Detection of Human Taurine Transporter and Production of Monoclonal Antibody

  • Taurine (2-ethaneaminosulfonic acid) is one of the major intracellular ${\beta}$ -amino acids in mammals and is required for a number of biological processes including membrane stabilization, osmoregulation, antioxidation, detoxification, modulation of calcium flux and neurornodulation. The taurine transporter (TAUT) which contains 12 hydrophobic membrane-spanning domains has been cloned from dog kidney, rat brain, mouse brain, human thyroid, placenta and retina. In this study, The TAUT cDNA from the human intestinal epithelial cell, HT-29 was cloned and sequenced. Reverse-transcription polymerase chain reaction (RT-PCR) was performed to amplify partial cDNA encoding human intestinal TAUT. The coding region of the PCR product was 732 bp long. The primers were designed to encode highly conserved amino acid sequences near the transmembrane domains III (IPYFIFLF) and Ⅵ (KYKYNSYR) both in human and mouse. The TAUT cDNA amplified was ligated into the pGEX 4T-1 expression vector. The resulting sequence of human intestinal TAUT cDNA (Accession number of NCBI Genebank is AF346763) was identical to the sequences of the TAUTs previously determined in the human placenta and retina except 3 base pairs from that of the reported human thyroid. TAUT specific antibodies were generated to use them as biological tools in the studies of the biological role of TAUT. Peptides of 149-162 amino acid residue (14 amino acids) of the TAUT were synthesized. The synthetic peptide used in this study was LFQSFQKELPWAHC. This region was chosen not only to avoid putative glycosylation sites but also to exclude regions of known homology with GABA transporters in the extracellular hydrophilic domains. The synthetic peptide, TAUT-1 was conjugated with carrier protein, kehole lympet hemocyanin (KLH) to use as an antigen. When used for immunization on a rabbit to produce polyclonal antiserum, the conjugates elicited high -titered specific anti-TAUT-1 antibodies, which reacted well with the ovalbumin (OVA) conjugated peptides in ELISA. The KLH-conjugated peptide was also used as immunizing antigen in BALB/c mice to produce TAUT specific monoclonal antibodies. From the culture supernatant of the hybridoma, the specificity of anti-TAUT-1 monoclonal antibodies was confirmed by ELISA. Further applications of more tools in TAUT expression analysis will be performed such as western blotting and flow cytometry.

Ionomycin Recovers Taurine Transporter Activity in Cyclosporin A Treated macrophages

  • Taurine is a major $\beta$-amino acid in various tissues. Taurine transporter (TAUT) is responsible for the transportation of taurine in the cell. The transporter is affected by various stimuli to maintain its cell volume. Macrophage cell volume varies in its activated states. In our experiment, it was found that the murine macrophage cell line, RAW264.7, expressed TAUT protein in its membrane. Its transportation activities could be blocked by a $\beta$-amino acid such as $\beta$-alanine, but not by $\alpha$-amino acids in this cell line. When assessed in RAW264.7 under the influence of immunosuppressive reagents, the activity of the TAUT was decreased by the treatment of rapamycin (RM) or cyclosporin A (CsA). However when ionomycin (IM) was added to this system, TAUT activity was recovered only in CsA-treated cells in a concentration-dependent manner. In order to inhibit the voltage gated {TEX}$Ca^{+2}${/TEX} channel, calmidazolium was added to the RAW264.7 cell line. Treatment of the cell with calmidazolium completely blocked TAUT. Furthermore, addition of IM to this system recovered the activity of TAUT again. When we added phorbol myristate acetate (PMA) to the cell line, secretion of nitric oxide (NO) was increased 4-fold and the TAUT activity was decreased 5-fold. However, the addition of N-nitro L-arginine methyl ester (L-NAME), an inducible NO synthase (iNOS) inhibitor, to the PMA-treated cells, resulted in the recovery of TAUT activity. These results showed that the activity of TAUT was sensitive to the intracellular concentrations of both {TEX}$Ca^{+2}${/TEX} and NO.

The Influence of Socialistic Ideology on Modern Architecture - Case Study of Bruno Taut - (사회주의 이념이 근대건축에 미친 영향에 관한 연구 - 브루노 타우트의 사례를 중심으로 -)

  • The formation of modern architecture during the Weimar Republic in Germany is seen through the works by Bruno Taut, one of the leading architects during that period. Through the extensive literature readings on Bruno Taut and critical analysis of Bruno Taut's ideologies, his works and the circumstances that he was in, the contention of this paper is that the root of modern architecture has less to do with functionalism and new technologies, but more to do with need to express new ideologies and expressions of their view of a new society. Although the socialistic idea did not account for sole reason for influencing their architecture, it did play a considerable part in deciding what architectural expressions are preferred. For this discourse this paper limits Bruno Taut's years from 1913 to 1932 prior to his exile from Germany; 1913 to 1923 being the years that constitute theoretical activities and 1923 to 1932 being the golden years for social housing when he participated on many public housing projects. It was during this period that his architecture put importance of practicality, and utilitarian; however the transition was not an abrupt departure from his expressionistic architecture but an inclusive act on his part.

Expression of Taurine Transporter in Cell Lines and Murine Organs (세포주와 마우스 조직에서 타우린수송체의 발현분석)

  • Taurine (2-ethaneaminosulfonic acid, $^+{NH}_3{CH_2}{CH_2}{SO_3^{-}}$) is endogenous amino acid with functions as modulator of osmoregulation, antioxidation, detoxification, transmembrane calcium transport, and a free radical scavenger in mammalian tissues. Taurine transporter(TAUT) contains 12 transmembrane helices, which are typical of the $Na^+$- and $Cl^-$-dependent transporter gene family, and has been cloned recently from several species and tissues. To analyze the expression of TAUT mRNA, one step RT-PCR was performed from human and mouse cultured cell lines and from various mouse tissues. The primers were designed to encode highly conserved amino acid sequences at the second transmembrane domain and at the fourth and fifth intracellular domains. RT-PCR analysis showed both of the human intestine HT-29 and mouse macrophage RAW264.7 cell lines expressed mRNA of TAUT. To define the expression patterns of the TAUT mRNA in the murine organs, RT-PCR was performed to detect cDNA representing TAUT mRNA from seven different mouse tissues. The TAUT was detected in all of the mouse tissues analyzed such as heart, lung, thymus, kidney, liver, spleen and brain. A large amount of transcript was fecund from heart, liver, spleen, kidney, and brain, while lung contained a very small amount of transcript.

  • The German Werkbund, which was founded in 1907, played an important role in the history of modern architecture. Its exhibition 1914 in Cologne is estimated as a meaningful event in the development of modern architecture. Especially two examples, among which were built at that time, are worthy of notice. The one is the 'Modelfactory' by Walter Gropius and the other 'Glashaus' by Bruno Taut. Generally in the Textbook on the history of modern architecture, the Taut's Glashaus is rarely mentioned or described as a early example of some expressionistic architecture, while the Modelfactory by Gropius is regarded as an essential workpiece in the early stage of modern movement. the time of searching alternative not only from rationalistic modernism but also postmodernism and today in the time of plurality, Taut's Glashaus could bring us more interesting and meaningful aspects in architectural design. Through investigating the background in the planning stage and analysing the composition of space, construction, circulation etc. it is to try to understand the building as really as what it was. Furthermore, historical meanings of the building in the modern architecture is reinterpreted in the following aspects; what should be reflected in architecture and how could 'Zeitgeist' be architecturally adopted?

A Research of that Architectural Influence of Bruno Taut on Japanese Modern Architecture (건축가 브르노 타우트가 일본 근대건축에 미친 영향에 관한 고찰)

  • This paper aims at researching the characteristics in Bruno Taut of architecture and its influence In Japanese modern architecture. We also can study how to harmonize Japanese traditional architecture with western architecture through activities of Bruno Taut in Japan. Especially, Japanese architectural society had a dispute on their national architectural style in 1910. During the 1930's, however, the rationalist architects denied this 'Jaegwan' style architecture, and insisted a different new Japanese architectural style. Thus Japanese modem architecture adapted two axis of Western and Japan, it had been developed with their conflict and harmony by turns. This activities of Bruno Taut in Japan deeply and freshly influenced on the contemporary Japanese Architectural situation.

  • Using magnetorheological (MR) dampers in multiswitch open-loop control mode has been shown to be cost-effective for cable vibration mitigation. In this paper, a method for analyzing the damping performance of taut cables incorporating MR dampers in open-loop control mode is developed considering the effects of damping coefficient, damper stiffness, damper mass, and stiffness of the damper support. Making use of a three-element model of MR dampers and complex modal analysis, both numerical and asymptotic solutions are obtained. An analytical expression is obtained from the asymptotic solution to evaluate the equivalent damping ratio of the cable-damper system in the open-loop control mode. The individual and combined effects of the damping coefficient, damper stiffness, damper mass and stiffness of damper support on vibration control effectiveness are investigated in detail. The main thrust of the present study is to derive a general formula explicitly relating the normalized system damping ratio and the normalized damper parameters in consideration of all concerned effects, which can be easily used for the design of MR dampers to achieve optimal open-loop vibration control of taut cables.

Estimation of damping induced by taut mooring lines

  • A moored floating structure may exhibit resonant motion responses to low-frequency excitations. Similar to the resonant responses of many vibration systems, the motion amplitude of a moored floating structure is significantly affected by the damping of the entire system. In such cases, the damping contributed by the mooring lines sometimes accounts for as much as 80% of the total damping. While the damping induced by catenary mooring lines is well-investigated, few studies have been conducted on the damping induced by taut mooring lines, especially one partly embedded in soil. The present study develops a simple but accurate model for estimating the damping contributed by mooring lines. A typical type of taut mooring line was used as the reference and the hydrodynamic drag force and soil resistance were taken into consideration. The proposed model was validated by comparing its predictions with those of a previously developed model and experimental measurements obtained by a physical model. Case studies and sensitivity studies were also conducted using the validated model. The damping induced by the soil resistance was found to be considerably smaller than the hydrodynamic damping. The superposition of the wave frequency motion on the low-frequency motion was also observed to significantly amplify the damping induced by the mooring lines.
