Common Ragweed-Derived Phenolic Compounds and Their Effects on Germination and Seedling Growth of Weed Species (돼지풀의 페놀화합물 동정 및 이들 화합물이 잡초의 유식물 생장에 미치는 영향)

  • Phenolic compounds, which are products of secondary metabolism, have been demonstrated to be widespread growth substances in plants. The objectives of this study were to identify the phenolic compounds in common ragweed (Ambrosia artemisiifolia var. elatior) by HPLC and to evaluate their effects on germination and seedling growth of three weed species. Under controlled conditions in Petri dishes at $25^{\circ}C$, $10^{-3}$ and $10^{-4}$ M solutions of phenolic compounds were evaluated in seed germination tests. Four phenolic compounds (caffeic acid, O-coumaric acid, ${\rho}$-coumaric acid and ferulic acid) in common ragweed plant were identified and their concentration was increased from the stage before flowering through full flowering stage. Treatment of O- and ${\rho}$-coumaric acids delayed the seed germination of Digitalia ciliaris, while the treatment of caffeic acid delayed the seed germination of Echinochloa crus-galli. In time to 50% germination ($T_{50}$), phenolic compounds at $10^{-4}$ M promoted in Cyperus microiria and E. crus-galli but the level of $10^{-3}$ M delayed the $T_{50}$ of those weeds. The O-coumaric acid inhibited seed germination and seedling growth of the tested weeds and especially it perfectly inhibited the root growth of E. crus-galli.

Effect of Boric Acid on In Vitro Pollen Germination in Transgenic Plants Expressing Monoclonal Antibodies (단일항체를 발현하는 형질전환 식물체의 In Vitro 화분발아에 대한 Boric Acid의 영향)

  • Pollen germination viability is an essential factor to produce seeds from pollination and fertilization, which are required to maintain plant generation. In this study, we tried to identify the effect of boric acid on pollen germination and tube grouch in non-transgenic and transgenic plants expressing monoclonal antibodies (anti-colorectal cancer mAb CO17-1A, anti-breast cancer mAb BR55, and anti-rabies virus mAb57). The pollen of non-transgenic plant was treated with different concentration of boric acid (0, 5, 10, 15, 20, $40{\mu}g/mL$) in germination buffer to investigate its effect on in vitro pollen germination. At $20{\mu}g/mL$ of boric acid, tile pollen germination rate was the highest (49.5%) compared to other concentrations. In general, the germination rate significantly increased 3-10 folds in boric acid ($20{\mu}g/mL$) treated group in non-transgenic and transgenic plants. Also, the pollen tube length increased in boric acid ($20{\mu}g/mL$) treated groups. In the treated group, the pollen tube length increased until 3 h boric acid treatment and decreased after the 3 h, indicating that the 3 h is the most appropriate incubation time period. Western blot analysis showed that the mAb transgene expression was more stable in leaf than pollen in transgenic plants. This study suggested that $20{\mu}g/mL$ of boric acid is ideal concentration to induce in vitro pollen germination of transgenic plants expressing therapeutic monoclonal antibodies, indicating stable pollination and fertilization in transgenic plants.

A Basic Study on the Development of the Plant-Mat for Ecological Restoration (생태복원용 식물매트 개발에 관한 기초연구)

  • This study was conducted to develop the plant-mat for ecological restoration which was formed as the thin multi-layers of woven nets, soil media, seeds, etc. The experimental site was selected at a slope of 240$m^2$ at the rear of Design College of Sangmyung University and divided into 8 sub-experimental plots (20$m^2$ each) according to mat types. (forest-type and grass-type mats with and without seed attachment, respectively) Also, the existing vegetation plot and the no-treatment plot were included in this experiment for comparison. The experiment construction was conducted on September 3, 2002. Mat covered plots regardless of seed attachment showed nearly no signs of erosion even if rainy season in summer compared to the no-treatment plot. Vegetation monitoring result showed that germination rates were somewhat high in both forest and grass-type mats without seed attachment in the initial germination stage. Although germination rates of seed attached mats were somewhat slower than non-seed attached mats in the germination stage, but over time, it showed a good rooting and shooting environment for germinated tree and grass species because of soil media existence within the mat. This indicates that while non seed-attached mats require follow-up maintenance, seed-attached mats does not require maintenance after rooting and germination.

Comparison of Cardinal Temperatures of Lettuce Using Bilinear, Parabolic, and Beta Distribution Functions (선형, 쌍곡선과 Beta 함수를 이용한 상추의 주요 온도 비교)

  • The objective of this study was to estimate cardinal temperatures for germination of lettuce (Lactuca sativar L.) using bilinear, parabolic, and beta distribution functions. Seeds of lettuce were germinated in a growth chamber at 7 constant temperatures: 10, 14, 16, 20, 24, 28, and $32^{\circ}C$. Four replicates of 100 seeds were placed on two layers of filter paper in a 9 cm petri-dish. Radicle emergence of 1 mm was scored as germination. The time course of germination was modeled using a logistic function. These minimum, optimum, and maximum temperatures were estimated by regression of the inverse of time to 50% germination rate against the temperature gradient. In bilinear function, minimum, optimum, and maximum temperatures were $7.9^{\circ}C$, $23.3^{\circ}C$, and $28.0^{\circ}C$, respectively. In parabolic function, minimum, optimum, and maximum temperatures were $9.7^{\circ}C$, $19.5^{\circ}C$, and $29.4^{\circ}C$, respectively. In beta distribution function, minimum, optimum, and maximum temperatures were $3.7^{\circ}C$, $20.7^{\circ}C$ and $32.0^{\circ}C$, respectively. Minimum, optimum, and maximum ranges of temperatures were $3.7{\sim}9.7^{\circ}C$, $19.5{\sim}23.3^{\circ}C$, and $28.0{\sim}32.0^{\circ}C$, respectively.

Effect of Various Fungicides on Pollen Germination and Pollen Tube Growth in Some Fruit Trees (각종 살균제(殺菌劑)가 주요과수(主要果樹)의 화분발아(花粉發芽) 및 화분관신장(花粉管伸長)에 미치는 영향(影響))

  • The experiment was carried out in order to choose fungicides which are proper to be used in flowering time of fruit trees. The effect of fungicides on pollen germination and pollen tube growth was investigated in vitro. The results obtained are as follows : 1. Difolatan-Wp, Lime-sulfur, Captan-Wp and Bordeaux mixture greatly inhibited pollen germination and pollen tube growth. 2. The effect of Daconil-Wp and Zimaneb-Wp on pollen germination and pollen tube growth was not so great as that of Difolatan-Wp, Lime-sulfur, Captan-Wp or Bordeaux mixture. 3. Polyram-combi-Wp, Topsin-Wp and Benlate-Wp did not affect pollen germination but these fungicides slightly inhibited pollen tube growth. 4. Polyoxin-Wp significantly increased pollen tube growth but did not show promotive effect on pollen germinaiton.

Effects of Pre-sowing Treatments of Tobacco Seed on Germination and Seedling Uniformity (담배종자의 파종 전처리가 발아 및 묘의 균일성에 미치는 영향)

  • Pre-sowing treatments of tobacco seeds (cv. NC82) were evaluated as a means of improving the speed of emergence, early seedling growth and uniformity of the seedlings. Seeds were treated through the use of polyethylene glycol 6000(PEG), solid matrix priming(SMP), chilling and dark-preincubati(DPI). Under the laboratory condition of 15$^{\circ}C$, all the treated seeds germinated more rapid and the time to 50% germination(T50) reduced than control. Among the treatments, SMP was the best to reduce the T50 for germination. All the treatments did not affect the final germination percentage, while the chilling showed of little benefit for germination and T50. Under the greenhouse condition, seedling from SMP emerged earlier and more uniformity and also greater mean fresh and dry weight showed than other treatments. The advantage of T50 from SMP was affected more in temperature than in water content of Agro-Lig during the treatment.

  • These experiment were conducted to evaluate the environmentally acceptable polymers, and 10 polymers were used in these study, and to investigate conductivity, germination percentage, water uptake of polymeric coating soybean seed. The conductivity of polymeric coating seed is higher than that of none coating seed and the highest conductivity was obtained with waterlock coating seed among the 10 polymer coating seed. As the soaking time was long, the conductivity was increased. The conductivity of large seed was higher than that of small seed, and that of long period storage seed was higher than that of short period storage seed. The effects of seed coating polymers on uptake water were various, and daran 8600 inhibited uptake water of low quality seed. The waterlock, captan, klucel and sacrust was rised germination percentage, and daran 8600 was declined germination percentage, and the effect of coating polymers on germination percentage of low quality seed was higher than that of high quality seed.

Priming Effect of Rice Seeds on Seedling Establishment under Adverse Soil Conditions

  • An experiment was carried out to find out the priming effects of rice seeds, Oryza sativa L. (cv. Ilpumbyeo) on. the seedling establishment and early emergence under excess soil moisture conditions. Seeds were primed by soaking in -0.6 MPa polyethylene glycol (PEG) solution at $25^{\circ}C$ for 4 days. The primed seeds were sown in soils with various soil moistures (60, 80, 100, 120, and 140% field capacity) at 17 and $25^{\circ}C$, respectively. Germination and emergence rates, plumule height, and radicle length of primed seeds were higher than those of untreated seeds at any soil moisture and temperature examined. The time from planting to 50% germination ($T_{50}$) of primed seeds was less than that of untreated seeds by 0.9~3.7 days. Germination rate, emergence rate, plumule height, and radicle length were highest at the soil moisture of 80% field capacity among the soil moistures. Priming effects of rice seeds on germination and emergence rates were more prominent under the unfavorable soil moistures (60, 100, 120, and 140% field capacity) than those under the optimum soil moisture condition (80% field capacity). However, priming effects on seedling growth were greater at near optimum soil moisture compared with too lower or higher soil moistures. Therefore, these findings suggest that priming of rice seeds may be a useful way for better seedling establishment under the adverse soil conditions.

  • Laboratory experiements were conducted to evaluate water uptake and osmotic potential of soybean seeds associated with germinability. Bonus, Wayne, Essex and Pickett were selected for this study. Large and small seeds from the four varieties were included in water uptake measurements at $25^{\circ}C$ There were significant differences in water uptake during germination due to seed size and cultivars at optimum moisture condition, -6 and -15 bars soil moisture. As water stress increased, the water uptake of Pickett and Essex were superior to Bonus and Wayne, and small seeds were superior to large seeds. The seed moisture content at germination was 60.8% on a fresh weight basis under optimum moisture condition. The minimum moisture contents necessary for the germination were 50.2% at -6 bars and 50.9% at -15 bars. There were significant differences among varieties in seed osmotic potential during germination, although these differences depended on imbibition time. The average osmotic potential ranged from -32.0 bars after 4 hours imbibition to -11.2 bars at the beginning of germination. The correlation coefficient between seedling length and osmotic potential was not significant after any period of imbibition, suggesting that osmotic potential is not directly associated with seedling growth. However, osmotic potential is closely related to water uptake capacity of soybean seed.

Effects of Scarification and Soaking Treatment on Germination of Sword Bean Seed (작두콩 종자의 종피파상과 침지처리가 발아에 미치는 영향)

  • Seed coat of sword bean (Canavalia gladiata) is very thick and hard, it is difficult to absorb water during germination and it requires much time that cotyledon come out from seed coat since seminal root appearanced. Therefore this experiment was carried out to increase the germination rate by easing water absorption through mechanical scarification on seed coat. Non-scarification seed did not germinated at 7 days after treatment but scarification seed showed germination rate of 98%. Therefore mechanical scarification of seed coat was increased germination rate. Non-scarification seed absorbed less than 10% water of dry weight at 8 hours after treatment but scarification seed absorbed about 90% water at 7 hours after treatment and more than 150% water at 8 hours and swelled about 1.4-1.5 times in length and width of seed. Germination rate of scarification seed was high under high temperature and it was 96% and 93% at $25^{\circ}C$ and 3$0^{\circ}C$, respectively. The rate of cotyledon emergence on clay loam soil was the highest among bed soils. Soaked seed after mechanical scarification in distilled water was germinated faster than non-soaked seed on early in bed soil. The rate of cotyledon emergence was more than 92% at 9 days after sowing but non-scarification and soaked seed was germinated late and showed very low germination rate as 67%.
