Change in Sugar Composition of Ginseng Extract During Heat Treatment (인삼정의 추출 및 열처리 중 유리당의 함량변화)

  • The changes in free sugar composition were investigated with respect to the kinds of dried ginseng for extraction, the various ethanol concentrations used for ginseng extract manufacture and the conditions of heating temperature and time under which the ginseng extract was stored . The results are as follows: 1) The free sugar content of dried ginseng was 6.02-8.02% and the sucrose and maltose content in the free sugar was 70-80%. 2) The free sugar content was 13.82-26.29% in the Sanggunsam (dried ginseng of whole root) extract and it had a tendency to increase with increase in ethanol concentration. In addition, when a higher ethanol concentration was used, the sucrose content was in- creased but the maltose content was decreased. 3) The glucose, sucrose and maltose content in ginseng extract, decreased, in the order, as heating temperature and time were increased. On the other hand the opposite results were neted for xylose and fructose. Keywords Panax ginseng, ginseng extract, Sanggunsam.

  • In this study we tried to optimize the enzyme reaction conditions for the synthesis of highly branched isomaltodextrin (Mw > 2.5 kDa) using two dextransucrases from L. mesenteroides B-742CB and B-512FMCM that are dextransucrase constitutive mutants. As the concentration of sucrose or the ratio of maltose to sucrose increased, the amount of dextran decreased and the number and the amount of acceptor-products (of sucrose or maltose) increased. With high sucrose concentration (over 34%), there was more branched isomaltodextrin (as acceptor products) than dextran. When the ratio of sucrose to maltose was 2.5, there produced 86.7% of isomaltodextrin were produced. The Mw of dextrans, however, was over 2${\times}$10(sup)6 and there was no significant amounts of branched clinical dextran or high molecular weight oligosaccharides. With the combined activities of B-742CB dextransucrase and B-512FMCM dextransucrase we could synthesize high molecular weight branched isomaltodextrin (Mw>2.5 kDa). The high molecular weight dextran was composed of high branches as B-742CB dextran.

Monitoring of Roast Color Formation in Ginseng Using Threonine and Sucrose (Threonine과 Sucrose를 이용한 인삼의 색상 발현 모니터링)

  • The study investigated the roast color formation properties of ginseng upon soaking in threonine/sucrose solution followed by roasting. To determine operational parameters, including threonine concentration ($X_1$, 0.1~0.9%), sucrose concentration ($X_2$, 1.0~3.0%), and roasting temperature ($X_3$, $130{\sim}170^{\circ}C$), response surface methodology was applied to monitor color properties, including brown color intensity, Hunter's colors, and organoleptic color. Coefficients of determinations ($R^2$) of the models were above 0.8758 (P<0.05) in terms of brown color intensity and Hunter's color parameters. Brown color intensity of roasted ginseng extract was maximized in 0.70% threonine and 2.32% sucrose soaking solution under roasting conditions of 25 min at $166.03^{\circ}C$. a values of roasted ginseng were maximized in 0.74% threonine and 2.19% sucrose soaking solution under roasting conditions of $165.40^{\circ}C$. b values of roasted ginseng were maximized in 0.61% threonine and 2.28% sucrose soaking solution under roasting conditions of $159.16^{\circ}C$. The maximum organoleptic color score of roasted ginseng extract was 7.27 in 0.53% threonine and 1.01% sucrose soaking solution under roasting conditions of $146.96^{\circ}C$.

Studies on the Factors Influencing Survival Rates of Frozen Bovine Demi-Embryos (소 동결분할배의 생존선에 영향을 미치는 요인에 관한 연구)

  • This study was carried out to investigate the effects of concentration and kinds of cryoprotectants, equilibraction time, thawing temperature and time, sucrose concentration on the survival rates of frozen bovine demi-embryos. The bovine demi-embryos following dehydration by cryoprotectants a various concentration of sucrose were freezed by cell freezer and thawed in 3$0^{\circ}C$ water bath. Survival and in vitro developmental rates was defined as development rates on in vitro culture or FDA-test. The results are summarized as follows : 1. The high survival rates of demi-embryos after frozen-thawing in freezing medium was attained 2.0M glycerol. The high survival rates of demi-embryos after frozen-thawing in freezing medium was obtained using single cryoprotectant(25.0~30.0%) than mixed cryoprotectants(16.7~19.0%). 2. The survival rates of demi-embryos after frozen-thawing in freezing medium added 1.5M, 2.0M glycerol+0.25M sucrose(37.5~33.3%) were higher survival rates than those of sucrose concentration of 0.50, 0.75M(12.5~26.7%). 3. The equilibration time on the survival rates of demi-embryos was attained after short period of time(30.0~35.0%) in the freezing medium higher than long period of time(21.1%). 4. The thawing temperature on the survival rates of demi-embryos was attained at 3$0^{\circ}C$ of thawing temperature(26.7~40.0%) higher than $25^{\circ}C$ or 37$^{\circ}C$ of thawing temperature(13.3~20.0%). 5. The thawing time on the survival rates of demi-embryos was attained at 1~5 minutes of thawing time(26.7~33.3%) in the freezing medium higher than 10 minutes of thawing time(13.3~18.8%).

Optimization of main factors using response surface method for the enhanced production of hGM-CSF from transgenic Nicotiana tabacum cell suspension cultures

  • Response surface methodology was employed to study the interactive effect of sucrose, nitrogen, temperature and to optimize their levels to enhance the production of human granulocyte-macrophage colony-stimulation factor from Nicotiana tabacum cell suspension cultures. A 15-runs Box-Behnken design including three center points was the response surface method selected for the initial set of experiments. The analysis of the data from the Box-Behnken experiments showed interactive effects of sucrose:nitrogen, sucrose:temperature and nitrogen:temperature. The optimal combinations of sucrose, nitrogen and temperature for hGM-CSF production from surface plot were sucrose 90 g/L, nitrogen 41 mM and 22$^{\circ}C$, respectively. The optimization of there factors enhanced the hGM-CSF production by 2 times because high sucrose concentration stimulated the secretion of hGM-CSF and low temperature prevented hGM-CSF degradation in media by pretenses.

Effect of Different Rumen-degradable Carbohydrates on Rumen Fermentation, Nitrogen Metabolism and Lactation Performance of Holstein Dairy Cows

  • Four multiparous lactating Holstein cows fitted with rumen cannulae were fed diets varying in the amount and source of rumen-degradable carbohydrates (starch vs. sucrose) to examine their effects on rumen fermentation, nitrogen metabolism and lactation performance. A $4{\times}4$ Latin square with four diets and four periods of 28 days each was employed. Corn starch and sucrose were added to diets and corn starch was replaced with sucrose at 0 (0 S), 2.5 (2.5 S), 5.0 (5.0 S) 7.5% (7.5 S) of diet dry matter in a total mixed ration (TMR) containing 60% concentrate and 40% forage (DM basis). Replacing corn starch with sucrose did not affect (p>0.05) ruminal pH which averaged 6.41, but the ruminal pH for 7.5 S decreased more rapidly at 2 h after morning feeding compared with other treatments. Sucrose reduced ($p{\leq}0.05$) ruminal $NH_3-N$ concentration (13.90 vs. 17.09 mg/dl) but did not affect peptide-N concentration. There was no dietary effect on total volatile fatty acids (110.53 mmol/L) or the acetate to propionate ratio (2.72). No differences (p>0.05) in molar proportion of most of the individual VFA were found among diets, except for the molar proportion of butyrate that was increased ($p{\leq}0.05$) with the inclusion of sucrose. Total branched chain volatile fatty acids tended to increase ($p{\geq}0.051$) for the control treatment (0 S) compared with the 7.5 S treatment. Dry matter intake, body weight changes and digestibility of DM, OM, CP, NDF and ADF were not affected by treatments. Sucrose inclusion in the total mixed ration did not affect milk yield, but increased milk fat and total solid percentage ($p{\leq}0.05$). Sucrose tended ($p{\geq}0.063$) to increase milk protein percentage (3.28 vs. 3.05) and reduced ($p{\leq}0.05$) milk urea nitrogen concentration (12.75 vs. 15.48 mg/dl), suggesting a more efficient utilization of the rapidly available nitrogen components in the diet and hence improving nitrogen metabolism in the rumen.

Effect of Combined Osmotic Dehydration and Hot-air Drying on the Quality of Dried Apple Products (삼투건조와 열풍건조의 조합이 사과 건조제품의 품질에 미치는 영향)

  • This study was conducted to investigate the effects of combined osmotic dehydration and hot-air drying on the quality of dried apple products. Apple cylinders were steeped in 30% and 50% sucrose solutions at different steeping times. During the osmotic dehydration, as the concentration of the sucrose solution and steeping time increased, weight reduction and water loss increased, and the solid gain showed similar results. Osmotic dehydration in the sucrose solutions was followed by hot-air drying at 50 and $70^{\circ}C$. The experimental data were fitted successfully using the modified Page model. At the drying temperature of $50^{\circ}C$, the drying time increased from 4.15 hr for the control to 5.78 hr and 6.42 hr for the 30 and 50% sucrose solution treatments, respectively. Similar results were shown at the $70^{\circ}C$ drying temperature. The k and n values of the apple cylinders decreased by osmotic dehydration, and the k and n of the apple cylinders steeped in the 50% sucrose solution were lower than those of the samples steeped in the 30% sucrose solution. The qualities of the dried apple products were compared to samples that did not undergo osmotic dehydration. The shrinkage and rehydration capacity of the apple products decreased via osmotic dehydration, and decreased as the concentration of the sucrose solution increased. The compressibility ratios of the apple products to raw apple cylinders increased by osmotic dehydration, and increased as the concentration of sucrose solution increased. The sensory evaluation results for the apple products rehydrated in yoghurt indicate that osmotic dehydration greatly enhances the palatability of apple products in terms of appearance, taste, and texture.

Effect of Cryoprotectant Concentration and Equilibration Time on Volume Change and In Vitro Development of Intact and Bisected Mouse Embryos following Rapid Freezing (동결보호제의 농도와 평형시간이 생쥐의 정상배 및 분할배의 용적 변화와 체외 발달에 미치는 영향)

  • This study was carried out to investage the effect of cryoprotectant concentration and equilibration time on volume change and in vitro development of intact and bisected mouse embryos by rapid freezing. When compacted morulae were rapidly frozen in 3.0 to 4.0 glycerol or DMSO with 0.25M sucrose solution, the superior(P<0.05) post-thaw survival rate was obtained at the glycerol concentration of 4.0M(89.4%) than 3.0M(71.4%) or 5.0M(42.4%), but at the DMSO concentration of 3.0M(84.5%) than 4.0M(51.1%) or 5.0M(0.0%). The optimal equilibraton time for rapid freezing of ZP-free or bisected morulae in 4.0M glycerol with 0.25M sucrose was found tobe 3 minutes. The minimal volume of compacted morulaewhich corresponded with 61 to 62% of pre-equilibrated embryo volume was obtained from equilibration for 3 minutes in both 3.0 and 4.0M glycerol solutions with 0.25M sucrose.

Cryopreservation of Embryo by Concentration of Ethylene Glycol and Day 6, 7, 8, 9 Embryo in Korean Native Cattle (Hanwoo)

  • This study was carried out to effects of ethylene glycol concentration, sucrose and culture day of in vitro production embryo on slow-down freezing in Hanwoo. 6, 7, 8 and 9 day embryos produced in vitro were frozen using 1.8M EG+0.1M sucrose, 1.8M EG+0.5% BSA and 1.5M EG+0.1M sucrose media. Survivability was confirmed after frozen-thawed 24 and 48h and ICM, TE cell number were counted by Hoechst 33342 and PI staining after frozen-thawed 24h. As a result, 1.8M EG+0.1M sucrose group was most significantly (p<0.05) higher compared with the other treatment groups on survivability, TE and total cell number after frozen-thawed 24h ($94.2{\pm}2.6%$, $94.67{\pm}3.4$ and $129.67{\pm}5.5$). ICM number did not found significant (p<0.05) differences between the three treatment groups. in 6, 7, 8 and 9 day of embryos using three types of freezing media, frozen-thawed, 1.8M EG+0.1M sucrose groups with embryos cultured 8 day was significantly (p<0.05) highest survivability to $98.3{\pm}1.7%$ after frozen-thawed 24h. 1.5M EG+0.1 sucrose group with embryos cultured 9 day was significantly higher survivability than group of embryos cultured 8 day after frozen-thawed 24 and 48h. In conclusion, 1.8M EG+0.1M sucrose media is considered to be effective to cryopreservation of embryos cultured 8 and 9 day.

Taxol Production in Taxus Cell Cultures : 2. Effects of Sugar Concentration (주목 세포배양에 의한 Taxol 생산:2. 당농도의 영향)

  • The effects of sucrose on cell growth and anticancer drug taxol production were investigated in cell suspension cultures of Taxus cuspidata. The cells were cultured at various concentrations of sucrose (20 to $100g/\ell$). The highest specific growth rata was achieved at $40g/\ell$ of sucrose as 0.076 day-1 and the highest final cell density, 34 g DCW/$\ell$ after 25 days of culture, was obtained at $60g/\ell$ of sucrose. The cell yield(Yx/s) was found to be 0.55g cell/g sucrose and doubling time (Td) was 9.12 day. High concentration of sucrose (80, $100g/\ell$) and the addition of osmoticum enhanced the production of taxol significantly. The maximum taxol production was $1.36mg/\ell$ at $80g/\ell$ of sucrose, which was 0.01% as a specific content.
