Test strip과 chlorophyll meter를 이용한 토마토의 신속한 영양진단 (Rapid Nutrient Diagnosis of Tomato by Test Strips and a Chlorophyll Meter)

  • 본 연구는 지금까지 제시된 여러 가지 간이 식물영양 진단 방법들 중 test strip과 chlorophyll meter의 이용가능성을 검토하고 실제 이용 방안을 모색하고자 수행되었다. 본 실험에서는 양액의 N, P, K수준을 달리하여 토마토를 재배하면서 생육시기별로 specific color difference sensor value (SCDSV), 엽병 즙액 중 $NO_3,\;PO_4,\;K$의 함량을 측정하였다. 실험 결과 양액중 N, P, K의 농도 변화에 따른 엽병 즙액 중 $NO_3,\;PO_4,\;K$ 함량 변화가 엽 중 total-N, E, K함량의 변화보다 더 민감하였다. Test strip을 이용하여 측정한 토마토 엽병 즙액 중 $NO_3,\;PO_4,\;K$ 함량은 엽 내 total-N, E, K의 농도와 고도의 일차상관관계를 나타내었다. 지금까지 여러 연구를 통해 확립된 토마토의 엽 중 total-N, E, K의 적정 함량을 희귀식에 대입하여 구한 즙액의 적정 $NO_3,\;PO_4,\;K$의 함량은 각각 3.4-5.9, 0.3-0.5, 3.6-6.5 g/l였다.. Chlorophyll meter를 이용하여 측정한 SCDSV는 엽 중 total-N와 높은 상관관계를 나타내었으며 기존에 연구된 토마토의 엽 중 total-N 함량을 희귀식에 대입하여 계산한 결과 적정 SCDSV는 36.0-40.0이었다. 본 연구 결과 test strip과 chlorophyll meter는 토마토의 신속한 영양 진단에 이용할 수 있을 것으로 판단된다.

바이오세라믹 분말의 엽면살포와 배양액내 처리가 분무경재배 토마토의 생장과 과실품질에 미치는 영향 (Effects of Bioceramic Powder in the Nutrient Solution and Foliar Spray on the Growth and Fruit Quality of Aeroponically Grown Tomato)

  • 본 연구는 서광 토마토(Lycopersicon esculentum MILL. cv. Seokwang)의 분무경재배시 신소재인 바이오세라믹 분말을 엽면살포(0.2%)와 배양액내에 처리(0.02%)하여 토마토의 생장 및 과실품질 특성을 비교하고자 수행하였던 바 그 결과를 요약하면 다음과 같다. 1. 지상부와 지하부의 바이오세라믹 처리시 대조구에 비해 초장은 감소하였으나 뿌리의 생장은 증가하였다. 2. 바이오세라믹 분말의 엽면살포시 과방의 생장이 대조구에 비해서 약 14% 정도 증가하였다. 바이오세라믹 분말의 지상부와 지하부 처리시 총건물중이 증가하였다. 3. 바이오세라믹 처리시에 T/R율은 대조구에 비해 낮았으며, 순동화율(NAR)과 상대생장율(RGR)은 바이오세라믹을 엽면살포했을 경우에 12.13g/d$m^2$/day와 0.05g/g/day로 가장 높았다. 4. 대조구에 비해 바이오세라믹 분말의 근권처리시에는 총누적생과중 및 건과중이 감소하였으나, 엽면살포시에는 증가하였다. 바이오세라믹 파우더의 근권 및 엽면살포시 대조구에 비해 과실의 수량이 감소하지 않고 당도가 높아졌다.

토마토 식물에 있어서 광합성이 유존유동성의 에틸렌 생성에 미치는 영향 (Effect of Photosynthesis on Ozone-Induced Ethylent Evolution from Tomato Plants)

  • The rate of evolution of ethylent by tomato plants was rapidly increased by ozone fumigation. In the present study, the mechanism of ethylent evolution by ozone was investigated in experiments with aminoethoxyvinylglycine (AVG) and tiron, which inhibit the formation of ethylene and peroxidation of lipids, respectively. Pretreatment with AVG significantly inhibited the ozone-induced ethylent evolution, but the treatment of plants with tiron did not inhibit. These results indicate that the induction of the evolution of ethylene by ozone involves the pathway via aminocyclopropane-1-carboxylate (ACC), while not released as a result of the peroxidation of lipids. Ozone-induced ethylent evolution was greater in dar- than light-incubated, intact tomato plants. The difference between dark- and light-ethylene evolution was examined with diuron, an inhibitor of photosynthetic electron transport. The inhibitor treatment promoted ethylent evolution. These results suggest that ethylent retention and metabolism in plants were regulated by internal $CO_2$ levels which, in turn, were controlled in large part by photosynthesis. Thus, ethylene was retained in illuminated leaf tissue under low intenal $CO_2$ concentration which may develop in a sealed container without exogenously supplied $CO_2$.

  • 국내 원예시설의 증가와 더불어 양액재배농가도 증가추세에 있어 저렴한 국산 양액재배 자동화기기가 농가에 필요한 실정이다. 이에 따른 양액재배의 EC와 pH를 자동조절하기 위하여 마이크로컴퓨터의 일종인 PLC(programmable logic controller)를 이용한 장치를 개발하고 재배실험을 실시한 결과를 요약하면 다음과 같다. 1. Ladder diagram언어를 사용하여 양액재배의 자동화 프로그램을 작성하였다. 2. PLC를 이용하여 양액의 EC, pH수준을 1.70-l.72mS/cm, 6.1-6.5로 전 생육기간 동안 유지시킬 수 있었다. 3. 대조구에 비하여 처리구가 상품과(당도 5.0 oBrix, 산도 0.4% 이상) 및 수량성이 높은 결과를 얻었다. 4. 식물체의 무기성분 함량은 처리구가 대조구에 비해 전체적으로 높았다.

Effects of Storage Duration on Physicochemical and Antioxidant Properties of Tomato (Lycopersicon esculentum Mill.)

  • This study explored the physicochemical and nutritional changes associated with storage duration of fresh tomatoes. Fruits of the 'TY Megaton' and 'Yureka' tomato cultivars were harvested at the pink stage and stored at $12^{\circ}C$ for 20 days. During storage, firmness, weight loss, skin color (Hunter L, a, b, a / b values), soluble solids content (SSC), titratable acidity (TA), pH, antioxidant contents (lycopene, ascorbic acid, and total phenolics) and antioxidant activity were evaluated. Firmness was above the minimum marketable limit and fresh weight loss was below maximum acceptable weight loss after 3 weeks of storage, and no deleterious effect on antioxidant contents or activities were observed. Significant differences in SSC, TA, and pH were seen between varieties, but not between fruits stored for different durations. In both varieties, Hunter a values increased more than five-fold after 8 days of storage; this correlated with a more than four-fold accumulation of lycopene after two weeks of storage. The antioxidant activity of tomatoes was highest at the beginning of the storage period, likely because of the effective DPPH - reducing power of ascorbic acid and total phenolics. Antioxidant activity increased after 12 days of storage because of increasing lycopene content. Hence, this study indicates that pink - stage tomatoes may be stored at $12^{\circ}C$ for up to 3 weeks without affecting marketability or nutritional value.

Ethylene-Induced Auxin Sensitivity Changes in Petiole Epinasty of Tomato Mutant dgt

  • The tomato (Lycopersicon esculentum Mill.) mutant diageotropica (dgt) lacking normal gravitropic response is known to be less sensitive to auxin compared with its isogenic parent VFN8. Straight growth as well as ethylene production in response to added auxin in hypocotyl segments of dgt was negligible. However, there was no significant difference between the two genotypes in auxin transport in petiole segments and its inhibition by the phytotropin N-1-naphthylphthalamic acid(NPA). Kinetic parameters of NPA binding to microsomal membranes were also non-distinguishable between the two. Its petiolar explants treated with ethylene developed epinastic curvature with the magnitude of response increased about 3 folds over non-mutant wild type. Ethylene-induced epinasty in both dgt and VFN8 was nullified by treatment of explants with the ethylene autagonist 2,5-norbonadiene. Lateral transport of 3H-IAA toward the upper side of ethylene-treated petioles in dgt, however, was not significantly more pronounced than in VFN8, the implications being that auxin sensitivity in the mutant was restored, or even rised above the wild type, by ethylene.

Improvement of Shelf-life and Quality in Fresh-cut Tomato Slices

  • Quality of fresh-cut tomato slices was compared during cold storage under various modified atmosphere packaging conditions. Chilling injury of slices in containers sealed with Film A was higher than with Film B; these films had oxygen transmission rates of 87.4 and 60.0 ml h-1 m-2 nun-1 at $5^{\circ}C$ and $99\%$ RH, respectively. While slices in containers with an initial atmospheric composition of air, $4\%$ CO2 + 1 or $20\%\;O_2, \;8\%\;CO_2+1$ or $20\%\;O_2$, or $12\%\; CO_2+\;20\%\;O_2$ showed fungal growth, slices in containers with $12\%\;CO_2 +\;1\%\;O_2$ did not. Low ethylene in containers enhanced chilling injury. Modified atmosphere packaging provided good quality tomato slices with a shelf-life of 2 weeks or more at $5^{\circ}C$. Experiments were conducted to compare changes in quality of slices of red tomato (Lycopersicon esculentum Mill. 'Sunbeam') fruit from plants grown using black polyethylene or hairy vetch mulches under various foliar disease management systems including: no fungicide applications (NF), a disease forecasting model (Tom-Cast), and weekly fungicide applications (WF), during storage at $5^{\circ}C$ under a modified atmosphere. Slices were analyzed for firmness, soluble solids content (SCC), titratable acidity (TA), pH, electrolyte leakage, fungi, yeasts, and chilling injury. With both NF and Tom-Cast fungicide treatments, slices from tomato fruit grown with hairy vetch (Vicia villosa Roth) mulch were firmer than those from tomato fruit grown with black polyethylene mulch after 12 days storage. Ethylene Production of slices from fruit grown using hairy vetch mulch under Tom-Cast was about 1.5- and 5-fold higher than that of slices from WF and NF fungicide treatments after 12 days, respectively. The percentage of water-soaked areas (chilling injury) for slices from tomato fruit grown.

  • 고형배지경에서 육묘기와 정식 후의 배양액농도의 차이가 토마토의 생육에 미치는 영향을 구명하고자 본 실험을 수행하였다. 배지는 펄라이트, 버미큘라이트 및 피트모스를 사용하였으며, 배양액 농도는 육묘기 때에는 0.5, 1, 2, 3 및 5mS/cm로 각기 다르게 처리되었고 정식후에는 1, 2 및 3mS/cm로 처리되었다. 모든 배지에서 총과수, 총수량, 상품과수 및 상품수량은 1.0mS/cm보다 2.0-3.0mS/cm에서 훨씬 더 높았다. 기형과율은 피트모스>펄라이트>버미큘라이트 순이었으며 비타민C 함량은 버미큘라이트>펄라이트>피트모스 순이었다. 모든 배지에서 육묘기에는 2.0-5.0mS/cm, 정식 후에는 2.0-3.0mS/cm로 관리했을 때, 상품수량이 높게 나타났다. 육묘기 뿐만 아니라 정식 후의 배양액농도는 총수량, 상품수량 및 당도에 결정적인 영향을 미쳤다. 그러나, 총과수와 상품과수는 정식 후의 배양액농도에 의해 큰 영향을 받았다.

When stored at $20^{\circ}C$, slices from light-red tomato fruit grown with black polyethylene or hairy vetch mulches both showed a rapid increase in electrolyte leakage beginning 6 hours after slicing. However, slices from tomato fruit grown using the hairy vetch mulch tended to have lower electrolyte leakage than those grown with black polyethylene mulch. These results suggest that tomato fruit from plants grown using hairy vetch mulch may be more suitable for fresh-cut slices than those grown using black polyethylene mulch. Also, use of the disease forecasting model Tom-Cast, which can result in lower fungicide application than is currently used commercially, resulted in high quality fruit for fresh-cut processing. Experiments were conducted to determine if ethylene influences chilling injury, as measured by percentage of slices exhibiting water-soaked areas in fresh-cut tomato slices of 'Mountain Pride' and 'Sunbeam' tomato (Lycopersicon esculentum Mill.). Ethylene concentration in containers without ventilation significantly increased during storage at $5^{\circ}C$, whereas little or no accumulation of ethylene occurred in containers with one or six perforations. Chilling injury was greatest for slices in containers with six perforations, compared to slices in containers with one perforation, and was over 13-fold greater than that of slices in control containers with no perforations. An experiment was also performed to investigate the effectiveness of including an ethylene absorbent pad in containers on subsequent ethylene accumulation and chilling injury. While ethylene in the no-pad controls increased continually during storage of both 'Mountain Pride' and 'Sunbeam' tomatoes at $5^{\circ}C$ under modified atmosphere conditions, no increase in accumulation of ethylene was observed in containers containing ethylene absorbent pads throughout storage. The ethylene absorbent pad treatment resulted in a significantly higher percentage of chilling injury compared with the no-pad control. In studies aimed at inhibiting ethylene production using AVG during storage of slices, the concentration of ethylene in control containers (no AVG) remained at elevated levels throughout storage, compared to containers with slices treated with AVG. Chilling injury in slices treated with AVG was 5-fold greater than that of controls. Further, we tested the effect of ethylene pretreatment of slices on subsequent slice shelf-life and quality. In slices treated with ethylene (0, 0.1, 1, or $10\;{mu}L\;L^{-1}$) immediately after slicing, ethylene production in non-treated controls was greater than that of all other ethylene pre-treatments. However, pretreatment of slices 3 days after slicing resulted in a different pattern of ethylene production during storage. Ihe rate of ethylene production by slices treated with 1 L $L^{-1}$ ethylene 3 days after slicing was greater during storage than any of the other ethylene treatments. With slices pre-treated with ethylene, both immediately and 3 days after slicing, the rate of ethylene production tended to show an negative correlation with chilling injury.
