Pharmacological Activities of Paecilomyces japonica, A New Type Cordyceps sp. (눈꽃 동충하초의 약물활성)

  • Cordyceps is reputed for its broad biological activities and as a tonic for replenishing vital function in Chinese traditional medicines. As an attempt to obtain fundamental data for the development of a new type Cordyceps, the effects of the fruiting bodies of cultivated fungus of Paecilomyces japonica grown on silkworm larvae on hyperglycemia induced by streptozotocin(STZ) and by epinephrine in rats and in mice as well as on immunological functions in mice were investigated. The 70% methanol extract of the fungus, when administered orally at 100 and 300 mg/kg in STZ-induced hyperglycemic rats, caused a significant decrease in blood glucose level 3hr after sample treatments. The methanol extract, when administered p.o. at the same dose levels in epinephrine-induced hyperglycemic mice, also caused a significant decrease in serum glucose levels as well as a significant reversal of the liver glycogen contents suggesting its hypoglycemic activity might be due to glycogen breakdown in the liver. Treatment of normoglycaemic mice with the methanol extract of the fungus exhibited a significant glucose tolerance up to 3hr after oral glucose load(2.0 g/kg). The methanol extract also showed immuno-stimulating activity as measured by carbon clearance in mice and a significant antifatigue effect as measured by weight loaded forced swimming performance in mice.

Comparing Acute and Swimming Endpoints to Evaluate the Response of Two Freshwater Midge Species, Chironomus yoshimatsui and Chironomus riparius to Heavy Metals (요시마쯔깔따구와 리파리깔따구(파리목: 깔따구과)의 중금속에 대한 급성독성 및 유영능력 비교)

  • The relative sensitivity of two freshwater non- biting midges, Chironomus yoshimatsui Martin and Sublette and C. riparius Meigan, was examined for lead, cadmium, and mercury in water- only exposures. Two endpoints were compared to assess toxicity 48 h and 96 h after exposure: Acute toxicity ($50\%$ lethal concentration: $LC_50$) and behavioral toxicity ($50\%$ effective concentration: $EC_{50}$). for the behavioral toxicity, reduction of swimming performance of two midge species in the treated conditions was compared to that in the untreated control. The sensitivities differed depending on the species and heavy metals, although some trends emerged. $LC_50$ values in C. yoshimatsui to cadmium and lead were always higher than those in C. riparius with increasing toxicity, regardless of the exposure times. The opposit was true for the mercury treatment. Similar trends were observed in the $EC_{50}$ values. The $EC_{50}$ values were always lower than the $LC_50$ values in all the treatment cases (midge species, heavy metals, and exposure times). These results indicate that the two midge species respond to the heavy metals differently: C. riparius is sensive to cadmium and lead and C. yoshimatsui to mercury. Behavioral toxicity such as swimming performance can be an effective endpoint for assessing heavy metal toxicity in water.

Development of Autonomous Bio-Mimetic Ornamental Aquarium Fish Robotic (생체 모방형의 아쿠아리움 관상어 로봇 개발)

  • In this paper, the designed fish robots DOMI ver1.0 is researched and development for aquarium underwater robot. The presented fish robot consists of the head, 1'st stage body, 2nd stage body and tail, which is connected two point driving joints. The model of the robot fish is analysis to maximize the momentum of the robot fish and the body of the robot is designed through the analysis of the biological fish swimming. Also, Lighthill was applied to the kinematics analysis of robot fish swimming algorithms, we are applied to the approximate method of the streamer model that utilizes techniques mimic the biological fish. The swimming robot has two operating mode such as manual and autonomous operation modes. In manual mode the fish robot is operated to using the RF transceiver, and in autonomous mode the robot is controlled by microprocessor board that is consist PSD sensor for the object recognition and avoidance. In order to the submerged and emerged, the robot has the bladder device in a head portion. The robot gravity center weight is transferred to a one-axis sliding and it is possible to the submerged and emerged of DOMI robot by the breath unit. It was verified by the performance test of this design robot DOMI ver1.0. It was confirmed that excellent performance, such as driving force, durability and water resistance through the underwater field test.

Improving the Performance of a Robotic Dolphin with a Compliant Caudal Fin (꼬리 지느러미의 유연성을 이용한 로봇 돌고래의 성능 향상)

  • Fish generates thrust with a compliant fin which is known to increase the efficiency. In this paper, the performance of a robotic dolphin, the velocity and the stability, was improved using an optimal compliant caudal fin under certain oscillating frequency. Optimal compliance of the caudal fin exists that maximizes the thrust at a certain oscillating frequency. Four different compliant fins were used to find the optimal compliance of the caudal fin at a certain frequency using the half-pi phase delay condition. The swimming results show that the optimal compliant fin increases the velocity of the robotic fish. The compliance of the caudal fin was also shown to improve the stability of the robotic fish. A reactive motion at the head of the robotic dolphin causes fluctuation of the caudal fin. This phenomenon increases with the oscillating frequency. However, compliant fin reduced this fluctuation and increased the stability.

  • Cutaneous injuries stung by jellyfish are not uncommon in human exposed to marine environments. Most of the cases occur to scuba divers, fishermen, and travelers swimming at the beach. The symptoms vary from mild dermatosis to fatal systemic reaction. Some group of jellyfish like Atlantic Portuguese man-of-war (Physalia physalis) provokes acute severe skin injuries with systemic symptoms of nausea, bradycardia, and rarely anaphylaxis. But it is unusual case that allergic dermatitis caused by Scyphistoma which is polyp stage of jellyfish (Aurelia aurita) happened to zoo keepers working at indoor dolphin pool. This case report is about dermatosis with symptoms such as painful, irritant, itching, and erupted skin lesions on the neck and face of zoo keepers working at dolphinarium in Seoul zoo, Korea.

The Effects of Different Type of Triglyceride Supplements on Exercise Performance Time, Energy Substrates, Insulin Hormone and Lipase Activity in the Trained Rats (서로 다른 형태의 지방산 투여가 훈련된 흰쥐의 지구성 운동수행력, 안정시기와 운동스트레스 시기의 에너지 기질, Insulin 호르몬과 Lipase 활성에 미치는 영향)

  • The purpose of this study was to investigate the effects of different type of triglycerides (MCT & LCT) on weight, survival time, energy substrate (FFA, TG, pyruvate, lactate), insulin and lipase in the trained rats. Fifty-four Sprague-Dawley rats were divided into 3 groups: control group (CG, n=18), MCT supplement group (MG, n=18), and LCT supplement group (LG, n=18). They also were divided into 3 periods: trained resting (R, n=6) and trained & exercise load (E, n=6), and survival time test was performed to know the supplemented effects. Body weight of all animals was checked every week, MCT group and LCT group received supplementary MCT and LCT orally and preliminary swimming training for 6 days before the start of main experiment. All animals received 15-minute swimming training 5 times during first week of experiment, and swimming training time was increased 15 minutes every week until it reached 90 minutes at last 9th week. After last swimming training, animals were fasted for 12 hours and blood samples were taken from abdominal aorta in the Department of Animal Medicine at the D university Animal Center. Among the CGE, MGE, and LGE groups, the MGE had the greatest increase in physical performance time. In the FFA levels, there was significant differences(p<.05) in CG, MG and LG groups, and also there was major difference of FFA levels in the MG and LG. In the lipase levels, there was signifi.ant differences (p<.05) in CG, MG and LG groups. MG was the greatest than the other groups. In the insulin hormone levels, there was the great differences (p<.05) in LG compare to CG groups, whereas there was no significant difference in the CG and MG. In conclusion, these results suggest that regular prolonged physical training with MCT supplementation, improves exercise performance time through the increase of energy substrate utilization, lipase activity and FFA levels, irrespective of insulin hormone responses.

Comparison of Power, Agility of Male and Female Fin Swimmers by Athletic Career

  • Purpose: The purpose of this study is to compare and analyze power and agility, which are factors that affect performance, by dividing adult fin swimmers into groups by male and female careers, and use them as basic data for improving performance during fin swimmer guidance and training. Research design, data, and methodology: Accordingly, 21 fin swimmers were selected and they were divided to four groups by gender and career to conduct experiments. Power and agility, which are factors related to performance of fin swimmers, were measured. Independent sample T-Test was conducted to verify differences between groups. Results: As a result of this study, there was no statistically significant difference in power and agility by the careers of male and female athletes. Conclusion: The results of this study can be used as basic data for organizing training programs suitable for the gen-der of fin swimmers and basic data for the guidance and training of fin swimmers. In future follow-up studies, based on the results of this study, it is expected that follow-up studies should be con-ducted to improve detailed performance according to age, fin swimming events, and gender. Further implications were discussed.

Effects of Starvation and Delayed Feeding on Growth and Survival of Pacific Cod Gadus macrocephalus Larvae (대구, Gadus macrocephalus, 자어의 첫 섭식 시 기아와 늦은 먹이 공급이 성장과 생존에 미치는 영향)

  • The present study aimed to investigate the influence of starvation on growth, survival and swimming ability of Pacific cod Gadus macrocephalus larvae. Notochord length, musculature height, body depth, gut height and volume of yolk of reared larvae were measured to determine the growth parameters. A significant difference was observed in all morphometric characteristics before 15 DAH (days after hatching). Body depth and volume of yolk of unfed larvae were significantly smaller than those of fed larvae from 9 DAH (P<0.05). Almost all yolk in fed group was consumed at 11 DAH. Survival and growth of larvae were observed to determine the effect of delayed initial feeding (2 DAH, 3 DAH, 4 DAH, unfed). All larvae in the unfed group died by 15 DAH and the larvae in other experimental groups survived until the end of the experiment to 21 DAH. Survival rate was not significantly different between the 2 DAH group ($17.5{\pm}4.27%$) and the 3 DAH group ($20.5{\pm}1.5%$) at 21 DAH (P>0.05). However, there was a significant difference in survival rate between the 3 DAH group and the 4 DAH group ($11.7{\pm}1.52%$) (P<0.05). There was no significant difference in notochord length among the groups fed from 2 DAH, 3 DAH and 4 DAH at 21 DAH (P>0.05). The swimming ability in fed group gradually increased in both cruising and burst swimming speeds, while those abilities in unfed group gradually decreased after reaching the peak at 6 DAH in both cruise ($18.7{\pm}6.56mm/s$) and burst swimming speed ($43.5{\pm}12.65mm/s$).

Morphology, molecular phylogeny, and pigment characterization of an isolate of the dinoflagellate Pelagodinium bei from Korean waters

  • The dinoflagellate genus Pelagodinium is genetically classified in distinct sub-clades and subgroups. However, it is difficult to determine whether this genetic diversity represents intra- or interspecific divergence within the genus since only the morphology of the type strain of the genus Pelagodinium, Pelagodinium bei, is available. An isolate associated with the genus Pelagodinium from Shiwha Bay, Korea, was recently cultured. This isolate was clustered with 3 to 4 strains from the Atlantic Ocean, Mediterranean Sea, and Indian Ocean. This cluster was distinct from the subgroup more closely associated with P. bei. The morphology of the isolate was analyzed using optical and scanning electron microscopy and was almost identical to that of P. bei except that this isolate had two series of amphiesmal vesicles (AVs) in the cingulum, unlike P. bei that has one series. When the pigment compositions of the isolate and P. bei were analyzed using high-performance liquid chromatography, these two strains had peridinin as a major accessory pigment and their pigment compositions were almost identical. In addition, the swimming behaviors of these two strains were very similar. The reexamination of the type culture of P. bei revealed two series in the cingulum as for the isolate. The new findings on the number of series of AVs in the cingulum, the pigment composition, and the swimming behaviors suggest that P. bei and the isolate are conspecific despite their genetic divergence. This study provides a basis to further understand the molecular classification within Pelagodinium combining genetic, morphological, pigment, and behavioral data.

Changes of Renal Peripheral Benzodiazepine Receptor in the Stress/Anxiety Response

  • Peripheral benzodiazepine receptor(PBR) has been indentified in various peripheral tissues including kidney. The physiological and pharmacological functions of PBR are still uncertain, althought it has been suggested that these are associated with the regulation of stress/anxiety response. Diazepam progeny, which were exposed to diazepam perinatally, was reported to be an animal model of chronic anxiety. However, PBR in the diazepam progenies are not known yet. In the present study, therefore, we examined the changes of PBR in the stress/anxiety response. Dams of rats were given injection of diazepam or vehicle during puerperium. Diazepam progenies showed increased level of anxiety on the performance of elevated plus maze, and increased Bmax of PBR. Saturation experiments followed by scatchard analysis of the results showed that the increase in the density of PBR and the affinity of the PBR remained unchanged. Forced swim stress increased anxiety on the plus maze in both groups of rats. In contrast to control, diazepam progenies did not show further upregulation of renal PBR immediately after swimming stress, but still higher than control. From the above results, it may be concluded that upregulation of renal PBR is associated with chronic anxiety as well as stress-induced response.
