant status of laying hens. Methods: A total of 960, 56-wk-old laying hens were divided into 12 dietary groups with eight replicates per group (10 birds per replicate). A completely randomized design with factorial arrangement 2×3×2 consisted of two levels of lysine supplement (0% and 10% over requirement), three SSM levels (0%, 10%, and 20%) with or without phytase (0 and 300 g/ton). The feeding trial lasted 10 weeks. Results: Birds fed diets with 10% SSM had higher feed intake than groups fed 0% and 20% SSM. The addition of phytase to experimental feeds, improved feed conversion ratio, increased egg weight and mass (p<0.01). Egg quality criteria was not affected by supplementing phytase; however, supplementing 300 g/ton phytase to hens diet, led to a significant (p<0.05) increase in egg shell strength. Egg yolk cholesterol and serum low-density lipoprotein cholesterol, atherogenic index and total cholesterol were decreased (p<0.01) by diet containing 20% SSM. The high-density lipoprotein cholesterol was increased (p<0.05) in serum of hens fed 20% SSM than the other groups. It was also observed that total antioxidant capacity and total superoxide dismutase content of hens fed 20% SSM was significantly higher than control group (p<0.05). Conclusion: As from results, dietary supplementation of SSM and phytase had no negative effects on laying hens performance or egg quality while improving the egg oxidative stability.
Objectives: Samul-tang is commonly used to alleviate the side effects of chemotherapy. This study aimed to establish an efficient method of extracting ACTS002 based on Samul-tang using the yield, high-performance liquid chromatography (HPLC), and antioxidant assay. Methods: ACTS002 was extracted from each extraction solvent, and the contents of 5-hydroxymethyl-2-furaldehyde (5-HMF), paeoniflorin, and ferulic acid were quantitatively analyzed and compared using HPLC. Moreover, the antioxidant activities of ACTS002 were measured using total flavonoids, total phenolic compounds, 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), and Ferric reducing/antioxidant power (FRAP). Results: All of the components were set as the index contents because they were easy to process. The antioxidant activity of total flavonoids was the highest in 70% ethyl alcohol extracts, and total phenolic compounds were the highest in 50% ethyl alcohol extracts. In DPPH, 50% ethyl alcohol extracts showed the highest activity, and in ABTS 70% ethyl alcohol extracts were the highest. In FRAP, 70% ethyl alcohol extracts showed the highest activity. Conclusions: ACTS002 can control quality by setting 5-HMF, paeoniflorin, and ferulic acid as the index contents. The antioxidant activity measurement was relatively high in the 50% and 70% ethyl alcohol extracts. Our results can predict the possibility of a pharmacological activity and the standardization of ACTS002.
Total phenolic content, total anthocyanin content and antioxidant activity were analyzed from rice samples collected in Korea, Japan and China. The results showed that the total phenolic content and free-radical scavenging activity differed significantly in these rice lines. The correlation between content and activity was subsequently investigated. The results showed that in black rice, anthocyanin was the major phenolic component and that both phenolic content and anthocyanin content were closely correlated with free-radical scavenging activity. Reverse Phase High Performance Liquid Chromatography (RP-HPLC) data showed that cyanidin-3-O-glucoside and peonidin-3-O-glucoside composed about 90% of the total anthocyanin content in black rice and in red rice. In the red rice extract, the total phenolic content produced a high correlation coefficient with antioxidant activity but correlated very poorly with the total anthocyanin content. The $OD_{458}$ and the $OD_{500}$ values which represent the proanthocyanidin content of the rice extract, produced high correlation coefficients with antioxidant activity and total phenolic content. These results suggest that the $OD_{458}$ and the $OD_{500}$ values can be used to evaluate the quality of red rice. In addition, based on the data obtained, a competitive accumulation model of anthocyanin and proanthocyanidin in black and red rice was proposed.
