Micromorphometric change of implant surface conditioned with tetracycline-HCI;HA and oxidized surface (표면처리 시간에 따른 임플란트 미세구조의 변화;HA와 양극산화 표면 임플란트)

  • The present study was performed to evaluate the effect of tetracycline-HCL on the change of implant surface microstructure according to application time. Implant with pure titanium machined surface. HA-coated surface and TiUniteTM surface were utilized. Implant surface was rubbed with 50mg/ml tetracycline-HCL solution for $\frac{1}{2}min.$, 1min., $1\frac{1}{2}min.$, 2min., and $2\frac{1}{2}min.$ respectively in the test group. Then, specimens were processed for scanning electron microscopic observation. The results of this study were as follows. 1. Both test and control group showed a few shallow grooves and ridges in pure titanium machined surface implants. There were not significant differences between two groups. 2. In HA-coated surfaces, round particles were deposited irregularly. The roughness of surfaces conditioned with tetracycline-HCL was lessened and the cracks were increased relative to the application time. 3. The anodic oxidized surfaces showed the craterous structures. The surface conditioning with tetracycline-HCl didn't influence on its micro-morphology. In conclusion, the detoxification with 50mg/ml tetracycline-HCL must be applied respectively with different time according to various implant surfaces.

The influence of tetracycline-HCI for micromorphology of Thermal dual acid etched surface implants (염산 테트라싸이클린이 이중 산부식 임플란트 표면 구조에 미치는 영향)

  • The present study was performed to evaluate the effect of Tetracycline-HCI on the change of implant surface microstructure according to application time. Implants with thermal dual acid etched surface were utilized. Implant surface was rubbed with $50mg/m{\ell}$ Tetracycline-HCI solution and sterilized saline for O.5min, 1min, 1.5min, 2min, 2.5min and 3min. respectively in the test group. Then, specimens were processed for scanning electron microscopic observation and measured surface roughness by optical interferometer. The results of this study were as follows. 1. The thermal dual acid etched surfaces showed many small peaks and valleys distributed overall surface. 2. The surface conditioning with Tetracycline-HCI and saline didn't influence on its micromorphology. In conclusion, the implant with thermal dual acid etched surface has a protective micromorphology from the detoxification with $50mg/m{\ell}$ Tetracycline-HCI and a scrubbing with cotton pellet. Therefore, the detoxification with $50mg/m{\ell}$ Tetracycline-HCI is an effective method for peri-implantitis in case implants with thermal dual acid etched surface.

  • Lactadherin (formerly known as BA46), a major glycoprotein of the human milk fat globule membrane, is abundant in human breast milk and breast carcinoma cells and is known to prevent symptomatic rotavirus infections. In this study, we tried to transfer the human lactadherin gene to the Chinese Hamster Ovary (CHO) cells using retrovirus vector system and tested inducible expression of the gene under the tetracycline-controllable promoter. At first, tetracycline-mediated inducibility was tested using E.coli LacZ marker gene. NIH3T3 cells co-infected with RevTet-On and RevTRE-LacZ retrovirus vectors showed that the cells responded to doxycycline (a derivative of tetracycline) in a dose-dependent manner, and prominent induction of the lacZ gene expression was observed from 1 $\mu\textrm{g}$/ml of doxycycline concentration. Based on the results of the pilot experiment, inductional expression of the human lactadherin gene was conducted using RevTet-On and RevTRE-Ltd retrovirus vectors. Analysis with the RT-PCR demonstrated successful inductional expression of the lactadherin gene in the Chinese Hamster Ovary (CHO) cells. Considering that constitutive overexpression of the exogenous genes in the target cells causes serious physiological imbalance, the results obtained in this study will be very useful especially in the studies of gene therapy and transgenic animal production.

Antibiotic Susceptibility to Isolated Bacteria and Fungi from the Indoor-air (실내 공기중에서 세균 및 진균의 분포와 분리균의 항생물질 감수성)

  • This investigation was performed to isloate and identify the total bacteria, Staphylococcus spp. and fungi from the indoor air exposed for 30 minutes on the blood agar plate at the 27 places in a hospital. Antibiotic susceptibility tests of the isolated bacteria were also studied. The mean numbers of total bacteria, Staphylococci spp. and fungi were 26, 17, and 2 in the summer and 19, 8, and 2 in the winter, respectively. Staphylococcus epidermidis was the most common isolated bacteria, and the next was Staphylococcus aureus, Aerococcus spp., Micrococcus spp., and Bacillus spp. from the indoor-air of hospital. Aspergillus spp., Cephalosporum spp., Curvularia spp., penicillium spp., and Phialophora spp. was frequently isolated from the indoor-air of hospital. The 109 strains of isolated Staphylococcus epidermidis sho-wed resistance to tetracycline(45.0%), methicillin(40.2%), erythromycin(31.2%), and kanamycin(24.8%). The 76 strains of isolated Staphylococcus aureus showed resistance to erythromycin(71.7%), methicillin(63.2%), kanamycin (44.7%), tetracycline(39.5%), and ampicillin(32.9). The 67 strains of isolated Aerococcus spp. showed resistance to erythromycin(26.9%), methicillin(25.4%), kanamycin(22%), and tetracycline(22.4%). The 48 strains of isolated Micrococus spp. showed resistance to tetracycline(27.0%), methicillin(22.9%), erythromycin(22.9%), and kanamycin(20.8%).

Examination of Improved Tetracycline Inducible Gene Expression System In Vitro (새로운 Tetracycline 유도적 유전자 발현 System의 In Vitro 검정)

  • Until recently the most popular tetracycline-inducible gene expression system has been the one developed by Gossen and Bujard. In this study, we tested the latest version of same system and the results are summarized as follows: Compared with previous one, the difference of new system are minor changes of nucleotide sequences in transactivator and tetracycline response element (TRE) regions. Sensitivity to the doxycycline (a tetracycline derivative) was improved. Leakiness of GFP marker gene expression in non-inducible condition was significantly decreased. Higher expression of the marker gene was observed when the cells were fed with doxycycline-containing medium. Optimal insertion site of woodchuck posttranscriptional regulatory element (WPRE) sequence which was known to increase gene expression was different depending on the origin of cells. In chicken embryonic fibroblast, location of WPRE sequence at 3' end of TRE resulted in the highest GFP expression. In bovine embryonic fibroblasts, 3' end of transactivator was the best site for the GFP expression.

First report of tetracycline-resistant Aeromonas veronii infection in Amur catfish (Silurus asotus) cultured in Korea

  • Mass mortality in commercially cultured Amur catfish (Silurus asotus), showing symptoms of dermal ulcerations, occurred on a private farm in Mar 2019 in Korea. β-hemolytic bacteria were isolated from the ulcers and kidneys of the fish and identified as Aeromonas veronii. The isolate was resistant to tetracycline and possessed cytotoxic heat-labile enterotoxin (aerolysin/hemolysin). We investigated the genetic determinants associated with tetracycline resistance, and the isolate has been confirmed to simultaneously possess tetA and tetE genes. This is the first report on the occurrence of tetracycline-resistant A. veronii infection related to mass mortality in commercially cultured Amur catfish in Korea.

Molecular Characterization of Antibiotic Resistant Escherichia coli Strains Isolated from Tap and Spring Waters in a Coastal Region in Turkey

  • A hundred and seventeen antibiotic-resistant Escherichia coli strains were isolated from public tap and spring waters which were polluted by fecal coliforms. There were no significant differences between two water sources as to the coliform pollution level (p> 0.05). All E. coli isolates were detected to be resistant to one or more antibiotics tested. Nearly 42% of the isolates showed multiresistant phenotype. Three (2.5%) of these isolates contained class 1 integron. Sequencing analysis of variable regions of the class 1 integrons showed two gene cassette arrays, dfr1-aadA1 and dhfrA17-aadA5. Resistance to ampicillin, tetracycline or trimethoprim-sulfamethoxazole was transferable according to the results of conjugation experiments. The rate of tetracycline resistance was 15%. tet(A)-mediated tetracycline resistance was widespread among tetracycline-resistant E. coli isolates. Genotyping by BOX-polymerase chain reaction (BOX-PCR) showed that some of the strains were epidemiologically related. This is the first report on the prevalence and characterization of class 1 integron-containing E. coli isolates of environmental origin in Turkey.

Prevalence and molecular characterization of tetracycline-resistant Enterococcus isolates from livestock (가축에서 테트라사이클린 내성 장구균 조사 및 분자생물학적 특성규명)

  • BIn the present study, Enterococcus isolates originating from livestock were studied for the phenotypic and genotypic assessment of tetracycline resistance. A total of 74 isolates encompassing the species Enterococcus faecalis (n=12) and E. faecium (n=62) displayed phenotypic resistance to tetracycline. Tetracycline resistance gene [tet (M), 1,886bp] were sequenced by dye terminator cycle sequencing method and compared with tet (M) sequences available from the GenBank database. Sequencing analysis of PCR amplicons showed high homology to the reference strains ranging 97.2~100%. The tet (M) genes were divided into three major subgroups according to phylogenetic analysis. The genetic information obtained from this study could be useful for the molecular study of enterococci.

The Effects of Additional Tetracycline Pleurodesis during Thoracoscopic Procedures for Treating Primary Spontaneous Pneumothorax (일차성 자연 기흉의 흉강경 수술 시 추가로 시행한 Textracycline 흉막 유착술의 효용)

  • Background: This study was performed to evaluate the safety and efficacy of performing additional tetracycline pleurodesis during the thoracoscopic treatment of primary spontaneous pneumothorax. Material and Method: Between March 2004 and December 2007, 91 cases of primary spontaneous pneumothorax were treated by video-assisted thoracoscopic surgery. The thoracoscopic procedures included resection of the blebs and mechanical pleurodesis by scrubbing the parietal pleura. For 27 cases (Tetracycline group, group I), 20 mg/kg tetracycline was instilled into the pleural space through a trocar before closing the chest. The control group (group II) consisted of 64 cases of primary spontaneous pneumothorax for which the same thoracoscopic procedures alone were performed during the same study period. Result: There was no significant difference between the two groups in terms of the demographic data, the operative findings and the operation time. The percentage of cases that needed intravenous analgesics and the duration of intravenous analgesics were comparable in both groups. There was no significant difference in the duration of air leaks and complications between the two groups. The patients treated with tetracycline pleurodesis had a longer period of postoperative chest drainage (4.2 days vs 3.5 days, respectively, p=0.03) and hospitalization (5.0 days vs 4.0 days, respectively, p=0.006). During the follow up period, the ipsilateral recurrence rate was much lower for the patients who were treated with tetracycline pleurodesis (0% vs 10.9%, respectively, p=0.099), and freedom from recurrence tended to be more favorable for group I (p=0.077), although this was not statistically significant. Conclusion: Additional tetracycline pleurodesis during thoracoscopic treatment for primary spontaneous pneumothorax caused prolongation of chest drainage and a prolonged hospital stay. However, further investigations are needed because tetracycline pleurodesis can be performed safely without serious complications and it showed a distinct tendency to reduce the rate of recurrence.

The Effect of Pleurodesis with Doxycycline in the Rabbit (토끼에서 Doxycycline을 이용한 흉막유착 효과)

  • Background: The intrapleural instillation of tetracycline for pleural sclerosis had been most commonly used in patients with symptomatic malignant pleural effusion or recurrent pneumothorax for a long time. Unfortunately, at a time of expanding use of this agent, the production of injectable tetracycline hydrochloride used for pleurodesis was discontinued by its sole manufacturer in mid-1991 because the manufacturer was unable to meet US Food and Drug Administration purity standards. So we performed a preliminary study of doxycycline, as a alternative pleural sclerosant on rabbit pleura and compared its efficacy with that of tetracycline. Method: Eighteen New Zealand white rabbits weighing 2 to 3kg(mean 2.6kg) were devided into three groups. In each groups, one tetracycline(20 mg/ml/kg) and two doxycycline solutions(7 mg/ml/kg and 20 mg/ml/kg) instillated into the right pleural space through an 18-gauge angiocath with care to prevent pneumothorax. All rabbits were sacrificed after 28 days. Results: 1) In the group of tetracycline 20 mg/ml/kg(six rabbits), five rabbits showed partial pleural symphysis with several fibrous bands, and one rabbit died on 22th day. 2) In the group of doxycycline 7 mg/ml/kg(six rabbits), three rabbits showed partial pleural symphysis and the other three rabbits showed complete pleural symphysis without necrosis of underlying parenchymal lung tissue. 3) In the group of doxycycline 20 mg/ml/kg(six rabbits), two rabbits showed complete pleural symphysis without lung necrosis, another two rabbits showed complete pleural symphysis with lung necrosis, and the other two rabbits died on 4th and 13th day, respectively. Conclusion: We concluded that doxycycline is a highly effective sclerosing agent having stronger pleurodesis effect with that of tetracycline by dose base and its optimal dosage was considered as 7 mg/ml/kg with minimal complications.
