Selection of stress-tolerant ginseng lines in fields is very difficult because it is almost impossible to control properly the environmental conditions of soil. On the contrary, it can be studied with ease to search for stress-tolerant ginseng lines through in vitro culture because of easy manipulation of stress conditions. This study was conducted for the selection of ginseng pure lines tolerant to salt stress. Murashige & Skoog(MS) media with 2.5 folds of KNO$_3$, NH$_4$NO$_3$, MgSO$_4$.7$H_2O$, KH$_2$PO$_4$, and CaC1$_2$.2$H_2O$ was established for the selection of ginseng pure lines tolerant to salt stress in vitro. Among 88 ginseng pure lines bred by Korea Ginseng and Tobacco Research Institute, Punggi Hwangsuk, 78093, 82886, 78135, 86024 and KG104 lines was tolerant to salt stress. For the stable production of quality Korean ginseng, genetic tolerance to salt stress is one of important factors since relatively high salt concentrations in the ginseng nursery soil environment of Korea. Ginseng inbred pure lines were tested for their tolerance to salt stress through in vitro culture technique.
Tolerance against mercuric chloride in mutants of Pyricularia oryzae Cavara induced by ultraviolet(UV) irradiation has been investigated. The tolerant isolates obtained using ultraviolet(UV) irradiation were maintained a high level of tolerance even after 15 times transfer to the chemical free media. Each isolate of mutants UM-1, UM-2, and UM-3 on the successive monoconidial culture has genetically homogeneous for tolerance. The tolerant isolates sporulated less and showed a higher percentage of germination of conidia on media without the chemical than the parental isolate. The xotins released from the parental and the tolerant isolates have been identified as piricularin and $\alpha$-picolinic acid by paper-chromatography. The tolerant isolates produced more piricularin and its virulence on seedlings of rice varieties were higher than the parental isolates. Both piricularin production and virulance on rice were highest in the UM-2 isolate.

In the present study we evaluated the efficacy of a bioformulation of Streptomyces cameroonensis for control of black pod disease in cocoa and enhancement of seedling growth. The formulation developed using talc powder and cassava starch as carriers showed high shelf-life of 1.07 × 106 CFU/g after six months storage at 4℃. The formulation was tested for inhibition of spore germination in Phytophthora megakarya and showed 100% inhibition at 10% (w/v) of formulation. To determine the efficacy of the formulation, we performed an in planta assay in the greenhouse on two hybrids of cocoa seedlings, the tolerant SNK413 × (♂) T79/467 and the susceptible UPA 134× (♂) SCA 12. Detached leaf assay showed a significant reduction in the disease severity index of about 67% for the tolerant hybrid and 55% for the susceptible hybrid compared to non-treated plants. A significant enhancement in stem length, leaf surface area and root weight was observed. Analysis of biochemical markers of defense showed a significant increase in total polyphenol, flavonoid, and total protein contents. There was also significant upregulation of PR-proteins such as chitinases, peroxidases and β-1, 3-glucanases following treatment of both tolerant and susceptible hybrids, though with a higher level of synthesis in the tolerant hybrids. A significant increase was also observed in polyphenol oxidase activities in plants treated with the formulation. This work demonstrated the stability and effectiveness of the S. cameroonensis powder formulation in suppressing black pod disease in cocoa and subsequently enhancing the growth of seedlings.

Grafting using a pumpkin (Cucurbita sp.) rootstock is an effective way to improve cucumber (Cucumis sativus) resistance to a combination of chilling and salinity stresses. We evaluated the tolerance of 15 pumpkin cultivars to chilling, salinity, and combined stresses at the germination and seedling stages. Selected plant characteristics, including germination rate, germination potential, germination index, plant height, stem thickness, fresh weight, and dry weight, were analyzed. We used the unweighted pair group method with arithmetic mean for cluster analyses to determine the stress tolerance levels of the pumpkin cultivars. The 15 cultivars were divided into three clusters: tolerant, moderately tolerant, and susceptible to stress treatments. The stress tolerances of all cultivars were variable in the germination and seedling stages, and most cultivars were not tolerant to individual treatments of chilling or salinity stresses at both stages. These results suggest that identifying suitable cultivars for use as rootstock during cucumber grafting should involve the evaluation of stress tolerance during different growth stages. Additionally, cultivars tolerant to chilling stress may not be tolerant to salinity stress; therefore, the choice of pumpkin rootstock should depend on where the grafted plant will be grown. Cultivars tolerant to a combination of chilling and salinity stresses may be useful as rootstock for cucumber grafting. Our findings may serve as reference material for choosing appropriate pumpkin rootstocks for cucumber grafting.
Using salt-tolerant bacteria to protect plants from salt stress is a promising microbiological treatment strategy for saline-alkali soil improvement. Here, we conducted research on the growth-promoting effect of Brevibacterium frigoritolerans on wheat under salt stress, which has rarely been addressed before. The synergistic effect of B. frigoritolerans combined with representative salt-tolerant bacteria Bacillus velezensis and Bacillus thuringiensis to promote the development of wheat under salt stress was also further studied. Our approach involved two steps: investigation of the plant growth-promoting traits of each strain at six salt stress levels (0, 2, 4, 6, 8, and 10%); examination of the effects of the strains (single or in combination) inoculated on wheat in different salt stress conditions (0, 50, 100, 200, 300, and 400 mM). The experiment of plant growth-promoting traits indicated that among three strains, B. frigoritolerans had the most potential for promoting wheat parameters. In single-strain inoculation, B. frigoritolerans showed the best performance of plant growth promotion. Moreover, a pot experiment proved that the plant growth-promoting potential of co-inoculation with three strains on wheat is better than single-strain inoculation under salt stress condition. Up to now, this is the first report suggesting that B. frigoritolerans has the potential to promote wheat growth under salt stress, especially combined with B. velezensis and B. thuringiensis.
The global commercial cultivation of transgenic crops, including glyphosate-tolerant soybean, has increased widely in recent decades with potential impact on the environment. The bulk of previous studies showed different results on the effects of the release of transgenic plants on the soil microbial community, especially rhizosphere bacteria. In this study, comparative analyses of the bacterial communities in the rhizosphere soils and surrounding soils were performed between the glyphosate-tolerant soybean line NZL06-698 (or simply N698), containing a glyphosate-insensitive EPSPS gene, and its control cultivar Mengdou12 (or simply MD12), by a 16S ribosomal RNA gene (16S rDNA) amplicon sequencing-based Illumina MiSeq platform. No statistically significant difference was found in the overall alpha diversity of the rhizosphere bacterial communities, although the species richness and evenness of the bacteria increased in the rhizosphere of N698 compared with that of MD12. Some influence on phylogenetic diversity of the rhizosphere bacterial communities was found between N698 and MD12 by beta diversity analysis based on weighted UniFrac distance. Furthermore, the relative abundances of part rhizosphere bacterial phyla and genera, which included some nitrogen-fixing bacteria, were significantly different between N698 and MD12. Our present results indicate some impact of the glyphosate-tolerant soybean line N698 on the phylogenetic diversity of rhizosphere bacterial communities together with a significant difference in the relative abundances of part rhizosphere bacteria at different classification levels as compared with its control cultivar MD12, when a comparative analysis of surrounding soils between N698 and MD12 was used as a systematic contrast study.
In order to clarify the structure and seasonal dynamics of warm tolerant benthic marine algal community in Korea, the species composition and biomass of marine algae at the discharge canal of Uljin nuclear power plant on the East Coast of Korea were investigated seasonally from February 1992 to October 2000. 107 species of marine algae were found at the discharge canal during the past nine years. In general, the number of species observed was abundant in spring or summer and less in autumn or winter. 27 species (4 blue-green, 5 green, 6 brown and 12 red algae) of marine algae occurred more than 1/ 6 frequency and thus can be categorized as warm tolerant species. Among these, one brown (Dictyota dichotoma) and four red algae (Gelidium amansii, Anphiroa ephedraea, Hydrolithon sargassi, Marginisporum crassissimum) are recorded as warm tolerant marine algae for the first time in Korea. Padina arborescens, Anphiroa zonata and Corallina pilulifera were common species found more than 75% frequency. Seasonal fluctuations of mean biomass were 0-1,330 g dry wt m^(-2) and dominant species in biomass were Corallina pilulifera (contribution to a total biomass proportion 34%), Anphiroa zonata (23%), Padina arborescens (18%) and Sargassum micracanthum (11%). The red algae appeared as predominant algal group at the discharge canal of Uljin nuclear power plant in the qualitative and quantitative aspects. The green algae such as Enterornorpha compressa appeared rather frequently at the discharge canal, but the biomass proportion was very low, in contrast to Kori nuclear power plant where there was definite green algal dominance. Differences in algal communities developed at the discharge canals of Uljin and Kori nuclear power plant on the East Coast of Korea, particularly biomass proportions of green algae, can probably be related to local environmental factors such as water velocity through the canal and natural seawater temperatures.

The objectives were to compare the salt tolerance levels in the parental rice cultivar, Dongjinbyeo, and induced mutagenesis derived its lines for plant height, MDA, ATPase, POD, and 2-dimensional protein electrophoresis pattern in NaCl-containing hydroponic nutrient solutions. Rice plants isolated from a population of rice (Oryza sativa L. cv. Dongjinbyeo) mutation lines, which were generated in combination with in vitro selection and gamma-ray, exhibited salt tolerance. Line No. 18 had the longest plant, whereas NaCl-sensitive line (No. 25) had the shortest plant. The parent, and the sensitive line showed severe damage from salt stress. Tolerant lines (No. 18, 50) had a lower malonaldehyde (MDA) content than the sensitive one (Dongjinbyeo, No. 25) during salt stress. Several proteins showed significant quantitative variation through 2DE; phosphoribulokinase, peroxidase, oxygen evolving enhancer 1 and the $H^+-ATPase$, which are known to be involved in salt tolerance. The effect of salt on peroxidase and $H^+-ATPase$ activity in the seedlings of two groups with contrasting genotypes of rice was studied. A greater activity was recorded in the tolerant lines as compared to the sensitive ones (P<0.05, Duncan's test). The results indicate that salt tolerant lines expressed more salt stress-inducible proteins associated with salt tolerance than the sensitive lines during salt stress.
Marine algal flora and community structure were examined seasonally at four study sites around Taean Power Plant, Korea from Jan. to Nov. 2006. A total of 73 algae species (12 green, 9 brown, 52 red) and 1 marine plant were identified. The number of species was maximal at the power plant Discharge (57 species) site followed by Hakampo (46 species), Intake (28 species) and Breakwater (15 species) sites during the study period. The average biomass in dry weight varied from 13.12g/m2 at Intake to 69.60g/m2 at Hakampo. Dominant and sub dominant species in terms of biomass were Gelidium divaricatum - Ulva pertusa at Intake, Chondria crassicaulis - Ulva pertusa at Discharge, Corallin a pilulifera - Chondrus ocellatus at Breakwater, and Corallina pilulifera - Sargassum thunbergii at Hakampo. Species richness of warm tolerant and green algae were greater at Discharge site than Hakampo, showing similar species richness. However, community indices were not distinguishable between Discharge and other study sites. In conclusion, species richness and biomass of seaweeds were greater at Discharge site compared to intake and breakwater sites, and the abundance of warm tolerant and green algal species were higher than Hakampo.
In order to examine the cross-tolerance of two chilling-tolerant cultivars (Donganbyeo and Heukhyangbyeo) and two chilling-susceptible cultivars (Hyangmibyeo and Taekbaekbyeo) to salt, paraquat, and drought, changes of physiological response and antioxidant enzymes were investigated. The seedlings were grown in a growth chamber until the 4-leaf stage. The seedlings were exposed to chilling at $5^{\circ}C$ for 3 days. For drought treatment, the seedlings were subjected to drought by withholding water from plants for 5 days. For paraquat study, plants were sprayed with $300{\mu}M$ paraquat. For the salt stress, the seedlings were transferred to the Hoagland's nutrient solution containing 0.6% (w/v) NaCl for 4 days. Chilling-tolerant cultivars showed cross-tolerant to other stresses, salt, paraquat, and drought in physiological parameters, such as leaf injury, chlorophyll a fluorescence, and lipid peroxidation. The baseline levels of antioxidative enzyme activities, catalase (CAT) and peroxidase (POX) activities in chilling-tolerant cultivars were higher than in the chilling-susceptible cultivars. However, there were no differences in ascorbate peroxidase (APX) and glutathione reductase (GR) activities between chilling-tolerant and -susceptible cultivars in untreated control. CAT activity in chilling-tolerant cultivars was higher than that in chilling-susceptible cultivars during chilling, salt, and drought treatments, but not during paraquat treatment. However, other antioxidative enzymes, APX, POX, and GR activities showed no significant differences between chilling-tolerant and -susceptible cultivars during chilling, salt, paraquat, and drought treatments. Thus, it was assumed that CAT contribute to cross-tolerance mechanism of chilling, salt, and drought in rice plants.
