Mutual Separation of Am(III) and Eu(III) by Solvent Extraction with Cyanex 301 (Cyanex 301에 의한 Am(III)과 Eu(III)의 상호분리)

  • The present paper describes the solvent extraction behaviors and the mutual separation of Am and Eu by sulphur bearing Cyanex 301 acidic extractant in sodium nitrate solutions. Original Cyanex 301 was not able to separate the Am and Eu because of their similar extraction behaviors. The saponification of commercial Cyanex 301 was performed by small amounts of 8 M NaOH solutions and saponified Cyanex 301 was able to separate of Am from Eu with high selectivity in sodium nitrate aqueous solution. The separation factors ($SF_{Am/Eu}$) were increased with saponified ratio of commercial Cyanex 301, pH of sodium nitrate solution and initial concentration of Eu. To obtain the homogeneous saponified Cyanex 301, heterogeneous saponified Cyanex 301 was treated with addition of octyl alcohol or filtration. The observed $SF_{Am/Eu}$ was 32.3 for the former and 930 for the latter. Finally, the stripping behaviors of Am and Eu were similar and stripping yields showed 96.1% for the 1 M $NaNO_3$(pH=1.3) and 99% for the mixture solution of 0.05 M DTPA and 1.5 M lactic acid.

Characterization on Fatty Acids and Amino Acids of Quality Protein Maize Lines (Quality Protein Maize 육성계통의 지방산 및 아미노산 특성)

  • This study was carried out to evaluate the nutritional properties of quality protein maize (QPM) and to select the elite lines for corn breeding. Two laboratory procedures for simultaneous identification of QPM and lysine content analysis were performed. The $BC_{1}F_{2}$ lines of KS5/QPM and KS135/QPM were analyzed with opaque-2(o2) specific SSR marker in order to differentiate the opaque-2 carrying QPM lines from the non-opaque genotypes. Although no significant difference in protein content, significant differences in lipid, ash, and crude fiber contents were observed. The composition of unsaturated fatty acid of QPM lines was slightly lower than non-QPM lines, but there was no significant difference. Sulphur-containing amino acids such as methionine and cystine showed no difference between QPM and non-QPM lines. However, lysine content of QPM lines was 38% higher than that of non-QPM lines, and the essential amino acid content of QPM lines (28.1%) was higher compared to non-QPM lines (27.1%).

Accumulation of Heavy Metals in Soil Growing for Red Pepper (Capsicum annuum) with using Lime Bordeaux and Lime Sulphur Mixture

  • Lime bordeaux mixture (LBM) and lime sulfur mixture (LSM) are representative environmental friendly organic materials for prevention of insect pests in South Korea. Recently, those have been widely used as an alternative for chemical pesticides in eco-friendly farms. However, South Korea has not established even recommendation of LBM and LSM considering the stability of heavy metals in soil. The aim of this study was to evaluate the accumulation of hazardous heavy metals in soil and plant with long-term application of LBM and LSM. Firstly, we investigated the amount of LBM and LSM used per year in several eco-friendly farms to determine a standard application rate of both materials. The pepper plant was grown on the pot in greenhouse for 14 weeks. Both materials were applied at 0, 1, 3, and 9 times of standard application rates (2.56 and $1.28L\;ha^{-1}$ of LBM and LSM per year, respectively). Dry matter yield of pepper and heavy metals (As, Cd, Cu, Hg, Ni, Pb, and Zn) concentration in soil and pepper plant were measured after 14 weeks. Yield of pepper plant did not significantly chang with up to application rate of 1 times, thereafter it markedly decreased with more than 3 times. With increasing LBM and LSM application, the concentration of Cu and Zn in soil significantly increased. Especially, Zn concentration in pepper significantly increased with increasing application rates of both materials. This might resulted in significant decrease in dry matter yield of pepper. The concentrations of those heavy metals in soil did not exceed safety levels ($150mg\;kg^{-1}$ for Cu and $300mg\;kg^{-1}$ for Zn) established by the Korean Soil Environmental Conservation Act as well as concentration of heavy metals in pepper plant by Korean Ministry of Food and Drug Safety. However, particular attention should be paid for heavy metal safety and crop productivity when using LBM and LSM in the organic farm.

Crystallographic and Magnetic Properties of Cu0.1Fe0.9Cr2S4 (Cu0.1Fe0.9Cr2S4의 결정학적 및 자기적 성질에 관한 연구)

  • Cu$_{0.1}$Fe$_{0.9}$Cr$_2$S$_4$ has been studied with Mossbauer spectroscopy, x-ray diffraction, vibrating sample magnetometer (VSM), and magnetoresistance (MR) measurement. The crystal structure was determined to be a cubic spinel with lattice parameter a$_{0}$=9.9880 $\AA$. The MR measurements show a semiconductor behavior below 110 K and metal behaved above 100 K. The temperature dependence of magnetization of Cu$_{0.1}$Fe$_{0.9}$Cr$_2$S$_4$ was reported. In addition to a large irreversibility between the zero-field-cooling (ZFC) and the field-cooling (FC) magnetization at applied field H=100 Oe, a cusp-like anomaly was observed in both the FC and ZFC curves. It shifted toward the lower temperature region with increasing magnetic field, and then showed convex type maximum at 110 K, under the applied field of 5 kOe. The Mossbauer spectra were measured from 15 K to room temperature. The asymmetric line broadening was observed for the sample Cu$_{0.1}$Fe$_{0.9}$Cr$_2$S$_4$, and it was considered to be dynamic Jahn-Teller relaxation. The charge state of Fe ions was ferrous in character. The unusual reduction of magnetic hyperfine field below 110 K was interpreted in terms of cancellation effect between the mutually opposite orbital current field (H$_{L}$) and Fermi contact field (H$_{C}$).

Effect of SO2 Generating Pad Treatments on the Quality of Dried Persimmons during Storage (SO2 발생패드처리가 곶감의 저장 중 품질에 미치는 영향)

  • The effects of $SO_2$ generating pads ($SO_2$ 0, 0.5, 1, and 2 g/kg) on the quality of dried persimmons slices were investigated. The $SO_2$ generating pads for storage did not affect to weight, moisture loss rate, and soluble solid contents of dried persimmons. The color change(E) after storage for 12 weeks was the highest (value = 3.5) in control ($SO_2$ 0 g/kg), whereas that was the lowest (value = 2.6) under $SO_2$ 2 g/kg condition. When we measured the browning degree after 12 weeks, they showed O.D. 0.15, 0.14, 0.10, and 0.05 in serial dilution treated pads with $SO_2$ 0, 0.5, 1, and 2 g/kg, respectively. The decaying rate was the highest (9.0%) in control after 12 weeks storage, whereas it did not show any spoilage in $SO_2$ 2 g/kg treated condition for whole storage period. The concentration of residual $SO_2$ in dried persimmons was detected within a safe range of 3.3~97.0 ppm. Therefore, the shelf-life of dried persimmons was lengthened in $SO_2$ generating pads (especially in $SO_2$ 2 g/kg) for inhibiting of browning and decaying.

Nutritional Quality and Variation of Meat and Bone Meal

  • Meat and bone meal is a valuable protein and mineral source in diets of production animals and contributes to the protein, energy and mineral component of diets. The aim of the present study was to more accurately characterise the apparent ileal amino acid digestibility of meat and bone meals produced in New Zealand and evaluate routine in vitro assays used in practise to measure meat and bone meal quality. A total of 94 commercial meat and bone meals from 25 New Zealand rendering plants over a two and a half year period were analysed for proximates, gross energy, gross amino acid content (incl. hydroxyproline, hydroxylysine and lanthionine), apparent ileal amino acid digestibility, pepsin nitrogen digestibility, protein solubility and bone content. The mean crude protein content of the 94 meat and bone meal samples was 56.8% with a range of >35% units and a coefficient of variation of 9.8%. The mean crude fat and ash content were 10.0 and 28.4% respectively. These latter components showed a large range (16 and 43%, respectively) with coefficients of variation above 22%. Amino acid digestibility between samples was highly variable with lysine and sulphur amino acids digestibility ranging between 45.8-89.0 and 38.2-85.5%, respectively. Pearson correlation coefficients are presented between crude protein content and individual gross amino acids, crude protein content and individual digestible amino acid content, and pepsin N digestibility and individual digestible amino acid content. There was a significant relationship between the digestible amino acid nitrogen content and the crude protein content while pepsin nitrogen digestibility was not correlated to ileal amino acid nitrogen digestibility (r=-0.06). Meat meals with a high protein content had relatively low hydroxyproline and hydroxylysine levels something that was attributed to the levels of collagen from bone. The data indicated that lanthionine (formed upon heat treatment of cysteine with a hydroprotein) is not a good indicator of the heat treatment employed to meat and bone meals. Step-wise multiple regression equations to predict the apparent digestible content of amino acids from rapid in vitro assays are presented. The most selected variables included ash and crude fat content. In general the equations derived for the essential amino acids had a higher degrees of fit (R2) compared to the non-essential amino acids. The R2 for the essential amino acids ranged from 0.43 for histidine and 0.68 for leucine. These equations provide a means of more rapidly estimating the apparent ileal digestible amino acid content (protein quality) of meat and bone meal using standard analyses.

Input, Output and Budget of Nitrogen and Sulphur in Forested Watershed Ecosystems (산림 소유역 생태계에서 질소와 황의 유입량, 유출량과 물질수지)

  • In order to elucidate the budget and cycling of Nitrogen and Sulfur, esential elements and principal constituents of acid rain, their input through precipitation, and their output by streamflow were quantified in coniferous and deciduous forested watersheds, using combination of nutrient concentration and hydrological analysis, in Kwangnung Experimental Forest from July 1991 to Decmeber 1993. Amount of annual mean precipitation was 12.916 ton·ha/sup -1/·yr/sup -1/, annual mean runoff 5,094 ton·ha/sup -1/·yr/sup -1/(39%), 7,647 ton·ha/sup -1/·yr/sup -1/(59%) in coniferous and deciduous forest watersheds, respectively. Amounts of annual input of N(NO₃/sup -/+NH₄/sup +/) and SO/sup 2-/₄ through preciptation were 12.5, 81.7 kg·ha/sup -1/·yr/sup -1/, respectively. Annual output via runoff of N(NO₃/sup -/+NH₄/sup +/) and SO/sup 2-/₄were 0.06, 39.23 ton·ha/sup -1/·yr/sup -1/ in the coniferous forest watershed ecosystem, and 0.15, 55.46 ton·ha/sup -1/·yr/sup -1/ in the deciduous one, respectively. On the basis of annual nutrient input and output, the annual budget of N(NO₃/sup -/+NH₄/sup +/) and SO/sup 2-/₄were +12.46, +42.49 ton·ha/sup -1/·yr/sup -1/ in the coniferous forest watershed, and +11.35, +26.26 ton·ha/sup -1/·yr/sup -1/ in the deciduous one. Thus N(NO₃/sup -/+NH₄/sup +/) and SO/sup 2-/₄were accumulated in both forested watershed ecosystems.

The Effect of Feeding Different Levels of Sardine Fish Silage on Broiler Performance, Meat Quality and Sensory Characteristics under Closed and Open-sided Housing Systems

  • Two experiments were conducted to evaluate the use of fish silage prepared from Indian oil sardines, Sardinella longiceps, as partial replacement of soybean meal as a sole source of protein for growing broiler chickens. The main objective of Experiment 1, an ileal digestibility assay, was to assess the nutritional value of fish silage compared with soybean meal for feeding broiler chickens. The two test ingredients, soybean meal and dried fish silage, were incorporated into semi-synthetic diets, as the only component containing protein. The ileal digestibility coefficients of amino acids of fish silage were considerably higher than those of soybean meal (p<0.001). The lower digestibility of amino acids from soybean meal was related to the presence of anti-nutritional factors such as trypsin inhibitors. Fish silage had higher levels of sulphur-containing amino acids than soybean meal. The objective of Experiment 2, a growth study, was to evaluate the effect of feeding fish silage on performance and meat quality characteristics of broiler chickens raised under closed and open-sided housing systems. Four diets containing various levels of fish silage (0, 10, 20 and 30%) were evaluated. Daily feed intake, body weight gain and feed conversion ratio were measured. At the end of Experiment 2, 96 birds were randomly selected and slaughtered to evaluate meat quality characteristics. Housing type had significant effects on feed intake and body weight gain (p<0.01). Birds in the open-sided house consumed 4.7% less amount of feed and gained 10.6% less than their counterparts in a closed house. Birds in both houses fed diets containing 10 and 20% fish silage gained more than birds fed 30% fish silage. The current study produced evidence that fish silage can replace up to 20% of soybean meal in broiler diets without affecting either growth performance or the sensory quality of broiler meat.

Acute Toxicity Evaluation of Loess-sulfur Complex in different pH (황토유황합제의 pH차이에 따른 급성독성평가)

  • Loess-sulfur complex has been widely used as an environmental friendly organic materials for insecticides and fungicides in crop cultivation. However, there are high concerns about skin and eye irritation for farm workers due to the high alkaline properties of loess-sulfur complex. The acute toxicity evaluation was conducted with three samples of loess-sulfur complex in different pH (pH 9, 10, 11) in order to supply the evidentiary data for selecting the optimal product among the test materials. The results of acute oral toxicity using rats showed $LD_{50}$ of over 2,000 mg/kg b.w. for all three samples of loess-sulfur complex. The calculated acute dermal $LD_{50}$ of all tested materials was over 4,000 mg/kg b.w.. The Skin and eye irritation indicated that all tested materials have no irritation. Consequently, it was suggested that loess-sulfur complex be low in acute toxicity at all different pH values (pH 9~11).

Transgenic Plants with Enhanced Tolerance to Environmental Stress by Metabolic Engineering of Antioxidative Mechanism in Chloroplasts (엽록체 항산화기구 대사조절에 의한 환경스트레스 내성 식물)

  • Injury caused by reactive oxygen species (ROS), known as oxidative stress, is one of the major damaging factors in plants exposed to environmental stress. Chloroplasts are specially sensitive to damage by ROS because electrons that escape from the photosynthetic electron transfer system are able to react with relatively high concentration of $O_2$ in chloroplasts. To cope with oxidative stress, plants have evolved an efficient ROS-scavenging enzymes such as superoxide dismutase (SOD) and ascorbate peroxidase (APX), and low molecular weight antioxidants including ascorbate, glutathione and phenolic compounds. To maintain the productivity of plants under the stress condition, it is possible to fortify the antioxidative mechanisms in the chloroplasts by manipulating the antioxidation genes. A powerful gene expression system with an appropriate promoter is key requisite for excellent stress-tolerant plants. We developed a strong oxidative stress-inducible peroxidase (SWPA2) promoter from cultured cells of sweetpotato (Ipomoea batatas) as an industrial platform technology to develop transgenic plants with enhanced tolerance to environmental stress. Recently, in order to develop transgenic sweetpotato (tv. Yulmi) and potato (Solanum tuberosum L. cv. Atlantic and Superior) plants with enhanced tolerance to multiple stress, the genes of both CuZnSOD and APX were expressed in chloroplasts under the control of an SWPA2 promoter (referred to SSA plants). As expected, SSA sweetpotato and potato plants showed enhanced tolerance to methyl viologen-mediated oxidative stress. In addition, SSA plants showed enhanced tolerance to multiple stresses such as temperature stress, drought and sulphur dioxide. Our results strongly suggested that the rational manipulation of antioxidative mechanism in chloroplasts will be applicable to the development of all plant species with enhanced tolerance to multiple environmental stresses to contribute in solving the global food and environmental problems in the 21st century.
