A transmission electron microscopic study was performed on the ultrastructure of the tegumental layer of GymophoLloines seoi (Digenea: Gymnophallidae) metacercarlae and adults. The metacercariae were obtained from naturally infected oysters, Crcssosoea gigas, and the adults from experimentally infected C3H mice. The tegumental layer generally revealed a small number of foldings, numerous small vacuoles, sines, and muscle bundles. Beneath the muscle layer, nuclei of the tegumental cells were located. There was little difference in the structure of the tegument between the metacercariae and adults. The oral sucker, having well-developed muscle layers, showed a similar structure to the ventral sucker except numerous foldlngs in the ventral sucker. The ventral pit was surrounded by a thin spcpiu layer, where a number of microtubules and mitochondria were seen. Around the ventral pit located well-developed circular and longitudinal muscles. The results showed that the ultrastructure of the tegumental layer of G. seoi metacercariae and adults revealed little difference from other trematodes in general. The ventral pit, a peculiar structure of this trematode, seems to function as a sphincter or an accessory adhesive organ.

Albendazole is a broad spectrum anthelmintics with high activity against intestinal nematodes as well as tremtodes and cestodes infections. But so far wormicidal effect of albendazole against Paragonimus westermani is uncertain. The present study undertaken to observe the morphological changes of Paragonimus westermani which was obtained from experimentally infected with metacercariae isolated from Cambaroides similis and 9 of them were given albendazole 2 doses of 25mg/kg daily for the 2 days and 4 days from the day of 20 weeks after infection. The Paragonimus werms were collected from the lung of the cats which were autopsied at 24 hours after treatment. The fine structures of the collected worms were studied by means of scanning and transmission electron microscopy. The findings of the observations were compared with those of untreated worms. The results are as follows : 1) In the scanning electron microscopic(SEM) observations, the worms obtained from 2 days treated group showed many blebs which were formed on the surface of damaged tegument in between two suckers. Protrusion of oral sucker typically observed and surrounding tegumental ridges were damaged. 2) The worms obtained from 4 days treated group showed marked contraction revealing half size of normal worms. 3) The transmission electronmicroscopic(TEM) observations indicated that large blebs of irregular shapes were formed by the destruction of tegumental ridge of tegumental syncytium and also showed degeneration of mitochondriae. 4) Destruction of tegumental syncytium formation of blebs were usually observed in the ventral side of the worms. 5) Differentiation of tegumental layer and basement layer muscular layer and parenchymal layer around two suckers of worms obtained at 4 days treated group was difficult. 6) Many host cells invaded and destroyed the degenerated tegument by the occurrence of blebs or the exposed muscular layer of the worm after exfoliation of the tegument.
An electron microscopic study was performed to know the basic tegumental structure of Cysticercus cellulosae. The scolex and bladder portions of cysticerci (human and porcine strains) were prepared for transmission and scanning electron microscopy by conventional procedures. In general, the tegument of C. cellulosae showed the basic ultrastructure of cestode tegument on electron micrographs. The teguments of both scolex and bladder portions consisted of such components, i.e., an outer vesicular layer with numerous microtriches and inner filroug layer. Below the fibrous layer, there were layers of muscle bundles and tegumental cells. The microtriches which covered the surface of cysticercus revealed two distinctly different shapes. The characteristic bladder-like, elongated pyramid shaped "tetrahedrial form" was observed on the surface of the scolex portion, whereas the elongated cylindrical "filamentous form" was distributed on the stirface of bladder portion. In spite of the difference of isolated host and location, the cysticerci showed tole same result. But dimensional variations of the tegument according to topography of the worm were observed. The possibility of application in making differential diagnosis from other larval cestodes and possible functions of this larval tegument were discussed.
An electron microscopic study was performed to observe the ultrastructure of the tegument of U seoulensis. The outer surface of the tegument was covered with a tnlaminated plasma membrane. The electron-dense cytoplasmic layer was $2.5{\;}\mu\textrm{m}$ wide In the anterior portion and contained numerous vacuoles, mitochondriae and granular materials in its matrix. The basement layer was 330 nm wade or so, and Its numerous extensions protruded into the cytoplasmic layer. The sensory organ was composed of a small vesicle of $1.7{\;}{\times}{\;}1.1{\;}\mu\textrm{m}$ in dimensions, which possessed a cilium of $1.2{\;}{\times}{\;}0.19{\;}\mu\textrm{m}$ in size. The pharynx was composed of the epithelial layer of about $0.5{\;}\mu\textrm{m}$ wide, well developed muscle layer and basement layer. The tegument of the oral sucker was composed of a cytoplasmic layer of $0.4-0.5{\;}\mu\textrm{m}$ width, a narrow basement layer, a well developed muscle layer and tegumental cells. Some kinds of secretory granules that seemed to be originated from the cells of the oral sucker were observed In the parenchymal portions of the adjacent cells. The tribocytic organ consisted of numerous microvilli. The microvilli were 5 nm wide and heptalaminated. Two types of secretory granules originated from the gland cells of tribocytic organ were observed In the tegument and parenchyme. The tegumental cells were irregular in shape, and of which nuclei were multifarious.
This study was undertaken to characterize the properties of a 100 kDa somatic antigen from Metagonimus yokogawai. Monoclonal antibodies (mAbs) were produced against this 100 kDa antigen, and their immunoreactivity was assessed by western blot analysis with patients' sera. The mAbs against the 100 kDa antigen commonly reacted with various kinds of trematode antigens, including intestinal (Gymnophalloides seoi), lung (Paragonimus westermani), and liver flukes (Clonorchis sinensis and Fasciola hepatica). However, this mAb showed no cross-reactions with other helminth parasites, including nematodes and cestodes. To determine the topographic distribution of the 100 kDa antigen in worm sections, indirect immunoperoxidase staining was performed. A strong positive reaction was observed in the tegumental and subtegumental layers of adult M. yokogawai and C. sinensis. The results showed that the 100 kDa somatic protein of M. yokogawai is a common antigen which recognizes a target epitope present over the tegumental layer of different trematode species.
Present study was performed to observe the tegumental ultrastructures by the developmental stages which derived from the experimental life cycle of Spirometra erinacei in laboratory conditions. In SEM view, coracidium was spherical in shape with numerous cilia, and its surface was covered with long cilia, tuberclelike projections with millet-like processes, and small holes. The body surface of procercoid was covered with numerous pointed microtriches except that of frontal pit with stout spine-like ones. However that of cercomer was covered with somewhat sparse blunt-tiped microtriches. Plerocercoids of 3 days old resembled the mature procercoid in shape, and their frontal pits were covered with numerous stout spine-like microtriches. However frontal pit and body surface in more than 5 days old ones were covered with conoid microtriches. On the surface of adult scolex, hairly long filamentous and stout short microtriches were mixedly distributed. Filamentous microtriches were more densely distributed in the anterior portion than in the posterior of scolex. The neck and immature proglottid were covered with only stout short conoid microtriches. In TEM view of coracidia, embryophore and oncosphere were obviously distinguished. The embryophore contained numerous glycogen particles, mitochondria and lipid granules. The cilia on the surface of embryophore rooted in the coracidial sheath, and consisted of 9 pairs of microtubules and 2 core complex. The oncosphere was covered with a thin and unarmed tegument, and was multi-nucleated. The protoplasmic layer of procercoid and plerocercoid consisted of disc-shaped bodies, vacuoles and mitochondria. Their tegumental cells commonly retained a nucleus, granular endoplasmic reticulums and secretory granules. The protoplasmic layer of plerocercoid was more compacted than that of procercoid. From the above results, it was confirmed that the tegumental ultrastructures are something different according to the developmental stages of S. erinacei.
In order to determine the antigenic localization in the tissues of the adult Metagonimus yokegawai, immunogoldlabeling method was applied using serum immunoglobulins (IgG) of cats which were infected with isolated metacercariae from Plecoglossus altivelis. The sectioned worm tissue was embedded in Lowicryl HM 20 medium and stained with infected serum IgG and protein A gold complex (particle size: 12 nm) , It was observed by electron microscopy at each tissue of the worm. The gold particles were observed on the tegumental syncytium as well as cytoplasm of tegumental cells and epithelial lamella of the caecum. The gold particles were not observed on the basal lamina of the tegument, interstitial matrix of the parenchyma, the muscle tissue and mitochondria of the tegument. The gold particles were specifically labeled in the secretory granules in the vitelline cells. They were also labeled on the lumen of bladder and egg shell. The above findings showed that antigenic materials in the tissue of adult worms were specifically concentrated on the tegumental syncytium as well as cytoplasm of tegumental cells and epithelial lamella of the caecum.
We observed ultra-structure of the tribocytic organ of Firicola seoulensis with transmission electron microscope. Microvilli are observed on the surface of the tribocytic organ. Below the muscle layer, we can find three types of cells. Type I cell has electron lucent cytoplasm with a few granules, while type II cell shows electron dense cytoplasm and the particulate granules. Type III cell's electron dense cytoplasm possesses many granules. Of the above three cells, Type I and II cells are believed as tegumental cells. Type III cells are thought as glandular cells specific to the tribocytic organ. This finding on also explain the secretory function of the tribocytic organ of f seoulensis.
The present study was performed to observe the ultrastructure of the integument of adult Paragonimus westermani. Dogs experimentally infected with 60 metacercariae of F. westermani were autopsied 4 months after the infection. Adult p. westermani were extracted from the dogs and the fine structure was studied by means of scanning and transmission electron microscope. The findings are as follows: 1. Scanning electron microscopic findings showed that the spines and the papillae are distributed at whole body surface but the well developed spines or papaillae are shown around the oral sucker and ventral sucker. 2. At the end of the body, excretory pore was found, the shape was irregular. 3. Transmission electron microscopic findings showed that plasma membrane, tegument, basal lamina, connective tissue, circular muscle layer, longitudinal muscle layer. nerve axon and tegumental cell were observed. 4. In higher magnification, plasma membrance and bar-shaped granules were found at the outer surface of the tegument.
The present study was performed to observe the infection status of several kind of animals with indigenous Gnnthostomn in Korea, and morphological characteristics of gnathostome larvae detected from pit-viper, Askistronon breuiccudus, for the species bullfrogs. several kinds of snakes, j.e., 55 Elnphe rufodorsata, 2 Dinodon rufozonatum rufzonatum. 62 Rhnbdophis tisrinus tigrinus and 87 Askistronon app., and 438 cats were examined. A total of 21 larval gnathostomes was detected from 12 pit-vipers, A. breuicnudus. They were 2.233 x 0.343 mm in average size and covered with about 210 transverse rows of minute cuticular spines. Their characteristic head bulbs were provided with 4 rows of hooklets of which average numbers in each row were 36.8, 39.0, 41.7 and 44.3. posteriorly. In the cross sections of midgut level, the intestinal wall consisted of a single layer of 19-25 elongate epithelial cells with a single nucleus. SEM observation of the larvae revealed unique features of head bulb , cuticular spines on transverse striations and a cervical papilla. On the basis of above morphological characteristics, they were identified as the advanced third-stage larvae of Gnothostomo hispinum. It was first confirmed that the pit-viper. ABkistronon breuicaunus is the snake intermediate host of G. hispidun.
