Particle-motion-tracking Algorithm for the Evaluation of the Multi-physical Properties of Single Nanoparticles (단일 나노입자의 다중 물리량의 평가를 위한 입자 모션 트랙킹 알고리즘)

  • The physical properties of biomaterials are important for their isolation and separation from body fluids. In particular, the precise evaluation of the multi-physical properties of single biomolecules is essential in that the correlation between physical and biological properties of specific biomolecule. However, the majority of scientific equipment, can only determine specific-physical properties of single nanoparticles, making the evaluation of the multi-physical properties difficult. The improvement of analytical techniques for the evaluation of multi-physical properties is therefore required in various research fields. In this study, we developed a motion-tracking algorithm to evaluate the multi-physical properties of single-nanoparticles by analyzing their behavior. We observed the Brownian motion and electric-field-induced drift of fluorescent nanoparticles injected in a microfluidic chip with two electrodes using confocal microscopy. The proposed algorithm is able to determine the size of the nanoparticles by i) removing the background noise from images, ii) tracking the motion of nanoparticles using the circular-Hough transform, iii) extracting the mean squared displacement (MSD) of the tracked nanoparticles, and iv) applying the MSD to the Stokes-Einstein equation. We compared the evaluated size of the nanoparticles with the size measured by SEM. We also determined the zeta-potential and surface-charge density of the nanoparticles using the extracted electrophoretic velocity and the Helmholtz-Smoluchowski equation. The proposed motion-tracking algorithm could be employed in various fields related to biomaterial analysis, such as exosome analysis.

Emulsifying Properties of Gelatinized Octenyl Succinic Anhydride Modified starch from Barley (호화 옥테닐 호박산 전분의 유화 특성)

  • The present study was carried out to investigate the emulsifying properties of heat-treated octenyl succinic anhydride(OSA) starch and the interfacial structure at oil droplet surface in emulsions stabilized by heat-treated OSA starch. First, the aqueous suspensions of OSA starch were heated at $80^{\circ}C$ for 30 min. Oil-in-water emulsions were then prepared with the heat-treated OSA starch suspension as sole emulsifier and their physicochemical properties such as fat globule size, surface load, zeta-potential, dispersion stability, confocal laser scanning microscopic image(CLSM) were determined. It was found that fat globule size decreased as the concentration of OSA starch in emulsions increased, showing a lower limit value ($d_{32}:0.31{\mu}m$) at ${\geq}0.2wt%$. Surface load increased steadily with increasing OSA starch concentration in emulsions, possibly forming multiple layers. In addition, fat globule sizes were also influenced by pH: they were increased in acidic conditions and these results were interpreted in view of the change in zeta potentials. The dispersion stability by Turbiscan showed that it was more unstable in emulsions at acidic condition. Heat-treated OSA starch found to adsorb at the oil droplet surface as some forms of membrane (not starch granules), which might be indicative of stabilizing mechanism of OSA starch emulsions to be steric forces.

Effect of Fouling Reducing Additives on Membrane Filtration Resistance of Activated Sludge (막오염 감소제가 활성슬러지의 여과저항에 미치는 영향)

  • Effect of three additives, chitosan, ferric chloride, and MPE50 on membrane fouling reduction was studied. They were introduced with various dosing rate into activated sludge, and changes in filtration resistance measured by the batch cell filtration test were evaluated. Both the filtration resistance and the specific cake resistance were minimized at 20 mg/g-MLSS with chitosan, 70 mg/g-MLSS with ferric chloride, and 20 mg/g-MLSS with MPE50 addition, respectively. Introduction of the additives into the activated sludge resulted in reduction of not only cake resistance, but also fouling resistance. However, the chitosan addition to three different activated sludge resulted in three different optimal dose of 10, 20, 30 mg/g-MLSS, respectively. This implies that the optimal dose is dependent on sludge characteristics rather than a constant value. Overdose above the optimal dosage always aggravated filterability in all cases. Zeta potential of sludge flocs, relative hydrophobicity, floc size distribution, soluble EPS concentration and supernatant turbidity were measured in order to analyze fouling reduction mechanism. Nearly neutral surface charge along with the largest particle size was observed at the optimal dose. This could be explained by particle destabilization and restabilization mechanism as positively charged additives were injected into sludge flocs of negative surface charge. Both soluble EPS concentration and supernatant turbidity also showed the lowest value at the optimal dose. These foulants are believed to be coagulated and entrapped in sludge flocs during flocculation. Chitosan and MPE50 which are cationic polymeric substances showed higher reduction in both soluble EPS and fine particles comparing with ferric chloride.

Characterization of Behavior of Colloidal Zero-Valent Iron and Magnetite in Aqueous Environment (나노크기의 교질상 영가철 및 자철석에 대한 수용상의 거동특성)

  • Nano-sized iron colloids are formed as acid mine drainage is exposed to surface environments and is introduced into surrounding water bodies. These iron nanomaterials invoke aesthetic contamination as well as adverse effects on aqueous ecosystems. In order to control them, the characteristics of their behaviour should be understood first, but the cumulative research outputs up to now are much less than the expected. Using zero-valent iron (ZVI) and magnetite, this study aims to investigate the behaviour of iron nanomaterials according to the change in the composition and pH of background electrolyte and the concentration of natural organic matter (NOM). The size and surface zeta potential of iron nanomaterials were measured using dynamic light scattering. Characteristic behaviour, such as aggregation and dispersion was compared each other based on the DLVO (Derjaguin, Landau, Verwey, and Overbeek) theory. Whereas iron nanomaterials showed a strong tendency of aggregation at the pH near point of zero charge (PZC) due to electrostatic attraction between particles, their dispersions became dominant at the pH which was higher or lower than PZC. In addition, the behaviour of iron nanomaterials was likely to be more significantly influenced by cations than anions in the electrolyte solutions. Particularly, it was observed that divalent cation influenced more effectively than monovalent cation in electrostatic attraction and repulsion between particles. It was also confirmed that the NOM enhanced the dispersion nanomaterials with increasing the negative charge of nanomaterials by coating on their surface. Under identical conditions, ZVI aggregated more easily than magnetite, and which would be attributed to the lower stability and larger reactivity of ZVI.

Electrostatic Formation of Chitosan-Polyacrylate Polyplex for the Preparation of Cross-Linked Hydrogel Particles (이온성 상호작용을 통한 키토산-폴리아크릴산 Polyplex의 형성 및 이를 이용한 하이드로젤 특성 분석)

  • Despite the great potential for the versatile applications in food industry and medical area, chitosan as a biocompatible cationic polysaccharide has suffered from the limited solubility under physiological condition. Herein, we demonstrated the electrostatic formation of chitosan-based polyplex particles, counterbalanced by polyacrylate as an anionic polyelectrolyte. The resulting polyplex exhibited pH- and composition-dependent changes in their surface charges as measured by zeta potential, which can be employed to provide the interparticle repulsive forces for enhanced colloidal stability in homogeneous solution. Subsequently, amide coupling between the acrylates and glucosamine residues of chitosan inside the polyplex further generated the hydrogel particles, which showed the temperature-sensitive swelling property. This aspect can be attributed to the partial formation of acryl amide residues, which have been generally known to possess the lower critical solution temperature (LCST).

Preparation of Fe2O3 Coated on Mica for Infrared Reflectance Red Pigment and Thermal Property of Its Isolation-Heat Paint (Fe2O3가 코팅된 판상 mica의 적외선 반사용 적색안료 제조 및 차열도료의 열특성)

  • $Fe_2O_3$ coated plate mica($Fe_2O_3$/mica) for infrared reflectance red pigment was prepared under hydrothermal treatment. $Fe_2O_3$ was perfectly coated on mica via the difference of surface charge between $Fe_2O_3$ and mica particles at pH 3. $Fe_2O_3$/mica was then calcined at $800^{\circ}C$ to stabilize the coated layer on mica. The infrared(IR) reflectance pigments were characterized by X-ray diffraction, FE-SEM, zeta potential, and a UV-Vis-NIR spectrophotometer. In particular, the CIE color coordinate and IR reflectance properties of $Fe_2O_3$/mica pigments were investigated in relation to the thickness variation of the $Fe_2O_3$ layer coated on mica of various lateral sizes. The isolation-heat red paints containing the pigments were prepared and optimized with a thinner, settling agent, and dispersant. Then, the films were made. The thermal property of isolation-heat on these films was observed through the relationship of the IR reflectance value, which was based on the variation of the $Fe_2O_3$ layer's thickness coated on mica and mica's lateral size as IR reflectance pigment. With an increase in IR reflectance on these films, the thermal property of isolation-heat was effectively enhanced.

Preparation of ZnO@TiO2 nano coreshell structure by the polymerized complex and sol-gel method (착체중합법과 sol-gel법에 의한 ZnO@TiO2 나노 코아쉘 구조의 제조)

  • Nano core shell structures of $TiO_2$ particles coated on surface of ZnO nanoparticles were prepared by the polymerized complex and sol-gel method. The average particle size of ZnO by the polymerized complex method showed 100 nm and the average particle size of $TiO_2$ by the sol-gel method showed below 10 nm. The average particle size of $ZnO@TiO_2$ nano core shell struture represented about 150 nm. The agglomeration between the ZnO particles using the polymerized complex method was highly controlled by the uniform absorption of $TiO_2$ colloid on the spherical ZnO surfaces. The driving force of heterogeneous bonding between ZnO and $TiO_2$ was induced by the Coulomb force. The ZnO and $TiO_2$ particles electrified with + and - charges, respectively, resulted in strong bonding by the difference of iso-electric point (IEP) when they laid neutrality pH area, depending on the heterogeneous surface electron electrified by the different zeta potential on the pH values.

Development and Characterization of a Hydrolyzed Goat Milk Protein/Chitosan Oligosaccharide Nano-Delivery System (산양유 단백질 분해물/키토올리고당 나노 전달체 제조 및 물리화학적 특성연구)

  • The aims of this study were to manufacture a hydrolyzed goat milk protein (HGMP)/chitosan ologisaccharide (CSO) nano-delivery system (NDS) and to investigate the effects of production variables, such as sodium tripolyphosphate (TPP), HGMP, and CSO concentration levels, on the formation and physicochemical properties of the NDS. An HGMP/CSO NDS was produced using the ionic gelation method at pH 5.5. Transmission electron microscopy and a particle size analyzer were used to determine the morphological and physicochemical properties of NDSs, respectively. The size of the HGMP/CSO NDS decreased from 225 to 138 nm as HGMP and CSO concentration levels decreased. The NDS had a positive surface charge, with a zeta-potential value of +23 mV. The encapsulation efficiency (EE) of docosahexaenoic acid was enhanced as the HGMP concentration level increased. Additionally, increasing the concentration level of CSO resulted in an increase in the EE of resveratrol. The HGMP/CSO NDS exhibited good physical stability during freeze-drying. Thus, our findings showed that the HGMP/CSO NDS was successfully manufactured and that HGMP and CSO concentration levels were key factors affecting the physicochemical properties of the NDS.

Influence of Ionic Strength, pH, and Complex-forming Anions on the Adsorption of Cesium-137 and Strontium-90 by Kaolinite (카올리나이트에 의한 세슘-137 및 스트론튬-90 흡착에 대한 이온강도, pH, 복합체-형성 음이온의 영향)

  • The effects of the major cations ($Ca^{2+}$, $Mg^{2+}$, $K^+$, $Na^+$), complex-forming anions ($SO_4{^{2-}}$, $HCO_3{^-}$), and solution pH on the adsorption of $^{137}Cs$ and $^{90}Sr$ by kaolinite in groundwater chemistry were investigated. Three-dimensional Kd modelling designed by a statistical method was attempted to compare the relative effect among hydrated radii, charge and concentration of competing cations on the adsorption of Cs and Sr. The modelling results indicate that the hydrated radii of competing cations is the most important factor, and then their charges and concentrations are also important factors in order. The property of zeta potential of kaolinite particles was discussed in terms of the amphoteric reactions of a kaolinite surface affecting the adsorption of Cs and Sr. The ionic strength of competing cations on the adsorption of Cs and Sr exerts a greater effect than the solution pH. The sorption behaviour of Sr on kaolinite is also highly dependent on the concentration of bicarbonate. The speciation of Sr and the saturation state of a secondary phase were thermodynamically calculated by a computer program, WATEQ4F. This indicates that the change in solution pH with the concentration of bicarbonate and the precipitation of a strontianite ($SrCO_3$) are major factors controlling Sr adsorption behaviour in the presence of bicarbonate ion.

A Study on Fouling Characteristics and Applicability of Fouling Reducer in Submerged MBR Process (침지형 MBR공정에서 파울링 특성과 파울링 완화제의 적용성에 관한 연구)

  • Though MBR process has many advantages, the greatest risk factors in operating MBR process are occurrence of membrane fouling and decrease of flux. It is very difficult to find exact mechanism due to complex influence by many effects, although there have been recently many studies of membrane fouling. The purposes of this study are firstly evaluating bioreactor of lab-scale and micro-filtration hollow fiber membrane, secondly investigating correlation between foulants affecting membrane performance and membrane fouling, and lastly evaluating various parameters affecting fouling and applicability of membrane fouling reducer. This study found that TMP was increasing rapidly and showed 0.32 bar and the average of flux was 88 LMH. EPS concentration tends not to change much above MLSS concentration (6,000 mg/L). However, EPS concentration variation is wide below MLSS concentration (6,000 mg/L). Also, from results of membrane surface condition and element analysis using SEM/EDX, carbon and fluorine were founded to be the highest percentage in membrane because of characteristics of membrane material. In operating continuously, inorganic fouling was generated by increase of these inorganic substances such as $Al^{3+}$ and $Mg^{2+}$. Lastly, the best filtration performance was obtained for 0.03 mg MFR/mg MLSS by results of particle size, zeta potential, $SCOD_{cr}$, EPS and MLSS concentration.
