Transition Substitution of Desired Bases in Human Pluripotent Stem Cells with Base Editors: A Step-by-Step Guide

  • The recent advances in human pluripotent stem cells (hPSCs) enable to precisely edit the desired bases in hPSCs to be used for the establishment of isogenic disease models and autologous ex vivo cell therapy. The knock-in approach based on the homologous directed repair with Cas9 endonuclease, causing DNA double-strand breaks (DSBs), produces not only insertion and deletion (indel) mutations but also deleterious large deletions. On the contrary, due to the lack of Cas9 endonuclease activity, base editors (BEs) such as adenine base editor (ABE) and cytosine base editor (CBE) allow precise base substitution by conjugated deaminase activity, free from DSB formation. Despite the limitation of BEs in transition substitution, precise base editing by BEs with no massive off-targets is suggested to be a prospective alternative in hPSCs for clinical applications. Considering the unique cellular characteristics of hPSCs, a few points should be considered. Herein, we describe an updated and optimized protocol for base editing in hPSCs. We also describe an improved methodology for CBE-based C to T substitutions, which are generally lower than A to G substitutions in hPSCs.

Synthesis of Dihydroxylated Chalcone Derivatives with Diverse Substitution Patterns and Their Radical Scavenging Ability toward DPPH Free Radicals

  • A series of dihydroxylated chalcone derivatives with diverse substitution patterns on a phenyl ring B and the para-substituents on a phenyl ring A were prepared, and their radical scavenging activities were evaluated by simple DPPH test to determine quantitative structure-activity relationship in these series of compounds. The chalcone compounds with the ortho- (i.e. 2',3'- and 3',4'-) and para- (i.e. 2,5'-) substitution patterns show an excellent antioxidant activities (80-90% of control at the concentration of 50 $\mu$M) which are comparable to those of ascorbic acid and $\alpha$ -tocopherol as positive reference materials. On the contrary, the compounds with meta- (i.e. 2',4'-, 3',5'-) substitution pattern demonstrate very dramatic decrease in activities which are around 25% of the control even at the concentration of 200 $\mu$ M (IC50 > 200 $\mu$ M). These dramatic differences could be interpreted in terms of the ease formation of fairly stable semiquinone radicals from the ortho- and parasubstituted chalcone molecules through facilitating electron delocalization. Our results indicate that the substitution patterns of two hydroxyl groups on ring B are very important structural factors for their radical scavenging activity enhancement. Meanwhile, the substituents at para-position of the phenyl ring A of chalcones have no influence on the activity.

항추가 및 보정을 적용한 대입에 의한 논리식 간략화 (Logic Substitution Using Addition and Revision of Terms)

  • 2개 논리식에 대해서 어떤 논리식 F의 일부가 다른 논리식 G 전체를 포함하고 있을 때, 논리식 F의 일부분을 논리식 G로 대치한 식을 대입식이라고 한다. 논리식 사이에 대입 관계가 성립되면 전체 논리식에 사용된 리터럴 개수를 대폭 줄일 수 있는 장점이 있으나, 대입 관계가 성립하지 않는 경우 대입식으로부터 얻을 수 있는 간략화 효과가 없게 되어 상대적으로 리터럴 개수를 줄이는 효과가 줄어들게 된다. 지금까지의 연구들이 주어진 논리식들 자체에 대해서 논리식들 사이의 대입 관계를 찾고, 대입이 가능하면 대입식을 산출하기 위한 방법을 제안하였는데, 본 논문에서는 논리식들 사이에 대입식이 만들어지도록 필요한 항을 추가하고, 다시 추가된 항들에 대한 보정을 통해 대입식을 산출하는 논리합성 방법을 제안한다. 최적화하고자 하는 2개의 논리식들로부터 항추가를 위한 행렬을 만들고, 행렬에서 항이 추가 가능한 묶음 찾고 추가된 항에 대해 보정을 하여 대입식이 완성된다. 실험결과 여러 벤치마크 회로에 대하여 제안한 방법이 기존 합성도구보다 리터럴 개수를 줄일 수 있음을 보였다.

S곡선 기반 기술적 불연속성(Technological discontinuity)의 정의 및 측정 : 로직 반도체의 기술대체 사례 (Definition and measurement of S-curve based technological discontinuity : case of technological substitution of logic semiconductors)

  • 기존 기술과 신기술의 확산 및 대체 과정에서 발생하는 기술적 불연속성 현상은 단일 기술 및 복수 기술의 확산 및 대체 현상의 거동을 이해하는데 중요하다. 본 연구에서는 기술적 불연속성 구간의 개념에 대해 정의하고, 이 구간을 측정할 수 있는 정량적 지표들에 대한 측정 모형을 개발하였다. 문헌리뷰 및 모형 도출을 바탕으로 기술적 불연속성 구간에 대해 정의 및 측정 모형을 제시하였고, 도출한 모형의 정합성을 반도체 산업의 기술대체 사례를 바탕으로 검증하였다. 기술적 불연속성 구간은 기존 기술과 신기술의 S곡선이 시간에 따라 동시에 존재하면서, 기존 기술의 성능이 신기술의 성능보다 높은 구간으로 정의된다. 또한 기술적 불연속 구간은 불연속 시간 및 불연속 성능으로 측정가능하며, 불연속 시간 및 불연속 성능지표는 불연속 구간에서의 기존 기술과 신기술의 시간 차이 및 성능 차이로 모형화 된다. 본 연구는 기술적 불연속성 현상에 대한 이해뿐만 아니라 기술 확산 및 대체 현상의 전체적인 거동의 이해에 유용할 것이다.

Effects of Substitutions of Gln16 and Asp18 with Phe or Tyr in HP(2-20) on its Structure and Antimicrobial Activity

  • HP (2-20), a 19-residue peptide derived from the N-terminus of Helicobacter pylori Ribosomal Protein L1, has antimicrobial activity but is not cytotoxic to human erythrocytes. Previously, we have synthesized several analogue peptides to investigate the effects of substitutions on the structure and antimicrobial activity. Substitution of $Gln^{16}$ and $Asp^{18}$ with Trp (Anal 3) caused a dramatic increase in bacterial and fungal lytic activities. In this study, analogue peptides were synthesized to investigate the effects of substitution of Gin and Asp with Phe (Anal 6) or Tyr (Anal 7) in HP (2-20) on its structure and antimicrobial activity. Substitution of Gin and Asp with hydrophobic aromatic residues at position 16 and 18 of HP (2-20) caused increase in antibiotic activity without hemolytic effect. Substitution of Gin and Asp with Trp and Try increased antibiotic activity of HP (220) twice more compared to substitution with Phe. The tertiary structures of Anal 6 and Anal 7 in SDS micelles has been investigated using NMR spectroscopy. The structures revealed that substitutions of the aromatic residues at C-terminus resulted in longer and well defined alpha-helix and improved their antibacterial activities

굴착복구용 속경성 경량기포 시멘트 모르타르의 최적 배합 도출을 위한 기초 물성 연구 (Optimized mix design of rapid-set lightweight-formed mortar for backfill)

  • PURPOSES : The objective of this study is to develop an optimized method of mix design for rapid-set lightweight-formed mortar mix. To achieve this objective, the workability, setting time, and compressive strength of mixes under various conditions of mix design were evaluated. METHODS : The water-bonder ratio, fly-ash substitution ratio, and forming agent injection amount were selected as design variables in the study. The fluidity, setting time, density, and strength of the mortar mix were considered as major evaluation criteria of the mixture, and were subsequently utilized to evaluate the characteristics of the mortar mix under various conditions. RESULTS : The observations made from the mix design process are as follows: 1) the air content and fluidity increase as the forming agent ratio and forming agent ratio increase, respectively; 2) the maximum air content is approximately 20%; 3) the accelerating agent decreases the fluidity of the mortar mix by 15% on average; 4) the forming agent injection ratio and fly-ash substitution ratio yield significant effects on the initial and final set times of the mortar mix; 5) as the forming agent injection ratio and fly-ash substitution ratio increase, the compressive strength of the mortar mix decreases; and 6) the 28-day compressive strengths of the forming agent injection ratio and fly-ash substitution ratio yield the most significant effects. CONCLUSIONS : It is concluded that the governing design variables for the rapid-set lightweight-formed mortar mix are the forming agent injection ratio and fly-ash substitution ratio.

Whey Protein Isolate(WPI)의 대체비율을 달리한 스폰지 케이크의 품질 특성에 관한 연구 (The Quality Characteristics of Sponge Cake with Varied Levels of Whey Protein Isolate)

  • The substitution effects of whey protein isolate(WPI) for egg in the preparation of sponge cake were determined by objective and subjective tests. Milk whey is drained from milk curd as a by-product of cheese manufacture. Whey protein is known as a good nutritional source and a functional material for many processed foods, especially baked goods. WPI contains above 90% whey protein. The specific gravity and viscosity of sponge cakes tend to be affected by WPI substitution. The cooking loss of sponge cakes with WPI substituted for egg(abbreviated as WPI cake) during oven baking was smaller than that made with egg(abbreviated as egg cake) and the specific loaf volume of WPI cake was larger than that of egg cake. The number of pores was highly increased and the size of pores was more uniformly and finely distributed in the cross section of WPI cake than those of egg cake, as observed by scanning electron microscopy(SEM). The hardness, gumminess and chewiness of WPI cake made with 10-20% WPI substitution were the lowest among all the tested cakes, including egg cake, thereby confirming the considerable improvement in their cake qualities. By the results of sensory evaluation, appearance, pore uniformity, softness, chewiness, moistness, flavor, mouth feeling, and overall acceptability of 10-20% WPI substitute cakes were evaluated as being significantly superior to those of all other cakes(p<0.05). These results support the better physicochemical characteristics and sensory evaluations of sponge cake prepared with 10-20% of WPI substitution for egg.

목제품 시장에서 국산품과 수입품의 아밍턴 대체 탄성치 추정 (Estimating Armington Substitution Elasticity between Domestic and Imported Wood Products in Korea)

  • 이 연구는 수입 목제품과 국산 목제품의 수요가 불완전 대체관계에 있다고 가정하고 아밍턴 대체 탄성치를 추정하였으며 이에 근거하여 수입 목제품의 관세 인하가 국내 목제품 생산에 미치는 영향을 분석한 것이다. 분석 대상 목제품은 합판, 섬유판, 파티클보드로 하였다. 분석결과, 국산 목제품과 수입 목제품은 완전 대체관계에 있지는 않은 것으로 나타났다. 합판과 섬유판에서 국산품과 수입품의 대체 탄성치가 높게 나타나 관세를 인하하였을 때 그 영향이 적지 않을 것으로 생각된다. 이에 비해 파티클보드 시장에서 국산품과 수입품은 차별화되어 있으며 대체 관계가 낮은 것으로 보인다. 섬유판은 국내 숲가꾸기 산물을 원료로 사용하고 있기 때문에 섬유판 생산의 위축은 국내 임업에도 부정적 영향을 미치게 될 것이다. 따라서, 시장개방에 대응하여 국내 목제품 산업의 경쟁력 강화를 위한 대책이 필요하다.

PMW-PNN-PZT계 세라믹스의 유전및 압전특성 (Dielectric and Piezoelectric Properties of PMW-PNN-PZT System Ceramics)

  • In this paper the structural dielectric and piezoelectric properties of Pb[(M $g_{1}$2// $W_{1}$2/)$_{x}$-(N $i_{1}$3//N $b_{2}$3/)$_{0.15-x-(Zr_{0.5})}$ $Ti_{0.5}$)$_{0.85}$$O_3$ (x=0.0~0.10) ceramic were investigated with the substitution of Pb(M $g_{1}$2// $W_{1}$2/) $O_3$. According to the substitution of Pb(M $g_{1}$2//W/1/2/) $O_3$ curie temperatures were slightly decrease due to the decrease of the tetrag-onality of crystal structure and coercive fields were decreased. Up to the substitution of Pb(M $g_{1}$2// $W_{1}$2/) $O_3$ 3mol%,remnant polarization dielectric constant piezoelectric constant were increased. Dielectric constant and electro-mechanical coupling factor( $k_{p}$, $k_{31}$ ) were appeared the highest value of 2230, 0.64, and 0.38 and piezoelectric constant( $d_{33}$ , $d_{31}$ ) was the largest value of 418, 202($\times$10$^{-12}$ /C/N), respectively, when the substitution amount of Pb(M $g_{1}$2// $W_{1}$2/N) respectively, when the substitution amount of Pb(M $g_{1}$2// $W_{1}$2/) $O_3$ was 3mol%.s 3mol%.%.

Cut-out이 있는 Box형 차체의 하부구조 소재대체 경량화 설계 방법 (A Weight-reduction Design Method by Underframe Material Substitution in a Box-type Bodyshell with Cut-outs)

  • In this paper, a theoretical weight-reduction method was suggested to substitute an underframe material of a box-type bodyshell having cut-outs with an alternative light-weight material. To utilize the material substitution method previously developed for a box-type hybrid bodyshell not having cut-outs, we derived a box-type baseline model without cut-outs which is similar to the stiffness condition of a box-type bodyshell having cut-outs. To do this, the thicknesses of roof and walls of the baseline model were determined such that the deflection of the baseline model under a distributed vertical load condition is equal to the sum of the theoretical section deflections of the original box model with cut-outs. Next, to derive a hybrid bodyshell by under-frame material substitution, the material substitution method for a box-type hybrid bodyshell without cut-outs was applied to the box-type baseline model. Finally, we compared the FE simulation results of the derived hybrid bodyshells having cut-outs for various materials with the theoretical results of the suggested method, and we obtained their good correlations.
