Dongchimi Fermentation for Nangmyeon - Changes of Chemical Property during Fermentation - (냉면육수용 동치미 제조-발효과정 중 화학적 성질의 변화-)

  • One percent starch-added Dongchimi, and Dongchimi which 1% starch was not added. each using, 70% radish cut in 2cm size were fermented at 4$^{\circ}C$ for 30 days. HPLC and TLC analyses showed that the no starch-added Dongchimi had glucose, fructose, sucrose. The starch-added Dongchimi produced, mal-tose. maitotriose and maltotetraose by the hydrolysis reaction of amalyse. And the sugar contents were reduced in the process of fermentation. No starch-added Dongchimi showed 361 ${\mu}$g/ml of reducing sugar, 0.012 unit /ml of amylase activity, 3.82 of pH, 3.0 of acidity. The 1% starch-added Dongchimi showed 329 ${\mu}$g/ml of reducing sugar, 0.04 unit/ml of amylase acidity, 3.8 of pH. 3.15 of acidity.

Effect of Controlled Atmosphere and Modified Atmosphere Storage on the Chemical Properties of Fresh and Red Ginseng (CA(controlled atmosphere storage) 및 MA(modified atmosphere storage)저장이 수삼 및 홍삼의 화학적 특성에 미치는 영향)

  • In order to stabilize the price of fresh ginseng by extension of seasonal variation and marketing structure, and to reduce the cost of production by works of intensive and short term, studies were carried out. As fresh ginseng of 4 years old by the MA (modified atmosphere storage) and CA (controlled atmosphere storage) was stored for 12 week at 4$^{\circ}C$, samples were collected after every 1, 2, 3, 4, 6, 8, 12 weeks for processing red ginseng. Chemical characteristics on ginseng steamed red for the quality evaluation are summarized as follows. The composition of free sugar was consisted of fructose, glucose, sucrose, and maltose in fresh ginseng. And small change was revealed in free sugar composition during the storage period. Glucose and sucrose were largely increased in both treatments of MA (modified atmosphere storage) and CA (controlled atmosphere storage) after 4 week, wherein maltose was decreased. Fifteen different amino acids were identified, and total amount of histamine and arginine were over 40%. Phenylalanine was slightly decreased. As the volatile components in control red ginseng, six different ones including triisopropyl benzene were detected. B-1 ($CO_2$ : $O_2$ : N2=6 : 4.90, DF-100) of CA and E-1 (CPP, DF-100) of MA did not shown triisopropyl benzene as the volatile components. And ${\gamma}$-muurolene, guaiaene, and beta-patchoulene were reduced, but the other components showed the trend of increase.

Changes in Physicochemical Properties of Packaged Backsulgi during Storage (포장한 백설기의 저장과정 중 이화학적 변화)

  • Packaged Backsulgi cooked by steam/convection oven and then rapidly chilled was examined by research of microbiological test and sensory evaluation while storing them at the temperatures of 3$^{\circ}C$ and 3$0^{\circ}C$ for 0, 2, 4, 6, 10 days . The pH and reducing sugar content were seemed to change little at 3$^{\circ}C$. However the pH was rapidly reduced until 4 days and then decreased a little at 3$0^{\circ}C$, the reducing sugar content was inclosed little by little. In the microbiological test, any microbial growth in total aerobic, psychrophilic, anaerobic, spore forming bacteria, yeast and molds was not observed until 10 days at 3$^{\circ}C$, but microbial changes of aerobic, psychrophilic and anaerobic bacteria increased to 6 logCFU/g until 10 days at 3$0^{\circ}C$. However microbial changes of them decreased from 6 logCFU/g to 5 logCFU/g. As a result of the sensory evaluation, appearance, taste, color, softness, chewiness and overall Quality were significantly decreased during storage times(p<0.05), but scores of taste and overall quality on 6th days were 7.38${\pm}$1.06, 7.00${\pm}$0.93. Therefore we concluded that there was no problem about stability of storage 6 days at 3$^{\circ}C$.

Daegu citizens' perceptions and factors affecting behavioral intentions to reduce sugars in the coffee shop beverages (커피전문점 음료의 당류 줄이기에 대한 대구시민의 인식 및 행동의도에 영향을 미치는 요인)

  • Purpose: This study aimed to provide baseline data for establishing a sugar reduction policy at coffee shops by analyzing the factors that affect a coffee shop user's perception and behavioral intention of reducing sugar intake. Methods: An online survey was conducted involving 1,274 Daegu citizens aged 19-49 years, who had visited coffee shops within the last month. Results: When visiting a coffee shop, the purchase of sweet drinks was higher in the younger age group, and the addition of syrup or sugar was higher in the older age group. Of the total respondents, 42.1% were aware that some coffee shops accommodate reduced sugar requests, 57.9% perceived the need to reduce sugar in coffee shop beverages and 22.3% had purchased beverages intending to reduce their sugar intake. In addition, 59.7% knew about sugar nutrition labeling, and 68.8% perceived the need for nutrition labeling for sugar. When purchasing beverages, 35.6% checked the nutrition labeling, and 77.2% purchased alternative drinks when the sugar content was high. Guiding the choice of sweetness levels in coffee shop orders was seen to have the highest effectiveness and intention to reduce sugar intake. Moreover, the perceived need to reduce sugar intake had the most positive effect on the behavioral intention to reduce sugars in coffee shop beverages (β = 0.558, p < 0.001). Conclusion: Although the overall awareness and practice of reducing the sugar intake in coffee shop users were low, the behavioral intention to reduce sugars was positive, and this was most affected by the perception of the need to reduce sugars. Therefore, there is a need for differentiated education and promotion for each age group for recognizing the necessity and outlining methods for reducing sugar intake. Furthermore, coffee shops should reflect customer's sugar reduction needs.

  • Dihaploid lines of tobacco (Nicotiana Tabacum L.) were developed from vitro anther culture of single cross F 1, "SC 72"X "Hicks". All genetic materials were evaluated for agronomic performance and chemical composition. Compared with midparent value, pollen derived dihaploid progenies showed reduced vigor in all measured features of plant growth, and increased total alkaloids as well as decreased reducing sugar content.

An Effect of Red-ripe Persimmon on Fermentation and Sensory Characteristics of Kimchi (연시첨가가 김치의 발효와 기호성에 미치는 영향)

  • This paper presents an investigation of chemical and sensory properties of Kimchi with red-ripe persimmon. Kimchi with different levels(0, 5, 10, 15%) of real-ripe persimmon was fermented at 4$^{\circ}C$ for 20days. During the fermentation, pH decreased in all Kimchi samples and pH of Kimchi with red-ripe persimmon was a little lower than that of Kimchi without red-ripe persimmon. Acidity increaased continuously and saltiness was maintained at 1.2∼2.2% levels during fermentation. The reducing sugar of Kimchi with red-ripe persimmon was relatively higher than that of the other sample without red-ripe persimmon. Total vitamin C(Vit. C) in Kimchi added red-ripe persimmon increased in the early stage of fermentation and reduced gradually after 4days of fermentation. In the sensory evolution, Kimchi with 10% red-ripe persimmon recorded high scores in taste, flavor, texture and overall acceptability during all fermentation period.

  • Some metabolites and embryo growth of primed tobacco (Nicotiana tabacum L. cv. ‘KFI09’) seeds were observed during priming. The seeds were primed at 15 and $25^{\circ}C$ for 1, 2, 3, 5, 10 and 15 days in a -0.8 MPa polyethylene glycol 6000 (PEG) solution. The time to 50% seed germination (T$_{50}$) was greatly reduced when the seeds were primed at $25^{\circ}C$ when compared with 15$^{\circ}C$. The $\alpha$-amylase activity and sugars and amino acid contents in the seeds primed at $25^{\circ}C$ greatly increased, while $\alpha$-amylase activity was similar, and sugar and amino acid contents increased slightly in the seeds primed at 15$^{\circ}C$. When the seeds were primed at $25^{\circ}C$, growth of the embryo which was enclosed by endosperm was detected, while the endosperm became thinner as the priming duration was extended.d.

Ethanol Production from the Seaweed Gelidium amansii, Using Specific Sugar Acclimated Yeasts

  • For the production of ethanol from seaweed as the source material, thermal acid hydrolysis and enzymatic saccharification were carried out for monosugars production of 25.5 g/l galactose and 7.6 g/l glucose using Gelidium amansii. The fermentation was performed with Pichia stipitis KCTC 7228 or Saccharomyces cerevisiae KCCM 1129. When wild P. stipitis and S. cerevisiae were used, the ethanol productions of 11.2 g/l and 6.9 g/l were produced, respectively. The ethanol productions of 16.6 g/l and 14.6 g/l were produced using P. stipitis and S. cerevisiae acclimated to high concentration of galactose, respectively. The yields of ethanol fermentation increased to 0.5 and 0.44 from 0.34 and 0.21 using acclimated P. stipitis and S. cerevisiae, respectively. Therefore, acclimation of yeasts to a specific sugar such as galactose reduced the glucose-induced repression on the transport of galactose.

  • The component and pharmacologic action on Dianthus sinensis L. were investigated as follows: 1) The components of Dianthus sinensis L. were suggested 8 kind of components by means of thin layer chromatography and paper chromatography. The one of them were suggested as a steroidal saponin by means of IR-spectrum and Liebermann-Burchard, Salkowiski, $SbCl_3$ reaction. 2) Sodium level in serum were reduced by administration of the sample to the rabbits, while in urine were elevated. 3) Blood sugar level of normal rabbits administered with sample, no significant variation were detected. 4) When the hyperglycemic rabbits treated with alloxan were administered sample, blood-sugar level were significantly decreased than control group.

  • Glycopeptide antibiotics, teicoplanin was purified from a mutant strain of Actinoplanes teichomyceticus ATCC31121, A. teichomyceticus MSL2211. We developed a simple procedure to separate and purify the teicoplanin from the fermentation broth. Teicoplanin was purified by two-step purification system, hydrophobic adsorption and sugar affinity chromatography in combination with HPLC analysis based on the properties of hydrophobic acyl chain and sugar moiety in teicoplanin. Teicoplanin was separated from the culture broth by Diaion HP-20 and further purified by concanavalin A affinity column chromatography. As an adsorbent resin, Diaion HP-20 in broth eliminated toxic effects on growth, reduced feedback repression of teicoplanin production, and assisted In rapid recovery of teicoplanin. The teicoplanin displayed the final yield of 80% and 95% of purity.
