압출성형 산삼배양근의 발효 특성 (Fermentation Characteristics of Extruded Tissue Cultured Mountain Ginseng)

  • 본 연구에서는 산삼배양근과 압출성형 산산배양근의 발효특성을 알아보기 위해 발효액의 pH 및 산도, 당도, 총당, 환원당, 알코올 함량의 변화를 홍삼발효액과 비교하였다. Saccharomyces cerevisiae와 Aspergillus usamii, Rhizopus japonicus를 함유한 누룩을 접종하여 15일간 발효시킨 알코올 발효액의 pH, 당도, 환원당, 총당은 발효 시간 5일 전에 급격히 감소하여 이후 완만하게 감소하여 안정된 상태를 보여주었다. 또한, 산삼배양근을 압출성형 시킴으로써 환원당, 총당, 알코올 함량이 2배 이상 증가함을 확인할 수 있었다. 발효액의 최종 산도는 홍삼 $1.12{\pm}0.06%$, 산삼배양근 $1.19{\pm}0.06%$, 압출성형 산삼배양근 $0.97{\pm}0.01{\sim}1.02{\pm}0.01%$로 측정되었다. 알코올 발효를 시킨 발효액의 최대 알코올 함량 은 홍삼 $3.82{\pm}0.10%$ 산삼배양근 $0.91{\pm}0.06%$, 압출성형 산삼배양근 $1.86{\pm}0.10{\sim}2.18{\pm}0.06%$로 측정되어 산삼배양근을 압출성형 시킴으로써 최대 2.5배 알코올 함량을 향상시킬 수 있었다. 압출성형물 중에서도 배럴온도 $140^{\circ}C$, 수분함량 25%로 압출성형 시킨 산삼배양근이 환원당, 총당, 알코올함량이 가장 높게 측정되었다. 결론적으로 본 연구에서 압출성형공정을 통하여 산삼배양근 발효적성의 향상과 함께 새로운 산삼발효제품과 식품개발의 가능성을 발견하였다.

Relationship between Water Content and Osmotic Potential of Lentinula edodes

  • This study was conducted to understand how osmotic potentials in Lentinula edodes tissues are related to water contents and how they change while a mushroom matures. Water content and osmotic potential of L. edodes mushroom tissues from log cultivation and sawdust cultivation were measured and the relationships were analyzed. Osmotic potentials in the tissues were exponentially proportional to their moisture contents and there were strain differences in the potentials. Strain 290 has lower osmotic potential than strain 302, in the tissues at the same water content. As the mushrooms mature, tissue water content maintained ca 94% in head tissues and ca 90% in gills, but significantly decreased from ca 90% to 82% in the stipe tissues. Osmotic potential changes were similar to the tissue water content changes as the mushrooms mature. While osmotic potentials maintained -0.25 to -0.45 MPa in head and gill tissues, the potentials greatly decreased from -0.65 to -1.33 MPa in stipe tissues. Our results show that osmotic potentials in L. edodes tissues are exponentially proportional to tissue water contents, that strains differ in osmotic potential related to water, and that stipe tissues can still have nutritional value when they mature.

노령 흰쥐의 대뇌 조직에서 알루미늄 투여에 대한 녹차 추출물이 인지질 함량에 미치는 효과 (Effects of Green Tea Extract Diet on the Phospholipid Content of Aluminum-Induced-Damaged Cerebral Tissue of Old Rats)

  • This study was designed to investigate the effects of green tea extract on aluminum-induced damage to phospholipid content in old rat cerebral tissue. The aim of this study was to investigate the possibility that aluminum is the cause of Alzheimer's disease. Forty Sprague-Dawley old male rats weighing 350$\pm$10 g were divided into four groups, consisting of a control group (CON), 40 ppm aluminum sulfate group (Al-40), green tea water extract group (GTWE), and 40 ppm aluminum sulfate and green tea water extract groups (Al-40+GTWE) and kept on their respective diets for 12 weeks. In order to discover the influence of aluminum on cerebral tissue of old male rats, the serum aluminum concentration and phospholipid composition were compared between the aluminum-treated group and the normal group. The results showed that the serum aluminum concentration was higher in the aluminum sulfate-treated group than in the normal group. The cerebral tissue phospholipid concentration decreased significantly in the aluminum sulfate treated group as compared to the normal group. The results of this experiment show that increase of aluminum concentration in experimental animals causes the rise of serum aluminum and phospholipid concentrations, phenomena that are very similar to those shown in Alzheimer's disease., The results of this experiment, together with reports that aluminum is a cause of neurofibrillary tangles in cerebral tissue, therefore demonstrate the possibility that aluminum is the cause of Alzheimer's disease. Green tea water extract is also shown to be an effective therapeutic candidate for the treatment of Alzheimer's disease.

구기자혹응애(Eriophyes ruro KISHIDA) 및 그 혹(Gall)에 관한 연구 II. 혹의 성장에 따르는 효소활성 및 화학성분에 대하여 (Studies on Eriophyes kuko KISHIDA and its Galls -On the Activity of Enzymes and the Chemical Constituents in Growing Galls-)

  • 구기자혹응애(Eriophyes kuko KISHIDA)의 침입기에 의하여 구기자나무(Lycium chineens MILL.)외 잎형성된 혹(mite gall)의 생화학적 성질을 구명할 목으로 몇 가지 효소의 활성도, 화학성분의 함량 및 호량 등에 관하여 실험한 바 다음과 같은 결과를 얻었다. Catalase와 peroxidase 활성도는 건전한 잎의 직보다 혹 조직에서 높았으며 혹이 점차로 커짐에 따라 활성도도 높아졌다. 2. 조효소의 phosphorylase 활성도는 큰 혹(matured gall) 조직과 건전한 잎의 조직에 있어서 차이가 없었으나 활성 측정에 미치는 다른 효소의 영향을 고려하면 건전한 잎의 조직이 높은 것 같으며, phosphomonoesterase의 활성도는 건전한 잎의 조직보다 큰 혹 조직이 5배 가까이 높았으며 $\beta-amylase$와 invertase 등의 활성도는 큰 혹 조직이 2배 가까이 높았다. 3. 조단백질 함량은 혹의 발생 형성초기인 작은 혹일때는 $20\%$ 정도 건전한 잎의 조직보다 증가하였으나 혹이 점차 커 짐에 따라 감소하는 경향을 나타내었으며, 환원당은 자은 측일 에는 약간 감소하는 경향이었으나 혹이 점차 커짐에 따라 증가하였다. 인과 tannin의 함량도 혹이 점차커짐에 따라 증가하여를 혹이 건전한 잎보다 2배정도 많았다. 4, 호흡에 있어서 $QO_2$는 혹 조직이 건전한 잎보다 높았으나, $QCO_2$는 반대로 건전한 잎이 높았으며, RQ는 혹 조직이 낮았다.

고지방 식이를 섭취시킨 흰쥐에서 고추장의 항비만효과 (Anti-obesity Effects of Kochujang in Rats Fed on a High-fat Diet)

  • The purpose of the present study was to investigate effects of kochujang and red pepper on energy intake, body fat content and energy expenditure in rats fed on high-fat(30%) diet. Kochujang and red pepper power were added in the high-fat diet, adjusting the level to 95 and 22g/kg diet, respectively, The level of red pepper addition was corresponding was corresponding to the level of the content of red pepper powder in the kichujang-added diet. Kochujang induced a 30% reduction in body fat gain which was associated with a significant increase in energy expenditure. However, red pepper reduced body fat gain by only 15%. Furthermore, energy expenditure was not affected by red pepper. Metabolizable energy intake, apparent digestibility and body protein gain were not affected by either kochujang or red pepper. It has been known that capsaicin, a pungent component of red pepper, enhances activity of brown adipose tissue through increasing protein content. In the present study, in addition of protein content, DNA content of interscapular brown adipose tissue was also increased by kochujang. Therefore, it appeared that the anti-obesity effects of kochujang was greater than those of red pepper, indicating more than red pepper was involved in the expression of the anti-obesity effects of kochujang(Korean J Nutrition 33(8) : 787-793, 2000)

크리핑 벤트그라스 훼어웨이에서 관수회수.예지물과 질소시비수준이 엽조직 및 토양 질소함유량에 미치는 효과 (A Three-year Study on the Leaf and Soil Nitrogen Contents Influenced by Irrigation Frequency, Clipping Return or Removal and Nitrogen Rate in a Creeping Bentgrass Fairway)

  • Responses of 'Penncross' creeping bentgrass turf to various fairway cultural practices are not well-established or supported by research results. This study was initiated to evaluate the effects of irrigation frequency, clipping return or removal, and nitrogen rate on leaf and soil nitrogen con-tent in the 'Penncross' creeping bentgrass (Agrostis palustris Huds.) turf. A 'Penncross' creeping bentgrass turf was established in 1988 on a Sharpsburg silty-clay loam (Typic Argiudoll). The experiment was conducted from 1989 to 1991 under nontraffic conditions. A split-split-plot experimental design was used. Daily or biweekly irrigation, clipping return or removal, and 5, 15, or 25 g N $m-^2$ $yr-^1$ were the main-, sub-, and sub-sub-plot treatments, respectively. Treatments were replicated 3 times in a randomized complete block design. The turf was mowed 4 times weekly at a l3 mm height of cut. Leaf tissue nitrogen content was analyzed twice in 1989 and three times in both 1990 and 1991. Leaf samples were collected from turfgrass plants in the treatment plots, dried immediately at 70˚C for 48 hours, and evaluated for total-N content, using the Kjeldahl method. Concurrently, six soil cores (18mm diam. by 200 mm depth) were collected, air dried, and analyzed for total-N content. Nitrogen analysis on the soil and leaf samples were made in the Soil and Plant Analyical Laboratory, at the University of Nebraska, Lincoln, USA. Data were analyzed as a split-split-plot with analysis of variance (ANOVA), using the General Linear Model procedures of the Statistical Analysis System. The nitrogen content of the leaf tissue is variable in creeping bentgrass fairway turf with clip-ping recycles, nitrogen application rate and time after establishment. Leaf tissue nitrogen content increased with clipping return and nitrogen rate. Plots treated with clipping return had 8% and 5% more nitrogen content in the leaf tissue in 1989 and 1990, respectively, as compared to plots treated with clipping removal. Plots applied with high-N level (25g N $m-^2$ $yr-^1$)had 10%, 17%, and 13% more nitrogen content in leaf tissue in 1989, 1990, and 1991, respectively, when compared with plots applied with low-N level (5g N $m-^2$ $yr-^1$). Overall observations during the study indicated that leaf tissue nitrogen content increased at any nitrogen rate with time after establishment. At the low-N level treatment (5g N $m-^2$ $yr-^1$ ), plots sampled in 1991 had 15% more leaf nitrogen content, as compared to plots sampled in 1989. Similar responses were also found from the high-N level treatment (25g N $m-^2$ $yr-^1$ ).Plots analyzed in 1991 were 18% higher than that of plots analyzed in 1989. No significant treatment effects were observed for soil nitrogen content over the first 3 years after establishment. Strategic management application is necessary for the golf course turf, depending on whether clippings return or not. Different approaches should be addressed to turf fertilization program from a standpoint of clipping recycles. It is recommended that regular analysis of the soil and leaf tissue of golf course turf must be made and fertilization program should be developed through the interpretation of its analytic data result. In golf courses where clippings are recycled, the fertilization program need to be adjusted, being 20% to 30% less nitrogen input over the clipping-removed areas. 지황 조직배양주의 수량성과 성분함량 특성 (Characteristics in Tissue Cultured Plants of Chinese foxglove(Rehmannia glutinosa))

  • 지황 (Rehmannia glutinosa) 우량종묘 생산을 위하여 조직배양에 의해 생산된 종묘의 포장수량성을 평가하고 지표성분인 Catalpol과 유리당 함량 특성을 비교한 결과는 다음과 같다. 조직배양주의 종근저장성은 재래종에 비하여 약간 양호한 경향이었다. 또한 근장, 근경 등의 뿌리생육이 왕성하여 조직배양주의 수량성은 서천재래종 384kg/10 a에 비하여 1년차 묘는 112%, 2년차 묘는 246%까지 증수되는 결과를 보였다. 성분함량 특성비교에서는 총당함량이 서천재래 1.86%에 비하여 조직배양 1, 2년차 묘는 각각 1.21, 1.10%를 보였고, 지표성분인 Catalpol도 서천재래 0.46%에 비하여 조직배양 1, 2년차묘는 0.4%로 약간 감소되는 경향이었다. 이상의 결과에서 지황 조직배양주의 수량성은 재래종에 비하여 증수되는 특성을 보였다. 그러나 유리당과 Catalpol 함량은 약간 감소되는 경향이었는데 모본식물과 큰 차이를 보이지는 않았다.

젖소의 난소 황체에 있어서 중심강의 유무에 따른 Protein, DNA, RNA 함량의 비교 (Comparison of Protein DNA, and RNA Contents in Corpus Luteum without and with Central Cavity in Dairy Cow)

  • 젖소 난소 황체에 있어서 중심강의 유무에 따른 황체 조직 중의 total protein, DNA 및 RNA 함량을 조사하여 중심강이 있는 황체와 중심강이 없는 황체간의 기능성을 구명하고자 수행한 시험에서 다음과 같은 결과를 얻었다. 1. 중심강이 없는 황체의 total, supernatant 및 Pellt protein의 함량은 각각 32.83, 16.87 및 15.96 mg/g wet tissue이었고, 중심강이 있는 황체의 그것들은 각각 29.62, 16.10 및 13.52 mg/g wet tissue으로서 중심강이 없는 황체와 중심강이 있는 황체간에 유의적인 차이를 나타내지 않았다(p〉0.05). 2. DNA 함량은 중심강이 없는 황체가 1.99mg/g wet tissue이었고 중심 강이 있는 황체가 1.32mg/g wet tissue으로 중심강이 없는 황체와 있는 황체간에 유의적인 차이(p<0.05)를 나타내었다. Protein : DNA ratios에 있어서도 중심강이 없는 황체가 16.63mg/g wet tissue이었고 중심강이 있는 황체가 22.99mg/g wet tissue으로 중심강이 없는 황체와 중심강이 있는 황체간에 유의적인 차이(p<0.05)를 나타내었다. 3. 중심강이 없는 황체의 RNA함량, Protein: RNA 및 RNA:DNA ratios는 각각 2.87, 12.24및 1.43mg/g wet tissue이었고, 중심강이 있는 황체의 그것들은 각각 2.47, 13.73 및 1.89mg/g wet tissue으로서 중심강이 없는 황체와 중심강이 있는 황체간에 유의적인 차이를 나타내지 않았다(p〉0.05).

Automatic Electronic Cleansing in Computed Tomography Colonography Images using Domain Knowledge

  • Electronic cleansing is an image post processing technique in which the tagged colonic content is subtracted from colon using CTC images. There are post processing artefacts, like: 1) soft tissue degradation; 2) incomplete cleansing; 3) misclassification of polyp due to pseudo enhanced voxels; and 4) pseudo soft tissue structures. The objective of the study was to subtract the tagged colonic content without losing the soft tissue structures. This paper proposes a novel adaptive method to solve the first three problems using a multi-step algorithm. It uses a new edge model-based method which involves colon segmentation, priori information of Hounsfield units (HU) of different colonic contents at specific tube voltages, subtracting the tagging materials, restoring the soft tissue structures based on selective HU, removing boundary between air-contrast, and applying a filter to clean minute particles due to improperly tagged endoluminal fluids which appear as noise. The main finding of the study was submerged soft tissue structures were absolutely preserved and the pseudo enhanced intensities were corrected without any artifact. The method was implemented with multithreading for parallel processing in a high performance computer. The technique was applied on a fecal tagged dataset (30 patients) where the tagging agent was not completely removed from colon. The results were then qualitatively validated by radiologists for any image processing artifacts.

고추냉이 수경재배시 배양액의 EC 수준이 Allyl-isothiocyanate 함량과 생리적 반응에 미치는 영향 (Allyl-isothiocyanate Content and Physiological Responses of Wasabia japonica Matusum as Affected by Different EC Levels in Hydroponics)

  • 본 연구는 고추냉이 '달마' 품종을 배양액의 EC 수준을 달리하여 수경재배하였을 때, 기능성 성분인 AITC의 조직별 함량, 비타민 C 함량 및 생리적 반응들에 미치는 영향을 구명하고자 하였다. 배양액의 EC는 Yamasaki 배양액 EC $0.5dS{\cdot}m^{-1}$를 기준으로 1N-NaCl로 조절한 5수준(EC 0.5, 1, 2, 3, $5dS{\cdot}m^{-1}$)으로 5주간 담액 수경재배하였다. 고추냉이의 총 AITC 함량은 처리 1주에는 EC $5dS{\cdot}m^{-1}$에서, 처리 5주에는 EC $3dS{\cdot}m^{-1}$에서 가장 높았다. 처리 5주 후 고추냉이 AITC 함량은 처리 1주에 비하여 EC 0.5-$2dS{\cdot}m^{-1}$에서 1.2-1.4배 증가하였으나, EC $3dS{\cdot}m^{-1}$에서는 6%, EC $5dS{\cdot}m^{-1}$에서는 56% 감소하였다. AITC의 상대적 비율이 EC 0.5-$2dS{\cdot}m^{-1}$에서는 처리 1주와 처리 5주 모두 엽신과 뿌리에 비해 엽병(평균 53.4%, 49.5-61.1%)에서 높았다. 그러나 EC 3과 EC $5dS{\cdot}m^{-1}$에서 뿌리의 AITC 상대적 비율이 처리 1주에는 평균 45.1%(43.7-46.1%)로 엽신과 엽병에 비해 높은 반면 처리 5주에는 평균 16.6%(16.0-17.1%)로 낮아져, 엽신에서는 5배, 뿌리에서는 6.8배의 AITC 함량이 감소하였다. 처리 1주 비타민 C 함량은 83.7-$107mg{\cdot}100g^{-1}FW$로 처리에 따른 차이가 없었으나, 처리 5주 후, EC 0.5-$2dS{\cdot}m^{-1}$에서는 117.8-$137.5mg{\cdot}100g^{-1}FW$로 27% 증가하였다. 고추냉이 잎 전해질 유출은 EC $5dS{\cdot}m^{-1}$에서 EC 0.5-$2dS{\cdot}m^{-1}$에 비해 3배 증가하였다. 그러나 엽록소 함량, 광합성 및 증산율은 EC $2dS{\cdot}m^{-1}$ 이상에서, 수분함량과 비엽면적은 EC $5dS{\cdot}m^{-1}$에서 감소하였다. 처리 5주 후 고추냉이 생육은 EC 3과 $5dS{\cdot}m^{-1}$에서 엽수와 지상부 생체중이 감소하였다. 이상의 결과, 기능성 채소로서 고추냉이 수경재배시 품질과 생육을 유지하기 위한 적정 배양액의 EC는 $3dS{\cdot}m^{-1}$ 이하가 적합하리라 판단된다.
