제지슬러지를 이용한 인공상토가 토마토와 오이 플러그묘의 생육에 미치는 영향 (Growth Responses of Tomato and Cucumber Plug-seedlings Grown for the Paper-sludge Substrates)

  • 본 시험은 폐기되고 있는 제지슬러지의 자원 재활용을 위한 육묘용 상토로서 이용 가능성을 알아보기 위해 수행되었다. 상업 용토 토실이를 대조구로 하여 제지슬러지와 토실이의 혼합 상토 (1:1, v:v), 그리고 제지 슬러지 단용 상토를 처리로 하였다. 토마토의 육묘 결과, 제지슬러지 혼합 상토에서의 생육은 토실이 상토와 비교하여 초장, 근장, 생체중과 건물중 등에서 그 차이 없었다. 반면, 제지슬러지 단용 상토에서의 생육은 현저히 억제되는 것으로 나타났다. 이러한 결과는 오이묘의 생육결과에서도 반영 되어 제지슬러지 단용 상토의 오이묘 생육은 현저히 억제되었다. 본 시험의 결과, 제지슬러지의 자원 재활용을 높이기 위해서는 미 가공 제지슬러지의 높은 pH와 EC의 안정화, 그리고 Zn의 함량을 낮추는 등의 처리가 선행 되어야 할 것으로 판단된다.

Lipase Production by Limtongozyma siamensis, a Novel Lipase Producer and Lipid Accumulating Yeast

  • Lipase is a well-known and highly in-demand enzyme. During the last decade, several lipase optimization studies have been reported. However, production costs have always been a bottleneck for commercial-scale microbial enzyme production. This research aimed to optimize the conditions for lipase production by Limtongozyma siamensis DMKU-WBL1-3 via a One-Factor-At-a-Time (OFAT) approach combined with statistical methods while using a low-cost substrate. Results suggest that low-cost substrates can be substituted for all media components. An optimal medium was found, using response surface methodology (RSM) and central composite design (CCD), to consist of 0.50% (w/v) sweet whey, 0.40% (w/v) yeast extract (food grade), and 2.50% (v/v) palm oil with the medium pH adjusted to 4 under shaking flask cultivation. From an economic point of view, this work was successful in reducing production costs while increasing lipase productivity. The medium costs were reduced by 87.5% of the original cost while lipase activity was increased by nearly 6-fold. Moreover, lipase production was further studied in a 2-L stirred-tank fermentor. Its activity was 1,055.6 ± 0.0 U/ml when aeration and agitation rates were adjusted to 1 vvm and 170 rpm, respectively. Interestingly, under this optimal lipase production, the yeast showed accumulated lipids inside the cells. The primary fatty acid is a monounsaturated fatty acid (MUFA) that is typically linked to health benefits. This study hence reveals promising lipase production and lipid accumulation by L. siamensis DMKU-WBL1-3 that are worthy of further study.

$Pt/SrTiO_3/Pb_x(Zr_{0.52}, Ti_{0.48})O_3/SrTiO_3/Si$ 구조의 전기적 특성 분석 및 $SrTiO_3$박막의 완충층 역할에 관한 연구 (Electrical Properties in $Pt/SrTiO_3/Pb_x(Zr_{0.52}, Ti_{0.48})O_3/SrTiO_3/Si$ Structure and the Role of $SrTiO_3$ Film as a Buffer Layer)

  • $Pt/SrTiO_3/Pb_x(Zr_{0.52}, Ti_{0.48})O_3/SrTiO_3/Si$ structure was prepared by rf-magnetron sputtering method for use in nondestructive read out ferroelectric RAM(NDRO-FEAM). PBx(Zr_{0.52}Ti_{0.48})O_3}$(PZT) and $SrTiO_3$(STO) films were deposited respectively at the temperatures of $300^{\circ}C and 500^{\circ}C$on p-Si(100) substrate. The role of the STO film as a buffer layer between the PZT film and the Si substrate was studied using X-ray diffraction (XRD), Auger electron spectroscopy (ASE), and scanning electron microscope(SEM). Structural analysis on the interfaces was carried out using a cross sectional transmission electron microscope(TEM). For PZT/Si structure, mostly Pb deficient pyrochlore phase was formed due to the serious diffusion of Pb into the Si substrate. On the other hand, for STO/PZT/STO/Si structure, the PZT film had perovskite phase and larger grain size with a little Pb interdiffusion. the interfaces of the PZT and the STO film, of the STO film and the interface layer and $SiO_2$, and of the $SiO_2$ and the Si substate had a good flatness. Across sectional TEM image showed the existence of an amorphous layer and $SiO_2$ with 7nm thickness between the STO film and the Si substrate. The electrical properties of MIFIS structure was characterized by C-V and I-V measurements. By 1MHz C-V characteristics Pt/STO(25nm)/PZT(160nm)/STO(25nm)/Si structure, memory window was about 1.2 V for and applied voltage of 5 V. Memory window increased by increasing the applied voltage and maximum voltage of memory window was 2 V for V applied. Memory window decreased by decreasing PZT film thickness to 110nm. Typical leakage current was abour $10{-8}$ A/cm for an applied voltage of 5 V.

논토양미생물의 Diazinon과 부식물질에 대한 기질 친화성 (Affinity of Diazinon and Humic Substances as a Substrate of Microorganisms in Paddy Soil)

  • Diazinon과 부식물질인 humin, HA, FA에 대한 토양미생물의 기질 선호성을 알아보기 위하여 한정비재에 diazinon과 부식물질을 각각 처리하여 항온($30^{\circ}C{\pm}1$)으로 유지하면서 토양미생물의 균수와 MO 활성, ${\alpha}$, -${\beta}$-ES 활성을 측정하였으며, 각각의 부식물질 첨가에 따른 diazinon의 분해속도를 조사하였다. 1. 토양미생물의 균수가 부식물질 처리구에서는 초기 1일부터 급격히 증가한 반면 dizainon 처리구에서는 3일후 부터 증가하였으며, 10일후 부식물질 처리구가 diazinon 처리구보다 약 1.5배 많았다. 2. MO 활성은 부식물질 처리구에서 diazinon처리구보다 3일까지 높았으며, HA > FA > humin의 순이었다. 3. ${\alpha}$-ES와 ${\beta}$-ES 활성은 비슷한 경향이었으며, humin과 HA에서는 diazinon보다 5일이후 높았지만, FA는 diazinon과 거의 비슷한 수준이었다. 4. 부식물질을 첨가함으로써 diazinon의 분해속도는 지연되었으며, 10일후의 분해율은 humin, HA, FA 첨가구에서 각각 51.4%, 58.9%, 62.4%이었고, 무첨가구에서는 71.9%이었다.
