MicroRNA-126 Regulates the Expression of Stem Cell Transcription Factors (Sox2 and Lin28) in Various Ovarian Tumors (MicroRNA-126은 난소 종양세포의 줄기세포 전사인자 (Sox2와 Lin28) 발현을 조절한다)

  • Stem cell-like tumor cells are reported to be the main reason for tumor recurrence and metastasis. As one of the new approaches to overcome cancer, studies are emerging to inhibit the expressions of stem cell transcriptional factors (Oct4, Sox2, Klf-4, and Lin28) in cancer cells. MicroRNAs are master genetic regulators that can control development and differentiation of stem cells. In this study using various ovarian tumors (Skov3, Ovcar3, Tov112D, Tov21G, PA-1 and Hsc832(c)T), we examined the expressions of stem cell-related transcription factors, and the biological changes in cell survival and growth by miR-126 that targets stem cell transcriptional factors. We observed that treatment of miR-126 induced the morphological changes and cell suspension in most cells. In addition, miR-126 induced gradual regression of cell division except Skov3 cells, especially significant time-dependent reduction in Tov112D, Tov21G and PA-1. When we examined the expression of stem cell transcriptional factors, Sox2 was shown to be down-regulated after miR-126. Our results demonstrate that miR-126 treatment can provide the reversible environment to regulate cell division and to induce cell death of ovarian tumors, suggesting the molecular biological clues for clinical usage.

Development of Questionnaire for Automobile Seat Comfort Evaluation (자동차 시트의 안락도 평가를 위한 문항개발에 관한 연구)

  • The automotive seat comfort evaluation was to begin a key aspect in seat design. It depended largely on the basic mechanical aspect such as geometric parameters of seat, choice of suspension system and cushion material used. Until recently, seat comfort evaluation advanced to evaluate subjective sensitivity of human. The external literatures showed in the last decade, there have been very few attempts to establish and document automotive seat comfort evaluation. In 2006 Smith, D. proposed the statistically reliable tool in giving a numeric rating for set comfort and the tool was used in the many country. On the other hands, we, in Korea, had not the reliable tool for the automotive seat comfort evaluation. So that, we studied to develope the questionnaire for seat comfort evaluation based on Smith, D.(2006) and some studies. As a result, we developed 36 contents for the automotive seat comfort evaluation with the help of professional in Korean automotive industry. Here, 36 contents were identified as the dimensions that represent the human sensitivity and psychological feeling on comfortable seat. Also, we derived the priorities for the 36 contents by using analytic hierarchy process (AHP), based on the judgments of 30 experts and drivers. This study will help the designers and developers clarify the conceptual and abstract aspect of the design evaluation by proposing a more systematic and process-oriented method.

Effects of Plant Water Extract Codonopsis Lanceolatae on Mouse Immune Cell Activation Ex Vivo (더덕 물 추출물의 경구 투여가 마우스 면역 세포 활성에 미치는 효과)

  • Codonopsis lanceolatae has been used as one of the traditional remedies as well as food source. However, few studies on their immunomodulating effects have been reported. We previously reported that ex vivo supplementation of Codonopsis lanceolatae water extracts enhanced splenocyte proliferation compared to the control group. In order to elucidate its ex vivo effect, six to seven week old balb/c mice were fed ad libitum on a chow diet and water extracts of Codonopsis lanceolatae were orally administrated every other day for four weeks at two different concentrations (50 and 500 mg/kg B.W). After preparing the single cell suspension, the proliferation of splenocytes was determined by MTT (3- [4,5-dimethylthiazol-2-y] -2,5-diphenyl terazolium bromide) assay. The production of cytokine (IL-1${\beta}$, IL-6, TNF-${\alpha}$), secreted by macrophages stimulated with LPS or not, was detected by ELISA assay using a cytokine kit. After 48 hrs of incubation with the mitogen (ConA or LPS) stimulation, the mice splenocyte proliferation in experimental group was statistically increased at two different concentrations than that in control group. The cytokines production was more significantly enhanced at the lower supplementation (500 mg/kg B.W.) group rather than higher concentration (500 mg/kg B.W.) compared to the control group. The results of this study may suggest that the supplementation of water extract of plant mixture could regulate the immune function by increasing the splenocyte proliferation and enhance the immune function through regulating cytokine production capacity by activated macrophages in mice.

Viability of eggs, filariform larvae and adults of Stronglyloides venezuelensis (Nematoda: Strongyloidea) maintained in vitro (베네수엘라분선충 (Strongvloides venezuelensis)의 충란, 감염자충 및 성충의 실험관 내 배양)

  • The present study was performed to check the viability of eggs, filariform larvae and adults of Strongvloines venezueLensis exposed to various conditions for an in vitro maintenance. The eggs in the feces remained viable for about 25 days at $4^{\circ}C$ and 15 days at room temperature. However, the isolated eggs in sterile saline lost their viability within 24 hr at $4^{\circ}C$. The eggs in morula stage were very sensitive to air drying and rapidly lost their viability (=12 hrs. Filariform larvae survived for a maximum period of 45 days in fecal suspension and 28 days in 0.12% nutrient broth in polyvinyl culture bags maintained at $20^{\circ}C$. On the other hand, those isolated from nutrient broth cultures survived for a maximum period of 32 days in tap water and 22 days in sterile saline at $20^{\circ}C$. The mature adult worms obtained from experimentally infected rats survived maximally for 9 days in serum supplemented (10% rat-serum) 0.12% nutrient broth and 4 days in serum free nutrient broth at $37^{\circ}C$ while the culture media were changed at an alternate day. The adult female worms deposited fertile eggs in serum supplemented and serum free nutrient broth cultures, however, the hatched larvae (Ll) were not able to develop to the filariform stage in the culture media and found to die within 24 hr of maintenance. The present findings on an in vitro maintenance of different stages of 5. uenezueLetis may provide useful information for biological and biochemical studies with Strongyloines species.   • In polymer exchange membrane fuel cells, it is crucial to fabricate a highly active and thin Pt catalyst layer for the smooth mass transport of dissolved oxygen and water. Although a highly loaded platinum (Pt) catalyst based on the hydrothermal synthesis has been reported in several studies, its growing behaviors and kinetics were yet to be understood. In this study, we investigated the growth of Pt crystal in suspension after the reduction step depending on a stirring time and evaluated the electrochemical activity. For only a couple of hours in the early stage, Pt colloids were adsorbed on the Pt-carbon catalyst and the Pt crystal was grown. After that, the small Pt colloid was formed by another nucleation step, which did not involve the growth of Pt crystal. We reveal that the Pt-Carbon catalyst with stirring for 6 h showed a high activity toward the oxygen reduction reaction.

Studies on the Processing of Yeast Extract by Waste Brewery Yeast (맥주효모박을 이용한 효모추출물의 제조에 관한 연구)

  • The optimum condition for production of yeast extract by autolysis and enzymatic method were studied with waste brewers\` yeast. The major components of brewer\`s yeast in proximate compositions were carbohydrate(46.0%) and protein(41.7%). There was a little appreciable difference in the promaximate composition of waste brewers\` and bakery yeast. The major minerals were identified as K, P, Mg and Ca, and their contents were about 1,659.7%, 1,197.4mg%, 210.4mg% and 105.6mg%, respectively. The major vitamins were identified as vitamin C and vitamin B group, and their contents were about 7.9IU/100g and 4.2IU/100g, respectively. The optimum yeast concentration in the reaction suspension was 10% (w/v) waste brewer\`s yeast slurry. The protein content in brewer\`s yeast extract was increased with addition of NaCl as plasmolyzers. However, the plasmolyzer effect of ethanol a little showed as the concentration increasd from 3% ∼7%. The 5\`-nucleotides(5\`-GMP and 5\`-IMP) content in brewer\`s yeast extract was increased with addition of glucanase. The total content of 5\`-nucleotides (5\`-GMP and 5\`-IMP) increased by 160% than those by the autolysis of waste brewer\`s and bakery yeast.

Prediction of Structural Performance of an Automotive Ball Joint (자동차용 볼조인트의 구조적 성능 예측)

  • An automotive ball joint connects the suspension system to the steering system and helps to enable rotational and linear motion between the two elements for steering. This study examines a ball joint used in medium and large-sized pickup trucks. Ball joints consist of a stud, socket, bearing, and plug. The main structural performance metrics of ball joints are the pull-out strength and push-out strength. These structural parameters must meet certain criteria to avoid serious accidents. Test and simulation methods are used to investigate the design requirements, but tests are time-consuming and costly. In this study, we modeled ball joints in SolidWorks and performed a finite element analysis in Abaqus to predict structural performance. The analysis was used to obtain the structural performance required for the static analysis of a 2D axisymmetric model. The uncertainties in the manufacturing of the ball joint were assumed to be the manufacturing tolerances, and the dimensional design variables were identified through case studies. The manufacturing tolerances at each level were defined, and the results were compared with experimental results.

Factors Affecting the Conflict between Grandparents Raising Grandchildren and Adult Children: Focusing on Grandparents' Characteristics (손자녀 양육 조부모와 자녀와의 갈등에 영향을 미치는 요인: 조부모의 특성을 중심으로)

  • The purpose of this study is to examine factors affecting the conflict between grandparents raising grandchildren and adult children. The data were based on the panel survey to explore korean retirement and income study conducted by National Pension Service in 2009. For this study, 287 parenting grandparents rearing grandchild are selected from the survey. Included variables are a demographic factor, a economic factor, a caregiving-related factor, a health factor, and a family relationship factor. The data were analyzed using descriptive statistics, t-test, co-relation and logistic regression, with SPSS WIN 18.0 program. The results are as follows: First, when grandparents are male, older, highly educated, having no religion, and unemployed, a conflict with their adult children grows. Second, when grandparents do not have earned income or financial income, but have private income transfers, a conflict with their adult children is high. Third, when grandchildren are younger, there are no caregiving rewards, economic activities suspension or reduction because of caregiving, a conflict with their adult children is strong. Also, when grandparents' physical health is good, but their mental health is poor, a conflict with their adult children increases. Fourth, when grandparents' satisfactions with family relationship and spouse relationship are low, but their satisfaction with adult children relationship is high, a conflict with their adult children rises. This study suggests that unlike in the past, grandparents raising grandchildren can no longer make sacrifice themselves for their adult children, but they expect proper exchange between parents and children. Therefore, there needs to be understanding of grandparents raising grandchildren and further studies of a conflict between grandparents raising grandchildren their adult children.

Studies on Mass Production of Intracellularly-Produced Secondary Metabolite, Cyclosporin A by Use of Immobilized Fungal Cells in Stirred-Tank Immobilized Perfusion Reactor System(IPRS) (교반식 perfusion 생물반응기(IPRS)에서 고밀도 고정상 곰팡이 세포를 이용한 세포내 축적 이차대사산물인 Cyclosporin A 대량생산에 관한 연구)

  • Immobilized bioprocess was carried out for continuous production of cyclosporin A (CyA) produced intracellularly as a secondary metabolite by a filamentous fungus, Tolypocladium inflatum. Immobilization procedure for entrapping conidiospores of the producer was significantly simplified by use of a modified immobilization technique. A newly-designed immobilized perfusion reactor system (IPRS) showed good process benefits as demonstrated by the role of the high density immobilized cells as an efficient biomass generator, continuously supplying highly active CyA-producing free cells (1.0g/$\ell$/hr) even at very high dilution rate ($0.1hr^{-1}$). IPRS bioprocess was possible since efficient decantor system developed in our laboratory separated the sloughed-off free cells from the immobilized biomass effectively, thus overcoming wash-out phenomenon frequently encountered in continuous free cell cultures. Furthermore the released-free cells remaining in the bulk solution did not appear to cause substrate mass transfer limitation which was often experienced in suspended mycelial fungal cell fermentations. The primary reason for this was that the suspension broth of the IPRS mainly consisted of roundshaped short mycelial fragments and conidiospores, still remaining Newtonian even at high cell density. In parallel with IPRS bioprocess development, other key factors to be considered necessarily for significant increase in CyA productivity would be strain improvement and medium optimization for the immobilized cells.

Partial Purification and General Properties of Yeast Acetolactate Synthase (효모 Acetolactate Synthase의 부분 정제와 일반 특성 연구)

  • Acetolactate Synthase (ALS) was partially purified from the yeast and its basic biochemical studies were carried out. Yeast was grown in the minimum media containing 0.5% glucose, 51 mM $K_2HPO_4$, 22 mM $KH_2PO_4$, 8 mM $(NH_4)2SO_4,\;0.4\;m M\;MgSO_4$ for 18 hours at 37 $^{\circ}C$. The cell was ruptured in the buffer (20 mM phosphate buffer pH 7.0, 0.1 mM TPP, 0.5 mM DTT, 1 ${\mu}M$ FAD, and 1 mM MgCl_2$) following an overnight suspension. The supernatant fraction was collected from $10,000{\times}g$ and the enzyme was further purified by ammonium sulfate fractionation, DEAE-Sephacel chromatography and leucine-agarose chromatography. The enzyme activity was measured under the various conditions by the function of protein concentration, time, temperature, pH, and substrate. The optimum temperature was found to be 50$^{\circ}C$, optimum pH 8.0∼8.5. The kinetic parameters, $K_m\;and\;V_{max}$ were 8.4 mM and 17.9 nmol/mg/min respectively. Stability of the enzyme was studied with ethylene glycol and glycerol added to the enzyme solution. Both ethylene glycol and glycerol improved the enzyme stability up to 50%. The study of feedback inhibition showed that valine was a strong inhibitor while leucine was a weak inhibitor.
