In this study, a poly(lactic-co-glycolic acid) (PLGA) microspheres was fabricated using emulsion solvent evaporation technique. During the procedure fabrication, some parameters process have effected on the formation of micro-carriers. The structure and morphology of micro-carriers were evaluated by SEM observation. Beside, heparin incorporated into microspheres was determined using toluidine blue method. Specifically, the effects of some parameters process such as ultrasonic levels, PLGA concentrations and freeze-dry times on the size, structure, porous formation and heparin entrapment of micro-carriers were studied carefully. We found that, the morphology and structure of carriers were influenced by the all above parameters. The diameter of the carriers varied from 20 to 400 ${\mu}M$ depending on experimental conditions. At suitable freeze-dry time, the pores were automatically formation on surface of microspheres with a significantly in the numbers of pore. After heparin incorporated porous PLGA microspheres, it was suggested that the highly heparin incorporated into porous PLGA microspheres could enhance of angiogenesis for tissue regeneration easily.

Generally, plants defend themselves against pathogens by structural and biochemical reactions. Defense structures act as physical barriers and inhibit the pathogen from gaining entrance and spreading through the plant. Xanthomonas axonopodis pv glycines, the causal pathogen of bacterial pustule of soybean, causes hypersensitive response (HR). When pepper fruits were inoculated with X. axonopodis pv. glycines, in situ, time-series defense-related structural changes occurred in the inoculated sites. Early responses were programmed cell death (PCD), characterized by condensation and vacuolization of the cytoplasm, condensation of nuclear materials, and fragmentation of the nuclear DNA, which were observed by transmission electron microscopy. Nuclear fragmentation was proven by TUNEL method under confocal laser scanning microscopy and DNA laddering through eletrophoresis. At later stages, plant responses were cell elongation and cell division, forming a periderm-like boundary layer that demarcated healthy tissues from the inoculation sites. Using several stains such as toluidine blue, sudan IV, annexin V, and phloroglucinol-HCl, defense-related materials and structural changes were also examined.
치근흡수 연구에서 치아재식을 위한 적당한 동물모델을 찾는일은 대단히 중요하다. 본 실험에서는 쥐 대구치의 발치 및 재식을 통해서 치근흡수가 발현되는 모양을 관찰했다. 모두 20마리의 쥐(30일생)에게 0.4% ${\beta}$-APN(aminoproprionitrile)가 포함된 분말용쥐사료를 5일간 먹인후 케타민 마취후 상악좌우측 제일대구치를 손상을 최소화하여 발치 하였다. 발거된 쥐치아는 0.02M Tris-HCl로 표면처리하여 부착치근막을 제거한후 파라핀팍스로 치근단을 밀폐 하여 재식하였다. 재식후 1, 2, 4, 6, 8 및 12일이 경과된 후 심장관혈을 통하여 쥐를 희생시킨후 3% glutaraldehyde가 포함된 EDTA에서 탈회한후 Epon 포매 하였다. 모든 시편은 유리칼을 사용하여 1미크론 두께로 절편한후 veronal acetate buffer에 녹인 1% toluidine blue로 염색하였다. 파치세포양 세포에 의한 치근흡수는 재식 5-7일후 처음 관찰되었다. 재식 12일후에는 치근 전체에 걸친 광범위한 치근흡수가 관찰되었다. 이상의 결과 ${\beta}$-APN을 포함한 쥐사료 식이요법은 쥐 상악제일대구치의 발치와 재식을 용이하게 하여 일정한 치근흡수를 발현시키는데 많은 도움이 된 것으로 판단되었다.
Background: In the maxillofacial region, giant cell granulomas occur in 2 clinical forms, central and peripheral. Despite histopathological similarity between these 2 forms totally different clinical behaviors have been reported. The present study was undertaken to compare mast cell and vascular concentrations in these pathologic lesions. Materials and Methods: In this cross-sectional descriptive study, 20 pathological samples of central giant cell granuloma (CGCG) and 20 samples of peripheral giant cell granuloma (PGCG) were selected and examined through toluidine blue staining for mast cell assessment and immunohistochemical staining by VEGEF antibody for comparing the number of mast cells. T-test, chi-squared test and backward multivariate linear regression were used for statistical analysis using SPSS 20. Statistical significance was set at P<0.05. Results: This study showed significantly greater VEGF expression and mast cell concentrations in CGCG compared to PGCG cases. Also there was a significant correlation between VEGF expression and the concentration of mast cells. No relation was found between age, sex and site of the lesion and concentration of mast cells or VEGF expression. Conclusions: It is feasible that higher concentrations of mast cells in CGCG versus PGCG samples might lead to more aggressive clinical behavior via vascular proliferation and angiogenesis. However, other biologic mechanisms should be considered in this situation.
Structural changes of the endosperm of Panax ginseng C.A. Meyer from fertilization to germination were investigated by light microscope. The endosperm of the ginseng seed is cellular type. Since endosperm cells adjacent embryo continuously breakdown and disappear with the elongation of embryo, the real of endosperm is gradually decreased. As the anatropous ovules of immature seed with green seed coat developes more and more, ovary cells adjacent ovary cavity become abundant by the periclinal division, their size is decreased, hypotrophy of cell wall discern, and they are gradually differentiated in seed coat. Though embryo responds strongly to basic dye at the stage of completion of endosperm formation, tissue of endosperm responds to acidic dye positively Cell wall of embryo and endosperm are composed of primary cell wall not lignified. Endosperm cells adjacent embryo begin to breakdown in the endosperm tissue of indehiscent seed before the beginning of the after-ripening. Dehiscent seed of which seed coat is opened through after-ripening represent the form as a seedling in the result of embryo developments with the formation of organs; radicle, cotyledon, plumule. Umbilifom layer represents strong positive response to the toluidine blue and the basic function. Umbiliform layer that endosperm cells breakdown and disappear is observed clearly at the periphery of the embryo cotylemon, while slightly at the periphery of the radicle.
This study reports significant improvement of atopic dermatitis condition as a result of experiment using Xanthium strumarium L. extract (XS-E) at the dorsal skin of induced atopic dermatitis Nc/Nga mice. Skin clinical score has decreased ($2.75{\pm}0.85$, *p<0.05), showing visible change of skin condition. IgE (***p<0.001) and IgG1 ($2522.00{\pm}32.80$, ***p<0.001) in plasma also decreased significantly. mRNA (gene expression) level increased ($RQ=2.75{\pm}0.10$, ***p<0.001) within skin tissue of CD4+CD25+Foxp3+ Treg cell that's activated by XS-E dosage, thereby discovering that there is an effect of suppressing proliferation and viability of Th2 cell, eosinophils, mast cell and inflammatory cell. Upon examining cells permeated with H&E and toluidine blue staining technique, thickness of epidermis and mast cell's permeation decreased, and the result of examining the distribution of CCR 3+ eosinophils within ALN showed that it's level fell down to that of wild type (normal group, NC/Nga-WT). This study was performed in order to evaluate the accuracy and the usefulness of the cytology of crush preparation in central nervous system (CNS) lesions. Forty four intraoperative biopsies were performed at the time of craniotomy including 34 benign and 10 malignant lesions. Crush preparations were prepared from tiny tissue fragments of craniotomy products. All cases were stained with toluidine blue. Intraoperative diagnoses made on cytologic examination were compared with the final paraffin section diagnoses. Comparison between the results of the cytologic and histologic findings revealed an overall diagnostic accuracy of 88.6%. This study attests to the diagnostic accuracy of cytologic examination in CNS lesions. The detailed cytologic features are described and important criteria for the cytodiagnosis of CNS lesions are discussed. This review leads us to think that cytologic examination by crush preparation in CNS lesions is considered as a simple, safe and highly diagnostically accurate method.
Objectives : In this study, we investigated the effect of modified-Okbyungpoongsan (mOP) on mast cell-mediated allergic response in basophilic leukemia cell line, RBL-2H3 mast cells. Methods : Cells were stimulated with anti-DNP-IgE after the treatment of DNP-HSA (AI/D), and then incubated with different concentrations of mOP (0.1, 0.2, 0.5 and 1 mg/$m{\ell}$) in RBL-2H3 cells. Cell toxicity was determined by WST-1 assay. The degranulation of mast cells was observed by microscope with toluidine blue staining and also the levels of beta-hexosaminidase, histamine and TNF-alpha were measured in culture supernatants by enzyme-linked immunosorbant assay. Results : mOP inhibited anti-DNP-IgE-imduced degranulation of mast cells in RBL-2H3 cells. mOP also significantly decreased the levels of histamine and inflammatory cytokine, TNF-alpha in RBL-2H3 cells, but slightly decreased the level of beta-hexosaminidase. Conclusions : These results indicate that mOP, an oriental prescription could be inhibit the allergic response through suppressing the mast cell activation.

In order to investigate the efficacy of a combination of JBB as topical and HTGMB as internal treatment method, changes in various immune related factors and histological changes in NC/Nga induced animal model was studied. Combined treatment of topical JBB and internal HTGMB significantly reduced the atopic dermatitis clinical index, the number of immune cells such as CD19+, CCR3+, B220+/IgE+, and Gr-1+/CD11b+ in DLN and dorsal skin, compared to the control group. Otherwise increased CD3+, CD4+/CD25+, CD8+ and CD4+ cells in the DLN. And also combined treatment of topical JBB and internal HTGMB suppressed the lymphocytes and mast cells from infiltrating into the skin tissues when stained with H&E and toluidine blue. Based on the results above, it is strongly suggested that the combined treatment of topical JBB and internal HTGMB significantly induced anti-allergic activities through immune modulation. The findings can be applied to developing a more sustainable treatment for atopic dermatitis and be helpful in practicing combined treatments in clinical treatments in the future.
The aim of the present study is to demonstrate the usefulness of intraoperative fine needle aspiration (FNA) of pancreatic lesions in 30 patients. A conclusive diagnosis was done in 27 patients and the diagnoses of three patients were deferred. No complications followed the procedure. Based on histologic findings of the resected specimens in 20 cases and of cell blocks in 10 cases, the final diagnoses were adenocarcinoma in 19 cases, chronic pancreatitis in nine cases and tuberculosis in two cases. The sensitivity, specificity and diagnostic accuracy were 95%, 100% and 96% resepectively and there were no false positives. The smear of aspirate was stained with toluidine blue and examined by light microscope. The presence of there-dimensional clusters of disoriented cells and the increased nuclear/cytoplasmic ratio with large prominent nucleoli were the most helpful criteria for a diagnosis of malignancy in the pancreas. The intraoperative FNA of pancreatic lesions was considered as a simple, safe, and highly specific and sensitive tool in differentiating benign from malignant lesions. The intraoperative FNA can be recommended as the first tool of choice of intraoperative diagnostic procedure in lesions of the pancreas.
