Helicobacter pylori is the etiologic agent of human gastritis and peptic ulceration and produces urease as the major protein component on its surface. H. pylori urease is known to serve as a potent immunogen as well as major virulence factor. In order to express the recombinant urease in tobacco plants, a DNA fragment containing the minimal H. pylori urease gene cluster was subcloned into a plant expression vector. The recombinant vector was transformed to tobacco plants. The integration of the recombinant plasmids into tobacco chromosomal genome was verified by genomic PCR. Expression to mRNA was confirmed by Northern blot analysis, and expression to recombinant urease protein was observed by Western blot analysis. These results showed that the recombinant urease can be produced in tobacco plants and will be tested for immune response to use as an edible vaccine.
Effect of several nitrogen fertilizer sources on the chlorine absorption by the burley tobacco plants was investigated under the field and pot condition. The nitrogen sources included compound fertilize.(containing 3.9% NH4-N and 6.1% NH2-N), (NH4)2SO4, NaNO3. (NH2)2CO and NH4NO3. The chlorine content of leaf during growing stage was high in (NH4)2SO4 plot , and the differences among nitrogen sources was remarkable at maximum growing stage. The chlorine content of cured leaf was high in (NH4)2SO4 plot. When the (NH4)2SO4 was applied, the total alkaloid content of cured leaf was increased and the color of cured leaf became undesirable with the increment of leaf chlorine. The yield, quality and value of cured leaf were high in NaNO3 plot , while low in (NH4)2SO4 plot.
To classify the inheritance of resistance to potato virus Y, crosses between susceptible flue-cured tobacco variety NC 95 and resistant variety McNair 30 were conducted. The parents, $F_1$ plants, $F_2$ populations, and haploid plants derived from anthers of $F_1$ plants were screened for a resistance of two potato virus Y strains (PVY-VB and PVY-VN) isolated in Korea. The Chi-square values for the $F_3$ populations and haploids of $F_1$ fitted 1 :3 and 1 :1 ratios of resistant to susceptible for two strains, respectively. Therefore, it was found that the resistance of McNair 30 for the potato virus Y was controlled by a single recessive gene. Moreover the resistance to two strains screened was inherited dependently.
The ecophysiological changes occurring upon cold stress were studied using cold tolerant transgenic and wild-type tobacco plants. In a previous study, cold tolerance in tobacco was induced by the introduction of a gene encoding the zinc finger transcription factor, PIF1. Gas-exchange measurements including net photosynthesis and stomatal conductance were performed prior to, in the middle of, and after a cold-stress treatment of $1{\pm}2^{\circ}C$ for 96 h in each of the four seasons. In both transgenic and wild-type plants, gas-exchange parameters were severely decreased in the middle of the cold treatment, but had recovered after 2-3 h of adaptation in a greenhouse. Most t-test comparisons on gas-exchange measurements between the two plant types did not show statistical significance. Wild-type plants had slightly more water-soaked damage on the leaves than the transgenic plants. A light-response curve did not show any differences between the two plant types. However, the curve for assimilation-internal $CO_2$ in wild-type plants showed a much higher slope than that of the PIF1 transgenic plants. This means that the wild-type plant is more capable of regenerating Ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) and has greater electron transport capacity. In conclusion, cold-resistant transgenic tobacco plants demonstrated a better recovery of net photosynthesis and stomatal conductance after cold-stress treatment compared to wild-type plants, but the ecophysiological recoveries of the transgenic plants were not statistically significant.
Transgenic tobacco plants expressing calmodulin derivative($lys{\rightarrow}ile$ 115 calmodulin) and hygromycin resistance genes or hygromycin resistance gene alone(control) were generated by Agrobacterium-mediated DNA transfer. Seeds obtained from the transgenic plants($F_o$) were tested for resistance to hygromycin and the expected 3 : 1 ratio was observed. The expression of calmodulin derivative in the tobacco plants was identified by a combined method of Western blot and Chemiluminescence. The effects of surface sterilizers on the germiation of seeds from transgenic tobacco plants were tested in Murashige and Skoog agar medium. Seeds obtained from transgenic tobacoo plants expressing the calmodulin derivative showed no fungi contamination with normal germination by treating with sterilized water alone or sodium hypochlorite(2% effective chlorine). In contrast, seeds from the control transgenic tobacco plants showed severe contamination with fungi by treating with sterilized water alone and showed no contamination with normal germination by treating with sodium hypochlorite(2% chlorine). The effects of calcium concentration on the germination of seeds from transgenic tobacco plants were tested in Murashige and Skoog agar medium. Seeds obtained from transgenic tobacco plants expressing the calmodulin derivative showed better germination frequency than that of the control transgenic tobacco seeds in the medium containing 30 mM $CaCl_2$. The data raise the possibility that the expression of calmodulin derivative gene in tobacco plants could increase adaptability of the seeds to environmental stresses.

CHilling tolereance of plants are closely correlated with the degree of fatty acid unsaturation of membrane lipids. We studied the effects of low-temperature photoinhibition on the photochemical efficiency of photosystem II in terms of fatty acid unsaturation of thylakoid membranes lipids isolated from chilling -sensitive plants and chilling -resistant ones. To directly test the chilling tolerance of photosynthetic machinery in relation to membrane lipids, we further compared wild type tobacco plants with that of transgenic tobacco plants, in which the sensitivity to chilling had been enhanced by genetic modification of fatty acid unsaturation of chloroplast membrane lipids. The transgenic tobacco plants were found to contain reduced levels of unsaturated membrane fatty acids after being transformed with cDNA for glycerol-3-phophate acyltransferase from squash. The functional integrity of photosystem II during and recovery of photosynthesis from low-temperature photoinhibition will be discussed in connection with the degree of fatty acid unsaturation of chlorophast membranes lipids.
In order to transform pokeweed antiviral protein cDNA to tobacco plant, total RNA was extracted from Phytolacca americana. PAP-II cDNA was synthesized from purified total RNA via RT-PCR and subcloned to recombinant vector pBluescript II SK-. 10 deletion mutant PAP-II cDNA fragments which were sequentially deleted from N-terminal by 90bp were synthesized from PAP-II cDNA except leading frame by PCR with primers designed in our laboratory. To select non-cytotoxic clone, pAc55M was constructed with yeast expression vector pAc55 and multicloning site(MCS). Sequentially deleted mutant PAP-II cDNAs were cloned on downstream of gall promoter of pAc55M. 6 non-cytotoxic deletion mutant PAP-II cDNA were selected. Selected cDNAs were cloned into plant expression vector pKGT101BH for transformation of these clones to plant through Agrobacterium tumefacience. After cloning, recombinant pKGT101BH carrying deleted mutant PAP-IIcDNA were transformed to Nicotiana tabacum cv. NC567. Transformed tobacco plants cultured on shooting and rooting media were transfered to green-house. About four weeks later, these plants were infected with physically infection using carborandum with PVY-VN strain. After 4 weeks, plants resistant to virus were selected , and seeds of these plants were gathered. Southern blot hybridization showed deleted fragments by 220bp and 420bp, so resistant ability of these plants is due to mutant PAP-II cDNA.
Tobacco diseases have not been recorded until 1900s in Korea, where tobacco plants were introduced at early 1700s. Practical researches on the disease have been conducted since mid 1960s. Major ten tobacco diseases were mosaic caused by tobacco mosaic virus·potato virus Y·cucumber mosaic virus, bacterial wilt, hollow stalk, wild fire caused by angular leaf spot strain, black shank, brown spot, powdery mildew and fusarium wilt. But their annual occurrences were varied according to changes of tobacco varieties and their cultivating practices. As no useful chemicals, several biological tactics have been developed to control the viral or bacterial diseases that give significant economic damages on sustainable crop yield, but not practicable to field farming condition yet. Transgenic tobacco plants containing foreign disease resistant genes have been developed by current bio-technology, but not released to farmers yet. Though some disease-resistant tobacco varieties have been developed by the conventional breeding technology and currently used by farmers, their disease controlling efficacy have been diminished by occurrence of the new strain or race. Future research on tobacco diseases has been focused on technical development to produce high quality tobacco with less production cost, which leads Korean tobacco industry to keep its competence against foreign industry and decreasing overall market.
Fourteen isolates of Pseudomonas solanarearum virulent to tobacco cultivars were selected among isolates obtained from tobacco plants shelving wilt symptoms throughout the country during $1980\~1981$. The fourteen isolates were classified into two races based on reactions in eggplant, tomato, red pepper, potato and tobacco, and also classified into biochemical type 1 and type 4 according to physiological characteristics.
Leaf water and osmotic potential, chlorophyll content, photosynthetic rate, and electrolyte leakage were measured to evaluate tolerance to water stress in wild-type (WT) and transgenic tobacco plants (TR) expressing copper/zink superoxide dismutase (CuZnSOD) and ascorbate peroxidase (APX) in chloroplasts. Leaf water potential of both WT and TR plants decreased similarly under water stress condition. However, leaf osmotic potential of TR plants more negatively decreased in the process of dehydration, compared with WT plants, suggesting osmotic adjustment. Stomatal conductance (Gs) in WT plants markedly decreased from the Day 4 after withholding water, while that in TR plants retained relatively high values. Relatively low chlorophyll content and photosynthetic rate under water stress were shown in WT plants since $4^{th}$ day after treatment. In particular, damage indicated by electrolyte leakage during water stress was higher in WT plants than in TR plants. On the other hand, SOD and APX activity was remarkably higher in TR plants. These results indicate that transgenic tobacco plants expressing copper/zink superoxide dismutase (CuZnSOD) and ascorbate peroxidase (APX) in chloroplasts improve tolerance to water stress.
