As a scaffold, SLX4/FANCP interacts with multiple proteins involved in genome integrity. Although not having recognizable catalytic domains, SLX4 participates in diverse genome maintenance pathways by delivering nucleases where they are needed, and promoting their cooperative execution to prevent genomic instabilities. Physiological importance of SLX4 is emphasized by the identification of causative mutations of SLX4 genes in patients diagnosed with Fanconi anemia (FA), a rare recessive genetic disorder characterized by genomic instability and predisposition to cancers. Recent progress in understanding functional roles of SLX4 has greatly expanded our knowledge in the repair of DNA interstrand crosslinks (ICLs), Holliday junction (HJ) resolution, telomere homeostasis and regulation of DNA damage response induced by replication stress. Here, these diverse functions of SLX4 are reviewed in detail.

To be economically profitable, the poultry industry demands an increase in stocking density, which could adversely affect chicken welfare. The current study was performed to investigate the effect of stocking density on stress-related, heat shock protein genes (HSP70 and HSP90), 3-hydroxyl-3-methyl-glutaryl coenzyme A reductase (HMGCR) gene and telomere length in broiler chickens. Seven-day-old broiler chickens were housed at High (0.0578 $m^2$/bird), Standard (0.077 $m^2$/bird) and Low (0.116 $m^2$/bird) stocking densities with 8 replicates each until 35 d of age. The growth performance, such as body weight gain and average daily feed intake, was found to be significantly (p<0.05) higher in the Low density group, but these parameters did not show any difference between the High and Standard groups. Other growth performance, such as feed conversion ratio and final feed intake, showed no difference among the treated groups. The expression levels of HSP70 and HMGCR were found to be elevated with the increase of stocking density. The expression level of these genes was significantly (p<0.05) higher in the High density stocked group compared with the other groups, whereas the expression levels were not significantly different between the Low and Standard groups. The expression levels of HSP90 did not show any significant changes among the treated groups. The telomeric length of the birds housed in High density was reduced significantly (p<0.05) when compared to that of the birds in Low density. These results clearly indicate that birds stocked at high density show physiological adaptive changes indicative of stress at gene transcriptional and telomere levels.
Telomeres locate at the end of chromosomes and consist of a tandem repeat sequence of $(TIAGGG)^{n}$ and associated proteins. Telomerase is a ribonucleoprotein which act as a template for the synthesis of telomeric DNA. Telomeres are essential for chromosome stability and are related with cell senescence, apoptosis and cancer. This study was carried out to analyze the amount of telomeres and telomerase activity of chicken cells during embryonic and developmental stages. The whole embryos and prenatal tissues such as brain, heart, liver, kidney and testis at different developmental stages were obtained from Korean Native Chicken. The amount of telomeres on embryonic cells was analyzed by quantitative fluorescence in situ hybridization (Q-FISH) techniques using the chicken telomeric DNA probe. Telomerase activity was measured by telomeric repeat amplification protocol (TRAP) assay. Results indicated that the amounts of telomeric DNA on the most embryonic cells were gradually decreased during ontogenesis. Furthermore, the quantity of telomeres was quite different among embryonic tissues according to developmental origin. The relative amount of telomeres has more in regenerative cells such as embryonic disc and testicular cells than in non-regenerative cells such as liver, brain, heart and kidney cells. Cordyceps militaris is a medicinal fungus, which has been used for patient suffering from cancer in Oriental medicine. It was reported previously that C. militaris extracts are capable of inhibiting tumor growth, however, the anti-poliferative effects of human cancer cells have not been poorly understood. In this study, to elucidate the growth inhibitory mechanisms of human cancer cells by treatment of aqueous extract of C. militaris (AECM) we investigated the anti-proliferative effects of AECM in human leukemia U937 cell line. AECM treatment inhibited the growth of U937 cells and induced the apoptotic cell death in a concentration-dependent manner, which was associated with morphological changes. We observed the up-regulation of cyclin-dependent kinase (Cdk) inhibitor p21(WAF1/CIP1) by p53-independent manner and activation of caspase-3 in AECM-treated U937 cells, however, the activity of caspase-9 was remained unchanged. Additionally, AECM treatment caused a dose-dependent inhibition of the expression of telomere regulatory gene products such as human telomere reverse transcriptase (hTERT) and telomerase-associated protein-1 (TEP-1). Taken together, these findings suggest that AECM-induced inhibition of human leukemic cell proliferation is associated with the induction of apoptotic cell death via modulation of several major growth regulatory gene products, and C. militaris may have therapeutic potential in human lung cancer.
경북지방(慶北地方)의 소나무와 곰솔에 대한 Giemsa C-분염(分染)에 의한 유전변이(遺傳變異)를 조사(調査)한 바 다음과 같다. 1. Giemsa C-분염(分染)에 의한 핵형분석(核型分析)에서 소나무와 곰솔의 체세포(體細胞) 염색체수(染色體數)는 2n=24이고 2. 소나무는 arm ratio가 비슷한 M형(型)으로 개체간(個體間)에는 큰 차이가 없었으나 이차협착(二次狹窄)의 수(數)와 위치(位置)가 개체간(個體間)에 다소(多少) 차이(差異)가 있었으며 3. 곰솔은 염색체(染色體) 길이와 arm ratio가 M형(型)으로 소나무와 비슷하고 이차협착(二次狹窄)의 수(數)와 위치가 비슷하여 개체간(個體間)의 변이(變異)는 거의 없었다. 4. 염색체(染色體) C-분염(分染)에 의한 유전변이(遺傳變異)에서 소나무와 곰솔은 12쌍(雙)의 염색체(染色體)에 동원체(動原體) 부분(部分)과 이차협착(二次狹窄) 부위(部位)에 band가 나타났고 5. 소나무는 3번, 4번, 7번 염색체(染色體)에서는 개체(個體)에 관계(關係)없이 모두 이차협착(二次狹窄)에 band가 나타나고 1번, 2번, 5번 염색체(染色體)의 이차협착(二次狹窄)의 band는 개체간(個體間)에 변이(變異)가 있었다. 8번, 9번, 10번, 11번 염색체(染色體)에서는 telomere에 band가 나타나고 개체간(個體間)에 변이(變異)가 있었으며 6. 곰솔은 2번, 3번, 5번, 7번 8번 염색체(染色體)의 이차협착(二次狹窄)에 band가 모두 나타나고 개체간(個體間)에 변이(變異)가 없었으며 telomere에는 band가 나타나지 않았다. 7. C-분염(分染)에 의한 변이(變異)는 소나무에서는 개체간(個體間)에 변이(變異)가 있으나 곰솔에서는 변이(變異)가 없었고 소나무는 4번 염색체(染色體)에서, 곰솔은 8번 염색체(染色體)에서 이차협착(二次狹窄)에 band가 나타나고 있다. 이러한 것이 소나무와 곰솔의 C-분염(分染)에 의한 차이점(差異點)이라고 생각된다.
본 연구에서는 국산종계 개발을 위하여 5계통의 토종닭 순계를 이용하여 $5{\times}5$ 이면교배조합 시험을 수행하고, 공시된 25계통에 대한 스트레스 반응 정도를 비교 분석하였다. 스트레스 반응정도 표지로 텔로미어 함량 및 감축율, 열 스트레스 단백질 유전자(HSPs)로 HSP-70, $HSP-90{\alpha}$, $HSP-90{\beta}$의 발현율 및 DNA 손상율을 이용하여 분석하였다. 텔로미어 함량은 양적 형광접합보인법을 이용하고, HSPs 발현율은 quantitative real-time PCR 기법으로, DNA 손상율은 Comet assay법을 적용하였다. 분석 결과, 교배조합 25계통 간 텔로미어 함량과 감축율, HSPs 유전자 발현율 및 DNA 손상율 모두에서 유의한 차이를 보였고, 더불어 계통 간 체중 및 생존율에도 유의한 차이가 나타났다. 텔로미어 감축율, HSPs 유전자 발현율 및 DNA 손상율 모든 분석 값에서 W 및 Y 계통과의 교잡구가 다른 교잡구에 비해 유의하게 낮은 값을 보였고, 반면 G계통의 순계구가 가장 높은 값을 나타내었다. 생존율과 스트레스 표지 값들 간의 상관계수를 추정한 결과, 텔로미어 함량과는 저도의 정(+)의 상관, HSP-70, $HSP-90{\alpha}$, $HSP-90{\beta}$ 및 DNA 손상율과는 중도 및 저도의 부(-)의 상관을 나타내었다. 또한 폐사 개체와 생존 개체 간 HSP-70, $HSP-90{\alpha}$ 및 $HSP-90{\beta}$의 발현율을 비교 분석한 결과, 폐사 개체의 HSPs 발현율이 생존 개체에 비해 유의하게 높은 발현율을 보였다. 이상 스트레스 표지 값의 분석 결과를 종합할 때, 교잡계들이 순계에 비해 스트레스 저항성이 높은 것으로 판단되고, 더불어 W, Y와 같은 경량종 계통의 교잡계들이 외적 스트레스에 강한 반면, G, H, F와 같은 고체중 계통의 교잡계들은 스트레스에 민감한 것으로 사료된다.
그동안 폐암의 발생에 관여하는 여러가지 유전자의 이상이 보고되어 왔으나 모든 종류의 폐암에서 보이는 비억제적 성장(unconkolle6 growth)을 대변할 수 있는 유전자적 종양 표시자(molecular tumor marker)는 보고된 바 없었다. 최근 유전자(chromosome)의 말단부에 위치한 특이한 구조인 telomere가 세포의 노화나 증식정도 (proliferative activity)에 따라 그 구조, 특히 TrAGGG 반복 구조가 변한다는 것이 알려진 바 있다. 따라서 telomere의 반복 구조를 만드는 효소인 telomerase의 활성도는 대상 세포의 증식 상황이나 증식 가능성을 나타내는 표시 자로서의 기능이 있다고 추정된다. 따라서 이는 종양의 진단은 물론 대상 환자의 향후 예후를 판 단하는 데도 좋은 지표로 이용될 수 있다. 12개의 다양한 비소세포폐암 세포주와 수술로 적출된 41명의 비세포계암 환자의 종양 조직에 대해 nROolymerase chain reaction)을 기초로 한 TRAP assay를 이용하여 telomeiase활성도를 측정하였다. 대조군으로는 동시에 적출된, 종양으로부터 가장 먼 위치의 정상 폐조직을 이용하였다. 12개 전 종양 세포주는 물론 대부분의 종양 조직(94%)에서, 성별, 연령, 세포 병리학적 subtype, 암기(stage) 등과 관련이 언이, telomerase의 활성도가 측정되었으며 정상이라고 간주된 조직에서는 5명으로 부터 채취한 조직에서만 미약하게 telomerase활성도가 관찰되었다. 예후에 연관지어 telomerase활성도의 의미는 telmerase활성도가 보이지 않는 종글이 극히 적었고 또한 추적 관찰 기간이 짧아 판정할 수 없었다. Telomere의 길이의 변화는의미를 판정키 어려웠다. 이상의 결과는 비소세포폐암에 있어 telomerase활성도의 변화가 암발생에 아주 중요한 과정일 수 있다는 추정을 가능하게 하며 telomerase활성도의 측정이 종양의 진 단에 유용하게 이용될 수 있음을 확인시켜 주는 것이라 사료된다.
The objectives of the present study were to initiate cloning of Korean native goat by somatic cell nuclear transfer (NT) and to examine whether unovulated (follicular) oocytes can support the same developmental ability of NT embryos as ovulated (oviductal) oocytes after hCG injection in stimulated cycles of the goat. The in vivo-matured and immature oocytes were collected from the oviducts and follicles of superovulated does, respectively, and the immature oocytes were maturated in vitro. Ear skin fibroblasts derived from a 3-yr-old female Korean native goat were used as the donors of nuclei or karyoplasts. Following fusion, activation and in vitro culture to a 2- to 4-cell stage, 49 in vitro-derived and 105 in vivo-derived embryos were transferred to 6 and 17 recipient does, respectively. One doe and three does of the respective groups were identified as pregnant by ultrasonography on day 30 after embryo transfer. However, only one doe, which had received in vivo-derived embryos, delivered a normal female kid of 1.9 kg on d 149. The cloned kid gained more weight than her age-matched females as much as 87% during the first 4 mo after birth (17.7 vs. $9.4{\pm}0.8$ kg) and reached puberty at 6-mo age a few months earlier than normal female does. The telomere length of the kid, which was similar to that of the donor fibroblast at 2-mo age, decreased 8% between 2- and 7-mo ages. Moreover, at 7-mo age, she had 21% shorter telomere than her age-matched goats. To our knowledge, this is the first case in which a cloned animal born with a normal weight exhibited accelerated growth and development. The unusually rapid growth and development of the cloned goat may have resulted from SCNT-associated epigenetic reprogramming involving telomere shortening.
