닭 전염성 기관지염에 관한 연구: II. 간접 enzyme-linked immunosorbent assay(ELISA)에 의한 항체가 측정 (Studies on avian infectious bronchitis: II. Standardization of an indirect enzyme-linked immunosorbent assay (ELISA) for antibody measurement)

  • Critical parameters affecting sensitivity and specificity of an enzyme-linked immunosorbent assay(ELISA) for detection of antibodies to avian infections bronchitis virus(IBV) were standardized. By adopting the optimized conditions an equation calculating ELISA antibody titers from the observations at single serum dilution was formulated. The purified antigen of IBV Mass-41 strain was dispensed into polystyrene microplate wells at a concentration of 300ng per well($100{\mu}l$) and the plates were coated by completey drying at $37^{\circ}C$. Diluted chicken serum and horseradish peroxidase conjugated goat anti-chicken IgG were added in order in $100{\mu}l$ volumes per well and allowed to react for 30 minutes each at room temperature. Just before use and after each reaction the plates were washed three times with distilled water. Finally o-phenylenediamine solution was added as an enzyme substrate. After incubation for another 15 minutes at room temperature absorbances were read at 492nm. Hyperimmune serum against Mass-41 strain was used as internal reference positive(IRP) serum. After repeated titration of IRP and negative serum, a constant titer of IRP was determined. Serum titrations were carried out for various sample sera together with IRP and negative sera and the observed titers of sample sera were corrected by reflecting the ratio between observed and constant titers of IRP serum. These corrected titers of the sample sera were plotted against sample/positive(S/P) OD ratios. All the OD's measured in the serum titrations were also corrected by substracting negative serum OD. The following equation was formulated from the above data; $Log_{10}$ ELISA titer=$5.568({\log}_{10}S/P)+4.161$ Thus it was possible to calculate ELISA titer by measuring absorbance at 1/400 single serum dilution. Titer measured by cross ELISA tests employing Mass-41 strain and three local IBV isolates were similar. These results suggest that the ELISA tests standardized in this study can be used for evaluating not only vaccinal immunity but also for infection status against fields IBV's.

분심기음의 항암작용 및 면역기능에 관한 연구 (Experimental Studies on Antitumor Effect and Immune Responses of Bunsimgieum)

  • This study was performed to investigate the effect of Bunsimgieum on antitumor effect after sarcoma-180 cells transplantation into peritoneal cavity or left groin and immune responses on the depressed immunity induced by methotrexate in mice. The Bunsimgieum extract of 10mg/kg was orally administered 14 days for antitumor effects and 21 days for immune responses. 50% inhibitory concentration($IC_{50}$) of SUN-1, SUN-C4, and SUN-396 cancer cell, mean sunvival days and body weight of tumor bearing mice, and growth of tumor mass for antitumor effect; delayed type hypersentivity, hemagglutinin titer, hemolysis titer, rosette forming cells, natured killer cell activity, lymphocyte transformation, productivity of interleukin-2, and phagocytic activity for their immune responses were measured in ICR mice. Significance in antitumor effect is noted in the enlongation of mean life days and inhibition of tumor growth(p<0.01, respectively). Significance of immune responses is also noted in hemolysis titer, lymphocyte transfumotion, IL-2 productivity, phagocytic activity, and natural killer cell activity at E/T ratio 100:1(p<0.01, respectively). Significant in rosette cell formation was seen at dosage of 20mg/kg(p<0.01). However, Difference of body weight as antitumor effect, delayed type hypersensitivity, and hemagglutinin titer were not shown significantly. According to the above results, it could be suggested that Bunsimgieum has prominent antitumor and immunity enhancing effect.

돼지에 있어서 Sareocustis와 Toxoplusma 감량의 혈청학적 교차반응 시험 (Serological Cross-Reactivity between Sarcocystis and Toxoplasma in Pigs)

  • 돼지에 $1.5{\times}10^6$개의 Sarcocystis suicanis sporocyst와 10,000개의 Toxcplasma gondii oocyst(미국 Georgia 대학교 수의과대학 보존주)를 각각 경구감염 시킨후 9주간에 걸쳐 혈중 IgG 항체의 소장과 Sarcocystis와 Tohoplasma 사이의 혈청학적 교차반응성을 IFA 검사와 ELISA법을 이용하여 검사하였다. IFA검사와 ELISA를 위한 Sarcocystis의 전충체 항원과 수용성 항원은 감염돈의 확육을 인공소화액으로 소화시켜 준비하였다. Toxoplasma의 전충체 항원과 수용성 항원은 감염 mouse 복강에서 충체를 분리하여 사용하였다. Sarcocystis와 Togoplasma를 각각 인공감염시킨 돈혈청의 IgG 항체는 IFA검사와 ELISA에서 모두 감염 2주째에 처음으로 검출되었으며 Tcxoplasma에 대한 항체가는 감염 6주째에 최고치에 도달하였다가 그 이후에는 하강하였다. Sarcocystis에 대한 항체가는 감염 9주까지 서서히 상승하였다. Toxoplasma에 감염된 돈혈청은 Sarcocystis 항원에 대하여 IFA검사에서는 1 : 16까지 교차반응이 나타났으며 ELISA에서는 1 : 32까지 나타났다. 대조군의 돈혈청은 모두 1 : 4 이하이었다.

Tissue Transglutaminase Antibody and Its Association with Duodenal Biopsy in Diagnosis of Pediatric Celiac Disease

  • Purpose: This study aimed to evaluate a possible association between the anti-tissue transglutaminase antibody (anti-tTG) titer and stage of duodenal mucosal damage and assess a possible cut-off value of anti-tTG at which celiac disease (CD) may be diagnosed in children in conjunction with clinical judgment. Methods: This observational study was conducted at a gastroenterology clinic in a tertiary hospital from April 2012 to May 2013. Seventy children between 6-months and 18-years-old with suspected CD underwent celiac serology and duodenal biopsy. Statistical analyses were done using SPSS 16. Diagnostic test values were determined for comparing the anti-tTG titer with duodenal biopsy. An analysis of variance and Tukey-Kramer tests were performed for comparing the means between groups. A receiver operating characteristics curve was plotted to determine various cut-off values of anti-tTG. Results: The mean antibody titer increased with severity of Marsh staging (p<0.001). An immunoglobulin (Ig) A-tTG value at 115 AU/mL had 76% sensitivity and 100% specificity with a 100% positive predictive value (PPV) and 17% negative predictive value (NPV) for diagnosis of CD (p<0.001, 95% confidence interval [CI], 0.75-1). Conclusion: There is an association between the anti-tTG titer and stage of duodenal mucosal injury in children with CD. An anti-tTG value of 115 AU/mL (6.4 times the upper normal limit) had 76% sensitivity, 100% specificity, with a 100% PPV, and 17% NPV for diagnosing CD (95% CI, 0.75-1). This cut-off may be used in combination with clinical judgment to diagnose CD.

폐쇄성 무호흡의 치료시 지속적 기도 양압치의 예측 (Prediction of Continuous Positive Airway Pressure Level for Treatment of Obstructive Sleep Apnea)

  • 연구배경: 지속적 기도 양압 치료는 폐쇄성 수면 무호흡증 환자의 가장 효과적인 내파적 치료법으로 알려져 있다. 폐쇄성 무호흡증 환자에 사용되는 지속적 기도 양암 치료는 양압이 상기도에서 단순히 물리적인 공기 부목과 같은 역할로 작용한다. 폐쇄성 무호흡 환자에서 지속적 기도 양암 치료시 최적 양압의 결정에 영향을 미치는 변수를 파악하고 이러한 변수들로 다중 선형 회귀 분석을 하여 최적압치를 예측해 보기위하여 이 연구를 하였다. 방법: 폐쇄성 수면 무호흡증이 있는 72명의 환자를 대상으로 이틀동안 진단 및 치료적 수면 다원 검사를 실시하였다. 이들 중 지속적 기도 양압 최적지가 $5cmH_2O$ 이하였던 35명과 최적치가 $10cmH_2O$이상이었던 37명의 환자에서 나이, 키, 몸무게, body mass index(BMI)와 수면 다원 검사의 각 변수의 차이를 분석하고 이러한 변수들로 지속적 기도 양압의 최적치를 다중선형 회귀분석하였다. 결과: 지속적 기도 양압의 최적치가 $10cmH_2O$이상이었던 환자에서 body mass index(BMI)가 의의있게 더 높았고 무호흡 지수와 무호흡 저호흡 지수도 의의있게 더 높았으며 최저 산소 포화도는 의의있게 낮았다. 이러한 변수들로 구한 최소의 지속적 기도 양압의 공식은 CPAP titer = 8.382 + 0.064 x BMI + 0.077 x AI - 0.004 x AHI - 0.077 x lowest $SaO_2$였다. 수면 다원 검사로 실제로 측정한 지속적 기도 양압의 최적압은 평균 $7.93{\pm}4.00cmH_2O$였으며 다중 선형 회귀분석하여 공식으로 구한 예측 최적압치는 $7.80{\pm}2.96cmH_2O$로 이들 두 수치사이의 의의있는 차이는 없었다. 결론: 폐쇄성 수면 무호흡의 치료시의 지속적 기도 양압의 최적압은 신체의 비만도, 무호흡의 심한 정도와 최저 산소 포화도와 관계가 있었으며 이러한 변수들로 비교적 정확하게 최적압치를 예측할 수 있었다.

강원 영동지역 우 바이러스성 설사병의 혈청학적 조사 (Serological Survey of Cattle on Bovine Viral Diarrhea in Young Dong Province)

  • To investigate epidemological sitution of bovine viral diarrhea infection, serological survey in cattle being raised in Young Dong province were conducted. Bovine sera collected ramdomly from August 1990 to December 1990 were tested for bovine viral diarrhea virus serum neutralizing antibody titers. The results were as follows 1. BVDV SN antibody levels were considerably varies and positive rate was 58(108 heads out of 186) 2. BVDV SN antibodies to breeds of cattle was various and positive rates showed that diary cattle, beef, native cattle(Korean) were 67.52%, 59.38%, 27.00% respectively followed in that order. 3. In the regional prevalence of BVD SN antibodies in cattle, Alpine(92%) was the highest, Young Dong south(59%) middle(44%), and North 30% followed in that order 4. In the age relatated prevalence of BVD SN antibodies, the younger than 6 month old group was the highest 65.7%, and older than 25 month old group was also at 62.2%. Then, 7 to 12 moth old group and 13 to 24 month old group showed to 58.5%, 52.1% respectively. 5. The geometric mean titer (log2) of 108 cattle serum samples showing positive BVD SN antibodies was 4.3. 6. In the geometric mean titer(log2) according to age, younger than 6 month old group (5.2) was the highest, then 7 to 12 month old group 2.8(SD=1.94 standard deviation) was lowliest.

Seroprevalence of Toxoplasma gondii and Bartonella henselase infection in stray cats of the Daejeon City, Korea

  • In this study, the seroprevalence of Toxoplasma (T.) gondii and Bartonella (B.) henselae infection among stray cats in Daejeon City, Korea was surveyed. A total of seven samples were positive (7/118, 5.93%) for T. gondii including three samples from female cats (3/58, 5.2%) and four samples from male cats (4/60, 6.7%). There was no significant difference between the genders. A total 22 samples (22/118, 18.6%) were positive for B. henselae; nine were from females and 13 were from males. There was no significant difference between genders. Nineteen samples had a titer of 1 : 50, two samples had a titer of 1 : 100, and one sample had a titer of 1 : 200. The present study is the first to use serological tests to analyze B. henselae prevalence among stray cats in Korea.

  • 페디스토마 성충의 단백질을 정제하여 이를 항원으로 면역시킨 mouse의 spleen cell과 myeloma cell을 세포융합 시킨 결과 융합율은 56% 이었고. 3차 cloning한 후의 항체생산율은 89% 이였으며, 이들 융합세포로 부터 IgM을 생산하는 hybridoma clone과 IgG을 생산하는 hybridoma clone을 얻었다. IgM을 생산하는 hybridoma clone의 supernate는 antibody titer가 희석농도 1 : 64까지 양성이었고. IgG을 생산하는 hybridoma clone의 supernate는 1 : 256까지 양성이었다.

  • The humoral immune response of Eicosapentanoic acid(EPA) was investigated in mice. ICR male mice were divided into 8 groups and received intraperitoneal injection of EPA(5 mg, 10 mg, 20 mg/kg) for 4 weeks. Cyclophosphamide(5 mg/kg) was administered i.p. 2 days prior to secondary immunization. Humoral immune response was evaluated by antibody titer, hypersensitivity to SRBC (Arthus), plaque forming cell(PFC) and organ weight. The ontanined results were as followings: The increased rate of body weight, the ratio of liver weight, spleen weight to body weight were decreased by all EPA administration groups as compared to normal group. HA titer, HY titer and Arthus reaction were enhanced according to the increase of EPA doses as compared to normal group. PFC was significantly enhanced by EPA 10 mg administration group. These results suggest that EPA enhances humoral immune response to SRBC in mice, indicating that EPA may block a immunoglobulin synthesis inhibition of arachidonic acid.

귀비온담탕(歸脾溫膽湯)이 흰쥐의 항(抗)스트레스와 면역반응(免疫反應)에 미치는 영향(影響) (The Effect of Guibiondamtang on Immune Response and in Concertration of Catecholamine in Immobilization Stressed Rates)

  • In order to investigate the Anti-stress effect of Guibiondamtang in the immobilization stressed rats, the level of serum catecholamine, the change of body weight, the humoral and cellular immune response were studied. The results were as follows; 1. The decrese of the body weight was significantly inhibited in test group for Guibiondamtang, comparting to the control group. 2. The increase of the level of serum norepinephrine was significantly inhibited in test group, comparing to the control group. 3. The increase of the level of serum epinephrine was significantly inhibited in test group, comparing to the control group. 4. In the hemagglutinaton titer, the control group was decreased on the serum antibody titer but test group was inhibitory effect on the decrease of sereum antibody titer. 5. In the plaque formation test, the control and test group were not shown significant differences. 6. In the foot pad swelling respopnse, the control group was decreased on DTH response but test group was increased comparing to the normal group. 7. There was no change on the distribution of lymphocyte subset(CD4, CD8), grnulocyte and macrophage analyzed by flow cytometry.
