Optimum inoculum concentration for the production of taxol was determined in Taxus brevifolia and Taxus cuspidata cell suspension cultures. By fresh weight, 2.5, 5, 7.5, 10 g/flask of cells were inoculated and cell growth as well as taxol production were examined. In both Taxus cell cultures, the higher the inoculum concentration, the shorter the length of the lag period. The optimum inoculum concentration for taxol production was found to be 5 g/flask. To produce taxol in large quantity, utilization of proper medium was thought to be important. In case of using a production medium with 6% sucrose, taxol production was noticed. Its level reached the maximum at the 9th day of culture and decreased afterwards. However, taxol was not detected from cell cultures in growth medium.
Time course monitoring of FCW/DCW ratio, production of intra and extracellular protein, and peroxidase activity were performed during suspension culture of Taxus chinensis cells. The observed FCW/DCW ratio was 12 at day 14, which was the lowest value during cultivation, and the specific protein production, based on dry cell weight, was also the lowest at day 14, which showed 4.3 mg/g DCW. The pattern of POD activity was similar to that of protein production. The results in this report were obtained using actively growing cells in flasks, therefore it is possible to use those results to control the process and indicate the stresses imposed on cells during large-scale cultivation.
The suspension cells of Taxus cuspidata were transformed with Agrobacterium tumefaciens harboring binary vector pCAMBIE1302 encoding mgfp. Transient transfection efficiency was compared by using the fluoremetric measurement. The transient transfection efficiency was improved by transformation with DMSO and/or sonication treatment. Optimum conditions for DMSO and sonication treatment were 3% and 30sec, respectively. selection and maintenance of transformed cells were continued for 3 months. An insertion of the mgfp gene in transformed cells was detected by PCR and an expression of GFP confirmed by the western blot analysis.
This study was executed to examine helical structure and other feature, of Taxus cuspidata S. et Z. and Larix gmelinii var. principis-ruprechtii Pilger through photomicroscopic and scanning electron microscopic observation, and the obtained result, were summarized as follows: 1. Tracheid transition from springwood to summerwood was abrupt, mel slightly gradual in Taxus cuspidata S. et Z. and summerwood width of Larix gmelinii var. principis-ruprechtii Pilger was wider than that of Taxus cuspidata S. et Z. 2. Normal vertical and horizontal resin canals and surrounding thick epithelial cells were present in Larix gmelinii var. principis-ruprechtii Pilger but not present in Taxus cuspidata S. et Z. 3. Instead of helical thickening, helical chicks were observed only in summerwood trachieds of Larix gmelinii var. principis-ruprechtii Pilger. However, helical thickenings forming S, Z., and horizontal helix perpendicular to long axis of tracheid were observed in springood and summerwood tracheids of Taxus cuspidata S. et Z. and these helical thickenings were considered as analogous to the innermost layer of secondary wall. 4. Uniseriate and fusiform rays were appeared on tangential surface of Larix gmelinii var. principis-ruprechtii Pilger but oly uniserate ray was observed on tangential surface ot Taxus cuspidata S. et Z. The fusiform rays of Larix gmelinii var. principis-ruprechtii Pilger usually contained one horizontal resin canal bot occasionally two horizontal resin canals were contained in a fusiform ray. 5. Trabeculae and crassulae were observed in the tracheids of Larix gmelimii var. principis-ruprechtii Pilger but not observed in the tracheids of Taxus cuspidata S. et Z.
The effects of nitrogen, phosphate in modified B5 medium and cell immobilization on cell growth and taxol production were investigated using cell cultures of Taxus cuspidata. The ratio of nitrate to ammonium was found to be an important parameter. The ratio of 1 increased taxol production 10-fold, compared to the original ratio of 20 in modified B5 medium. Reducing phosphate concentration inhibited cell growth, but increased taxol production noticeably. Immobilized cells produced a taxol concentration of ${\sim}120\;g/l$
The localization of paclitaxel was investigated in suspension culture cells of Taxus chinensis. Over 93% of the cell-associated paclitaxel were detected throughout the entire culture period. Intracellular localization of paclitaxel over the culture time was analyzed further by cell fractionation for days 21 and 42. Paclitaxel contents in intracellular organelles were decreased at day 42, while the content in the cell wall fraction was increased at day 42 compared to the value for day 21. The localization of paclitaxel in the cell wall was confirmed by using the immunocytochemical method with the aid of a confocal laser scanning microscope.
The growth promotion of a Taxus brevifolia cell suspension culture was investigated using conditioning factors. The conditioning factors produced and secreted from cultured cells usually stimulate cell division and the production of secondary metabolites. Therefore, the effective incubation time for the optimal secretion of conditioning factors was firstly determined for the promotion of cell growth. Conditioned media obtained by cultivating for 2 and 5 days showed the promotion of initial cell growth during the early cell growth period. However, the positive effect of the conditioning factors on the initial cell growth did not continue because of the depletion of the medium nutrients. Accordingly, the addition of a carbon source to the conditioned medium prolonged the positive effect on the cell growth. The addition of sucrose to the conditioned medium resulted in the maximum cell density being reached 4 days earlier compared to the control group and an increased substrate yield.
Ion fluxes across the plasma membrane activated by 1 mM $Ce^{4+}$, cell apoptosis and taxol biosynthesis in suspension cultures of Taxus cusp/data were studied. The extracellular pH sharply decreased upon the addition of 1 mM $Ce^{4+}$, then increased gradually and exceeded the initial pH value over a time period of 12 h. The extracellular $Ca^{2+}$ concentration decreased within the first 3 h after the addition of $Ce^{4+}$, then gradually decreased to one third of initial value in control at about 72 h and remained unchanged afterwards. Experiments with an ion channel blocker and a $Ca^{2+}$-channel blocker indicated that the dynamic changes in extracellular pH and the $Ca^{2+}$ concentration resulted from the $Ce^{4+}$-induced activation of W uptake and $Ca^{2+}$ influx across the plasma membrane via ion channels. A pretreatment of the ion channel blocker initiated $Ce^{4+}$-treated cells to undergo necrosis, and the prior addition of the $Ca^{2+}$-channel blocker inhibited $Ce^{4+}$-induced taxol biosynthesis and apoptosis. It is thus inferred that W uptake is necessary for cells to survive a $Ce^{4+}$-caused acidic environment and is one of the mechanisms of $Ce^{4+}$-induced apoptosis. Furthermore, the $Ca^{2+}$ influx across the plasma membrane mediated both the $Ce^{4+}$-induced apoptosis and taxol biosynthesis.
In this study, the characteristics and mechanism of microwave-assisted drying were investigated to improve the efficiency of the storage and extraction of biomass through the removal of moisture from plant cell Taxus chinensis. The efficiency of microwave-assisted drying increased with increasing microwave power. When the experimental data were fitted to typical drying kinetic models, the page and modified Page models were the most appropriate. The microwave-assisted drying was determined to be a spontaneous endothermic process, and randomness increased during the drying process. The effective diffusion coefficient (3.445 × 10-9~7.163 × 10-7 ㎡/s) and mass transfer coefficient (3.1529 × 10-5~1.2895 × 10-2 m/s) increased with increasing microwave power. The small Biot number (0.3890~0.7198) indicated that the mass transfer process was externally controlled.

To date many kinds of compounds have been obtained from plants kingdom as antineoplastic and anti-cancerous agents. However, there is no special type of compounds for ncancer therapy. Various types of substances are effective for various types of cancers and tumors: for instance, alkaloids, lignans, terpenes and steroids etc. Curcumol obtained from Curcuma aromatica was tested and noticed to be effective against cancer of the uterine cervix clinically Oridonin isolated from Rabdosia ssp.is now investigated for clinical trials in China. Moreover, camptothecine isolated from Camptotheca acuminata is also antineoplastic alkaloid, but is very toxic. Chemical modification has been tried to decrease its toxicity. This compound is now using as clinical agent. Harringtonin was investigated as an anticancerous drug in China. Taxol, a compound with a taxane ring isolated from the bark of Taxus brevifolia, has been demonstrated to have substantial anticancer activity in patients with solid tumors refractory standard chemotherapy. Supply of this drug has severely limited full exploration of its antineoplastic potential. Some efforts are continued in National Cancer Institute NCI) Washington for surveying various Taxus species for optimal taxol content, improvement in semi-synthesis from baccatin III, improvement in method of extraction, and development of alternative renewable resources. Further, there are many compounds which have been reported as antineoplastic agents. On the other hand, we have screened on higher plants collected in Japan, China, Korea, Southeast Asia and South America for antineoplastic activity, which has been done using Sarcoma 180 ascites in mice, P388 Iymphocytic leukemia in mice, Chinese hamster lung V-79 cells, P388 cells and nasopharynx carcinoma (KB) cells in our laboratory, as primary screening. In this meeting, I will present on antitumor and cytotoxic substances of the higher plants (Rubia cordifolia, Ailanfhus Vilmoriniana, Aster tataricus, Taxus cuspidata var. nana, Cephalotaxus harringtonia var drupacea, etc.) selected from above screening tests.
