Effect of different biochar formulations on the growth of cherry tomatoes

  • Biochar is a solid carbon material made by pyrolyzing a biomass under limited oxygen conditions. Biochar has been reported to confer various benefits, such as increased soil productivity, pollutant absorption, and reduced greenhouse gas. In this study, oak pyrolyzed at 600℃ for 3 hours was either powdered or pelleted. Each of the biochar types was added to the soil at a rate of 2%. The control did not receive any biochar while a combination of the biochar and NPK treatment (biochar 2% + NPK) was also included. The cherry tomatoes were grown in greenhouse pots for 50 days to compare the growth characteristics of the different treatments. The cherry tomato with the powdered biochar 2% + NPK treatment had the heaviest plant fresh shoot weight of 276.4 g and the highest chlorophyll content of 59.3 SPAD. The control had the lightest plant fresh shoot weight of 44.2 g and a chlorophyll content of 26.5 SPAD. Both forms of biochar affected the chemical properties of the soil, increased the pH, electrical conductivity, available phosphate, total carbon and total nitrogen and positively influenced the cherry tomato growth and productivity. From the above results, therefore, both biochar forms are suited for use as soil amendments.

Study on the Nutrient Solution Content and Growth of Cherry Tomato in Scoria Culture (제주 송이를 이용한 방울토마토 양액재배시 양액성분 및 생육에 관한 연구)

  • The main purpose of these studies were to clarify differences in Cheju - scoria and other solid media on quantity and quality of cherry tomatoes and on shift of component of the nutrient solution, and to use Practically Cheju-scoria as an excellent solid culture medium. The results obtained were summarized as follows ; 1. Among scoria plots, the rates of dry weights, fruits and their sugar-acid ratio were higher in the plot that was drained well with deep flow for one hour once a day. 2. Fresh fruit weights were lighter in rockwool and deep flow technique, but larger in scoria, Hyugashi (artificial gravel, $\Phi$10-12mm) and perlite in moving to higher flower cluster. 3. The results of analysis on microelement among solution components showed decrease of concentrations of P and K in the period of growth and development. 4. Yields and brix of cherry tomato showed a tendency to increase in rockwool and Hyugashi than anothers. 5. The concentration of fertilizer base was increased in general solution culture. Transpiration and absorption were similar in scoria plot and other media. 6. More studies of the Cheju-scoria development is required in order to use it as a solid medium for solution culture.

Effect of Immediate Setting with Lateral Branch Cuttings on Growth and Yield of Cherry Tomatoes in Soilless Culture (삽수 직접정식이 방울토마토 양액재배에 있어서 생육 및 수량에 미치는 영향)

  • The purpose of this research was to develop labor-saving cultural practices of cherry tomatoes in the deep flow technique (DFT) by omitting the process of raising seedling. Three cultivars, ‘Pepe’, ‘Pico’ and ‘Koko’ were propagated by the immediate setting of cuttings(lateral shoots) to the culture bed and by setting seedlings obtained by raising cuttings for 15 days, and grown in the standard and half - strength solution. 1. The immediate setting resulted in higher plants, larger number of leaves and larger leaf area during the early growth stage. And the harvest time was hastened by six days in the direct setting than that in the seedling setting. It was also earlier in ‘Pepe’, but not significant by different between two strengths of nutrient solution. 2. The average fruit weight was affected only by cultivars, but not by the type of cutting and the strength of nutrient solution. The highest yields were obtained from the immediate setting of ‘Pepe’ in other cultivars grown in the nutrient solution of standard strength. 3. Brix degree of treated cherry tomato juice was higher in ‘Pepe’ tomato cultured by immediate setting than others.

Expression of Antisense Polygalacturonase Gene in Transgenic Tomato (형질전환 토마토에서 Antisense Polygalacturonase 유전자의 발현)

  • A truncated Polygalacturonase (PG) cDNA was fused in reverse orientation to the CaMV 35S promoter of the binary vector pCA643, and introduced into tomato cells by Agrobaderium - mediated transformation. Transformed cells were selected using kanamycin as select agent then regenerated into plants. After selfed, one transgenic line (T9), was germinated and grown on MS medium containing 1 mg/mL of kanamycin Genomic Southern analysis of a T9 progeny with labelled PG2 cDNA probe showed a single antisense PC fragment as well as the endogenous PG2 gene, suggesting that PC antisense gene was integrated into tomato genome. Northern blot analysis demonstrated that the antisense RNA was produced from the transgene at much tiger level than the endogenous PG2 gene. Polygalacturonase activity analysis of the fruit from transgenic plants demonstrated that the antisense transgene expression caused 4 to 60% reduction of endogenous PG activity.

Durability of the Expanded Rice Hull as a Hydroponic Culture Medium (양액재배용 팽연화 왕겨의 적정 사용기간)

  • In an effort to evaluate the economic value and durability of the expanded rice hull as substrates, changes in the physical and chemical properties of material and plant growth in that substrate were studied. Using and electron microscope, the structure of used and new expanded rice hull substrate was examined. Considerable decomposition was found in the substrate which had been used one to three times. Compactness and lowered porosity in the used substrates were probably caused by decomposition. The results of cation analysis showed the possible destruction of cell wall of rice hulls. Abundant $Ca^{2+}$ in the substrates used for two to three times also indicated the possibility of decomposition. In tomato yield comparison, 15.2% more yield of tomato fruit in a new substrates indicated the negative effects of decomposition of one-time used substrates. Yield decreased in the substrates used for three times. if perlite substrates is used for three years before renewal and the cost of the perlite renewal is counted. 65.3% saving in the cost will be realized with the use of an expanded rice hull substrate. Another positive effect of the expanded rice hull substrate is the decrease of environmental contamination.n.

  • Smart-farm has been spreading across Korea to improve the labor efficiency and productivity of greenhouse crops. Although notable improvements have been made in the monitoring technologies and environmental-controlling systems in greenhouses, only a few simple decision-support systems are available for predicting the optimum environmental conditions for crop growth. In this study, a tomato growth model (GreenTom), which was developed by Seoul National University in 1997, was calibrated and validated to examine if the model can be used as a decision-supporting system. The original GreenTom model was not able to simulate artificial defoliation, which resulted in overestimation of the leaf area index in the late growth. Thus, an algorithm for simulating the artificial defoliation was developed and added to the original model. The node development, leaf growth, stem growth, fruit growth, and leaf area index were generally well simulated by the modified model indicating that the model could be used effectively in the decision-making of smart greenhouse.

Ameliorating Effect of $\textrm{Ca}({NO_3})_2$ or $\textrm{CaCl}_2$ on the Growth and Yield of NaCl-Stressed Tomato Grown in Plastic Pots Filled with Soil (NaCl 스트레스를 받은 토마토의 생육 향상을 위한 $\textrm{Ca}({NO_3})_2$$\textrm{CaCl}_2$ 처리 효과)

  • Enhanced supply of $Ca^{2+}$ as well as NO$_3$$^{[-10]}$ is known to restrict the uptake of the Na$^{+}$ and Cl$^{[-10]}$ ion and ameliorate growth under saline conditions. This test was conducted to investigate the ameliorating effects of Ca(NO$_3$)$_2$ or CaCl$_2$ on the growth and yield of NaCl-stressed tomato plants grown in plastic pot filled with soil. All treatments except for the control were supplied with 80 mM NaCl fur two weeks after transporting. The saline solutions with nutrient were supplemented with either 0, 10 or 20 mM Ca(NO$_3$)$_2$ and either 0, 10 or 20 mM CaCl$_2$ during harvesting time from two weeks after transporting. Ca(NO$_3$)$_2$ or CaCl$_2$ application enhanced the growth such as plant height, fresh weight, dry weight, fruit number, and fruit weight, and yield of NaCl-stressed tomato, and also their effects increased greater as concentration of supplemented Ca(NO$_3$)$_2$ or CaCl$_2$increased. Yield increased in 20 mM Ca(NO$_3$)$_2$ compared with the others except fur the control. Photosynthetic rate in Ca treatments was lower than that of the control, but higher than that of NaCl treatment. Leaf chlorophyll content was higher in Ca treatments compared with the others, especially in younger leaf, while that was not affected by concentration of supplemented Ca. Ca(NO$_3$)$_2$ or CaCl$_2$ supply increased the $K^{+}$ and $C^{2+}$ concentration of tomato plants, whereas the Na$^{+}$ transport to the leaves was inhibited. There was a strong increase in the $K^{+}$/Na$^{+}$ ratio in plants treated Ca(NO$_3$)$_2$, or CaCl$_2$. Cl$^{[-10]}$ content of plants was decreased by supplemental Ca(NO$_3$)$_2$ but Cl$^{[-10]}$ was increased in plants with CaCl$_2$compared with Ca(NO$_3$)$_2$. N concentration in plants of tomato increased with enhanced Ca(NO$_3$)$_2$ or CaCl$_2$supply, In conclusion, our study confirms the potential of Ca(NO$_3$)$_2$ or CaCl$_2$to alleviate NaCl-induced growth reductions in tomato.

Effect of Nitrate Concentration in Culture Solution on the Growth and the Uptake of Inorganic Elements of Tomato Plants(Lycopersicum esculentum Mill) (배양액(培養液)의 질산태질소농도가 토마토(Lycopersicum esculentum Mill)의 생육 및 무기원소(無機元素) 흡수에 미치는 영향)

  • This experiment was carried out to investigate the effects of nitrate concentration in culture solution on the growth and the uptake of inorganic elements in Tomato plant in the greenhouse. Tomato plants(cv. TVR-2) were grown with nitrate concentrations 8, 16, 24, 32cmol/l, based on Japan ENSI standard solution. Dry weights of lamina and petiole increased with the nitrate concentration. However, the dry weight of fruit was the highest in the treatment of nitrate concentration of 16cmol/l. The proportion of dry weights of vegitative organ to reproductive organ was the lowest in the treatments of nitrate concentrations of 16cmol/l and it increased with the nitrate concentration. The fruit yield was the highest at the treatment of nitrate concentration of 16cmol/l. With the increase of nitrate level the concentrations of N, $NO_3-N$, Ca and Na increased in lamina and petioles. The concentrations of K, P, S and Cl tended to decline in the nitrate concentration of 16 and 32cmol/l. These results indicate that optimum nitrate concentrations in a tomato grown by hydroponics change with growth stage, and the optimum concentrations for vegitative and reproductive stage were 8 and 16cmol/l, respectively. It also was proved that the nitrate concentrations in the culture solution affected antagonistically the uptake of inorganic anion in tomato : In low nitrate level $Cl^-$ uptake was affected much, while $SO_4{^{2-}}$ and $H_2PO_4{^-}$ uptake were affected in high nitrate level.

Recurrent parent genome (RPG) recovery analysis in a marker-assisted backcross breeding based on the genotyping-by-sequencing in tomato (Solanum lycopersicum L.) (토마토 MABC 육종에서 GBS(genotyping-by-sequencing)에 의한 RPG(recurrent parent genome) 회복률 분석)

  • Marker-assisted backcrossing (MABC) is useful for selecting an offspring with a highly recovered genetic background for a recurrent parent at early generation to various crops. Moreover, marker-assisted backcrossing (MABC) along with marker-assisted selection (MAS) contributes immensely to overcome the main limitation of the conventional breeding and it accelerates recurrent parent genome (RPG) recovery. In this study, we were employed to incorporate rin gene(s) from the donor parent T13-1084, into the genetic background of HK13-1151, a popular high-yielding tomato elite inbred line that is a pink color fruit, in order to develop a rin HK13-1084 improved line. The recurrent parent genome recovery was analyzed in early generations of backcrossing using SNP markers obtained from genotyping-by-sequencing analysis. From the $BC_1F_1$ and $BC_2F_1$ plants, 3,086 and 4868 polymorphic SNP markers were obtained via GBS analysis, respectively. These markers were present in all twelve chromosomes. The background analysis revealed that the extent of RPG recovery ranged from 56.7% to 84.5% and from 87.8% to 97.8% in $BC_1F_1$ and $BC_2F_1$ generations, respectively. In this study, No 5-1 with 97.8% RPG recovery rate among $BC_2F_1$ plants was similar to HK13-1151 strain in the fruit shape. Therefore, the selected plants were fixed in $BC_2F_2$ generation through selfing. MAS allowed identification of the plants that are more similar to the recurrent parent for the loci evaluated in the backcross generations. MABC can greatly reduce breeding time as compared to the conventional backcross breeding. For instance, MABC approach greatly shortened breeding time in tomato.

Selection and Characterization of Horticultural Traits of Tomato leaf curl virus (TYLCV)-resistant Tomato Cultivars (토마토 황화잎말림바이러스(TYLCV) 저항성 품종 선발 및 원예특성 분석)

  • This study was conducted to evaluate imported tomato $F_1$ cultivars as breeding materials for the resistance to Tomato yellow leaf curl virus (TYLCV) by molecular markers and bioassay. From marker genotyping and disease evaluation of 40 $F_1$ cultivars, most of the cultivars declared as TYLCV-resistance carried heterozygous marker genotype for the TYLCV resistance genes Ty-1, Ty-3, or Ty-3a, and showed low disease rates. Whereas, 4 of 5 $F_1$ cultivars declared as intermediate resistance showed marker genotype for susceptibility and disease rates ranged 18.1-33.3%. However, the xx cultivars showed inconsistency in marker genotype and disease rate. Characterization of horticultural traits of the $F_1$ cultivars with TYLCV-resistance indicated that large-size fruit cultivars were higher in yield and similar in sugar contents and solid-acid ratio compared to a control cultivar preferred in the domestic market, although hardness remained to be a problem. On the other hand, cherry tomato cultivars showed lower yield and brix, but longer internode compared to a control cultivar, indicating that breeding for TYLCV-resistance using these cultivars will require more efforts and time compared to large-sized.
