A clinical case of bovine anemia due to Theileria orientalis group in a non-grazed dairy cow in the upper part of South Korea

  • This study reports the detection and genotyping of the first clinical case of bovine anemia due to Theileria orientalis group (BATOG) in non-grazed dairy cow in upper South Korea. Blood and serum tests revealed anemia and hyperbilirubinemia from animal showing clinical symptoms, and later confirmed as piroplasmosis-positive. Follow-up surveillance on the herd revealed 2 asymptomatic cows with anemia. The three animals were confirmed theileriosis-positive and genotyping revealed the clinical and one of the asymptomatic cases have Chitose, while the other has Ikeda genotype. Clinical BATOG cases were rarely reported worldwide, and asymptomatic animals left untreated could serve as parasite reservoir.

  • 소 theileriosis의 태반감염 사실을 입증하기 위하여 Theileria sergenti에 자연감염된 암소의 비장, 태반 그리고 유산된 태자의 비장과 태반조직으로부터 T sergenti를 면역화학적으로 검색 입증하고자 T sergenti 표피의 34KD 항원의 단크론 항체를 활용하여 avidin biotin complex 방법으로 면역화학적으로 염색하였던 바, 이들 formalin 고정 조직표본에서 T sergenti의 특이 항원성 물질을 관찰함으로써 태반감염을 증명할 수 있었다.

  • Eighteen holstein-calves(4~5 months old) in a divided groups including the matched control were immunized with $100{\mu}g/dose$ of 34kDa, 45kDa polypeptide and T sergenti merozoite vaccine(protein content $100{\mu}g/dose$) respectively, previously mixed with aluminium hydroxide to elicit antibodies. All groups of calves were boosted with same dose and intervals. The animals were challenged by tick infestations in the endemic pasture of theileriosis from March to September 1994. The animals were monitored for the erythrocyte count, parasitemia, hematocrit and the specific antibody reactions elicited by immunization. The immunological responses demonstrated that vaccination with 34kDa polypeptide and T sergenti merozoite derived vaccine inhibited to produce the 75kDa band immunological responds even in the vaccinated calves after being challenged by tick infestations in the pasture. However, the specific antibody reactions were detected at the 32kDa band in the nonimmunized calves and T sergenti merozoite derived vaccine by the western blot. The 34kDa polypeptide vaccine and T sergenti merozoite derived vaccine were evaluated to be able to protect inducing anemia and to decrease parasitemias level. These vaccines have the efficacy of inhibition to produce a certain antigen corresponding 75kDa band antigen of parasite in the calves as challenged with tick infestations.

Production of Theileria sergenti recombinant protein by E coli expression system

  • As an attempt to develop an effective control method against theileriosis, recombinant antigen protein was produced. Thirty-two kDa membrane protein(MP) gene of T sergenti was amplified through RT-PCR from extracted total RNA of T sergenti isolated in Chonbuk, Korea. The amplified 869 bp of Korean T sergenti membrane gene was cloned and the base sequences were analyzed. The amplified gene was cloned into E coli expression vector, pQE32 plasmid vector, and the vector was introduced into E coli strain M15 to produce the recombinant membrane protein. For the induction of T sergenti membrane protein(KTs-MP), the plasmid harboring E coli strain M15 were cultured in the presence of IPTG, and the recombinant protein were purified by $Ni^+$-NTA agarose. Then, to confirm the authenticity of the produced membrane protein, molecular weight of expressed recombinant KTs-MP was analyzed by SDS-PAGE and Western blotting. The molecular weight of expressed recombinant protein was 32 kDa as expected. The recombinant KTs-MP was successfully recognized by anti-His Tag antibody, antisera of T sergenti infected cattle and monoclonal antibody of T sergenti membrane protein. Therefore, we concluded that the authentic 32 kDa membrane protein of T sergenti was produced as immunologically recognizable form.

소의 질병감염이 생산성에 미치는 영향 (Productivity Affected by Various Disease Conditions in Bovine)

  • Present investigations were undertaken in order to clarify the clinical status of Korean native cattle and dairy cattle(holstein). Blood, Feces and urine samples were collected from 247 Korean native cattle(222 adult and 25 calf), 224 dairy cattle(211 adult and 13 calf) at Chonbuk area and analyzed for clinical, serum chemical, hematological and urinary findings. In addition, we were examined the infection rate of Theileriosis, internal patasite and ring worm. The mean value for each component was calculated by statistical analysis using Excel computer program. From these investigations the following results were obtained. The mean values for RBC, PCV and etc in 433 adult cow(Korean native cattle and dairy cattle) were similar with other reports. But the mean values for MCHC of all species were lower than normal. Adult Korean native cattle and adult dairy cattle which showed hematologically normal levels were only 9.01% and 9.48%, respectively. Ahnormally high values for PCV, RBC and Hb were recorded in 7.66% of adult Korean native cattle, 20% of Korean native calf, 15.38% of dairy calf. Adult Korean native cattle and adult dairy cattle which showed hematologically anemia were 4.95% and 19.43% respectively, but all in Korean native calf and dairy calf showed normal values. Adult Korean native cattle, adult dairy cattle, Korean native calf and dairy calf which showed normal serum protein level were 84.0%, 90.8%, 50% and 44.4%, respectively. In present investigations, 50% of Korean native calf and 55.6% of dairy calf were decreased serum protein values under normal range. These abnormally decreased serum protein values mean the shortage of antibody, and these have a possibility to occrus to pneumonia and diarrhea. From these results, the economical loss caused by pneumonia was calculated as 124, 038, 833 won in the KNC and 742, 703, 430 won in the dairy calf rearing in Chonbuk area. Calculated economical loss caused by enteritis was 56, 658, 690 won in Korean native cattle. 476, 775, 799 won in dairy calf and the total loss amount to 533, 434, 488 won in Chonbuk area. Abnormally high values($21.7{\pm}4.0mg/dl) for serum calcium were recorded 49.6% in dairy cattle. The mean values of serum total cholesterol were $170.8{\pm}99.8mg/dl in Korean native cattle, $196.0{\pm}40.6mg/dl$ in Korean native calf, $202.9{\pm}86.0mg/dl$ in adult dairy cattle and $289.4{\pm}97.5mg/dl$ in dairy calf. The infection rate of internal parasite were as follows; adult Korean native cattle:21.2%, Korean native calf:80%, adult dairy cattle:67.8%. The estimated economical loss caused by internal parasites infection were 1, 120, 855, 837 won in Korean native calf, 4, 994, 959, 405 won in adult Korean native cattle, 3, 334, 751, 066 won in adult holstein, and the total loss amount to 9, 450, 566, 308 won. The infection rate of theileriosis were 1.4% in Korean native cattle and 6.6% in dairy cattle. The presumed Economical loss by T. sergenti infection were 154, 408, 482 won in Korean native cattle and 171, 577, 237 won in dairy cattle rearing at Chonbuk area. The infection rat of ringworm were 0.5% in Korean native cattle, 0.9% in adult dairy cattle and 7.7% in dairy calf. The presumed economical loss by dermatomycophyte were 12, 061, 532 won in Korean native cattle, 16, 895, 403 won in dairy cattle, and the total estimated loss amount to 28, 955, 935 won a year in Chonbuk area. The infection rate of ringworm were 0. 5% in Korean native cattle, 0.9% in adult dairy cattle and 7.7% in dairy calf. The presumed economical loss by dermatomycophyte were 12, 061, 532 won in Korean native cattle, 16, 895, 403 won in dairy cattle, and the total estimated loss amount to 28, 955, 935 won a year in Chonbuk area.

  • For the purpose to increase productivities of livestock the present investigations were undertaken in order to clarify the clinical and suclinical status of Korean native cattle, dairy cattle(holstein) and Korean native goat. Blood, feces and urine samples were collected from 247 Korean native cattle(222 adult and 25 calf), 224 dairy cattle(211 adult and 13 calf) and 142 Korean native goat rearing at Chonbuk area and analyzed for clinical, serum chemical, hematological and urinary findings. In addition, we were examined the infection rate of theileriosis, internal parasite and ring worm. The presumed economical loss by T. sergenti infection were 154,408,482 won in Korean native cattle and 171,577,237 won in dairy cattle rearing at Chonbuk area. Validation of Reference Genes for Quantitative Real-Time PCR in Bovine PBMCs Transformed and Non-transformed by Theileria annulata

  • Theileria annulata is a tick-borne intracellular protozoan parasite that causes tropical theileriosis, a fatal bovine lymphoproliferative disease. The parasite predominantly invades bovine B lymphocytes and macrophages and induces host cell transformation by a mechanism that is not fully comprehended. Analysis of signaling pathways by quantitative real-time PCR (qPCR) could be a highly efficient means to understand this transformation mechanism. However, accurate analysis of qPCR data relies on selection of appropriate reference genes for normalization, yet few papers on T. annulata contain evidence of reference gene validation. We therefore used the geNorm and NormFinder programs to evaluate the stability of 5 candidate reference genes; 18S rRNA, glyceraldehyde-3-phosphate dehydrogenase (GAPDH), ACTB (${\beta}-actin$), PRKG1 (protein kinase cGMP-dependent, type I) and TATA box binding protein (TBP). The results showed that 18S rRNA was the reference gene most stably expressed in bovine PBMCs transformed and non-transformed with T. annulata, followed by GAPDH and TBP. While 18S rRNA and GAPDH were the best combination, these 2 genes were chosen as references to study signaling pathways involved in the transformation mechanism of T. annulata.

경북지방(慶北地方)의 육성우(育成牛) 및 비육우(肥育牛)에 있어서 기생충(寄生蟲)의 감염(感染)과 질병실태조사(疾病實態調査) (Incidence of Parasitic Infections and Diseases in Rearing and Fattening Cattle Raising in Gyeongbug District)

  • A survey was made from March 1978 through Feb. 1979 to know infection rate of parasites and incidence of diseases in 1968 cattle of 240 rearing and fattening cattle herds (1746 Korean native cattle, 186 Holstein, 34 Charolais and 2 Hereford) raising in 28 cities and counties of Gyeongbug district. The results obtained are summarized as follows: 1. The rate of nematoda infection was about 49.0% in the examined cattle. These nematodes were identified as Mecistocirrus sp., Oesophagostomum sp., Bunostomum sp., Trichostrongylus spp., Strongyloides sp. and other 3 species. Infection rate of nematoda in fattening and breeding cattle was higher than that in rearing calves, 44.3% of the positive cattle were infected with 2 to 5 species, and incidence of mixed infection was high in fattening cattle. 2. The rate of coccidal infection was 10.9%. Eimeria $z{\ddot{u}}rni$, Eimeria bovis and Eimeria bukidnonensis were mainly found in the examined cattle. The infection rates of cocidia in Holstein, breeding Korean cows, rearing Korean calves and fattening Korean cattle were 27.3%, 15.8%, 11.2% and 9.1%, respectively. 3. The rate of trematoda was 14.6% with Fasciola spp. (11.2%), Eurytrema spp. (2.0%), and Paramphistomum spp. infections (1.4%). Breeding Korean cows, fattening Korean cattle, Holstein and rearing Korean calves indicated 47.4%, 26.4%, 18.2% and 6.3% incidence of trematoda infections, respectively. of trematoda positive cattle, 76.6% were infected with Fasciola spp., 4. The rate of Theileria and Babesia infection was 62.7% in the examined cattle. Of these positive cattle 5.4% were infected with both parasites, 57.3% with Theileria, and 1.3% of rearing Korean calves and 22.6% of Holstein were found to have parasitized erythrocytes over 1.0%. 5. The incidence of diseases in 1968 cattle of 240 herds was 564 cattle (28.7%) of 154 herds (64.2%). of the diseases observed of skin occurred in 354 cattle (18.0%) of 61 herds (25.4%), diseases of respiratory system in 121 cattle (6.1%) of 34 herds (14.2%), diseases of digestive system in 38 cattle (1.9%) of 22 herds (9.2%), diseases of eye in 21 cattle (1.1%) of 13 herds (5.4%), diseases of urogenital system in 8 cattle (0.4%) of 8 herds (3.3%) and the other diseases in 24 cattle (1.2%) of 22 herds (9.2%). 6. Diseases of skin in 1968 cattle of 240 herds werds were observed in the highest incidence. Ringworm was observed in 13.3% of the examined herds, rearing Korean calves and Holstein showed high incidence. Scabies caused by Chorioptes bovis and alopecia localis along with infestation of lice (Damalinia bovis, Linognathus vituli) were observed in fattening Korean cattlein winter. 7. Diseases of respiratory system occurred frequently in rearing Korean cattle and Holstein. In the diseases of digestive system acute indigestion occurred in fattening Korean cittle, enteritis in rearing Korean calves and Holstein, and coccidiosis in rearing Korean calves maninly. 8. Infectious keratoconjunctivitis occurred in a fattening Korean cattle herd, sterility (2.0%) in breeding Korean cows, and theileriosis in Holstein calves were also occurred. In addition, poisoning and heat stroke were observed in several cattle.
