Background: Tobacco contains agents which generate various potent DNA adducts that can cause gene mutations. Production of DNA adducts may be neutralized by glutathione S transferase (GST) along with other phase I and phase II enzyme systems. The existence of null type of GST among the population increases the susceptibility to various disorders and diseases. The present study focuses on the impact of high tobacco usage and possible null type mutation in GST loci. Methods: Genotypes of GST were detected by multiplex polymerase chain reaction in unrelated 504 volunteers of high tobacco using natives of Gujarat. Allelic frequencies were calculated using Statistical Package for Social Studies-16 software. Hardy Weinberg Equilibrium (HWE) was calculated using Chi square test. Two sided Fisher's significance test was used to compare allelic frequencies of different populations. Results: The frequency of homozygous null genotype of GSTM1 and GSTT1 were 20% (95% CI 16.7-23.9) and 35.5% (95% CI 31.4-39.9) respectively. The GSTM1 and GSTT1 null allele frequency distribution in the Gujarat population was significantly deviating from HWE. GSTT1 null frequency of Gujaratians was significantly higher and different to all reported low tobacco using Indian ethnics, while GSTM1 was not differing significantly. Conclusion: Tobacco usage significantly influences the rate of mutation and frequency of GSTT1 and M1 null types among the habituates. The rate of mutation in GSTT1 loci was an undeviating response to the dose of tobacco usage among the population. This mutational impact of tobacco on GSTT1 postulates the possible gene - environment interaction and selection of null genotype among the subjects to prone them under susceptible status for various cancers and even worst to cure the population with GSTT1 dependent drugs.
Tobacco wild fire caused by Pseudomonas tabaci(Wolf et Foster) Stevens is one of the most important diseases on tobacco plant in Korea. The objectives of the study were to investigate: 1) the relationships between environmental conditions and the disease development, 2) the effect of chemical spray and soil sterilization on the disease development, and 3) the proper date for chemical sprays during the growing season. The appearance of wild fire on tobacco plant was somewhat proportional to temperature and moisture in atmosphere. The first occurrence of the disease was in late part of June when the temperature was $18\;to\;20^{\circ}C$. with $65\;to\;70^{\circ}C$ of relative humidity. The disease was, then, gradually increased during early part of July with the temperature of $22\;to\;26^{\circ}C$. and $85\;to\;86^{\circ}C$ of. relative humidity. The rapid development of the disease occurred after heavy rain or storm during July. Two to three times of Phytomycin sprays(1:300-350) during the epidemic season reduced $28.3\%$ of disease damage when compared with control plot. Soil sterilization with chloropicrin. 10 liter/10a, Prior to transplanting of seedlings reduced not only wild fro damage but also the other tobacco diseases on field. The study showed that the damage caused by tobacco wild fro could be reduced with the use of early varieties and/or 2 or 3 times of Phytomycin sprays during early part of June, and soil sterilization with chloropicrin prior to transplanting.
Major diseases of tobacco and farmer's control practices were surveyed in the Kyeongbuk province area from 2005 to 2006. Mosaic caused by potato virus Y and bacterial wilt caused by Ralstonia solanacearum were most severe during the harvest season. Compared with the disease occurrence of ten years ago, the damage by tobacco mosaic virus reduced but bacterial wilt increased. These changes in the disease occurrences may probably be due to releasing the resistant tobacco cultivar to the mosaic virus but susceptible to the bacterial wilt pathogen. More than thirty percentage of the farmers have misused fungicides and also have applied the continuous mono-cropping system for more than ten years, and have chosen the incorrect crops for the rotation.
Bacterial wilt(Ralstonia solanacearum) is one of the major diseases of flue-cured tobacco (Nicotiana tabacum L.) in the world. This study was conducted to investigate degree of dominance, selection, and correlation between leaf shape and degree of bacterial wilt resistance in flue-cured tobacco. The degree of disease caused by bacterial wilt was evaluated in parents, F$_1$, F$_2$ and F$_3$ populations of two crosses, BY 4 x NC 95 and BY 4 x Coker 86, in the infected field. The leaf shape index was also measured in parents and F$_2$ population of BY 4 x NC 95. The incidence of bacterial wilt was observed in the middle of June and peaked in late July, when the highest value of pathogen density reached 1.0 x 10$^{6}$ colony forming unit per gram. It was concluded that the inheritance mode of risestance to bacterial wilt in the above two crosses of susceptible and resistant varieties was recessive and polygenic. The resistance to bacterial wilt was significantly correlated with leaf shape in F2 generation of BY 4 x NC 95. But certain plants having narrower leaves were also resistant to bacterial wilt. It is considered that the bacterial wilt resistant lines having narrower leaves could be selected. The selection for bacterial wilt resistance in the F$_2$ population might be effective.
The vein necrotic strain of Potato Virus Y (PVY - VN) and black shank (Phyto-phthora parasitica var. nicotianae) are the two major diseases causing severe damages especially in burley tobacco (N. tabacum L.) area in Korea. A new tobacco variety, KB 111, resistant to PVY and black shank disease, was developed by Korea Ginseng & Tobacco Research Institute in 1997. It is a male sterile(MS) F$_1$ hybrid of the cross between MS TC 613 and KB 108. KB 111 was compared to Burley 21 on the agronomic characteristics and disease resistances in performance tests: It possessed upright growth habit and flowered two days later than Burley 21. It was resistant to both PVY and black shank and yielded about 3% more cured leaf than Burley 21, but other characteristics are very simiar to those of Burley 21. The chemical composition and physical properties of the cured leaf of KB 111 were as much acceptable as those of Burley 21 while it produced average yield of good quality leaf and appeared to resistant to PVY and black shank disease on regional farm test in 1998.

Cigarette smoking is the potential risk factor for lung cancer and chronic pulmonary disease, as well as inflammatory bowel disease and reproductive malfunction. Nicotine and tar have been im plicated as a major factor in the pathogenesis of the diseases. Nicotine increases heart rate and blood pressure due to stimulation sympathetic neurotransmission and tar also accounts for the severe damage of peridontal diseases and osteoporosis in postmenopausal women. Portulaca oleracea, which contains significant amount of K+, noradrenaline and dopamine as well as various nutrients, has been used for many medicinal purposes and one of which is the detoxification of insect or snake toxins. The purpose of this study was to investigate the action of Portulaca oleracea extracts on the reduction of harmful materials of tabacco. The reduction percentages were measured in the presence and absence of each solvent extract of Portulaca oleracea using reversed C18 column of HPLC. Nicotine reduction effects were obtained from aqueous, methanol and chloroform extracts of Portulaca oleracea as 89%, 55% and 51%, respectively. The results suggest that the polar extracts of Portulaca oleracea affects the reduction of nicotine which is responsible for many diseases.
From the early 1,100 to 1,900, ginseng cultivation seemed to be practiced with management of the diseases which were, in general, referred to a "disaster" at that time. Farmers had individually developed their own methods to manage the disaster with a try and error from generations to generations. It was not determined until 1909, however, that plant pathogens as a new concept was involved in the disaster and thirteen ginseng diseases were reported in Korea by Japanese plant pathologists. The intensive researches have been carried out from 1978 when Korea Ginseng and Tobacco Research Institute was established. Among the ginseng diseases reported in ginseng growing countries, Altemaria Panax, Eotrytis sp., Cylindrocarpon destmctans, Colletotrichum gloeosporioides, Erwinia carotovora subsp. carotovora, Pythium app. Phytophthora cactomm, Sclerotinia sp., Sclerotium rolfsii, and root rot caused by nitylenchus destructor have been observed in Korea and the appropriate control methods for the major diseases were developed. However, the other nine diseases reported by former researchers have not been confirmed for their pathogenicities and/or occurrences on ginseng yetinseng yet.
Background: The scientific evidence relating to the burden of oral diseases attributable to tobacco use has been reviewed and the need for a well-structured dental teaching program concentrating on oral cancer education and tobacco cessation interventions has been emphasized. The aim of our study was to evaluate the awareness of oral cancer and perception of tobacco use cessation counseling among dental students at all study levels in India, Saudi Arabia, the United Arab Emirates, and Yemen. Materials and Methods: A structured, pre-tested, self-administered 15-item questionnaire was used to conduct a cross-sectional survey. Data analyses including percentages, frequency distributions and tests of chi-square were generated. Results: A total of 621 (97.6%) Indian, 493 (96.5%) Saudi, 194 (96.5%) Yemeni and 187 (98.4%) United Arab Emirates respondents recognized the association between oral cancer and cigarette smoking. Although more than 96% of the students surveyed recognized the association between oral cancer and cigarette smoking and about 55% reported cigarette smoking as one of the etiological factors of oral cancer, more than 66% of students who reported cigarette smoking as an etiological factor of oral cancer disagreed/strongly disagreed with all the statements concerning tobacco use cessation. Conclusions: A higher level of oral cancer awareness did not have a positive impact on the perception of tobacco use cessation counseling among the sample surveyed.
Chronic alcohol and tobacco abuse plays a crucial role in the development of different liver associated disorders. Intake promotes the generation of reactive oxygen species within hepatic cells exposing their DNA to continuous oxidative stress which finally leads to DNA damage. However in response to such damage an entangled protective repair machinery comprising different repair proteins like ATM, ATR, H2AX, MRN complex becomes activated. Under abnormal conditions the excessive reactive oxygen species generation results in genetic predisposition of various genes (as ADH, ALDH, CYP2E1, GSTT1, GSTP1 and GSTM1) involved in xenobiotic metabolic pathways, associated with susceptibility to different liver related diseases such as fibrosis, cirrhosis and hepatocellular carcinoma. There is increasing evidence that the inflammatory process is inherently associated with many different cancer types, including hepatocellular carcinomas. The generated reactive oxygen species can also activate or repress epigenetic elements such as chromatin remodeling, non-coding RNAs (micro-RNAs), DNA (de) methylation and histone modification that affect gene expression, hence leading to various disorders. The present review provides comprehensive knowledge of different molecular mechanisms involved in gene polymorphism and their possible association with alcohol and tobacco consumption. The article also showcases the necessity of identifying novel diagnostic biomarkers for early cancer risk assessment among alcohol and tobacco users.
Isaria javanica pf185 is an important entomopathogenic fungus with potential for use as an agricultural biocontrol agent. However, the effect of I. javanica pf185 on plant growth is unknown. Enhanced tobacco growth was observed when tobacco roots were exposed to spores, cultures, and fungal cell-free culture supernatants of this fungus. Tobacco seedlings were also exposed to the volatiles of I. javanica pf185 in vitro using I-plates in which the plant and fungus were growing in separate compartments connected only by air space. The length and weight of seedlings, content of leaf chlorophyll, and number of root branches were significantly increased by the fungal volatiles. Heptane, 3-hexanone, 2,4-dimethylhexane, and 2-nonanone were detected, by solid-phase micro-extraction and gas chromatography-mass spectrophotometry, as the key volatile compounds produced by I. javanica pf185. These findings illustrate that I. javanica pf185 can be used to promote plant growth, and also as a biocontrol agent of insect and plant diseases. Further studies are necessary to elucidate the mechanisms by which I. javanica pf185 promotes plant growth.
