Di- and Sesqui-Terpenoids Isolated from the Pods of Sindora sumatrana and Their Potential to Inhibit Lipopolysaccharide-Induced Nitric Oxide Production

  • Activity-guided fractionation of the n-hexane and ${CHCl_3}-soluble$ fractions of Sindora sumatrana using a bioassay based on the inhibition of lipopolysaccharide (LPS)-induced nitric oxide (NO) production by inducible nitric oxide synthase (iNOS) in murine macrophage RAW 264.7 cells led to the isolation of the known compound, $(+)-7{\beta}-acetoxy-15,16-epoxy-3$, 13(16), 14-clero-datriene-18-oic acid (2) as an active constituent. In addition, a new trans-clerodane diterpenoid, (+)-2-oxokolavenic acid (1), together with six known compounds, (+)-3, 13-clerodadiene-16,15-olide-18-oic acid (3), $(+)-7{\beta}-acetoxy-3$,13-clerodadiene-16,15-olide-18-oic acid (4), $(+)-7{\beta}-acetoxy-16-hydroxy-3$,13-clerodadiene-16, 15-olide-18-oic acid (5), ${\beta}-caryophyllene$ oxide (6), $clovane-2{\beta},9{\beta}-diol (7),{\;}and{\;}caryolane-1,9{\beta}-diol$ (8) were isolated and found to be inactive. The structure of compound 1 was determined using physical and spectroscopic methods such as 1D and 2D-NMR experiments. The known compounds 2-8 were identified by the spectroscopic data and by comparison with the published values. Of eight isolates (1-8), only compound 2 exhibited an iNOS inhibitory activity with $IC_{50}$/ value of $51.6{\;}\mu\textrm{m}M$.

Terpene-Strengthened Ginkgo biloba Extract as a Platelet-Activating Factor Antagonist

  • Since platelet-activating factor (PAF) is involved in inflammation, allergic response and anaphylactic shock, PAF receptor antagonists may have potential for controlling these disease conditions. The extract of the leaves of Ginkgo biloba having a higher content of terpenoids (12%) with flavonoids (24%) (YY1224) was prepared in order to obtain the increasing PAF antagonistic activity. As expected, YY1224 showed a higher PAF antagonistic binding affinity ($IC_{50}\;=\;0.09\;{\mu}g/ml$) using $[^3H]PAF$ and rabbit platelets as ligand and receptor source, compared with an $IC_{50}$ of $>\;100\;{\mu}g/ml$ by Egb 761, a standardized extract. YY1224 also showed a higher inhibitory activity against PAF-induced platelet aggregation and NO production from lipopolysaccharide-treated RAW 264.7 cells. In addition, it protected PAF-induced death in mice by oral administration at 15 mg/kg. All these results suggest that YY1224 may show favorable effects on PAF-related disorders.

Emission of Biogenic Volatile Organic Compounds from Trees along Streets and in Urban Parks in Tokyo, Japan

  • Ozone concentration in Tokyo Metropolitan area is one of the most serious issues of the local air quality. Tropospheric ozone is formed by radical reaction including volatile organic compound (VOC) and nitrogen oxides ($NO_x$). Reduction of the emission of reactive VOC is a key to reducing ozone concentrations. VOC is emitted from anthropogenic sources and also from vegetation (biogenic VOC or BVOC). BVOC also forms ozone through $NO_x$ and radical reactions. Especially, in urban area, the BVOC is emitted into the atmosphere with high $NO_x$ concentration. Therefore, trees bordering streets and green spaces in urban area may contribute to tropospheric ozone. On the other hand, not all trees emit BVOC which will produce ozone locally. In this study, BVOC emissions have been investigated (terpenoids: isoprene, monoterpenes, sesquiterpenes) for 29 tree species. Eleven in the 29 species were tree species that did not emit BVOCs. Three in 12 cultivars for future planting (25 %) were found to emit no terpenoid BVOCs. Eight in 17 commonly planted trees (47%) were found to emit no terpenoid BVOC. Lower-emitting species have many advantages for urban planting. Therefore, further investigation is required to find the species which do not emit terpenoid BVOC. Emission of reactive BVOC should be added into guideline for the urban planting to prevent the creation of sources of ozone. It is desirable that species with no reactive BVOC emission are planted along urban streets and green areas in urban areas, such as Tokyo.

참두릅 잎의 Protein Tyrosine Phosphatase 1B와 α-Glucosidase 저해 활성 (Inhibitory Activity of Aralia elata Leaves on Protein Tyrosine Phosphatase 1B and α-Glucosidase)

  • Anti-diabetic potential of the leaves of A. elata through the inhibitory activity on PTP1B and ${\alpha}$-glucosidase has not been reported. In this study, the EtOAc fraction of methanolic extract from the leaves of A. elata showed potent inhibitory activity against the PTP1B and ${\alpha}$-glucosidase with $IC_{50}$ value of $96.29{\pm}0.3$ and $264.71{\pm}14.87{\mu}g/mL$, respectively. Three known triterpenoids, oleanolic acid, oleanolic acid-28-O-${\beta}$-D-glucopyranoside and oleanolic acid-3-O-${\beta}$-D-glucopyranoside were isolated from the most active EtOAc fraction. We determined the chemical structure of these triterpenoids through comparisons of published nuclear magnetic resonance (NMR) spectroscopic data. Furthermore, we screened these triterpenoids for their ability to inhibit PTP1B and ${\alpha}$-glucosidase over a range of concentrations ($12.5-50{\mu}M$). All three terpenoids significantly inhibited PTP1B in a concentration dependent manner and oleanolic acid effectively inhibited ${\alpha}$-glucosidase. In addition, these compounds revealed potent inhibitory activity with negative binding energies toward PTP1B, showing high affinity and tight binding capacity in the molecular docking studies. Therefore, the results of the present study clearly demonstrate that A. elata leaves and its triterpenoid constituents might be beneficial in the prevention or treatment of diabetic disease.

들깨(Perilla frutescens)와 쑥(Artemisia asiatics)잎으로부터 휘발성 타감 작용 성분의 분리 (Isolation of Volatile Allelochemicals from Leaves of Perilla frutescens and Artemisia asiatica)

  • 본 연구는 들깨와 쑥 잎의 휘발성 타감작용을 확인하고, 그 성분을 동정하려 한 것이다. 휘발성분을 벼, 무우, 녹두, 상치를 대상으로 생물검정하여 휘발성 타감작용을 확인했다. 휘발성분을 headspace cold trapping-Tenax GC 흡착으로 분리하여 들깻잎으로부터 terpenoid 4종, aldehyde 3종, alcohol 3종, 탄화수소류 3종, 기타 성분 1종을 동정하였으며, 쑥으로부터는 terpenoid 9종, aldehyde 3종, alcohol 1종, 기타 성분 1종을 검출하였다. 한편, 수증기 증류-추출로 포집한 경우에는, 들깻잎으로부터 terpenoid 2종, alcohol 2종의 동정되었으며, 쑥 잎으로부터는 terpenoid 6종, alcohol 1종, 탄화수소류 1종, 기타 성분 2종이 검출되었다. 수증기 증류-추출로부터 나누어진 각 분획을 무우를 대상으로 생물검정한 결과 중성 분획이 가장 타감작용 활성이 컸다. 휘발성 타감작용 성분을 분리하기 위해 이를 GC-MS로 분석한 결과, 들깨로부터 terpenoid 7종과 alcohol 2종 등 9종을 동정하였고, 쑥으로부터 terpenoid 15종과 alcohol 3종, aldehyde 1종, 탄화수소류 2종, 기타 성분 3종 등 24종을 동정하였다.

붉은 덕다리버섯 발효 대조(大棗)의 예쁜꼬마선충 수명연장효과 (Lifespan Extension of Fermented Zizyphus jujuba Fruits in Caenorhabditis elegans)

  • Objectives : Zizyphus jujuba fruits(ZJF), a traditional Korean medicine has various biological activities such as anti-inflammatory, anti-oxidative and neuro-protective effects. However, it is still unclear whether ZJF has any biological effect on anti-aging. In this study, we examined the effect of ZJF on lifespan and thermal stress in C. elegans. Methods and Results : ZJF water extracts were fermented for 7 days(F7-ZJF) and 14 days(F14-ZJF) by Laetiporus sulphureus to increase secondary metabolites such as aglycone of flavonoids and terpenoids. In the lifespan assay, ZJF water extracts and fermented ZJF were treated on the agar medium plate with age synchronized egg stage of C. elegans. Treatment of F7-ZJF-$200{\mu}g/mL$ with OP-50 E. coli and F14-ZJF-$200{\mu}g/mL$ with OP-50 E. coli significantly increased life span of C. elegans(N2) at thermal stress condition of $25^{\circ}C$. Moreover mRNA levels of lifespan associated HSP 16.1, HSP 70, and HSF-1 were increased at thermal stress condition of $25^{\circ}C$. However, in the equilibration temperature of $20^{\circ}C$ after stress condition of $35^{\circ}C$ for 2 hr, F-14-ZJF-$200{\mu}g/mL$ treatment decreased the levels of heat shock protein in hsp16.2/GFP C. elegans. Conclusions : Our study indicates that prolong role of fermented-ZJF in C. elegans is mediated by control HSPs production.

  • Platelet aggregation is a complex phenomenon that probably involves several intracellular biochemical pathways. When activated, platelets change shape, aggregate and release the contents of their intracellular granules. The interactions between platelets and blood vessel walls are important in the development of thrombosis and cardiovascular diseases. When blood vessels are damaged, platelet aggregation occurs rapidly to form haemostatic Plugs or arterial thrombi at the sites of vessel injury or in regions where blood flow is disturbed. These thrombi are the source of thromboembolic complications of atherosclerosis, heart attacks, stroke, and peripheral vascular disease. Therefore, the inhibition of platelet function represents a promising approach for the prevention of thrombosis. Plants constitute a rich source of bioactive chemicals such as phenolics, terpenoids and alkaloids. Plant extracts may be an alternative to currently used medicinal source because they constitute a rich source of bioactive chemicals. This study was performed to investigate the antiplatelet activity of extract of Tabebuia impetiginosa Martius ex DC (Taheebo) and find out which fractions to this activity in rabbit platelet. Taheebo was methanol extracted and solvent fractionated in to five fractions (hexane, chloroform, ethylacetate, butanol and water). And each fractions were investigated inhibitory effects on platelet aggregation induced by various agonists using washed rabbit platelets in vitro.

곤달비 잎과 뿌리의 생물 활성 (Biological Effects of the Leaves and Roots of Ligularia stenocephala)

  • 국내에서 식품으로 사용되고 있는 국화과 식물인 곤달비(Ligularia stenocephala) 잎과 뿌리의 생물활성을 과학적으로 검증하기 위하여 각 추출물에 대하여 항산화 활성 및 세포독성을 in vitro에서, 간보호 효과, 알코올 해독작용 및 기억증진 효능 등을 in vivo에서 평가하였으며 잎의 독특한 향기성분을 GC-MS로 분석하였다. 지질과산화 억제효과는 잎(20.4% 억제율)이 뿌리 보다 좋았으며, 유해 라디칼의 일종인 superoxide anion은 뿌리에서 생성 억제효과가 더 좋았고, DPPH 소거활성은 잎과 뿌리 모두 77~79%로 매우 뛰어났다. 사염화탄소로 유발된 급성 간독성 개선효과를 AST와 ALT 효소활성도를 지표로 확인한 결과, 잎의 ALT 억제활성이 대조군에 비해 약 78% 정도 감소하였으며 알코올을 투여한 mouse의 혈중 알코올 농도는 잎추출물 투여시 약 60% 가량 유의성 있게 감소되어 효과를 인정할 수 있었다. 세포독성은 뿌리에서 비교적 강하게 나타났는데, 흑색종의 경우, $IC_{50}=40.14mg/ml$이었으며, 잎의 세포독성은 비교적 약하였다. 기억증진 효과를 동물모델을 이용한 수동회피시험법으로 평가한 결과, 잎과 뿌리 모두 scopolamine에 의해 유도된 기억력 감소를 80% 이상 향상시킨 것으로 조사되었다. 곤달비 잎의 n-헥산 추출물을 GC-MS로 분석한 결과, 독특한 향기는 주로 terpene 화합물에서 유래되는 것으로 추정되었다.

Phytochemical Screening and Biological Studies of Boerhavia Diffusa Linn

  • Hexane, ethyl acetate and methanol extracts of whole plant of Boerhavia diffusa were screened for phytochemical and biological activities. Qualitative phytochemical screening via colorimetric method and the quantitative estimation of phenolic and flavonoid content were performed. Antioxidant assay using DPPH scavenging method was studied. Antimicrobial screening of plant extracts was done by cup diffusion technique. Cytotoxic activity of B. diffusa was studied by brine shrimp bioassay and anthelminthic activity was evaluated in vitro in Pheretima posthuma. This study revealed B. diffusa as a source of various phyto-constituents such as alkaloids, glycosides, saponins, tannins, carbohydrates, cardiac glycosides, flavonoids and terpenoids. Quantitative estimation of total phenol was found to be maximum in BEE i.e. $29.73{\pm}0.88$, BME $19.8{\pm}2.02$ and in BHE $9.15{\pm}0.304mgGAE/g$. Similarly, the total flavonoid content was found to be $17.44{\pm}0.75$ in BEE, $14.43{\pm}0.23$ in BHE and 3.678 mg QE/g in BME. Ethyl acetate extract showed its antibacterial activity against all tested pathogens except Escherichia coli whereas Staphylococcus aureus and Salmonella Typhi were resistant to methanol and hexane extract. The zone of inhibition (ZOI) of ethyl acetate extract against S. Typhi and B. cereus was found to be 18 mm and 14 mm respectively. The MIC value of BEE in S. Typhi was $3.125{\mu}g/ml$ and in B. cereus was $12.5{\mu}g/ml$. The preliminary screening of anticancer property of B. diffusa i.e. BSLT in methanol was found to be $165.19{\mu}g/ml$. B. diffusa was also found to contain anthelmintic property. The study helped in further exploration of medicinal properties of B. diffusa by phytochemical screening and biological activities paving the path for study and investigation in this plant.

추출 방법에 따른 여정실의 최종당화산물 생성 저해 및 라디칼 소거 활성 (Anti-Glyaction and Radical Scavenging Activities of Ligustri Fructus by Extraction Method)

  • Objectives : Ligustri Fructus has been used since ancient times as a medicinal usages in folk medicines against antitumor purpose. Many biological active constituents have been identified from this biomass such as several terpenoids and lignans. In current study, the properties of antioxidant and anti-diabetic complications using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)($ABTS^+$) radicals scavenging, and advanced glycation end products (AGEs) inhibition assays were evaluated by different extraction methods of Ligustri Fructus. Methods : In present continuing research for development of bioactive natural ingredients, antioxidant and AGEs formation inhibitory capacities of Ligustri Fructus extracts using different organic solvents were prepared and the biological potentials were investigated using in vitro bioassays. Antioxidant properties were evaluated employing radical scavenging assays using DPPH and $ABTS^+$ radicals. In addition, the anti-diabetic complications effects of Ligustri Fructus extracts were tested via AGEs formation inhibitory assay. The total phenolic contents were determined using a spectrophotometric method. Results : All the tested extracts exhibited dose-dependent radical scavenging and AGEs formation inhibitory activities. Among the tested samples, hot water extract of Ligustri Fructus was showed the most potent activity with $IC_{50}$ value of $494.8{\pm}6.7{\mu}g/m{\ell}$ against DPPH radical scavenging assay. Also, $ABTS^+$ radical scavenging activity of hot water extract was higher than those of other extracts. In addition, AGEs formation inhibitory effects of each extacts and total phenolic contents were evaluated. Conclusions : These results suggested that Ligustri Fructus can be considered as a new effective source of natural antioxidant and anti-diabetic complications resources.
