2,7-Phloroglucinol-6,6-Bieckol Increases Glucose Uptake by Promoting GLUT4 Translocation to Plasma Membrane in 3T3-L1 Adipocytes (2,7-Phloroglucinol-6,6-Bieckol의 3T3-L1 지방세포에서 GLUT4 활성화를 통한 포도당 흡수 증진 효과)

  • Type 2 diabetes occurs when there is an abnormality in the tissue's ability to absorb glucose. Glucose uptake and metabolism by insulin are the basic mechanisms that maintain blood sugar. Glucose uptake goes through various signaling steps initiated by the binding of insulin to receptors on the cell surface. In line with the foregoing, the purpose of this study was to investigate the effect of 2,7-phloroglucinol-6,6-bieckol (PHB), an active compound isolated from Ecklonia cava, on glucose uptake in 3T3-L1 adipocytes. Notably, PHB increased glucose uptake in a dose-dependent manner owing to the enhanced glucose transporter type 4 (GLUT4) expression in the plasma membrane of 3T3-L1 adipocytes. These effects of PHB were attributed to the phosphorylation of insulin receptor substrate-1 and protein kinase B (PKB or AKT), as well as to the phosphoinositide 3-kinase (PI3K) activation in the insulin signaling pathway. PHB also stimulated 5' AMP-activated protein kinase (AMPK) phosphorylation and activation. The phosphorylation and activation of the PI3K/AKT and AMPK pathways by PHB were identified using wortmannin (a PI3K inhibitor) and compound C (an AMPK inhibitor). In this study, we showed that PHB can increase glucose uptake in 3T3-L1 adipocytes by promoting GLUT4 translocation to the plasma membrane via the PI3K and AMPK pathways. The results indicate that PHB may help improve insulin sensitivity.

Fucoidan Stimulates Glucose Uptake via the PI3K/AMPK Pathway and Increases Insulin Sensitivity in 3T3-L1 Adipocytes (후코이단의 3T3-L1 지방세포에서 PI3K/AMPK 경로를 통한 포도당 흡수 촉진 및 인슐린 민감성 증진 효과)

  • Brown seaweeds have been shown to decrease blood glucose levels and improve insulin sensitivity previously. In this study, we investigated the effect of fucoidan, a complex polysaccharide derived from brown seaweeds, on glucose uptake to improve insulin resistance, and examined its mechanism of action in 3T3-L1 adipocytes. We observed that fucoidan significantly increased glucose uptake and it was related to an increased expression of plasma membrane-glucose transporter 4 (PM-GLUT4) in 3T3-L1 adipocytes. Fucoidan treatment increased the activation of phosphatidylinositol-3-kinase (PI3K) and the phosphorylation of insulin receptor substrate 1 (IRS1tyr) compared with that of the control cells. Fucoidan also promoted the phosphorylation of Akt and protein kinase C (PKC)-λ/ζ compared to that of the control cells. Moreover, fucoidan significantly upregulated acetyl-CoA-carboxylase (ACC) and adenosine monophosphate - activated protein kinase (AMPK) phosphorylation. As a result, translocation of GLUT4 was significantly enhanced in 3T3-L1 adipocytes, which significantly promoted glucose uptake via the PI3K/AMPK pathways. The elevation of glucose uptake by fucoidan was blocked by inhibitor of PI3K and inhibitor of AMPK in 3T3-L1 adipocytes. These findings indicate that fucoidan might ameliorate glucose uptake through GLUT4 translocation to the plasma membrane by activating the PI3K/Akt and AMPK pathways in 3T3-L1 adipocytes. Fucoidan is thought to be of high material value to diabetes treatments and functional foods.

  • From the on-line mass spectrometric analyese of the exhaust gaseous composition of fermentor and the material balance equations for oxygen and carbon dioxide, oxygen uptake rate (OUR) and carbon dioxide evolution rate (CER) were calculate using a personal computer (IBM PC-AT) interfaced to a quadrupole mass spectromter. The calculate OUR and CER were used for the indirect estimation of cell and substrate concentrations during the batch culture of Candida utilis. For the estimation of sustrate concentration, the yield model of Pirt was applied. It was found that the cell and substrate (glucose) concentration could be ssatisfactorily estimataed and the results showed the more accurate estimations of both fermentation state variables when OUR data were used than CER data.

  • Carbon catabolite repression (CCR) is a key regulatory system found in most microorganisms that ensures preferential utilization of energy-efficient carbon sources. CCR helps microorganisms obtain a proper balance between their metabolic capacity and the maximum sugar uptake capability. It also constrains the deregulated utilization of a preferred cognate substrate, enabling microorganisms to survive and dominate in natural environments. On the other side of the same coin lies the tenacious bottleneck in microbial production of bioproducts that employs a combination of carbon sources in varied proportion, such as lignocellulose-derived sugar mixtures. Preferential sugar uptake combined with the transcriptional and/or enzymatic exclusion of less preferred sugars turns out one of the major barriers in increasing the yield and productivity of fermentation process. Accumulation of the unused substrate also complicates the downstream processes used to extract the desired product. To overcome this difficulty and to develop tailor-made strains for specific metabolic engineering goals, quantitative and systemic understanding of the molecular interaction map behind CCR is a prerequisite. Here we comparatively review the universal and strain-specific features of CCR circuitry and discuss the recent efforts in developing synthetic cell factories devoid of CCR particularly for lignocellulose-based biorefinery.

  • Polymeric hydrogel particles were fabricated to demonstrate the scale-up possibilities with the Particle Replication In Non-wetting Templates (PRINT) process. A permanently etched, specifically designed master was made on a silicon wafer using conventional photolithography, then reactive ion etching. The master and substrate were used repeatedly to make a large number of identical elastomeric perfluoropolyethers (PFPE) replica molds. The PFPE replica molds were used to fabricate and harvest individual, monodisperse micron-sized particles using the PRINT process. A water-soluble polymer adhesive was used as a sacrificial layer for harvesting particles. Particles were composed of biodegradable poly (ethylene glycol) diacrylate (PEG-diA), and aminoethylacrylate (AEM) and 2-acryloxyethyltrimethyl ammonium chloride (AETMAC) were added to them for improving the uptake of the cells. This study suggested PRINT used to produce the uniformed and shape specific biodegradable polymer is the effective technique for the non viral vector for the drug and the gene delivery.

Analysis of the Substrate Removal Characteristics of TPA Using OUR and NUR Tests, and Simulation with ASM1 (호흡률과 탈질률 실험과 ASM1을 이용한 전산모사를 통한 TPA의 기질 분해 특성 평가)

  • In this study, nitrate uptake rate(NUR) and oxygen uptake rate(OUR) tests were conducted for the assessment of application of Terephtalic acid(TPA) by-product as an alternative external carbon source for sewage treatment plant(STP). With the ASM1 installed in GPS-X the substrate removal characteristic was investigated with simulation by obtained data from NUR and OUR test. As a result, the fraction of RBDCOD(readily biodegradable COD) was mort than 90% and specific denitrification rate was observed about 8.00 mg $NO_3^-$-N/g VSS/hr that was similar to conventional external carbon source. In the next step, sensitivity analysis for heterotrophic biomass in ASM1 was conducted. Optimized parameters of ${\mu}_{max,H}$, $K_s$, ${\eta}_g$, and $b_H$ were 6.60/day, 23.3 mg/L, 0.88, and 0.54/day, respectively. Then, relative mean squared error(RMSE) was reduced to about 40%. Optimized parameters value were well corresponded with the substrate removal characteristics of high maximum and final endogenous specific OUR and high specific NUR.

Application of QUAL2E Model for Water Quality Management in the Keum River(I) -Estimation of Model input Parameter and Autochthonous BOD- (금강수계의 수질관리를 위한 QUAL2E 모델의 적용(I) -모델입력인자 산정 및 자생BOD 평가-)

  • The Keum river is one of the important river in Korea and has a drainage area of 9,873$\textrm{km}^2$. The Keum river is deepening pollution state due to development of the lower city and construction of a industrial complex. The water quality of the Keum river come to eutrophication state and belong to III grade of water quality standard. The concentration BOD in river is affected by the organic loading from a tributary and the algae biomass that largely happen to under eutrophication state. In the eutrophic water mass such as the Keum river, the autochthonous BOD was very important part for making a decision of water quality management, because it was accounted for majority of the total BOD. The purpose of this study was to survey the chatacteristics of water quality in summer and to estimate reaction coefficient. Also, we studied to correlationship between chlorophyll a and BOD(COD) for estimation of the autochthonous BOD. The correlationship between chlorophyll a and BOD(COD) were obtained through the culture experiment of phytoplankton in the laboratory. The results of this study may be summarized as follows ; The characteristics of water quality in summer were belong to III~IV grade of water quality standard as BOD and nutritive condition is very high. The BOD, ammonia nitrogen and phosphate loadings in Miho stream which inflowing untreated sewage from Chungju city was occupied with 64.07%, 26.36%, 46.08%, respectively. Maximum nutrient uptake (Vmax) was 0.4400$\mu$M/hr as substrate of ammonia nitrogen, 0.1652$\mu$M/hr as substrate of phosphate. Maximum specific growth rate ($\mu$max) was 1.2525$hr^{-1}$ as substrate of ammonia nitrogen, 1.5177$hr^{-1}$ as substrate of phosphate. The correlation coefficient between chlorophyll a and BOD by the culture experiment were found to be 0.911~0.935 and 0.942~0.947 in the case adding nutrient and no adding nutrient, respectively. The correlation coefficient between chlorophyll a and COD through the culture experiment were found to be 0.918~0.977 and 0.880~0.931 in the case adding nutrient and no adding nutrient, respectively. The autochthonous BOD(COD) was estimated to the relationship between BOD(COD) and chlorophyll a. The regression equation were found to be autochthonous BOD=(0.045~0.073)${\times}chlorophyll$ a and autochthonous $COD=(0.137~0.182){\times}chlorophyll$ a.

Sludge Returned CMAS에 의한 전기부속품제조공장 폐수처리

  • Sludge Returned CMAS process was applied to treat the wastewater from electric accessory manufacturing company while this type of wastewater was usually treated by chemical process. This result show that the removal rate of TCOD was about 70-80% regardless of hydraulic retention time, On the contrary, the removal rate of BOD was abtained in a range of 77-92% depending on hydraulic retention time. In order to remove more than 80% of organic materials with the proposed process, the F/M ratio should be maintained below 0.17. In this case, the calculated value of organic removal rate, Km, was calculated to be 1.26 hr$^{-1}$, and the ratio of cell synthesis/total energy was 0.32 and 0.26 for COD and BOD base, respectively. The yield coefficient was calculated to be 0.242 and the half velocity coefficient was 0.3 hr$^{-1}$. The value of endogenous respiration coefficient was 0.02 hr$^{-1}$. The measured effluent BOD concentration, MLSS concentration in aeration tank, oxygen uptake rate, and sludge production were matched relatively well with the calculated values using above coefficients, In order to optimize the dewatering of sludge, the hydraulic retention time was recommended to be 15. 6 hrs. These results indicate that the wastewater from an eletric accessory manufacturing company can be treated safely with a biological process.

Determination of Mineral Components in the Cultivation Substrates of Edible Mushrooms and Their Uptake into Fruiting Bodies

  • The mineral contents of the cultivation substrates, fruiting bodies of the mushrooms, and the postharvest cultivation substrates were determined in cultivated edible mushrooms Pleurotus eryngii, Flammulina velutipes, and Hypsizigus marmoreus. The major mineral elements both in the cultivation substrates and in the fruiting bodies were K, Mg, Ca, and Na. Potassium was particularly abundant ranging 10${\sim}$13 g/kg in the cultivation substrates and 26${\sim}$30 g/kg in the fruiting bodies. On the contrary, the calcium content in the fruiting bodies was very low despite high concentrations in the cultivation substrates, indicating Ca in the cultivation substrates is in a less bio-available form or the mushrooms do not have efficient Ca uptake channels. Among the minor mineral elements determined in this experiment, Cu, Zn, and Ni showed high percentage of transfer from the cultivation substrates to the fruiting bodies. It is noteworthy that the mineral contents in the postharvest cultivation substrates were not changed significantly which implies that the spent cultivation substrates are nutritionally intact in terms of mineral contents and thus can be recycled as mineral sources and animal feeds.

Pear pomace ethanol extract improves insulin resistance through enhancement of insulin signaling pathway without lipid accumulation

  • BACKGROUND/OBJECTIVES: The anti-diabetic activity of pear through inhibition of ${\alpha}-glucosidase$ has been demonstrated. However, little has been reported about the effect of pear on insulin signaling pathway in obesity. The aims of this study are to establish pear pomace 50% ethanol extract (PPE)-induced improvement of insulin sensitivity and characterize its action mechanism in 3T3-L1 cells and high-fat diet (HFD)-fed C57BL/6 mice. MATERIALS/METHODS: Lipid accumulation, monocyte chemoattractant protein-1 (MCP-1) secretion and glucose uptake were measure in 3T3-L1 cells. Mice were fed HFD (60% kcal from fat) and orally ingested PPE once daily for 8 weeks and body weight, homeostasis model assessment of insulin resistance (HOMA-IR), and serum lipids were measured. The expression of proteins involved in insulin signaling pathway was evaluated by western blot assay in 3T3-L1 cells and adipose tissue of mice. RESULTS: In 3T3-L1 cells, without affecting cell viability and lipid accumulation, PPE inhibited MCP-1 secretion, improved glucose uptake, and increased protein expression of phosphorylated insulin receptor substrate 1 [p-IRS-1, ($Tyr^{632})$)], p-Akt, and glucose transporter type 4 (GLUT4). Additionally, in HFD-fed mice, PPE reduced body weight, HOMA-IR, and serum lipids including triglyceride and LDL-cholesterol. Furthermore, in adipose tissue, PPE up-regulated GLUT4 expression and expression ratio of p-IRS-1 ($Tyr^{632})/IRS$, whereas, down-regulated p-IRS-1 ($Ser^{307})/IRS$. CONCLUSIONS: Our results collectively show that PPE improves glucose uptake in 3T3-L1 cells and insulin sensitivity in mice fed a HFD through stimulation of the insulin signaling pathway. Furthermore, PPE-induced improvement of insulin sensitivity was not accompanied with lipid accumulation.
