Marine Algae of Cheju Island -The Leathesiaceae- (제주도산 해조류 -바위두둑과-)

  • Morphotaxonomic study was made on the Leathesiaceae of Cheju Island. One species of Petrospongium, P. rugosum, and three of Leathesia, L. difformis, L. sphaerocephala, and L. verruculiformis, were found in Cheju Island. Myriactula species was not found in the locality. L. verruculiformis was described as a new species of the genus Leathesia. L. verruculiformis was characterized by the less compact cortical layer, clavate assimilating filaments, and plurilocular sporangia born in tufts.

  • Thirty three species in six subfamilies of Staphylinidae are recognized from Ulleung-do through the survey in 2001. Among them, Thirty one species are newly reported from Ulleung-do. Two species, Anotylus lewisius(Sharp) and Megarthrus aino Cuccodoro, are reported for the first time from Korea.

Morphology of one Frontonia and two Spathidium ciliates (Ciliophora: Intramacronucleata) from Korea

  • Three ciliate species, Frontonia schaefferi Bullington, 1939, Spathidium curiosum Foissner, 2016, and S. muscicola Kahl, 1930, were collected from freshwater and terrestrial habitats in Korea. Their morphology was investigated based on live observations and protargol impregnation. Frontonia schaefferi was characterized by a cell size of $89-112{\times}45-53{\mu}m$ (after protargol impregnation) and three vestibular kineties. Spathidium curiosum was characterized by a cell size of ca. $125{\times}25{\mu}m$ in vivo and the unusual shape of the extrusomes. Spathidium muscicola was characterized by a cell size of ca. $135{\times}40{\mu}m$ in vivo and four to six macronuclear nodules. These three species are new records for Korea.

  • This study describes the optimum size of a reef set in Korean coastal waters. Data were analyzed in relation to the weight and variety of fish species caught with a three-layer gill net. The results did not indicate a significant difference in the mean catch and mean number of fish species among the three different reef sets, 800, 1,600 and 2400 $m^3$.

Random Amplified Polymorphic DNA (RAPD) Identification of Genetic Variation in Chlorella species

  • The random amplified polymorphic DNA (RAPD) technique was used to characterize 18 reference strains of microalgae, mostly Chlorella species, collected from various localities around Korea peninsular. Eighteen strains consist of four genera of the family marine Chlorella from 12 samples, two genera of fresh water Chlorella from three samples, and three genera on Nannochloris. Twenty 10-mer anonymous primers were screened for amplification of genomic DNA extracted from samples using the CTAB extraction method. Nineteen of these oligonucleotide primers were positive or band producing. Three of 20 random primers (OPA 10, OPA 12, and OPA 18) resulted in both clear band and a high degree of reproducibility and showed some potential to be used to discriminate individual samples of both genetically hetero-and homogeneous populations, in determining phylogenetic relationships between species within a genus and developing individual fingerprints for each samples.

  • Seasonal variations of GnRH were investigated by immunohlstochemicsl technique in three species of frog, Rona niEromaculutu, R. dvbowskii 8nd R. mgosa with different ovulation period in order to examine the relationship between GnRH expression and reproductive function. In all three species of frog, the intensity of GnRH immunoreactivitv and the number of GnRH neurons in the brawn were relatively high in frogs at the pre-ovulation period and markedly increased at the ovulation period. Those were then decreased after ovulation and further lowered during early hibernation period. These results indicate that GnRH experssion is closely related to specific phases of annual reproductive cycle in frog.

  • Sixty three species and 5 varieties belonging to 30 fungal genera were collected from 75 soil samples. Cultivated (29 genera and 58 species + 5 var.), desert (22 and 35 + 2 var.) and saline soil (21 and 41 + 1 var.) fungi were recovered on glucose-, cellulose- and 50% sucrose-Czapek's agar at $28^{\circ}C$. The most common genera were Alternaria, Aspergillus, Emerieella, Fusarium, Mycosphaerella, Nectria and Penicillium. The most prevalent species from the three types of soils on the three types of media were Alternaria alternata, Aspergillus flavus, A. fumigatus, A. niger, A. terreus, Emerieella nidulans, Fusarium oxysporum, Myeosphaerella tassiana, Nectria haematococca and Penicillium ehrysogenum. Chaetomium globosum was in the top of fungi in producing endo-$\beta$-1,4-glucanases among the 42 tested isolates obtained from soils on cellulose-Czapek's agar. Maximum production of this enzyme by C. globosum obtained after 6 days of incubation at $30^{\circ}C$ with culture medium containing maltose as a carbon source and ammonium nitrate as a nitrogen source and pH initially adjusted to 6.

  • The three yolk polypeptides have been isolated and partially characterized. Their molecular weights of YPI, YP2, and YP3 were 48, 000, 47, 000, and 46, 000, respectivelv, as judged by SDS-polyacrvlamide gel electrophoresis. They have different digestion products upon in situ peptide mapping by limited proteolysis. Two-dimensional gel electrophoresis showed that their isoelectric points were heterogeneous from 5.92 to 6.54. And thew showed three different antigen-antibody reactions when each polvpeptides is reacted with antisera made to a mixture of all of three. These data reported here indicate that the yolk proteins are consisted of distinctive polypeptides in Drosophlla sp. (robusta species group).

Three New Records of Penicillium Species Isolated from Insect Specimens in Korea

  • Three Penicillium species have been isolated from insect specimens in Korea; Penicillium sp., P. steckii, and P. polonicum. Penicillium sp. (KNU12-3-2) was isolated from Lixus imperessiventris, while P. polonicum (KNU12-1-8) and Penicillium steckii (KNU12-2-9) were isolated from Muljarus japonicas and Meloe proscarabaeus, respectively. The identification was based on the morphological characteristics of the fungi and in internal transcribed spacer analysis. This is the first report on the isolation of these three species of Penicillium from insects in Korea.

A New Report on Three Species of Sordariomycetes Class Isolated from Soil in Korea

  • Three fungal strains belonging to the class Sordariomycetes, namely BH-06, 17-039 and BE12-1, were isolated from soils in Korea and identified as Chaunopycnis alba, Myrothecium cinctum and Humicola olivacea, respectively. These species were confirmed according to their morphological characteristics and phylogenetic relationships determined based on internal transcribed spacer regions, as well as large subunit, small subunit, ${\beta}$-Tubulin and RNA polymerase II largest subunit sequences of ribosomal DNA. These three species are the first members of the Sordariomycetes reported in Korea.
