Influence of Food Dye on the Activity of Hepatic Azo Reductase and the Effect of Flavin (식용 Azo 색소가 Hepatic Azo Reductase에 미치는 영향과 Flavin의 효과)

  • With the market of food products, the use of food additives is on the increase. The dye as food additives, can be used for some foods which are difficult to preserve their own colors. It can be also classified as tar dye, vegetable dye and mineral dye. Because tar dye has dense toxicity, only 15 articles among them are legally allowed to be used. Among the allowed articles, the azo compound amaranth, tartrazine, sunset yellow, and allura red, were used in determining and comparing rat hepatic azo reductase activity and we observed the flavin's effects as follows: 1. Investigation with amaranth as substrate gave an apparent Km of $645\;\mu\textrm{M}$ and Vmax of 50 n mol/min/mg protein. 2. On investigation using a fixed amaranth concentration over a range of flavin concentration, FAD significantly increased the activity of the azo reductase compared with only minor increases in reaction mediated by the NADPH-generating system alone. 3. On investigation with amaranth, tartrazine, sunset yellow allura red as electron acceptor in the absence or presence of 300 mM-FAD, sunset yellow was reduced at a rat similar to amaranth, tartrazine was reduced at a slower rate and allura red was reduced a little more rapidly.

Investigation on the Current Labelling Trend of Tar Color Additives in Infant's Drugs Marketed in Korea (국내 시판중인 소아용 의약품에 함유된 타르색소의 표기실태 조사 연구)

  • Objective: This paper was aimed to investigate the current labelling trend of tar color additives in infant's drugs used in Korea and other countries. Method: The current labelling system of the tar color additives in infant's drugs was investigated using inserted papers for drug information of infant's drugs as well as three drug information internet sites popular in Korea including internet sites of foreign countries of USA, UK, Japan, and China for infant's drug information. Results: Among 325 drugs registered as syrup form for infant's drugs (sources, http://www.health.kr), 4 drugs indicated tar color related information on the internet and only 2 drugs indicated tar color additives' name. And from the investigated results of 76 inserted papers for infant's drug information, among 31 drugs containing tar dyes, in only 19 drugs the tar color additives' names were notated. Among 6 drugs which do not contain tar dyes, only 1 drug was notated as 'Dye-Free'. Conclusion: For advanced labelling system in Korea, we proposed the below guidelines. [1] The name of tar color additives used in infant's drugs should be notated even in drug information on internet sites. [2] All drug ingredients used in infant's drugs need to be separately notated as active and inactive species on both internet sites and inserted papers. [3] 'Dye-Free' notation should be mandatory for infant's drugs which do not contain tar color additives in drug information on the internet sites as well as inserted papers.

Pharmacokinetics and the Intestinal Permeability of Amaranth in Rats (적색식용색소인 아마란스의 약동학 특성 및 위장관 투과도 연구)

  • Although amaranth, a red-colored tar dye, is usually used in foods, cosmetics, and pharmaceutics, its bioavailability and intestinal absorption have not previously been investigated. Therefore, the purpose of this study was to investigate the pharmacokinetics properties and intestinal permeability of amaranth in rats following the intravenous and oral administration of this dye. Amaranth rapidly disappeared from the plasma following the intravenous injection, with a half-life of 38.8 minutes. However, the plasma concentration of amaranth was increased to 400 minutes following the oral administration of amaranth, and the absorption time and bioavailability of amaranth were calculated to be 356 minutes and 55.6%, respectively. This suggests that once amaranth exists in the gut, this dye may be efficiently and effectively absorbed. Consistent with this result, the intestinal permeability of amaranth was comparable to atenolol, a marker compound of moderate permeability, and to one-third of caffeine's intestinal permeability (a highly permeable compound). In conclusion, a significantly long absorption time and substantial intestinal absorption of amaranth was observed following the oral administration of amaranth at a dose of 300 mg/kg in rats, despite the rapid elimination of this dye from the plasma. These results may suggest the necessity of a careful and limited use of amaranth dye when it is added to food, lip-care cosmetics, and orally administered pharmaceutics.

A Study on the Development of Food Dye from Gardenia Fructus (치자(梔子)(Gardenia Fructus)를 이용(利用)한 식용색소(食用色素) 개발(開發)에 관(關)한 연구(硏究))

  • Crocin was extracted from Gardenia fructus in order to manufacture a natural food dye. In the extraction and purification process methanol and ethanol were used as the most Suitable solvent selected in preliminary test and 9.28%g/100g of Cretin product in 99.89% of purity was acquired. The lethal dose of Cretin administered in mouse by intra peridoneal was 5.36g/kg which is much lower toxic than any other. The toxic dose which caused diarrhoea in rat was 2.55g/kg and maximum no effect level was found to be 272mg/kg. From the $1%{\sim}2.5%$ content of Crocin in rat diet reduction of body weight appeared and GTP and GOT (Transaminase) value in creased significantly. Crocin showed a good properties of tolerating acid, alkali, sunlight, reductant, oxidant, and salt compared with tar dyes. The range from 100 ppm to 500ppm bring out bright $yellow{\sim}orange$ color, the most effective color, with a good solubility in water-slightly in oil. The acceptable daily intake of crocin was calculated as 2.72mg/kg based on maximum no effect dose (2% additive dose).

  • In a batch reactor, the characteristics of photocatalytic degradation of brilliant blue FCF in titanium dioxide suspension was studied under the irradiation of ultra-violet ray. Photocatalytic degradation in anatase type of TiO$_2$ was more effective than in rutile type of $TiO_2$ below the dosage of 5g. The degradation rate was slightly increased with decreasing initial pH of brilliant blue FCF aqueous solution, but rapidly increased with the addition of oxidant. Potassium bromate acted as more effective oxidant than ammonium persulfate. The photocatalytic degradation rate of brilliant blue FCF was pseudo-first order with rate constants of 0.012, 0.006 and $0.003min^{-1}$ at initial pH 3.1, 5.2 and 7.1 of brilliant blue FCF solution, respectively.

Components and Characteristics of Black Tea Colorants (홍차색소의 성분과 특성)

  • Colored components in black tea were extracted, freeze-dried, and analysed to investigate the possibility using as a natural dye. Fractionation of the colored components was carried out by gel permeation chromatography. The colored components in black tea were elected into seven fractions. Each fraction was analyzed by UV spectrophotometer. The early fluted fractions 1-4 did not show any absorption peaks in 320-700 nm and showed the increase in absorption as it approaches to short wavelength and are considered as highly polymerized colored substances. Fractions 5-6 showed tar at 350 m and are considered as thearubigins. Fraction 7 showed absorption peaks at 376 nm and 456 nm and is considered as theaflavin. IR spectra of each fraction show: Strong C=0 stretching band at 1650 cm-1 appears in fractions 1-4, but not in fractions 5-7. Strong C=0 stretching band at 1700 cm-L appears in fraction 3-7. C=0 stretching band at 1610 cm-1 appears as a shoulder in fraction 4 and progressively changes into strong peak in fraction 5-7. From these results, it is assumed that colored components in black tea consist of polyphenolic substances having different molecular weight which were formed during tea manufacturing process. The colorants from black tea infusion were applied to silk, wool, cotton and nylon fabrics. Black tea colorants showed high affinity to wool, silk and nylon, but very low affinity to cotton fabrics.

Removal of Quinoline Yellow by Granular Activated Carbon (입상 활성탄에 의한 Quinoline Yellow의 제거)

  • The adsorption characteristics of quinoline yellow by granular activated carbon were investigated experimently in the batch adsorber and packed column. The adsorptivity of activated carbon for quinoline yellow were largely improved by acidic pH and higher temperature. When the pH was 3 at $60^{\circ}C$, quinoline yellowcould be removed 97 percent of initial concentration(10 mg/L). It was estabilished that the adsorption equilibrium of quinoline yellow on granular activated carbon was successfully fitted by Freundlich isotherm equation in the temperature range from $25^{\circ}C$ to $60^{\circ}C$. The estimated values of k and ${\beta}$ are 38.71~166.60, 0.380~0.490, respectively. The breakthrough curve of activated carbon-packed column depends on the design variables such as initial concentration, bed height, and flow rate.

Equilibrium, Kinetic and Thermodynamic Parameter Studies on Adsorption of Allura Red from Aqueous Solution by Granular Activated Carbon (입상활성탄에 의한 수용액으로부터 오로라 레드의 흡착에 대한 평형, 동력학 및 열역학 파라미터에 관한 연구)

  • Allura Red (AR) is a water-soluble harmful tar-based food colorant (FD & C Red 40). Batch adsorption studies were performed for the removal of AR using bituminous coal based granular activated carbon as adsorbent by varying the operation parameters such as adsorbent dosage, initial concentration, contact time and temperature. Experimental equilibrium adsorption data were analyzed by Langmuir, Freundlich and Temkin isotherms. The equilibrium process was described well by Freundlich isotherm. From determined separation factor ($R_L$), adsorption of AR by granular activated carbon could be employed as effective treatment method. Temkin parameter, B was determined to 1.62~3.288 J/mol indicating a physical adsorption process. By estimation of adsorption rate experimental data, the value of intraparticle diffusion rate constant ($k_m$) increased with the increasing adsorption temperature. The adsorption process were found to confirm to the pseudo second order model with good correlation. Thermodynamic parameters like change of free energy, enthalpy, and entropy were also calculated to predict the nature adsorption in the temperature range of 298~318 K. The negative Gibbs free energy change (${\Delta}G$ = -2.16~-6.55 kJ/mol) and the positive enthalpy change (${\Delta}H$ = + 23.29 kJ/mol) indicated the spontaneous and endothermic nature of the adsorption process, respectively.
