Kainic Acid가 미성숙 오리 망막에 미치는 영향에 관한 전자현미경적 연구 (Electron Microscopic Study on the Effects of the Kainic Acid in the Duck Retina)

  • In order to identify the effects of the kainic acid in the retina kainic acid (120 nmol/$5{\mu}l$) were injected through the pars plana of the ciliary body into the duck eye ball at 7th day after hatching. The animals were sacrificed by decapitation on the 1st, 4th and 10th day after injection of the kainic acid. The specimen was processed for the light and electron microscopes respectively. The results obtained were as followings 1. The inner nuclear, inner plexiform and ganglion cell layers of the retina showed marked changes in all of the groups. 2. The cells in the inner nuclear layer showed marked degenerative changes of the organelles including the pyknoses and destruction of the nucleus on the 1st day after injection. These destructive changes of the cells were progressively disappeared until the 10th day after injection. 3. After injection of the kainic acid the neurites of the inner plexiform layer were swollen at the first, and thereafter those changes were gradually disappeared until the 10th day. 4. In the ganglion cell defected by the kinic acid, the enlargement and segmentation of the endoplasmic reticulum and pyknosis of the nucleus were-observed in all the groups, although the lipid droplets were only shown in the 4th day. From the above results, the evidence suggests that kainic acid gives to the toxic effect on the nuurons of the retina with exception of the photoreceptors.

고지방식이 섭식 횐쥐의 간조직에 미치는 한방초콩환의 영향 (Effect of Herbal Chokong Pill on the Microstructure of Liver Cell in Rats Fed High Fat Diet)

  • 본 연구는 한방초콩환의 고지방식이를 섭식한 흰쥐의 간조직 지방제거 효과를 관찰하기 위하여 서목태(Rhynchosia nulubilis)를 식초에 15일간 절인 후, 5종류의 한약재(감초, 산조인, 백출, 황기, 산수유)를 첨가하여 한방초콩환을 제조하고, 고지방식이로 흰쥐의 비만을 유도하면서 고지방식이에 한방초콩환을 1%와 5% 첨가하여 9주 동안 투여한 후 간장의 무게, 혈청 효소활성도 및 간세포의 미세구조를 투과형전자현미경(TEM)으로 관찰하였다. HF군은 정상군의 간 무게에 비하여 약 35% 증가된 반면에 T5군의 경우는 HF군에 비하여 약 13% 감소하였다. 혈청 중 GOT 활성도는 정상군과 HF군이 비슷한 수준이었으나 T1군과 T5군에서는 HF군에 비하여 각각, 6.1%, 17.8% 감소하였으며 혈청 중 GPT 활성도 역시 정상군과 HF군이 거의 비슷한 수준이었으나 T1군과 T5군에서는 HF군에 비하여 각각 25.4, 32.4% 감소하였다. 간세포조직의 지방축적 정도 및 형태적 특징을 전자현미경으로 관찰한 결과, 정상군의 모양은 원형 내지 다각형을 나타내고 핵은 크고 둥글며 세포의 중앙에 위치해 핵질이 고르게 발달해 있을 뿐만 아니라 세포질에는 미토콘드리아를 비롯한 각종 세포 소기관이 고루 분포되어 있었으며 세포 전반에 걸쳐 지방구는 보이지 않았다. HF군은 세포전반에 걸쳐 지방구에 뒤덮여 세포의 형태 관찰이 불가능할 정도였으며 지방의 과잉 축적으로 인한 전자밀도가 낮아 백색을 나타내어 세포소기관의 막과 형태가 불분명한 지방간의 심각한 양상을 볼 수 있었다. T1군은 HF군에 비하여 지방구가 많이 소멸되었으며, T5군은 T1군에 비하여 지방세포의 크기와 수가 현저히 줄어든 것이 관찰 되었고, 더욱더 선명한 핵과 핵막 및 세포질의 전자밀도가 높아 지방질의 감소를 볼 수 있었다.

오미자 에탄올 추출물이 Listeria monocytogenes의 균체 형태와 그 성분에 미치는 영향 (Effect of Treatment with an Ethanol Extract of Schizandra chinensis on Cell Composition of and Shape Change in Listeria monocytogenes)

  • 오미자 에탄올 추출물 처리에 의한 균체의 지방산과 단백질 분석 및 형태 변화에 미치는 영향을 비교하였다. 오미자 처리에 의한 L. monocytogenes를 전자 현미경으로 관찰한 결과 오미자 추출물을 처리하지 않은 대조구는 smooth한 모양을 나타내었으나, 오미자 추출물을 첨가한 처리구는 전체적인 균체 표면이 불규칙한 모양을 나타내었고, 세포벽과 세포막의 파괴, 균체의 팽윤현상을 나타내었다. 단백질 변화를 살펴본 결과 오미자 처리에 의한 L. monocytogenes의 균체 단백질은 거의 소실되었다. 균체 지방산은 L. monocytogenes Scott A와 L. monocytogenes ATCC 19111의 경우 13-methylpentadecanoic acid(i-15:0), 12-methylpentadecanoic acid(a-15:0)와 15-methylpentadecanoic acid (i-17:0)의 함량은 감소하였으나 Cis-9,12 octadecanoic acid($18:2^{9,12}$)와 cis-9-octadecanoic acid($18:1^9$)의 함량은 증가하였다. L. monocytogenes Brei I는 13-methylpentadecanoic acid(i-15:0)와 12-methylpentadecanoic acid(a-15:0)의 함량은 감소한 반면 Cis-9,12 octadecanoic acid($18:2^{9,12}$)와 cis-9-octadecanoic acid($18:1^9$)의 함량은 증가하였다. 특히, 세 균주 모두 12-methylpentadecanoic acid(a-15:0)의 함량은 뚜렷이 감소한 반면에 Cis-9,12 octadecanoic acid($18:2^{9,12}$)의 함량은 뚜렷이 증가하였다.

  • Candida 바이오필름은 숙주조직과 의료기기의 표면에 자라는 자가-조직화된 미생물의 군락으로 전통적인 항진균제에 대한 저항성이 높게 나타난다. 황금(Scutellaria baicalensis)의 뿌리는 극동지방에서 의료용 목적으로 널리 사용되어 왔다. 본 연구의 목적은 10 C. albicans 임상 분리균주에 의해 형성된 바이오필름에 대한 황금의 수용성 추출물의 효과를 평가하고, 항바이오필름 활성에 대한 메커니즘을 알아보는 것이다. 바이오필름에 대한 효과는 XTT 환원분석법을 사용하였으며, 조사된 모든 균주에 대한 대사활성은 MIC에서 유의하게 감소($57.7{\pm}17.3$%)하였다. 황금추출물은 (1,3)-${\beta}$-D-글루칸 합성효소의 활성을 저해하였고 C. albicans의 형태에 대한 황금의 효과는 글루칸 합성의 억제로 인한 생장의 변화와 관련이 있었다: 대부분의 세포는 둥글고 팽창되었으며 세포벽이 진하게 염색되거나 파열되었다. 항캔디다 활성은 살진균성이었고, 황금은 C. albicans를 $G_0/G_1$기에 머물게 했다. 데이터는 황금이 목표가 되는 균류에 다중의 치명적인 효과를 내며, (1,3)-${\beta}$-D-글루칸 합성효소의 활성을 저해함을 통해 궁극적으로는 세포벽의 파열과 죽음에 이르게 한다는 것을 나타낸다. 따라서 황금은 바이오필름과 관련된 캔디다의 감염을 치료하고 제거하기 위한 항진균제 개발 후보 물질로서의 가능성을 가진다.

고슴도치의 구강 편평상피암종의 세포학적 고찰 1증례 (Cytologic Aspects of Oral Squamous Cell Carcinoma in a Captive African Hedgehog (Altelerix albiventris))

  • 3세령의 암컷 고슴도치(Atelerix albiventris)가 심하게 종대된 하악 병변의 평가를 위해 내원하였다. 세포학적 검사를 위해 세침흡인을 실시하였으며, 도말표본을 제작 후 로마노프스키식 염색으로 염색하였다. 세포학적 검사에서 원형을 포함한 다양한 모양의 편평상피세포가 주로 관찰되었으며, 일부 세포는 유사 방추 모양 또는 매우 길쭉한 모양을 띄고 있었다. 세포학적 소견은 편평상피암종이었으며, 조직검사를 위해 수술적으로 제거하였다. 조직병리상 악성 편평상피암종세포들이 중등도의 콜라겐과 방추형 섬유모세포로 이루어진 간질에 둘러싸여 있었으며, 인접한 골격근과 골조직으로의 침습이 관찰되었다. 종양세포는 중등도의 세포 대소부동과 핵 대소부동을 보였으며, 중등도 이상의 각화상태를 보였다. 종양의 중심부에서는 극세포해리가 있었으며 림프형질세포 및 호중구성 염증소견이 동반되었다. 유사 분열상은 고배율에서 2-3개가 관찰되었다. 이와 같은 소견에 기초해서 편평상피암종으로 진단하였으며, 보호자는 종괴의 수술적 제거 후 더이상의 치료를 원치 않아 바로 퇴원하였다. 환자는 추가 처치 없이 3개월 생존 후 폐사하였다.

X-선(線) 조사(照射)가 Ehrlich 암세포(癌細胞)의 용적(容積), 단백양(蛋白量) 및 수종(數種) Sulfhydryl 기(基)에 미치는 영향(影響)에 관(關)하여 (Effect of X-Irradiation on the Levels of some Sulfhydryl Groups, Protein and Cell Volume of Ehrlich Ascites Tumour Cells)

  • It is well known that a number of -SH and -SS containing substances afford a certain measure of protection against radiation effects in many biological systems, and it is conceivable that inherent -SH levels in Ehrlich ascites tumour (ELD)cells may be of decisive improtance with respect to the development of cellular radiation injury. So far, little effort has been directed to elucidate the changes in levels of different -SH and -SS groups in ELD cells when the tumour-bearing whole animal was subjected to the sublethal dose of X-irradiation. The present study was designed to bring some lights in the possible changes of and relationship between various sulfhydryl levels, such as P-SH, NP-SH and NP-SS, as well as the content of protein and cell volume of ELD cells, after subjecting the ELD mice to 1,200 r of X-irradiation. The animals used in this experiment were all mixed bred mice of $20{\sim}25\;gm$ in body weight (approximately 2 months old) irrespective of sex. 12 mice in one experiment were inoculated intraperitoneally with 0.2 ml of ascites tumour cells $(2{\times}10^6\;cells)$, and on the 7th day of the tumour growth, they were X-irradiated with 1,200 r, using the conventional X-ray machine under the following conditions: 200 Kv at 15 mA, 0.5 mm Cu filter, target-skin distance: 50 cm. Radiation dose was measured with the the Philip integrating dosimeter. At 24, 36, 48 and 60 hours after the X-irradiation, the mice were killed by cervical dislocation, and the tumours were taken out. Freshly withdrawn ascites tumours were placed in ice, and immediately the cell concentration was measured with the Coulter Cell Counter (Model B), and the hematocrit of the tumour cells were also determined. Cell volume was thus calculated by the cell concentration and hematocrit value. P-SH content of ELD cells was measured potentiometrically according to the method of Calcutt & Doxey, and NP-SH and NP-SS contents were measured spectrophotometrically by the method described by Ellman. Protein content of ELD cells was determined with the Folin phenol reagent by Lowry et al. Altogether, 48 experimental mice were used, and 12 mice with the only exception of X-irradiation were used as the control. Results obtained indicate that the contents of all the cellular sulfhydryl groups as well as cell volume and protein content of the ELD cells increase significantly as time progresses after the sub-lethal X-ray dose of 1,200 r was given and that all the increase is in a lineal fashion. The regression lines of the relative values, (i. e., taking each control value as 1) of all the values obtained, and the regression lines of cell volume, protein and NP-SH are identical, whereas those of NP-SS and P-SH appear to be widely seperated. However, the difference of those two lines (NP-SS & P-SH) were found to be not significant statistically (p>0.05). Therefore, it can be concluded from the above results that all the values examined increase in a lineal fashion with no statistically significant difference among them. Also, with the radiation dose of 1,200 r, the ELD cell becomes enlarged and swollen progressively up to 60 hours post-irradiation and it becomes more than two times of the original normal size at 60 hours after the irradiation, and up to this stage, it seems apparent that the cell division has been slow due to the X-irradiation applied in this experiment. It is well understandable that the contents of NP-SH, NP-SS, P-SH and protein of the ELD cells increase in parallel with the increase of the cell volume by the X-ray does used, but it also seems interesting to note that all the cellular substances tested show no appreciable difference in the pattern of increase.

난소 섬유종과 포막종의 호르몬 분비능에 관한 연구 (Hormonal Activity of Ovarian Fibroma and Thecoma)

  • Six cases of ovarian fibrous stromal neoplasm were studied clinically, light microscopically and ultrastructurally for the clinico-pathological evidences of hormone production. Of the six cases, two cases were fibroma, three cases were fibrothecoma, and one case was thecoma. Two cases of fibroma and one fibrothecoma were associated with clinical history of menstrual abnormality, however fat staining of the tumor was negative or weakly positive. Two cases of fibrothecoma and one thecoma were negative for the clinical history of hormone imbalance. Fat stain of those cases revealed positive in varying intensity. Ultrastructural examination of fibroma-thecoma group revealed dark and pale cells by their nuclear characteristics. The dark cells had indented nucleus and abundant cytoplasmic organelles of rough ER, Golgi apparatus and mitochondria. Intracytoplasmic cisternal spaces were seen in the dark cell cytoplasm and some lipid droplets were seen around the cisternae. Pale cells had pale swollen nucleus and fine chromatins. Their cytoplasm showed scanty amount of organelles. Fibroma-thecoma spectrum showed varying degree of population of dark cells, light cells and intervening collagenous stroma. Lipid droplet was structurally associated with intracytoplasmic cisterna and they were frequently seen in thecoma and two of the fibrothecoma. But clinical history of hormone imbalance was poorly related to the light microscopic morphology and ultrastructural organization.

대두 함유 방사선 조사식이를 섭취한 Mouse의 산화적 스트레스 (Oxidative Stress of Mouse Fed with ${\gamma}$-Irradiated Soybean Diet)

  • Food irradiation has been steadily increased in many countries concomitantly with increasing international trades. Harmful contaminants naturally occurred from foods which contain high levels of unsaturated fatty acids that are easily oxidized can affect the human anti-oxidation system through the generation of free radicals. Moreover, previous studies proved that ${\gamma}$-irradiation may cause production of free radicals in food. We investigated the effect of ${\gamma}$-irradiated soybeans in relation to oxidative stress in mice. Oxidative index of mice was evaluated by TBARS, DNA fragmentation in various organs such as blood lymphocytes, liver and kidney. Forty male ICR mice were equally divided into 4 groups and fed control diet or ${\gamma}$-irradiated diet containing 50% soybeans (5, 10, and 20 kGy, respectively) for 8 weeks. Peroxide values of the irradiated diets were higher than that of the non-irradiated one and increased according to the storage period. There was no significant difference in weight gain as well as in TBARS value in plasma and kidney of all groups. Liver TBARS value of the group fed with irradiated diet at 20 kGy increased significantly compared with the control group (p < 0.05). DNA oxidative damage as measured by alkaline comet assay showed that % tail DNA in the blood lymphocytes of 5 kGy and 10 kGy groups increased significantly over the control group (p < 0.05). Also, tail moments of 5 kGy and 10 kGy groups were higher than that of the control group. Ultrastructural examination shows myeline figures and swollen mitochondria in parietal and intestinal epithelial cells of the group fed with irradiated diet. Therefore, considering unsaturated fatty acid content, consumption of soybeans ${\gamma}$-irradiated with over 20 kGy or repeatedly may decrease the body's antioxidant mechanism.

Purification and Characterization of a Novel Antifungal Protein from Paenibacillus macerans PM1 Antagonistic to Rice Blast Fungus, Pyricularia oryzae

  • An antifungal protein antagonistic to the rice blast fungus, Pyricularia oryzae was purified from Paenibacillus macerans PM-1 by ammonium sulfate fractionation, Q Sepharose Fast Flow column chromatography, Phenyl Sepharose CL-4B column chromatography and Superose 12 gen filtration. An apparent molecular mass of the purified antifungal protein was determined as 8 kDa by SDS-PAGE and 9 kDa by analytical gel filtration, respectively, suggesting that the purified protein is a monomer. The antifungal protein was stable at pH range from 7-12 and up to $100^{\circ}C$. The protein was also stable at 0.1-1% Tween 20 and Triton X-100. The N-terminal amino acid sequence of the antifungal protein was Thr-Glu-Leu-Pro-Leu-Gly-Ile-Val-Met-Asp-Lys-Tyr-Thr-Asp-Ala-Phe-Lys-Phe-Asp-Met-Phe. Comparison of the determined sequence with other peptide and DNA sequences did not reveal homology at all. Therefore, the purified antifungal protein was speculated to be a novel protein. The condidial germination in vitro of P. oryzae KJ301:93-39 by the purified protein ($5.9{\mu} g/ml$) was limited to $9{\pm}3.2%$ only, compared with $69{\pm}2.4%$ of the control. Ungerminated conidia were swollen at basa and mid cell by the purified protein. In vivo bioassay for inhibition of conidial germination of P. oryzae KJ 301, one of the most predominating racesin Korea. the purified protein ($5.9{\mu} g/ml$)strongly inhibited the conidial germination. The conidia, even though germinated, could not develop any further to produce appressoria efficiently.

Penicillium verruculosum 으로부터 분생자 원형질체의 생성과 재생 (Formation and Regeneration of Conidial Protoplast from Penicilliun verruculosum)

  • 섬유소 분해능이 우수한 P. verruculosum 의 분생포자로부터 원형질체의 생성 및 재생에 대하여 조사하였다. 균의 원형질체 생성을 위한여 2-deoxy-D-glucose(2-DG) 가 첨가된 최소배지에서 분생포자를 10-12 시간 전배양 시킨후 시판 세포벽분해효소들을 작용시켜 본 결과 그중 Novozyme 234 (1%, w/v) 에서 50% 이상의 가장 좋은 원형질체 생성 수율을 보였다. 전배양 되지 않은 포자로부터는 원형질체 생성이 이루어지지 않았으며 원형질체 생성율을 향상시키기 위한 첨가테로서 2-DG 의 효과가 무첨가에 비해 2배 이상의 수율을 증가시켰다. 포자 원형질체의 재생율은 49.2% 로 동 균주의 균사체 원형질체의 재생율4.6-27.8%(삼투압 조절체로서 0.6 M magnesium sulfate) 보다 높았다. 원형질체 생성 및 재생을 위한 상투압 안정제로서는 0.6M ammonium sulfate 와0.6 M magnesium sulfate가 가장 적절하였다. 광학 형미경을 통하여 부풀린 포자로부터 원형질체의 생성과정과 두 가지 각기 다른 양상의 재생과정이 관찰되었다.
