Heat-resistant microbial hosts are required for bioprocess development using high cell density cultivations at the industrial scale. We report that the thermotolerance of Escherichia coli can be enhanced by overexpressing ybeD, which was known to encode a hypothetical protein of unknown function. In the wild-type E. coli BL21(DE3), ybeD transcription level increased over five-fold when temperature was increased from $37^{\circ}C$ to either $42^{\circ}C$ or $46^{\circ}C$. To study the function of ybeD, a deletion strain and an overexpression strain were constructed. At $46^{\circ}C$, in comparison to the wild type, the ybeD-deletion reduced cell growth half-fold, and the ybeD-overexpression promoted cell growth over two-fold. The growth enhancement by ybeD-overexpression was much more pronounced at $46^{\circ}C$ than $37^{\circ}C$. The ybeD-overexpression was also effective in other E. coli strains of MG1655, W3110, DH10B, and BW25113. These findings reveal that ybeD gene plays an important role in enduring high-temperature stress, and that ybeD-overexpression can be a prospective strategy to develop thermotolerant microbial hosts.
Thermal stress due to extreme changes in the thermal environment is a critical issue in cattle production. Many previous findings have shown a decrease in feed intake, milk yield, growth rate, and reproductive efficiency of cattle when subjected to thermal stress. Therefore, selecting thermo-tolerant animals is the primary goal of the efficiency of breeding programs to reduce those adverse impacts. The recent advances in molecular genetics have provided significant breeding advantages that allow the identification of molecular markers in both beef and dairy cattle breeding, including marker-assisted selection (MAS) as a tool in selecting superior thermo-tolerant animals. Single-nucleotide polymorphisms (SNPs), which can be detected by DNA sequencing, are desirable DNA markers for MAS due to their abundance in the genome's coding and non-coding regions. Many SNPs in some genes (e.g., HSP70, HSP90, HSF1, EIF2AK4, HSBP1, HSPB8, HSPB7, MYO1A, and ATP1A1) in various breeds of cattle have been analyzed to play key roles in many cellular activities during thermal stress and protecting cells against stress, making them potential candidate genes for molecular markers of thermotolerance. This review highlights the associations of SNPs within these genes with thermotolerance traits (e.g., blood biochemistry and physiological responses) and suggests their potential use as MAS in thermotolerant cattle breeding.

The fermentative conversion of food wastes into feed was investigated by seeding of mixed inoculum YM (Youngjin Environmental co.), and thermotolerant yeast Kl. marxianus. For 6 days' fermentaion, the fermentation method of 2 days' aerobic followed by 4 days' anaerobic was better for the production of organic acids and increasing total microbial population than 6 days' continuous aerobic or anaerobic fermentation. By seeding YM, the total microbial count increased about 100 times of the control group. In addition, Kl. marxianus seeding together with YM increased total viable cell count, but did not increase yeast count significantly.

Cambodia is a typical country that a very famous tourist destination in South Asian regions, but water has some serious problems. As a public hygiene point of view, water problems in Cambodia can be roughly classified into two categories. A numbers of the report revealed that thermotolerant coliform(TTC) bacteria and arsenic(As) pollution representative species as a representative pollutant for surface water and ground water problems, respectively. In recent years, home water purifier systems for various filter systems including BSF(Bio-Sand Filter), MF and RO have been spread and adopted in rural areas. However, BSF and composite membrane processes are recognized as an undesirable and representative method due to unreliable efficiency, high cost and maintenance issues. In order to develop a small-scale ceramic filter as a home water purifier, the applicability of two types of gravity ceramic filter using clay and red soil in the part of the Northwest was made and compared in this research.

As a process to utilize agricultural residues, simultaneous hydrolysis-fermentation (SSF) was compared with fermentation of enzymic hydrolyzate using koji cultures of Trichoderma sp. KI 7-2 and a thermotolerant yeast Saccharomyces cerevisiae NCYC 716. Cellobiose was not detected in SSF broth whilst 15 mg/$m\ell$ of the disaccharide was found in enzymic hydrolysate of rice straw using the same enzyme source. It was found that converting glucose to ethanol in SSF process reactivated the cellobiase activity, which is inhibited by the accumulation of glucose in enzymic hydrolysis process. Cutting milled rice straw was fermented as effectively as ball milled one in SSF process. From tile results discussions are made on the product inhibition mechanism of cllulolytic enzyme system.
A systematic investigation was performed on the bacterial, Bacillus, fungal, and yeast communities of the three types of Daqu (mechanically prepared, manually prepared, and mixed prepared) used in Baiyunbian Company by reconditioning PCR-denaturing gradient gel electrophoresis (PCR-DGGE). The DGGE results showed that the microbes in the three types of Daqu were mainly thermotolerant and thermophilic microbes, and the most dominant bacterial species were Bacillus and Virgibacillus, followed by Lactobacillus and Trichococcus. Furthermore, the dominant fungi were found to be molds, such as Rasamsonia, Penicillium, Aspergillus, and Monascus, and the dominant yeasts were Saccharomyces cerevisiae, Saccharomycopsis fibuligera, Pichia anomala, and Debaryomyces hansenii. In general, the three types of Daqu showed slight differences in microbial communities, and the Shannon indexes (H') of the manually prepared and mechanically prepared Daqu were similar. The results suggest that mechanically prepared Daqu can replace manually prepared Daqu in liquor production, and this research provides useful information for liquor production and process improvement.
To investigate the function of chloroplast-localized small HSP in rice, the cDNA, Oshsp21, was introduced into rice plants via Agrobacterium-mediated gene transfer system. Calli induced from rice immature embryos were co-cultivated with a A. tumafaciens EHA101 that contained a plasmid, pIHSP21. The efficiency of plant regeneration from the calli co-cultivated with the Agrobacterium was about 30%. PCR and Southern blot analyses using genomic DNA revealed that gene for the chloroplast small HSP was introduced into the genome of rice. Expression of transgene was investigated by northern blot analysis. Results indicate that the transgene, Oshsp21, was constitutively expressed at normal growth temperature.
The cost of conventional cultivation of ginseng (Panax ginseng C.A. Meyer) is very expensive, because shadow condition should be maintained during cultivation periods owing to inherently weak plant for high-temperature. Therefore, application of plant biotechnology may be possible to overcome these difficulties caused by conventional breeding of ginseng. Transgenic plants were produced via Agrobacterium tumefaciens Gv3101, both carrying the binary plasmid pBI121 mLPISO with nptII and Iso (isoprene synthase) gene. Integration of the transgenes into the P. ginseng nuclear genome was confirmed by PCR analysis using nptII primers and Iso primers. RT-PCR result also demonstrated the foreign isoprene synthase gene in three transgenic plant lines (T1, T3, and T5) which was expressed at the transcriptional level. When whole plants of transgenic ginseng were exposed to high temperature at $46^{\circ}C$ for 1 h, a non-transformed plant was wilted from heat shock, whereas a transgenic plant appeared to remain healthy. We suggest that the introduction of exogenous isoprene synthase is considered as alternative methods far generating thermotolerance ginseng.

The waste from beer fermentation broth (WBFB) has been found an excellent and inexpensive resource for bioethanol production. We tried to evaluate the saccharification and fermentation capabilities of WBFB to confirm its effectiveness for bioethanol production. The saccharification potentials of the WBFB were evaluated at various temperatures (30, 40, 50, 60 and $70^{\circ}C$). It was found that the saccharification capabilities increased with temperature and highest reached maximum at $60^{\circ}C$ and $70^{\circ}C$ after 4h. Ethanol production from a mixture of WBFB and chemically defined media (CDM) without addition of any microbial species confirmed the fermentation capabilities of WBFB. Simultaneous saccharification and fermentation were performed using WBFB, starch solution and CDM as culturing media. The maximum yield of bioethanol production was obtained at $30^{\circ}C$. The saccharifying enzymes and the yeast cells present in WBFB were essential factors for the production of bioethanol from WBFB without any additional enzymes or microbial cells.

Sugar beet stillages were used as a substrate for the production of single cell protein by the thermotolerant yeasts Candida rugosa, Kluyveromyces marxianus and C. utilis. The biomass production increased in accordance with the increase of pH-value, but protein content decreased. C. rugosa showed the highest crude protein production as 3.68g/l and C. utilis 2.9g/l, Kl. marxianus 2.30g/l, respectively. The rate of COD reduction in stillage versus crude protein production of C. rugosa showed the highest value as 0.35~0.39g/l as a good strain for single cell protein production using sugar beet stillages.
