Optimized Serological Isolation of Lung-Cancer-associated Antigens from a Yeast Surface-expressed cDNA Library

  • The technique of serological analysis of antigens by recombinant cDNA expression library (SEREX) uses autologous patient sera as a screening probe to isolate tumor-associated antigens for various tumor types. Isolation of tumor-associated antigens that are specifically reactive with patient sera, but not with normal sera, is important to avoid false-positive and autoimmunogenic antigens for the cancer immunotherapy. Here, we describe a selection methodology to isolate patient sera-specific antigens from a yeast surface-expressed cDNA library constructed from 15 patient lung tissues with non-small cell lung cancer (NSCLC). Several rounds of positive selection using patient sera alone as a screening probe isolated clones exhibiting comparable reactivity with both patient and normal sera. However, the combination of negative selection with allogeneic normal sera to remove antigens reactive with normal sera and subsequent positive selection with patient sera efficiently enriched patient sera-specific antigens. Using the selection methodology described here, we isolated 3 known and 5 unknown proteins, which have not been isolated previously, but and potentially associated with NSCLC.

A study on the surface model and normal and tangential forces for power transmission flat belts (동력전달용 평벨트의 표면모델과 수직력 및 접선력에 대한 연구)

  • The normal and tangential belt forces for two types of flat belts are measured and compared. From friction theory, ti was assumed that tangential friction is proportional to the actual contact area $A_{a}$ and $A_{a}$ is proportional to normal pressure P; i.e., $A_{a}$ .var.P$^{n}$ . For a flat belt with cloth backing, the n=2/3 is obtained for the constant of belt surface model. For a flat belt with rubber backing, the n=0.9 to n=1.0 is suggested as a surface model constant. The theoretical equation developed in this paper showed agood with the experimental results.

Influence of soft drinks supplemented calcium to enamel remineralization (칼슘보강음료가 법랑질 재광화 효과에 미치는 영향)

  • Objectives: The purpose of this study was to identify the correlation between calcium in soft drinks and enamel remineralization. Method: Six soft drinks were used in this study. These were calcium milk, normal milk, calcium yoghurt, normal yoghurt, calcium orange juice, and orange juice. Enamel specimens which 300- 400Vickers Hardness Number (VHN) were selected. These samples were immersed in each soft drink for 12 hours in an in vitro remineralization model. All specimens were processed for SEM image of the enamel surface. Results were analyzed by SPSS 13.0 package program. Results: Calcium milk was the most influential and normal yoghurt was the least to enamel surface. There was not significant difference according to calcium supplement in milk although the difference of enamel hardness was ${\Delta}f13.4$ in calcium milk, and ${\Delta}f4.7$ in normal milk (P>0.05). Other soft drinks showed a little change about calcium but they were insignificant. Demineralization effect was remarkably observed in calcium yoghurt ($-{\Delta}f269.1$) and this effect was confirmed by SEM images. In conclusion, calcium supplemented soft drinks had little influences to hardness of enamel surface.

Study of moxibustion stimulation in the ovariectomized rat model

  • In order to examine the effectiveness of moxibustion stimulation for climacteric disturbance, we administered moxibustion stimulation to ovariectomized (OVX) rats, and compared the tail surface temperature, laboratory parameters, and the level of locomotor activity with those of untreated rats. Twenty-four female Wistar rats (8 weeks old, 160 - 180 g) were divided into three groups. The rats in the OVX-M group underwent moxibustion stimulation after ovariectomy. The rats in the OVX-C group underwent ovariectomy but did not receive moxibustion stimulation. The rats in the Normal group received neither ovariectomy nor moxibustion stimulation. The level of locomotor activity was determined by a metabolism measuring system. The tail surface temperature was significantly lower in the OVX rats before moxibustion stimulation than in the Normal group. In the OVX rats before moxibustion stimulation, there was no clear difference in the level of locomotor activity between the active and resting phases, and the pattern of locomotor activity was irregular. After moxibustion stimulation, the tail surface temperature of the OVX-M group did not significantly differ from that of the Normal group, and the pattern of locomotor activity of the OVX-M group became diphasic with clear active and resting phases, similar to that observed in the Normal group. These results demonstrate that moxibustion stimulation is effective for the treatment of climacteric disturbance.

Identification and Purification of a Normal Rat Liver Plasma Membrane Surface Protein which Disappears after Chemical Carcinogenesis

  • The electrophoretic patterns of plasma membrane surface proteins of normal rat liver cells and rat hepatomas were compared in 10% non-denaturing and 7-15% gradient non-denaturing gel. Chemical carcinogens, 2-Me DAB (2-methyl-4-dimethylaminoazobenzene) and DENA (diethylnitrosamine), were used to induce hepatoma in rats. One protein which disappeared in hepatoma was identified in normal rat liver by non-denaturing gel electrophoresis. Rabbit antisera were raised against this specific protein, and the protein was purified by Sephacryl S-200 column and immunoaffinity chromatography using the purified antibody. The purified protein showed two bands of molecular weights approximately 50 $kD_{\alpha}$ and 52 $kD_{\alpha}$ by SDS-polyacrylamide gel electrophoresis, which reacted specifically with the antibody. However only one band was observed in non-denaturing gel and also in isoelectric focusing with a pI value of 6.6. This study showed the existence of an unique protein on the plasma membrane surface of normal rat liver cells which disappeared in rat hepatomas induced by chemical carcinogens.

A Study on the Parameter Estimation of DURUMI-II for the Fixed Right Elevator Using Flight Test Data

  • The stability and control derivatives of DURUMI-lI UAV using the flight test are obtained. The flight test data is gathered from the normal flight condition (normal mode) and the flight condition assumed as the right elevator fixed (fault mode). Using real-time parameter estimation techniques, applied to Fourier transform regression method, simulates the aircraft motion. From the result, the fault of control surface is to be detected. In this paper, the results of the real- time parameter estimation techniques are compared with the results of the Advanced Aircraft Analysis (AAA). Using the aerodynamic derivatives, it provides the base line of normal/failure for the control surface by using the on-line parameter estimation of Fourier transform regression. In flight, this approach maybe helpful to detect and isolate the fault of primary control surface. It is explained how to perform the flight condition assumed as the right elevator fixed in the flight test. Also, it is mentioned how to switch between the normal flight condition and the assumed fault flight condition.

A new simple shear and normal deformations theory for functionally graded beams

  • In the present work, a simple and refined trigonometric higher-order beam theory is developed for bending and vibration of functionally graded beams. The beauty of this theory is that, in addition to modeling the displacement field with only 3 unknowns as in Timoshenko beam theory, the thickness stretching effect (${\varepsilon}_Z{\neq}0$) is also included in the present theory. Thus, the present refined beam theory has fewer number of unknowns and equations of motion than the other shear and normal deformations theories, and it considers also the transverse shear deformation effects without requiring shear correction factors. The neutral surface position for such beams in which the material properties vary in the thickness direction is determined. Based on the present refined trigonometric higher-order beam theory and the neutral surface concept, the equations of motion are derived from Hamilton's principle. Numerical results of the present theory are compared with other theories to show the effect of the inclusion of transverse normal strain on the deflections and stresses.

  • Nano adhesion and friction between a Sj$_3$N$_4$ AFM tip and thin silver films were experimentally studied. Tests were performed to measure the nano adhesion and friction in both AFM(atomic force microscope) and LFM(lateral force microscope) modes in various ranges of normal load. Thin silver films deposited by IBAD (ion beam assisted deposition) on Si-wafer (100) and Si-wafer of different surface roughness were used. Results showed that nano adhesion and friction decreased as the surface roughness increased. When the Si surfaces were coated by pure silver, the adhesion and friction decreased. But the adhesion and friction were not affected by the thickness of IBAD silver coating. As the normal force increased, the adhesion forces of bare Si-wafer and IBAD silver coating film remained constant, but the friction forces increased linearly. Test results suggested that the friction was mainly governed by the adhesion as long as the normal load was low.

  • In this paper an experimental study is presented of the problem of dynamic behavior of a water droplet impinging upon a heated surface. The experiments are mainly focused on the effects of impinging angle of a droplet and surface temperature on the impact dynamics of the droplet. It Is clarified that the droplet exhibits much different behavior depending on the normal momentum of an impinging droplet before impact. At surface temperature In the nucleate boiling regime. the disintegration of a droplet doesn't occur, whereas the deforming droplet adheres to the surface. The spreading and contraction of the liquid film is repeated a couple of times for the horizontal surface but the expanded droplet just slips without noticeable contraction for the inclined surfaces. In the film boiling regime, the impinging droplet spreads over the surface as a liquid film which is separated from the surface by produced vapor. Depending on the magnitude of the normal momentum of the droplet the disintegration into the several irregular shapes of liquid elements occurs for the horizontal and 30o-inclined surfaces, whereas the impinging droplet for the 60o-inclined surface doesn't break up and tends to recover the original spherical shape.

Capabilities of stochastic response surface method and response surface method in reliability analysis

  • The stochastic response surface method (SRSM) and the response surface method (RSM) are often used for structural reliability analysis, especially for reliability problems with implicit performance functions. This paper aims to compare these two methods in terms of fitting the performance function, accuracy and efficiency in estimating probability of failure as well as statistical moments of system output response. The computational procedures of two response surface methods are briefly introduced first. Then their capabilities are demonstrated and compared in detail through two examples. The results indicate that the probability of failure mainly reflects the accuracy of the response surface function (RSF) fitting the performance function in the vicinity of the design point, while the statistical moments of system output response reflect the accuracy of the RSF fitting the performance function in the entire space. In addition, the performance function can be well fitted by the SRSM with an optimal order polynomial chaos expansion both in the entire physical and in the independent standard normal spaces. However, it can be only well fitted by the RSM in the vicinity of the design point. For reliability problems involving random variables with approximate normal distributions, such as normal, lognormal, and Gumbel Max distributions, both the probability of failure and statistical moments of system output response can be accurately estimated by the SRSM, whereas the RSM can only produce the probability of failure with a reasonable accuracy.
