Background: Sancho oil extracted from Zanthoxylum schinifolium (Siebold & Zucc) is a useful edible oil that has been in use for a long time, but it is known to be susceptible to rancidity. Sancho oil purification can remove impurities to prevent rancidity. This study was performed in order to improve the quality of sancho oil and enhance its availability throughout the purification process. Methods and Results: Sancho oil extracted in Hadong, Korea was refined via the degumming and deoxygenation processes, following which we examined the changes in the polyphenol content, fatty acid content and antioxidant activity of the oil. Acetic acid was effective for deoxygenation of sancho oil and 2 N NaOH was effective for its deoxidation. The polyphenol content and antioxidant activity were reduced by the purification process. Saturated fatty acids contents did not vary with the degumming and deoxygenation processes, however the content of unsaturated fatty acids were slightly reduced. Conclusions: This study suggests that the process of sancho oil purification used in this study will contribute to the increased use and storage of sancho oil.

This study investigated temporal changes in forest physical variables and their effects on thermal comfort and physiological and psychological responses. Environmental factors (air temperature, mean radiant temperature, wind speed, and relative humidity), the predicted mean vote (PMV), and the predicted percentage of dissatisfied (PPD) visitors were continuously measured between 9:00 AM and 6:00 PM. We assessed the physiological and psychological responses (heart rate variability, heart rate, oral temperature, blood pressure, pulse rate, thermal sensation vote, comfort sensation vote, and subjective feelings) of 30 male university students (average age: 21.7±1.9 years), who closed their eyes and relaxed for 5 minutes every hour. Examination of correlations between environmental factors and physiological responses showed that ① the mean radiant temperature, wind speed, and relative humidity significantly changed with time; ② PMV and PPD also showed significant changes over time, and the thermal sensation vote corresponded with PMV; however, the comfort sensation vote did not correspond with PPD; ③ Among the physiological responses, parasympathetic nerve activity, sympathetic nerve activity, heart rate, and diastolic blood pressure significantly varied with time, with parasympathetic nerve activity having the lowest value and sympathetic nerve activity the highest at the highest air temperature (2:00-4:00 PM); and ④ Air temperature, mean radiant temperature, and wind speed showed a negative correlation with parasympathetic nerve activity and a positive correlation with sympathetic nerve activity. These findings indicate that the relaxation effects of the forest environment depend on what time of day people are exposed to it, and the changes occurring in forest environmental factors over time modulate these effects.

Antimicrobial activity and antioxidative activity of the organosoluble extractives from the leaves, bark and xylem of Quercus dentata were investigated. Antimicrobial activity was tested by paper disk method and bioautography methods, The most pronounced antimicrobial activities of leaves parts were ethyl acetate solubles fraction against the Klebsiella pneumoniae by the paper disk method. The strongest activities of bark parts were ethyl acetate solubles fraction against Bacillus subtilis, Klebsiella pneumoniae. Ethanol extractives from xylem parts showed high activities against Bacillus subtilis, Staphylococcus aureus, Klebsiella pneumoniae and Vibrio parahaemolyticus. These antimicrobial activities of ethanol extractives from Quercus dentata were in order to xylem > bark > leaves. The strong inhibition zones of the ethanol extractives and its fractions of xylem showed Rf values in 0.41~0.63. In leaves extractives, the petroleum ether solubles showed lower antioxidative activity and ethyl acetate insolubles showed higher antioxidative activity of 70% compared with the $EC_{50}$ values of the control. Antioxidative activity of bark and xylem extractives showed higher approximately 2 times than the control except the petroleum ether solubles.
Two-year-old seedlings of Ilex rotunda were grown under control (full sunlight) and three different shading condition. Those conditions were full sunlight (PPFD 1600${\mu}mol\;m^{-2}\;s^{-1}$), 30% (PPFD 400${\mu}mol\;m^{-2}\;s^{-1}$), 50% (PPFD 250${\mu}mol\;m^{-2}\;s^{-1}$) and 70% treatment (PPFD 100${\mu}mol\;m^{-2}\;s^{-1}$). Total chlorophyll contents were inverse proportion to light intensity. Seedlings under full sunlight showed the highest photosynthetic activity such as photosynthetic rate, intercellular $CO_2$ concentration and water use efficiency. Photosynthetic activity trend was increased at the higher light intensity than the lower treatment over PPFD 500${\mu}mol\;m^{-2}\;s^{-1}$. Especially, seedlings under 70% treatment showed the worst photosynthetic activity at high light intensity. That result was regular for adapted plant in low intensity environment. Leaf area was also inverse proportion to light intensity, while dry weight per leaf area was shown the opposite trend.
The Sundarbans mangrove forest has a rich biodiversity of flowering plants and many of these have been used in traditional medicine although the flora remains comparatively uninvestigated scientifically. Xylocarpus granatum, Xylocarpus moluccensis and Excoecaria agallocha methanolic extract showed a central nervous system depressant activity on the hole cross and open field test at 800 mg/kg dose level. The most significant depressant activity was observed in Xylocarpus granatum followed by Xylocarpus moluccensis and Excoecaria agallocha. There was no depressant activity observed in the models for Sarcolobus globosus. Further studies are required to confirm the activity and to explain the mechanism.
Seasonal and vertical variations of abiontic soil carboxymethylcellulase (CMCase) activities were assessed every other month for a year in two contrasting forest soils and evaluated the relationships between soil CMCase activity and environmental parameters. In climax deciduous soil, variations in CMCase activities caused by differences in sampling time were greater than those caused by differences in soil depth. On the other hand, counter phenomenon was obserned in coniferous soil at the stage of development. Correlation analyses showed that soil CMCase activities were significantly (p>0.01) correlated with microbial respiration rates ($O_2$ uptake) and all of the microbial population sizes. From these results, it is suggested that determination of abiontic soil CMCase activity is an useful additional index for evaluating the overall microbial growth and activity in soils.
The effects of soil acidification on the photosynthetic activity of Quercus acutissima seedlings were investigated. We measured the growth and photosynthetic activity of the seedlings in relation to soil acidification. The dry weights of the seedlings were reduced according to the amount of $H^+$ in the soil. The concentrations of Al in needles at the 90 meq $H^+$ were significantly higher than those at the control. The contents of chlorophyll in needles at 90 meq $H^+$ were significantly lower than those at the control. The net photosynthetic rates of the seedlings in the acidified soil were reduced by increasing the amount of $H^+$ to the soil. The carboxylation efficiencies(CE) of photosynthesis were reduced in the seedlings grown in the acidified soil. These results suggested that the soil acidification induced the inhibition of photochemical reactions and $CO_2$ fixation of photosynthesis.

Oak wilt disease caused by Raffaelea quercus-mongolicae has emerged obviously in Korea. We selected antifungal isolates against R. quercus-mongolicae among 368 endophytic fungal isolates from different parts of oak and pine trees. The experiment was conducted in the primary and secondary screenings by dual culture test. The antifungal activity of the selected isolates was assessed in culture filtrate test based on the inhibition rates in mycelial growth, sporulation, and spore germination of oak wilt fungus. Five isolates, E089, E199, E282, E409 and E415, showed strong antifungal activity in culture filtrate test, and their antifungal activity decreased on the culture media supplemented with heated culture filtrate. Higher mycelial growth inhibitions on the unheated media were recorded in E409 (Colletotrichum acutatum), E089 (Daldinia childiae), E415 (Alternaria alternata) and E199 (Daldinia childiae) with the inhibition rates of 79.0%, 70.1%, 68.9% and 64.5%, respectively. These isolates also had the higher sporulation inhibitions on unheated media with the rates of 96.8%, 84.2%, 82.8% and 80.5%, respectively. The spore germination of the oak wilt fungus was completely inhibited by E282 (Nectria balsamea) on both unheated and heated media. These results showed that a higher number of potent antifungal isolates against oak wilt fungus was isolated from the petiole compared to the other parts. This study could contribute to the development of biological control approaches for the management of oak wilt disease caused by R. quercus-mongolicae.
A metagenome is a unique resource to search for novel microbial enzymes from the unculturable microorganisms in soil. A forest soil metagenomic library using a fosmid and soil microbial DNA from Gwangneung forest, Korea, was constructed in Escherichia coli and screened to select lipolytic genes. A total of seven unique lipolytic clones were selected by screening of the 31,000-member forest soil metagenome library based on tributyrin hydrolysis. The ORFs for lipolytic activity were subcloned in a high copy number plasmid by screening the secondary shortgun libraries from the seven clones. Since the lipolytic enzymes were well secreted in E. coli into the culture broth, the lipolytic activity of the subclones was confirmed by the hydrolysis of p-nitrophenyl butyrate using culture supernatant. Deduced amino acid sequence analysis of the identified ORFs for lipolytic activity revealed that 4 genes encode hormone-sensitive lipase (HSL) in lipase family IV. Phylogenetic analysis indicated that 4 proteins were clustered with HSL in the database and other metagenomic HSLs. The other 2 genes and 1 gene encode non-heme peroxidase-like enzymes of lipase family V and a GDSL family esterase/lipase in family II, respectively. The gene for the GDSL enzyme is the first description of the enzyme from metagenomic screening.
To evaluate effects of ligninolytic enzyme type on the mycelial response and ligninolytic enzyme production during interspecific interactions among wood-rotting fungi, 4 fungal strains, Trichophyton rubrum LKY-7, Trichophyton rubrum LSK-27, Pycnoporus cinnabarinus, and Trichoderma viride, were selected. Regarding ligninolytic enzyme production, LKY-7 secreted laccase and manganese peroxidase (MnP), P. cinnabarinus secreted only laccase, and LSK-27 secreted only MnP in glucose-peptone medium, while T. viride did not produce any ligninolytic enzymes. In the co-culture of LKY-7 with P. cinnabarinus, the formation of aerial mycelium was observed and the enhancement of laccase activity owing to interspecific interaction appeared to be very low. In the co-culture of LKY-7 and P. cinnabarinus with LSK-27, a hypha-free clear zone was observed, which resulted in deadlock, and increased laccase or MnP activity was detected at the interaction zone. The interaction responses of LKY-7, P. cinnabarinus, and LSK-27 with T. viride were characterized by the formation of mycelial barrages along the interface. As mycelial barrages were observed at the T. viride territory and no brownish pigment was observed in the mycelial barrages, it is suggested that laccase and MnP are released as part of an offensive response, not as a defensive response. The co-culture of P. cinnabarinus with T. viride lead to the highest enhancement in laccase activity, yielding more than 14-fold increase in laccase activity with respect to the mono-culture of P. cinnabarinus. MnP activities secreted by LKY-7 or LSK-27 was generally low in interspecific interactions.
