Acifluorfen-tolerant callus lines of Solanum ptycanthum were isolated by stepwise selection. Growth of the unselected line was completely inhibited at 0.5 uM. while some selected lines grew at 8 uM acifluorfen. Twenty-two of twenty-five acifluorfen-tolerant callus lines regenerated shoots. Many of the regenerated somaclones were variants, differing in leaf shape, leaf color, number of flower parts, flower color, and fertility. The acifluorfen tolerant S. ptycanthum callus lines differed.

The fast-changing climatic conditions make plants to be vulnerable to many abiotic stresses. Drought stress is one of the limiting factors that affect pepper production in water deficient regions. It affects plant growth and development by altering physiological, morphological, and metabolic processes. Breeding drought tolerant varieties is one of the mitigation strategies to overcome the ever increasing drought disaster. Hence, screening of new drought tolerant pepper genotypes is essential. The current study was aimed to identify new drought tolerant genotypes among the collection of pepper genetic resources. In total, 70 pepper genotypes were screened for drought tolerance after exposure to drought stress condition. The pepper genotypes were classified as highly tolerant, intermediate, or severely sensitive to drought stress based on the phenotypic analysis. Consequently, 13 genotypes significantly exhibited higher recovery rate after drought stress and were classified as highly tolerant. Comparative analysis of morphological and physiological parameters and expression of drought responsive genes between tolerant and susceptible pepper genotypes will be presented and discussed. The identified tolerant genotypes will be useful resources for breeding drought tolerant pepper cultivars.
Saline or alkaline condition in soil inhibits growth of most crop plants and limits crop yields in many parts of the world. Augmenting an alkaline soil with alkali-tolerant bacteria capable of promoting plant growth can be a promising approach in expanding fertile agricultural land. Near-shore environments of Dokdo Island, a remote island located in the middle of the East Sea, appear to have patches of seawater-influenced haloalkaline soil that is unsupportive for growth of conventional plants. To exploit metabolic capacities of alkali-tolerant bacteria for promoting plant growth in saline or alkaline soils, we isolated of alkali-tolerant bacteria from near-shore soil samples in Dokdo and investigated properties of the isolates. Alkali-tolerant bacteria were selectively cultivated by inoculating suspended and diluted soil samples on a plate medium adjusted to pH 10. Fifty colonies were identified based on their $GTG_5$-PCR genomic fingerprints and 16S rRNA gene sequences. Most isolates were affiliated to alkali-tolerant and/or halotolerant genera or species of the phyla Firmicutes (68%), Proteobacteria (30%) and Actinobacteria (2%). Unlike the typical soil bacterial flora in the island, alkali-tolerant isolates belonged to only certain taxa of terrestrial origin under the three phyla, which have traits of plant growth promoting activities including detoxification, phytohormone production, disease/pest control, nitrogen-fixation, phosphate solubilization or siderophore production. However, Firmicutes of marine origin generally dominated the alkali-tolerant community. Results of this study suggest that haloalkaline environments like Dokdo shore soils are important sources for plant growth promoting bacteria that can be employed in bio-augmentation of vegetation-poor alkaline soils.
The significance of environment change and genetic safety has been recently recognized by many genetically modified (GM) plants. This study was to evaluate the safety of drought-tolerant rice and to identify the environment variance. The GM rice of drought-tolerant rice and four check cultivars were analyzed the data on agronomic characters and principal component in large-GM crop field. There was no significant difference in agronomic characters between the drought-tolerant rice and donor plant, 'Ilmi'. Grain yield showed the standard deviation of the difference, did not significant statistically. Related to grain characters, grain appearance were similar to the drought-tolerant rice and donor plant, 'Ilmi'. In Chemical characters, brown rice of the drought-tolerant rice and a donor plant, 'Ilmi' did difference in starch and protein, however, was similar as 'Ilpum'. These results indicated that drought-tolerant rice may perform to detect genetic safety in GM plants progeny.
Doubled haploids have long been recognized as a valuable tool in plant breeding since it not only offers the quickest method of advancing heterozygous breeding lines to homozygosity, but also increased the selection efficiency over conventional procedures due to better discrimination between genotypes within any one generation. Salt tolerant mutants were obtained in rice the variety, 'Hawsungbyeo', through in vitro mutagenesis of in vitro cultured anther-derived calli. Various doses (30, 50, 70 and 90 Gy) of gamma ray were applied to investigate the effect of radiation on callus formation on medium containing 1% NaCl, green plant regeneration, frequency of selected doubled haploid mutants and of the salt tolerant screen. It was demonstrated that the dose of 30 and 50 Gy gamma rays had significant effects on callus formation, regeneration and selection of salt tolerance. No tolerant lines were obtained from non-mutagenized cultures. From gamma ray irradiated cultures, five tolerant lines ($M_2$generation) at germination stage and 13 tolerant lines ($M_3$genoration) at seedling stage were obtained. The frequency of salt tolerant mutants indicates that anther culture applied in connection with gamma rays is an effective way to improve salt tolerance.
The four tolerant and one susceptible plant to oxyfluorfen were selected from 26 species and investigated for the inhibition of protox activity, the PPIX accumulation, and the activity of antioxidative enzymes by oxyfluorfen treatment. When treated, the oxyfluorfen-tolerant plant species showed a less decrease in fresh weight and height than the susceptible one. The susceptible barnyardgrass showed more inhibition of protox activity due to the treatment of oxyfluorfen than the tolerant species. Especially at the concentration of $10^{-6}M$, protox activity of the susceptible barnyardgrass was inhibited completely, but tolerant species maintained $25{\sim}45%$ of the activity. Under the light and dark condition, the susceptible barnyardgrass showed more PPIX accumulation than the tolerant.

An efficient method to select drought tolerant Korean native plants using in vitro culture system was established in this study. While the plant growths and root inductions of each plant were proportionately affected by concentrations of mannitol on in vitro culturing seven plant species to test tolerance to osmotic stress, growth index (GI) and number of root induction of Chrysanthemi zawadskii var. latilobum and Dianthus chinensis var. semperflorens plantlets were higher than the others in 125mM mannitol. In test with polyethylene glycol (PEG), plantlets of C. zawadskii var. latilobum and D. chinensis var. semperflorens showed higher GI and number of root induction than the others in 33.3mM. On testing whether the well grown plants under osmotic stress are tolerant to virtual drought stress, there were significant differences in the withering rates of C. zawadskii var. latilobum and D. chinensis and those of were Aster yomena and Centaurea cyanus after 12 days without watering. It was found that significantly lower stomata numbers were shown in both drought tolerant plants than the sensitive plants. Averages of the stomata circumferences and the stomata area in the plantlets of the tolerant species were larger than those of the sensitive plants D. chinensis var. semperflorens showed the lowest transpiration level per unit area. The highest stomatal area per unit area was found in C. zawadskii, followed by D. chinensis var. semperflorens, Aster yomena and C. cyanus. In conclusion, C. zawadskii var. latilobum and D. chinensis var. semperflorens were more tolerant to drought than other two species. Furthermore in vitro selection was successfully used to screen drought tolerance species of native plant species.

A fault tolerant control system, in which a digital back-up controller system is added on the existing analog control system, is developed for enhancing reliability of boiler control system in power plant. The digital back-up controller system(DBCS) has a multi-processor structure with capabilities of fault diagnosis, back-up control, self test, and graphic monitoring. Specifically, switching mechanism composed of expandable modules is designed so that back-up controller takes over any faulty control loops and the number of back-up control loops is determined as that of simultaneous faults. A process simulator that simulates the boiler analog control system is developed for safety test and performance evaluation prior to real plant application. DBCS is installed at the Ulsan thermal power plant, and fault tolerant control performance is assured under the faults that some controller modules are pulled out.
The combination of radiation technique with an in vitro culture system was initiated to develop salt tolerant rice. We established an in vitro culture system to select tolerant lines against salt stress. NaCl tolerant cell lines were selected from the callus irradiated with gamma ray on N$_{6}$ medium with 1.5% NaCl and 2 mg/L 2,4-D. Regenerants (M$_1$) were obtained from the tolerant callus which was cultured for 30 days auxin-free medium. The M$_2$seeds were harvested from M$_1$plants on an individual plant basis. Thirty seedlings from each 450 M$_2$lines were transplanted in a field and total 5,000 M$_3$lines were harvested with an average 90 percent of fertile grain. M$_3$lines were utilized for selection of salt tolerance. Salinity-tolerant lines (225) were selected among 5,000 M$_3$lines. Of the 225 lines tested, the morphological traits of two lines (120-10 and -11) were far superior to control (Donagjinbyeo) in agromomic traits such as plant height, root length and no. of roots. Control and tolerant lines were analyzed by RAPD markers. Three polymorphic bands were presented in only tolerant lines, demonstrating a genetic difference between control and the tolerant lines. Such tolerant lines could be used as genetic resources to improve salt tolerance.e.
