The Effect of Compost Application on Degradation of Total Petroleum Hydrocarbon in Petroleum-Contaminated Soil (유류오염 토양 내 석유계 탄화수소 화합물의 분해에 대한 퇴비의 시용 효과)

  • BACKGROUND: Petroleum-contaminated soil from leaking above- and underground storage tanks and spillage during transport of petroleum products is widespread environmental problem in recent years. Application of compost may be the most promising, cost-effective, and eco-friendly technology for soil bioremediation because of its advantages over physical and chemical technology. The objective of this study was to evaluate effect of compost application on degradation of total petroleum hydrocarbon (TPH) in petroleum hydrocarbon-contaminated soil.METHOD AND RESULTS: An arable soil was artificially contaminated by diesel, and compost was applied at the different rate of 0, 10, 30, and 50 Mg/ha. Concentration of TPH in the soil decreased as application rate of compost increased. Degradation efficiency was highest at compost 30 Mg/ha; however, it slightly decreased with compost 50 Mg/ha. Kinetic modeling was performed to estimate the rates of chemical reaction. The correlation coefficient (R2) values for the linear plots using the second-order model were higher than those using the first-oder model. Compost 30 and 50 Mg/ha had the fastest TPH degradation rate in the second-order model. Change of microbial population in soil with compost application was similar to that of TPH. Microbial population in the soil increased as application rate of compost increased. Increasing microbial population in the contaminated soil corresponded to decreased in TPH concentration.CONCLUSION: Conclusively, compost application for soil bioremediation could be an effective response to petroleum hydrocarbon-contaminated soil. The increase in microbial population with compost suggested that compost application at an optimum rate might enhance degradation of TPH in soil.

  • The small size food organism(under the size 150${\mu}m$) is needed as food for early stage of marine fish larvae of small mouse (e.g the group of grouper). This study was investigated to develop a method for copepod Apocyclops sp. culture in combination with the rotifer B. rachionus for stable culture of copepod species and harvest of various size food organisms. The culture conditions as temperature, salinity, culture volume, photo period, culture preiod and observation interval were 25${\circ}C$, 22ppt, 40ml, all dark except to observation time, 16 days and every two day during the experimental period, respectively. The Tetraselmis suecica was used as the food for the two testing orgtanisms. After every two day counting, theses two organisms were transferred to fresh culture tanks with Tetraselmis suecica of $7{\times}10^5$cells/ml. In the mixed culture of B. rotundiformis and A. sp., growth of rotifer was suppressed by mixed culture with A. sp. whereas the growth of copepod Apocyclops was promoted in the mixed culture with rotifer B. rotundiformis (the maximum density was 22 individuals/ml through the 16 culture days). Moreover, the number of copepod nauplius were promoted about 2 times in the mixed culture compared to the numbers in single species culture. With this combination culture, the havested two food organisms of variable sizes. This size variation of food organisms was useful tools for larval rearing of small mouse marine fish larvae and next step food organism size of post hatched larvae.

Studies on the Sources of Bacterial Contamination in U. H. T. Processed Milk (살균시유(殺菌市乳)의 세균오염원(細菌汚染源)에 관한 연구(硏究))

  • In order to elucidate the source of bacterial contamination during processing U. H. T. milk and to ensure its hygienic control, bacterial numbers were determined each step of the processes on the milks, water, tanks and pipe lines, and environments. The results obtained were as follows. 1. The viable numbers of mesophilic bacteria were $1.2{\sim}1.9{\times}10^7/ml$ of milk in the storage tank and in pipe line connected to the preheater. These were decreased to $7.0{\times}10cells{\sim}3.4{\times}10^2cells/ml$ after preheating and homogenization, and to $1.0{\times}10cells/ml$ after sterilization, then increased up to $1.2{\times}10^2cells/ml$ after packing. 2. The numbers of thermophilic bacteria were $5.0{\times}10cells{\sim}1.0{\times}10^2cells/ml$ of milk before preheating ; $3.0{\sim}5.0{\times}10cells/ml$ after homogenization ; none in the sterilizer and surge tank ; and $1.0{\sim}8.0{\times}10cells/ml$ after packing. 3. The levels of psychrophilic bacteria were $1.0{\sim}3.7{\times}10^6cells/ml$ of milk before preheating ; $1.0{\sim}4.0{\times}10cells/ml$ after homogenization ; $1.0{\times}10cells/ml$ after sterilization ; and $2.0{\times}10cells{\sim}2.5{\times}10^2cells/ml$ after packing. 4. No coliform bacteria were detected after sterilization, while the level before preheating was $2.1{\times}10^4cells{\sim}6.5{\times}10^5cells/ml$ of milk. 5. The level of mesophiles was $3.0{\times}10cells{\sim}7.4{\times}10^2cells$ in the environmental air, water supply, and unfilled packs and bottles ; that of thermophiles $1.0{\sim}3.0{\times}10cells$ in the air and water ; that of psychrophiles $1.0{\times}10cells{\sim}1.0{\times}10^2cells$ in the air, water, packs and bottles ; however no coliform was detected.

  • This study was conducted to investigate the effects of Kluyveromyces fragilis, Candida utilis and brewer's yeast as an additive in formulated diets containing different fish meal levels ($55{\%} and 45{\%}$) on growth and body composition of juvenile Korean rockfish (Sebastes schlegeli). Juvenile rockfish (average weight, 2.6 g) were distributed into 24 tanks with 3 replication groups and fed eight isoproteic ($41{\%}$) and isolipidic ($11{\%}$) diets containing each level ($3{\%}$) of K. frayilis, C. utilis and brewer's yeast as an additive. Survival rate of fish was not affected by dietary fish meal level and yeast kind (P > 0.05). Weight gain of fish fed the control and diets containing C. utilis and brewer's yeast was not influenced by fish meal level (P > 0.05), Weight gain of fish fed the $55{\%}$ fish meal diet containing C. utilis and $45{\%}$ fish meal diets containing K. fragilis and C. utilis was lower than that of fish fed the control diet (P < 0.05). Feed efficiency and protein efficiency ratio of fish fed the control and diet containing brewer's yeast were not affected by fish meal level, but these value of fish fed the diets containing K, fragilis and C. utilis were lower in $45{\%}$ fish meal diets than in $55{\%}$ fish meal diets. Hepatosomatic index and liver lipid content of fish fed the diet containing C. utilis were decreased regardless of fish meal level (P < 0.05), Hemoglobin, hematocrit, red blood cell, plasma total protein and plasma total glucose were not affected by dietary fish meal levels and yeast supplements (P > 0.05). From the above results, it can be concluded that yeast supplement in this formulated diet for growth of juvenile rockfish is not necessary.

An Investigation on the Optimal Ship Size for Chemical Tankers by Main Shipping Routes (케미컬 탱커선 운항노선별 최적선형에 관한 연구)

  • This study objects to find characteristics in chemical tanker markets and to determine optimal chemical tanker size using a total shipping cost in main trading route of asia chemical tankers .Precedent studies of determination of the optimal ship size and case studies about chemical tankers was carried out and tried to introduce a cost model which is applicable to chemical tanker. This study is dependant on numerical analysis and involves scenario analysis to minimize sensitivity of results. This analysis shows as follows. First, 12,000DWT tanker is an optimal size on the 'Far East-Middle East' services, 9,000DWT tanker is a most competitive on the 'Far East-South East Asia' services and 3,000DWT tanker is a most economic size on the 'Inner Far East' services at average market situation. Second, the bigger size of chemical tanker, the more competitive advantage the tanker will obtain when bunker fuel prices rise. Small size ship gets more competitive during bunker prices down. Third, market fluctuation of time charter rate for chemical tanker is less than 20% against its average time charter hire which means less volatile. And tanker's competitiveness per each size is remained mostly same when time charterer rates rise at same proportion. Fourth, bigger size chemical tankers have cost advantages when tanker's quantity of each part cargo increase. And small-sized tanks are more competitive when part cargo scales decrease. For the last, ship's port stay strongly influences on the determination of the optical tanker size. When vessel has shorter port stay, bigger-sized tanker will be more competitive and even can be competitive if applies in short voyage as well.

  • Stable Isotope Analysis(SIA) of carbon and nitrogen is useful tool for the understanding functional roles of target organisms in biological interactions in the food web. Recently, mixing model based on SIA is frequently used to determine which of the potential food sources predominantly assimilated by consumers, however, application of model is often limited and difficult for non-expert users of software. In the present study, we suggest easy manual of R software and package SIAR with example data regarding selective feeding of crustaceans dominated freshwater zooplankton community. We collected SIA data from the experimental mesocosms set up at the littoral area of eutrophic Chodae Reservoir, and analyzed the dominant crustacean species main food sources among small sized particulate organic matters (POM, <$50{\mu}m$), large sized POM (>$50{\mu}m$), and attached POM using mixing model. From the results obtained by SIAR model, Daphnia galeata and Ostracoda mainly consumed small sized POM while Simocephalus vetulus consumed both small and large sized POM simultaneously. Copepods collected from the reservoir showed no preferences on various food items, but in the mesocosm tanks, main food sources for the copepods was attached POM rather than planktonic preys including rotifers. The results have suggested that their roles as grazers in food web of eutrophicated reservoirs are different, and S. vetulus is more efficient grazer on wide range of food items such as large colony of phytoplankton and cyanobacteria during water bloom period.

Reduction of dissolved hydrogen sulfide and mortality of white leg shrimp, Litopenaeus vannamei by Bacillus spp. microorganisms (Bacillus속 미생물의 용존황화수소 저감효과와 흰다리새우(Litopenaeus vannamei)에의 영향)

  • The utility of Bacillus spp. organisms for reduction of dissolved hydrogen sulfide ($H_2S$) in white leg shrimp (Litopenaeus vannamei) culture was tested with different combinations of Bacillus spp. microorganisms: combination A (B. subtilis + B. licheniformis); combination B (B. licheniformis + B. amyloliquefaciens); combination C (B. subtilis + B. licheniformis + B. amyloliquefaciens). Of these 3 combinations, C was effective in few hours after addition whereas B needed longer time to be effective. The $H_2S-reducing$ effect of combination C was dependent on the amount of microorganisms added to $H_2S-containing$ test solution. Exposure of white leg shrimp to $H_2S$ at 8 mg/L for 7 days led to survival of 80% and 1 mg/L for 14 days it was 82.5%. The survival rate was 97.5% when combination C was simultaneously added to shrimp tanks during $H_2S$ exposure at 1 mg/L for 14 days. It was demonstrated that combination C microorganisms (B. subtilis + B. licheniformis + B. amyloliquefaciens) can reduce dissolved $H_2S$ concentrations, and this effect can be utilized to protect white leg shrimp from $H_2S$ toxicity.

Changes in Ion Balance and Individual Ionic Contributions to EC Reading at Different Renewal Intervals of Nutrient Solution under EC-based Nutrient Control in Closed-loop Soilless Culture for Sweet Peppers (Capsicum annum L. 'Fiesta') (EC 기준 파프리카 순환식 수경재배에서 양액 교체 주기에 따른 양액 중의 이온 균형 및 각 이온의 EC 기여도 변화)

  • Individual ion concentrations and ionic contributions to EC reading in the circulated nutrient solution are the important factors to be considered for stable EC-based closed-loop soilless culture. This study was conducted to determine appropriate ion-analysis intervals of the circulated nutrient solutions based on ion concentration, ion balance, and ion electrical conductivity under different renewal intervals in EC-based nutrient control systems for sweet peppers (Capsicum annum L. 'Fiesta') in early growth stage. Average node numbers of the plants were 13 and 18 when the experiment started and finished, respectively, and three plants were grown in each rockwool slab. Four different renewal intervals of circulated nutrient solutions such as 1, 2, 3, and 4 weeks were used as treatment. Nutrient solutions were supplied to the plants based on integrated radiation. Drainage was collected into drain tanks after irrigation ended in the day and then mixed with fresh water until the EC reaches 2.69 $dS{\cdot}m^{-1}$. The replenished nutrient solution was supplied to the plants in the next day. Ion concentrations of the individual ions periodically analyzed in the circulated nutrient solutions showed no significant differences among the treatments during the experimental period. Ion concentrations of $K^+$, $Ca^{2+}$, $Mg^{2+}$, $Na^+$, $NO_3{^-}$, ${SO_4}^{2-}$, ${PO_4}^{3-}$, and $Cl^-$ varied within 5-8, 11-14, 2.0-2.7, 0.5-0.6, 14-19, 4-5, 1-4, and 0.3-0.5 $meq{\cdot}L^{-1}$, respectively. Ion balance showed a consistent tendency over all the treatments and especially $K^+$ : $Ca^{2+}$ and ${SO_4}^{2-}$ : ${PO_4}^{3-}$ played great roles in the cation and anion balances in the nutrient solutions, respectively. Activity coefficients of ions such as $K^+$, $NO_3{^-}$, and $H_2PO_4{^-}$ varied within 0.8-0.9 and those of $Ca^{2+}$, $Mg^{2+}$, ${SO_4}^{2-}$ varied within 0.5-0.6, showing little changes with time. Ionic contributions of $K^+$ and $NO_3{^-}$ to EC reading were the greatest followed by $Ca^{2+}$, ${SO_4}^{2-}$, and $Mg^{2+}$ in the order. From the results, we thought that allowable ranges in ion concentration, ion balance, and subsequent individual ionic contributions to EC reading would be obtained within 4-week renewal interval of nutrient solution in EC-based closed-loop soilless culture for sweet pepper plants.

Characterization of Sedimentation and pH Neutralization as Pretreatment of Acid Contaminated Water (산 오염수 전처리용 침전 및 중화 특성)

  • Sedimentation and pH neutralization has been investigated as preteatment of acid contaminate water. The settling and neutralizing process derive more effective degradation efficiency as the pre-treatment process before the removal process of adsorption, volatile, biodegradation, or oxidation. Settling velocity, uniformity coefficient, coefficient of curvature, and grain size index can define in the sedimentation process for characteristics of the soil. The stainless steel sieve has been used to separate each particle size of the dry soil by assembling in order of 4, 10, 20, 40, 80, 100, and 200 mesh sizes. The soil from Gamcheon Port in Busan drops upper side of the sieve and shakes back and forth to separate each different size of the particle. The 1L of Imhoff cone and 200 mL of the mass cylinder were used as settling tanks to calculate settling velocity. Stokes' equation was used to figure out the average density of dry soil with a value from settling velocity. In the results, the average particle density and lowest settling velocity were 1.93 g/cm3 and 0.11 cm/s, respectively. These values can detect the range of settling points of sediment to prevent chemical accidents. In pH neutralization, the initial pH of 2, 3, 4, and 5 of nitric acid and sulfuric acid are used as an acid solution; 0.1, 0.01, and 0.001 M of sodium hydroxide and calcium hydroxide are used as a base solution. The main goal of this experiment is to figure out the volume percentage of the acid solution becomes pH 7. The concentration of 0.001 M of base solution exceeds all the conditions, 0.01 M exceeds partially, and 0.1 M does not exceed 5 v/v% except pH 2. Calcium hydroxide present less volume than sodium hydroxide at pH neutralization both sulfuric and nitric acid.

Project of Improving Good Agriculture Practice and Income by Intergrated Agricultural Farming (미얀마 우수농산물 재배기술 전수사업)

  • The objectives of the project are to increase farmers' income through GAP and to reduce the loss of agricultural produce, for which the Korean partner takes a role of transferring needed technologies to the project site. To accomplish the project plan, it is set to implement the project with six components: construction of buildings, installation of agricultural facilities, establishment of demonstration farms, dispatching experts, conducting training program in Korea and provision of equipments. The Project Management Committee and the Project Implementation Team are consisted of Korean experts and senior officials from Department of Agriculture, Myanmar that managed the project systematically to ensure the success of the project. The process of the project are; the ceremony of laying the foundation and commencing the construction of training center in April, 2012. The Ribbon Cutting Ceremony for the completion of GAP Training Center was successfully held under PMC (MOAI, GAPI/ARDC) arrangement in SAl, Naypyitaw on June 17, 2012. The Chairman of GAPI, Dr. Sang Mu Lee, Director General U Kyaw Win of DOA, officials and staff members from Korea and Myanmar, teachers and students from SAl attended the ceremony. The team carried out an inspection and fixing donors' plates on donated project machineries, agro-equipments, vehicles, computers and printer, furniture, tools and so forth. Demonstration farm for paddy rice, fruits and vegetables was laid out in April, 2012. Twenty nine Korean rice varieties and many Korean vegetable varieties were introduced into GAP Project farm to check the suitability of the varieties under Myanmar growing conditions. Paddy was cultivated three times in DAR and twice in SAl. In June 2012, vinyl houses were started to be constructed for raising seedlings and finished in December 2012. Fruit orchard for mango, longan and dragon fruit was established in June, 2012. Vegetables were grown until successful harvest and the harvested produce was used for panel testing and distribution in January 2013. Machineries for postharvest handling systems were imported in November 2012. Setting the washing line for vegetables were finished and the system as run for testing in June 2013. New water tanks, pine lines, pump house and electricity were set up in October 2013.
