Purification and Characteristics of Chitosanase from Bacillus sp. HW-002

  • Chitosanase from Bacillus sp. HW-002 was purified with CM-cellulose column chromatography, and HPLC with DEAE- TSK gel and YMC-pack Diol 120. The purified enzyme appeared as a single band on SDS-polyacrylamide gel. The molecular weight of the enzyme was estimated to be about 46 kDa on SDS-polyacrylamide gel, and was estimated to be about 23 kDa by GFC. The optimal pH of chitosanolytic activity was about pH 5.5-6.0, and the purified enzyme was most stable at pH 5.0. The optimal temperature of chitosanolytic activity was $65^{\circ}C$ and the enzyme was stable at $45^{\circ}C$ for 1 h. Chitosan was the most favorable substrate among various $\beta$-glucan. UVmax of the purified enzyme was 195 nmand was not noted around 280 nm. The main product of enzyme reaction with chitosan was chitobiose.

  • Lithium insertion has been studied in a number of vanadium oxides with special regard to their application as the active materials in rechargeable lithium cells. Very high stoichiometric energy densities for lithium insertion are found for several of these materials. Some vanadium oxides, e.g. V$_2$ $O_{5}$ and V$_{6}$ $O_{13}$, are now being used in commercially developed rechargeable Li batteries. Another material which is receiving remarkable attention for this kind of cells is LiV$_3$ $O_{8}$. In variety of ternary lithium-vanadium oxides, the lithium content can be varied between certain limits without major changes in the vanadium oxygen lattice. In our worts, the oxides which do net form these thermodynamically stable bronzes can still accommodate large amounts of lithium at ambient temperature, forming kinetically stable insertion compounds. These compounds owe their existence to the whereas lithium is easily introduced into these open structures. The oxides investigated are rather poor electronic conductors; the conductivity decrease with increase in the lithium content. Improvements in the electrode fabrication technique are needed to alleviate this Problem.oblem.

  • We conducted a test of a direct burning of crude Jatropha oil (CJO) in a commercial boiler system. The fuel, crude Jatropha oil is not biodiesel which comes from transeterification process of bio oil, but it is pure plant oil. The higher heating value (HHV) of the CJO is 39.3 MJ/kg (9,380 kcal/kg) and is higher than that of a commercial heating oil, 37.9 MJ/kg. The kinematic viscosity of CJO is 36.2 mm2/s at $40^{\circ}C$ and 8.0 mm2/s at $100^{\circ}C$. The burner used in the test is a commercial burner for a commercial heatingoil and its capacity is 140 kW (120,000 kcal/h). We did a preliminary test whether the combustion is stable or not. The preliminary test was a kind of open air combustion test using the commercial burner with crude Jatropha oil. We found that the combustion can be stable if the crude Jatrophaoil temperature is higher than $90^{\circ}C$. We measured the flue gas concentration by using a gas analyzer. The NOx concentration is $80{\sim}100\;ppm$ and CO concentration is nearly 0 ppm at flue gas O2 concentration of 3.0 and 4.5%.

Exhaust and Combustion Characteristics of Premixed Swirl Burner for Steam Reforming System (선회류 예혼합버너를 적용한 개질기용 연소시스템의 배기 및 연소특성)

  • The reformer system is a method for hydrogen production from hydrocarbon fuels such as natural gas under high temperature environment($about{\sim}1,000^{\circ}C$). The premixed swirl burner with mixing swirler and combustion swirler designed to deliver fuel cell electric output from 0.5 kW to 1.5 kW. Premixed swirl burner experiments using natural gas and mixture of natural gas and anode off gas were carried out to analyse flame patterns and stability by equivalence ratio respectively. The results show that the stable swirl flame can be established for all cases of fuels type using the premixed swirl burner. The swirl flame had a wide stability region and it showed very low CO(50 ppm) and $NO_x$(20 ppm) emission at different fuel type and various equivalence ratio conditions. The operating range of premixed swirl burner for stable swirl flame is found to exist between equivalence ratio of 0.70 to 0.90 for turn down ratio of 3:1.

  • A lab-scale Reverse Osmosis(RO) membrane reactor was installed to investigate the membrane permeability, characteristics of membrane fouling at each conditions, and performance of elimination at different trans-membrane pressure(TMP) in the liquid fertilizer accumulated system. Experimental setup was divided to three different TMP conditions. As a result of experiment, permeability of RO membrane was proportional to the increase of TMP and temperature. After experiment was completed, two types chemical cleaning(remove the organic foulant and inorganic foulant) was done, and recover rate of permeability was each 99.8, 99.7 and 99.7%, respectively. From this experimental data, membrane fouling could be determined that the most of it was recoverable in this system, and major reason of fouling was concentration polarization. Elimination rate of solute substance in the liquid fertilizer indicated very stable(above 99%), except ammonia nitrogen, and the most stable elimination rate was investigated at the highest TMP condition (Run 3).

Characterization of Bacteriocin Production by Lactococcus lactis LAB3113 Isolated from Kimchi

  • A lactic acid bacterium LAB3113, isolated from traditionally fermented Kimchi was found to produce bacteriocin whose activity was very specific toward lactobacilli and not effective against any other bacteria. Lactobacilli affected by the inhibitory substance included Lactobacillus delbrueckii-lactis, L. johnsonii, L. gsseri, and L. curvatus. Based upon biochemical and physiological characteristics, LAB3113 was classified as Lactococcus lactis, and its bacteriocin was named as lactococcin K3113. Lactococcus lactis. LAB3113 produced bacteriocin at th early stage of growth and the concentration of the bacteriocin did not decrease even after alt stationalry phase. Optimal temperature of bacteriocin production was $25^{\circ}C$ at the initial pH 7.0. Partially purified lactococcin K3113 was completely inactivated by protease, but not affected by lipase, lysozyme and RNase. The bacteriocin was very heat-stable even after autoclaving for 20 min. It was also stable in pH changes, an was not affected by th presence of solvents. lacotococcin K3113 appeared to act in bactericidal mode against L. delbrueckii-lactis ATCC4797. Molecular weight of lactococcin K3113 was calibrated as 10,500 dal by SDS-PAGE an activity staining. Lactococcus lactis LAB3113 had four residential plasmids of 3.7kb, 11.2kb, 15.5kb, and 48kh in molecular sizes. Plasmid profile analysis of mutant strain revealed that 15.5 kb plasmid was re-sponsible for the production of lactococcin K3113 and its immunity to the bacteriocin.

  • Continuous solid circulation test at high temperature and high pressure conditions and batch type reduction-oxidation tests were performed to check feasibility of a 0.5 MWth chemical looping combustion system. Pressure drop profiles were maintained stable during continuous solid circulation up to 16 hours. Therefore, we could conclude that the solid circulation between an air reactor and a fuel reactor could be smooth and stable. The measured fuel conversion and $CO_2$ selectivity were high enough even at high capacity and even after cyclic tests. Therefore, we could expect high reactivity of oxygen carrier at real operation condition.

  • The resistive memory switching characteristics of resistive random access memory (ReRAM) using the amorphous GeSe thin film have been demonstrated at Al/Ti/GeSe/$n^+$ poly Si structure. This ReRAM indicated bipolar resistive memory switching characteristics. The generation and the recombination of chalcogen cations and anions were suitable to explain the bipolar switching operation. Space charge limited current (SCLC) model and Poole-Frenkel emission is applied to explain the formation of conductive filament in the amorphous GeSe thin film. The results showed characteristics of stable switching and excellent reliability. Through the annealing condition of $400^{\circ}C$, the possibility of low temperature process was established. Very low operation current level (set current: ~ ${\mu}A$, reset current: ~ nA) was showed the possibility of low power consumption. Particularly, $n^+$ poly Si based GeSe ReRAM could be applied directly to thin film transistor (TFT).

  • The characteristics of diesel and biodiesel are similar like as cetane number and auto-ignition temperature. High cetane number of diesel and BD could make possible to compression ignition. but BD showed different atomization from diesel due to component like density, viscosity and iodine value etc. Because of this, the biodiesel requires validation. This study using diesel and BD20 investigated effect to durability injector. Durability test were used common rail and bosch solenoid type 5-hole injector. Total test was 672hr but actual running time was 200hr. Spray experiments for spray characteristics were carried out using constant volume combustion chamber. Spray characteristics of diesel and BD showed different result up to durability test time. After 100hr, diesel showed spray shapes were stable but BD was not. After 200hr, difference of diesel and BD spray shapes were grow serious.

  • This paper describes gas turbine combustion characteristics of synthetic natural gas which contains a small amount hydrogen content. By conducting ambient pressure high temperature combustion test at gas turbine relevant combustor geometry, the combustion characteristics such as combustion instability, NOx and CO emission, temperatures at turbine inlet, nozzle and dump plane, and flame structure from high speed OH chemiluminescence images were investigated when changing hydrogen content from zero to 5%. From the results, qualitative and quantitative relationships are derived between key aspects of combustion performance, notably NOx/CO emission and combustion instability. Natural gas containing hydrogen up to 5% does not show significant difference in view of all combustion characteristics except combustion instability. Only up to 1% hydrogen addition could not change the pressure fluctuation and phase gas between fluctuations of pressure and heat release. From the results, it can be concluded that synthetic national gas which contains 1% of hydrogen can be guaranteed for the stable and reliable operation of natural gas firing gas turbine.
