Reduction of Visceral and Body Fats in Mice by Supplementation of Conjugated Linoleic Acid with γ-Oryzanol (Conjugated linoleic acid와 γ-oryzanol 혼합물의 생쥐 체지방 및 복부지방 감소 효과)

  • The synergistic effect of conjugated linoleic acid (CLA) and $\gamma$-oryzanol (OZ) on the reduction of visceral and body fats was investigated in mice. Female ICR mice, 10 weeks of age, were acclimated for one week and then randomly divided into 5 treatment groups by body weights: Control (70 ${\mu}l$ olive oil + 30 ${\mu}l$ CLA), CLA-OZ 1 (70 ${\mu}l$ olive oil + 30 ${\mu}l$ CLA + OZ 0.5 mg), CLA-OZ 2 (70 ${\mu}l$ olive oil + 30 ${\mu}l$ CLA + OZ 1.0 mg), OZ (100 ${\mu}l$ olive oil + OZ 1.0 mg), and Olive oil (100 ${\mu}l$ olive oil). Samples were daily intubated, p.o., for 4 weeks. Food and water were ad libitum. Four weeks later, mice were sacrificed by neck dislocation, followed by measuring whole body weight, empty carcass weight (ECW), which is weight without organs and visceral fats, visceral fats, body fats and protein content. Mice treated with CLA (control) sample maintained significantly, p<0.05, lower whole body weight, ECW, visceral and body fats, relative to mice treated with olive oil sample, indicating that CLA reduces the visceral and body fats. The CLA-OZ 1 treatment significantly reduced, p<0.05, visceral and body fats as compared to OZ treatment, but not significantly different from control treatment.Meanwhile, CLA-OZ 2-treated mice maintained significantly, p<0.05, lower visceral and body fats than control and OZ-treated mice. Protein contents in mice were not affected by any other treatments. These results suggest that OZ enhanced the reduction of visceral and body fats in mice by CLA.

Antibacterial Effects of Sasangjasaebang Aqueous Extracts Against Gardnerella Vaginalis ATCC14018, and Combination Effects with Metronidazole (사상자세방(蛇床子洗方)의 Gardnerella vaginalis에 대한 시험관 내 항균력 및 Metronidazole과 병용효과)

  • Purpose: The objective of this study was to observe the antibacterial effects of Sasangjasaebang (SSJSB) aqueous extracts, traditionally used for treating various gynecological diseases including vaginitis in Korea and their individual components - Cnidi Fructus(CF), Zanthoxyli Fructus(ZF) and Alumen(AL) against Gardnerella vaginalis ATCC14018, and combination effects of SSJSB extracts and Metronidazole were also monitored in this study. Methods: Antibacterial activities against Gardnerella vaginalis ATCC14018 of SSJSB (CF : ZF : AL = 10 : 3 : 3) aqueous extracts and their individual components were detected using standard agar microdilution methods. In addition, the effects on the bacterial growth curve were also monitored at MIC and $MIC{\times}2$ levels. The combination effects of SSJSB aqueous extracts and Metronidazole were observed by Checkboard Microtiter Assay and effects on bacterial growth curve treated with SSJSB aqueous extracts MIC + Metronidazole MIC, 1/2MIC and 1/4MIC, respectively. In the present study, Gardnerella vaginalis ATCC14018 were incubated under $37^{\circ}C$, 10% CO2 and bacterial growth curves were calculated at 24, 48, 72, 96 and 120 hrs after incubations. Results: MIC of CF aqueous extracts, ZF aqueous extracts, AL powders and SSJSB aqueous extracts against Gardnerella vaginalis ATCC14018 were detected as $27.5\;{\pm}\;13.693$(12.5~50), $6.875\;{\pm}\;3.423$(3.125~12.5), $4.375\;{\pm}\;1.713$(3.125~6.25) and $2.188\;{\pm}\;0.856(1.563{\sim}3.125)mg/m{\ell}$, respectively. MIC of Metronidazole was detected as $7.6\;{\pm}\;5.367(2{\sim}16){\mu}g/m{\ell}$ at same conditions. In addition, Metronidazole, CF aqueous extracts, ZF aqueous extracts, AL powders, and SSJSB aqueous extracts showed marked dosage-dependent inhibition of bacterial growth, and more dramatical inhibitions were detected in Metronidazole 1/2 MIC + SSJSB aqueous extracts MIC treatment as compared with each of single Metronidazole MIC and SSJSB aqueous extracts MIC treatments, respectively. Especially, quite similar inhibitory effects on bacterial growth were detected in Metronidazole 1/4 MIC + SSJSB aqueous extracts MIC treatment as compared with single Metronidazole MIC treatment in the present study. FIC index were detected as $0.475\;{\pm}\;0.137$(0.375~0.625) at Checkboard Microtiter Assay. Conclusion: The results obtained in this study suggest that CF aqueous extracts, ZF aqueous extracts, AL powders, and SSJSB aqueous extracts showed antibacterial effects against Gardnerella vaginalis ATCC14018, and they also showed dosage-dependent inhibitory effects on the bacterial growth. More potent antibacterial effects were detected in SSJSB aqueous extract as compared with individual components, respectively. In addition, combination treatment of SSJSB aqueous extract and Metronidazole showed more potent inhibitory effects on the growth of Gardnerella vaginalis with FIC index $0.475\;{\pm}\;0.137$(0.375~0.625). According to these results, the combination of SSJSB aqueous extract and Metronidazole is synergistic, and it is expected that effective dosages of Metronidazole could be reduced to 1/4 levels in combination with SSJSB extracts. And it might be needed to make further studies to seek the herbs which have antibacterial effects on the Gardnerella vaginalis ATCC14018.

The Effects of Glucose, Insulin and Angiotensin II on Plasminogen Activator Inhibitor-1 Expression and Growth of Aortic Vascular Smooth Muscle Cell in Rats (포도당, 인슐린 및 Angiotensin II가 흰쥐 대동맥평활근세포의 Plasminogen Activator Inhibitor-1 발현 및 성장에 미치는 영향)

  • Background: Plasminogen activator inhibitor-1(PAI-1) is known as the primary physiological inhibitor of tissue-type plasminogen activator(t-PA) in the plasma, and is present within the atherosclerotic vessels. Increased plasma levels of PAI-1 are one of the major disturbances of the hemostatic system in patients with diabetes and/or hypertension, and may have multiple interrelations with the important risk factors in the development of atherosclerosis. This study was performed to determine whether altered gene expression of PAI-1 occurs within the arterial wall, and thereby potentially contributing to the increase of cardiovascular risks associated with diabetes and/or hypertension. Material and Method: The aortic vascular smooth muscle cells of the rat were exposed to 22 mM glucose, angiotensin II, and insulin increased PAI-1 mRNA expression with the use of Northern blotting were examined. Also examined were the effects of 22 mM glucose, angiotensin II and insulin on the growth of the rat's aortic smooth muscle cells by using MTT assay. Result: Twenty-two mM glucose treatment increased the PAI-1 mRNA expression in a time- and dose-dependent manner. Aniotensin II treatment synergistically increased the glucose-induced PAI-1 mRNA expression. In contrast, addition of insulin attenuated the increase of 22 mM glucose and angiotensin II induced PAI-1 mRNA expression. Furthermore, treatment of 22 mM glucose, angiotensin II and insulin resulted in a significant increase in cell numbers. This study demonstrated that 22 mM glucose and angiotensin II have a synergistic effect in stimulating the PAI-1 mRNA expression and in the cell growth of the rat's aortic smooth muscle cells. Conclusion: Elevation of glucose and angiotensin II may be important risk factors in impairing fibrinolysis and developing atherosclerosis in diabetic patients.

Enhancement of Erythrosine Photodynamic Therapy against Streptococcus mutans by Chlorhexidine (Streptococcus mutans에 대한 클로르헥시딘과 Erythrosine 광역동 치료의 상승효과)

  • The purpose of this study was to investigate the synergistic effects of erythrosine sensitized with a conventional halogen curing unit and sub-minimal bactericidal concentration(sub-MBC) of chorhexidine on bacterial viability of Streptococcus mutans in planktonic state. Sub-minimal bactericidal concentration of chlorhexidine was added into wells containing bacteria and erythrosine. The range of concentrations tested for chorhexidine was from 0.0000001% to 0.001%. The irradiation of the bacterial suspensions was performed for 15 sec with a conventional halogen curing unit light. In another set of experiment, the effects of 0.001% chlorhexidine were observed by adding chlorhexidine into wells containing the sub-minimal bactericidal concentration of erythrosine. At the concetration of 0.001% chlorhexidine, there were no antibacterial effects in the absence of erythrosine PDT(p < 0.05). At the concentraton of $1{\mu}M$ erythrosine, there was no photodynamic therapy effect in the absence of chlorhexidine(p < 0.05). But in the presence of sub-minimal bactericidal concentration of erythrosine with light exposure, the addition of 0.001% chlorhexidine increased the bactericidal rate(p < 0.05). A combination of erythrosine PDT with sub-MBC chlorhexidine resulted in a significant reduction in bacterial counts when compared to the case with the absence of chlorhexidine.

Risk Factors of Acute Renal Failure after Colorectal Surgery (대장수술 후에 발생한 급성신부전의 위험인자)

  • Background : Acute renal failure is one of the leading causes of postoperative morbidity and mortality. The purpose of this study was to determine the risk factors that are associated with acute renal failure after colorectal surgery. Materials and Methods : Five hundred seventy patients who operated colorectal surgery at the Yeungnam University Medical Center over three years from 2004 to 2006 were enrolled in this study. The effects of gender, age, ASA classification, concomitant disease, surgery type and duration, reoperation, urogenital manipulation, medication, hypotension, hypovolemia, transfusion, and postoperative ventilatory care on the occurrence of acute renal failure after colorectal surgery were studied. Results : The major risk factors of acute renal failure after colorectal surgery were age of patients (P=0.003), ASA classification (P<0.001), concomitant disease (P<0.001), duration of the time surgery (P=0.034), reoperation (P=0.001), use of intraoperative diuretics (P=0.005), use of postoperative diuretics (P<0.001), intraoperative hypotension (P=0.018), intraoperative transfusion (P<0.001), postoperative transfusion (P<0.001), and postoperative ventilatory care (P=0.001). Conclusion : Multiple factors cause synergistic effects on the development of acute renal failure after colorectal surgery. Therefore, efforts to reduce the risk factors associated with acute renal failure are needed. In addition, intensive postoperative care should be provided to all patients.

Apoptotic Effect of Co-Treatment with Valproic Acid and 17AAG on Human Osteosarcoma Cells (Valproic acid와 17AAG의 병용처리가 사람골육종세포에 미치는 세포자멸사 효과에 대한 연구)

  • Valproic acid (VPA) is a well-known anticonvulsive agent and has been used in the treatment of epilepsy for almost 30 years. VPA emerged in 1997 as an antineoplastic agent. And it is known that antitmor activity of VPA is associated with its targeted at histone deacetylases. 17AAG, Inhibition of HSP90 leads to the proteasome degradation of the HSP90 client proteins, such as Akt, Raf/Ras, Erk, VEGF, cyclin D and p53, and causes potent antitumor activity. It is reported that 17AAG-induced HSP90 inhibition results in prevention of cell proliferation and induction of apoptosis in several types of cancer. This study was undertaken to investigate the synergistic apoptotic effect of co-treatment with the histone deacetylases inhibitor, VPA and the HSP90 inhibitor, 17AAG on human osteosarcoma (HOS) cells. Cell viability was evaluated by trypan-blue exclusion. Induction and augmentation of apoptosis were confirmed by Hoechst staining, flow cytometry (DNA hypoploidy and MMP change), Westen blot analysis and immunofluorescent staining. In this study, HOS cells co-treated with VPA and 17AAG showed several lines of apoptotic manifestation such as nuclear condensations, the reduction of MMP, the decrease of DNA content, the release of cytochrome c into cytosol, the translocation of AIF onto nuclei, and activation of caspase-3, caspase-7 and PARP whereas each single treated HOS cells did not. Although the single treatment of 1 mM VPA or 0.5 ${\mu}M$ 17AAG for 48 h did not induce apoptosis, the co-treatment with them induced prominently apoptosis. Therefore our data in this study provide the possibility that combination therapy with VPA and 17AAG could be considered as a novel therapeutic strategy for human osteosarcoma.

Biological Control of Anthracnose (Colletotrichum gloeosporioides) in Red Pepper by Bacillus sp. CS-52 (Bacillus sp. CS-52를 이용한 고추 탄저병 (Colletotrichum gloeosporioides) 방제 특성)

  • This study was carried out in order to develop a biological control of anthracnose of red pepper caused by fungal pathogens. In particular, this study focuses on the Colletotrichum species, which includes important fungal pathogens causing a great deal of damage to red pepper. Antagonistic bacteria were isolated from the soil of pepper fields, which were then tested for biocontrol activity against the Colletotrichum gloeosporioides anthracnose pathogen of pepper. Based on the 16S rRNA sequence analysis, the isolated bacterial strain CS-52 was identical to Bacillus sp. The culture broth of Bacillus sp. CS-52 had antifungal activity toward the hyphae and spores of C. gloeosporioides. Moreover, the substances with antifungal activity were optimized when Bacillus sp. CS-52 was grown aerobically in a medium composed of 0.5% glucose, 0.7% $K_2HPO_4$, 0.2% $KH_2PO_4$, 0.3% $NH_4NO_3$, 0.01% $MnSO_4{\cdot}7H_2O$, and 0.15% yeast extract at $30^{\circ}C$. The inhibition of spore formation resulting from cellulase, siderophores, and indole-3-acetic acid (IAA), were produced at 24 h, 48 h, and 72 h, respectively. Bacillus sp. CS-52 also exhibited its potent fungicidal activity against anthracnose in an in vivo test, at a level of 70% when compared to chemical fungicides. These results identified substances with antifungal activity produced by Bacillus sp. CS-52 for the biological control of major plant pathogens in red pepper. Further studies will investigate the synergistic effect promoting better growth and antifungal activity by the formulation of substances with antifungal activity.

Evaluation of 1,1,2-trichloroethylene Removal Efficiency Using Composites of Nano-ZnO Photocatalyst and Various Organic Supports (다양한 유기계 지지체와 광촉매 Nano-ZnO 복합체를 활용한 1,1,2-trichloroethylene 제거 효율 평가)

  • In this study, the various organic supports (i.e., silicone, acrylonitrile-butadiene-styrene, epoxy, and, butadiene rubber) with great sorption capacity of organic contaminants were chosen to develop nano-ZnO/organic composites (NZOCs) and to prevent the detachment of nano-ZnO particles. The water resistance of the developed NZOCs were evaluated, and the feasibility of the developed NZOCs were investigated by evaluating the removal efficiency of 1,1,2-trichloroethylene (TCE) in the aqueous phase. Based on the results from water-resistance experiments, long-term water treatment usage of all NZOCs was found to be feasible. According to the FE-SEM, EDX, and imaging analysis, nano-ZnO/butadiene rubber composite (NZBC) with various sizes and types of porosity and crack was measured to be coated with relatively homogeneously-distributed nano-ZnO particles whereas nano-ZnO/silicone composite (NZSC), nano-ZnO/ABS composite (NZAC), and nano-ZnO/epoxy composite (NZEC) with poorly-developed porosity and crack were measured to be coated with relatively heterogeneously-distributed nano-ZnO particles. The sorption capacity of NZBC was close to 60% relative to the initial concentration, and this result was mainly attributed to the amorphous structure of NZBC, hence the hydrophobic partitioning of TCE to the amorphous structure of NZBC intensively occurred. The removal efficiency of TCE in aqueous phase using NZBC was close to 99% relative to the initial concentration, and the removal efficiency of TCE was improved as the amount of NZBC increased. These results stemmed from the synergistic mechanisms with great sorption capability of butadiene rubber and superior photocatalytic activities of nano-ZnO. Finally, the removal efficiency of TCE in aqueous phase using NZBC was well represented by linear model ($R^2{\geq}0.936$), and the $K_{app}$ values of NZBC were from 2.64 to 3.85 times greater than those of $K_{photolysis}$, indicating that butadiene rubber was found to be the suitable organic supporting materials with enhanced sorption capacity and without inhibition of photocatalytic activities of nano-ZnO.

Effects of Dietary Betaine and Protein Levels on Performance, Blood Composition, Abdominal Fat and Liver Amino Acid Concentration in Laying Hens (사료내 비태인과 단백질 수준이 산란계의 생산 능력, 혈액의 조성, 복강지방 및 간의 아미노산 함량에 미치는 영향)

  • An experiment was conducted to investigate the effects of dietary betaine and protein levels on performance, internal e99 Qualify, blood composition, abdominal fat and liver amino acid concentration in laying hens. A total of 432 ISA-Brown laying hens were randomly allotted to four treatments with six replications for twenty few weeks. Treatments were factorially designed with two levels of crude protein(14.5 and $16.5\%$) and two levels of betaine(0 and 600ppm). The egg production was increased by feeding $16.5\%$ protein-fed group compared to that of $14.5\%$ protein-fed groups(P<0.05), but was different from betaine supplementation. Increasing protein and betaine supplementation to diets improved egg weight, egg mass and fred conversion(P<0.05). Eggshell breaking strength, thickness and Haugh unit were not influenced by either dietary betaine or protein levels. Dietary betaine did not affect serum total protein, albumin and uric acid concentration, whereas uric acid concentration was increased in $16.5\%$ protein-fed groups(P<0.05). The deposition of abdominal fat was lower in 600ppm betaine groups compared to those fed on 0ppm betaine groups(P<0.05). Concentration of the most hepatic amino acids were affected by dietary protein levels and betaine supplementation. There were synergistic effect between dietary protein and betaine. These results suggested that dietary betaine can improve the egg weight, mass, feed conversion and decrease the abdominal fat, but increase the hepatic amino acid contents.

Effects of Single or Mixed Supplementation of Bacterial Phytase and Fungal Phytase on Laying Performance and Nutrient Digestibility (Bacterial 및 Fungal Phytase의 단일 및 혼합 급여가 산란계의 생산성과 영양소 이용에 미치는 영향)

  • This study was conducted to investigate the effect of single or mixed supplementation of bacterial and fungal phytase using 45-wk-old 450 Hy-Line Brown laying hens housed in individual cages for 12-wk period. The birds were reallocated to have similar egg productivity by examining the egg production for one wk before starting the experiment. Two sources of phytase, bacterial (BP) and fungal (FP), were used either in single or mixture to determine the effects of these phytase. Five dietary treatments consisted of control (BP 0, FP 0), T1 (BP 300, FP 0), T2 (BP 300, FP 300), T3 (BP 300, FP 3000), and T4 (BP 0, FP 3000). The DPU was used for phytase activity in this experiment. The nonphytate phosphorus (NPP) content of control was 0.30%, and those of phytase treatments were set to 60% of the Control. Experimental diets were fed ad libitum throughout the experimental period. The lighting schedule of 17L7D was employed. The egg production was not different between control and bacterial phytase treatments, but the T4 showed significantly low productivity compared to control (P<0.05). No difference was found in average egg weight among all treatments. The daily egg mass did not show any statistical differences among all treatments: however, it was significantly low in T4 compared to ther control during the latter half of the experiment (P<0.05). No significant difference was found among treatments in terms of feed intake, feed conversion and egg quality. The digestibilities of dry matter, crude protein, and fat digestibility were similar regardless of the treatments. No significant trends were detected in Ca and P availability. In conclusion, the BP level of 300 DPU contributed to achieve 40% reduction of recommended nonphytate phosphorus addition. The synergistic effect of bacterial and fungal phytase was not confirmed.
