Effects of different types of sub-freezing methods on quality of raw pork loin, beef loin and tuna were studied. Storage tests were undertaken as follows; the three different types of sub-freezing methods, such as regular cold chamber at $-1^{\circ}C$, air-blast sub-freezing at $-5^{\circ}C$ and sharp-plate sub-freezing at $-5^{\circ}C$, were evaluated for extending freshness of samples. Pork loin packed with polyethylene-film showed no significant differences (P<0.05) in physico-chemical properties among the above sub-freezing methods. Air-blast sub-freezing methods revealed faster evaporation of moisture from samples. The same sub-freezing method also showed increased VBN values of pork loin and tuna samples. Regular cold chamber resulted in increased TBA values and K values of beef loin and tuna, respectively. It appeared that sharp-plate freezing system kept the food samples with the least loss of freshness among the above three sub-freezing methods. These results indicated that freshness of raw pork, beef and tuna could be significantly extended by the sharp-plate sub-freezing method.
The observe relaxation phenomena and freezing behavior in Pb(Mg/sub ⅓/Nb/sub ⅔/O₃, relaxor ferroelectrics, weak electric-field properties as well as strong electric-field properties were investigated. The temperature dependence of the dielectric properties obtained using the low electric-field of 1 V/w was investigated. The dielectric properties obtained from the slope of the dielectric hysteresis loop and the temperature dependence of the pyroelectric properties were also investigated. When fitting all the experimental data with the Vogel-Filcher relation, a close agreement between the experimental data and equation was observed. The freezing temperature could be consistently calculated by the various methods.
PURPOSES : Nowadays, cavity phenomena occur increasingly in pavement layers of downtown areas. This leads to an increment in the number of potholes, sinkholes, and other failure on the road. A loss of earth and sand from the pavement plays a key role in the occurrence of cavities, and, hence, a structural-performance evaluation of the pavement is essential. METHODS: The structural performance was evaluated via finite-element analysis using KPRP and KICTPAVE. KPRP was developed in order to formulate a Korean pavement design guide, which is based on a mechanical-empirical pavement design guide (M-EPDG). RESULTS: Installation of the anti-freezing layer yielded a fatigue crack, permanent deformation, and international roughness index (IRI) of 13%, 0.7 cm, and 3.0 m/km, respectively, as determined from the performance analysis conducted via KPRP. These values satisfy the design standards (fatigue crack: 20%, permanent deformation: 1.3 cm, IRI: 3.5 m/km). The results of FEM, using KICTPAVE, are shown in Figures 8~12 and Tables 3~5. CONCLUSIONS: The results of the performance analysis (conducted via KPRP) satisfy the design standards, even if the thickness of the anti-freezing layer is not considered. The corresponding values (i.e., 13%, 0.7 cm, and 3.0 m/km) are obtained for all conditions under which this layer is applied. Furthermore, the stress and strain on the interlayer between the sub-grade and the anti-freezing layer decrease gradually with increasing thickness of the anti-freezing layer. In contrast, the strain on the interlayer between the sub-base and the anti-freezing layer increases gradually with this increase in thickness.
Background: Using cryovial for freezing dog spermatozoa provides a practical method to increase extended sperm volume and shorten the time required for equilibration by using a simple freezing techniques. The purpose of this study was to determine the optimal thawing condition for dog sperm cryopreservation using cryovials. Methods: For sperm freezing, cryovials with 200 × 106 sperm/mL were cooled after the addition of tris egg yolk extender (TEY) at 4℃ for 20 min, then TEY with 4% glycerol was added and equilibrated for another 20 min before being aligned over LN2 vapor for another 20 min and plunged directly into LN2. Spermatozoa were thawed in a water bath at 37℃ for varying times (25 sec, 60 sec, 90 sec, and 120 sec) in the first experiment. In the second experiment, spermatozoa were thawed in a water bath at various temperatures and times (37℃ for 1 min, 37℃ for 1 min with gentle stirring, 24℃ for 24 min, and 75℃ for 20 sec). In these experiments, the effect of thawing conditions on motility parameters, viability (SYBR-14/PI), and acrosome integrity (PSA/FITC) of spermatozoa were investigated. Results: The post-thaw sperm motility parameters, viability, and acrosome integrity were not significantly different across the experimental groups. Conclusions: In this study, the characteristics of spermatozoa frozen using cryovials were not significantly affected by various thawing conditions.
This study was done to find optimal cryopreservation medium and method to improve the post-thaw motility of human sperm. Thirty three semaen samples were included in the study. Of these, nineteen Samples showing normal semen profile were frozen using three cryprotectants (TYB, TYB+DTT and KS II)to compare post-thaw motility. Fourteen samples were frozen with vapor freezing and programmable freezing methods to compare post-thaw motility in correlation with freezing method. After 24 hrs of cryostorage , the vials were thawed and the post-thaw sperm motility was assessed by two kinds of computer-aededsperm analysis (CASA; SAIS and Hamilton Thorn). As a result, the post-thaw motility of the KS II group was higher than the TYB or TYB+DTT group in normal semen(34.8%, 28.3% and 23.0%, respectively). In the asthenospermia group, a significantly hither post-thaw motility was observed in the KS II (18.5%) compared to those of TYB or TYB+DTT group (13.6%, 10.0%, respectively). No difference was observed between vapor freezing group and computerized freezing group in normal semen (27.8%, 33.2%, respectively) and semen group showing asthenospermia (12.8%, 12.9%, respectively). In conclusion, our results indicate that KS II medium is reliable for cryopreservation of human sperm and vapor freezing and programmable freezing were equally effective in terms of the recovery of motile spermatozoa.

BACKGROUND: This study was to develop a analytical reference material including twenty pesticides in tomato. To use it for proficiency test, the suitability of homogeneity, storage stability, assigned values and uncertainty for analytical reference material were important.METHODS AND RESULTS: In order to develop a analytical reference material of tomato for multi-residue analysis of pesticides, twenty-pesticides were treated in tomato and the samples were frozen and homogenized. The homogeneity, stability, assigned value and uncertainty were calculated according to the requirements of the KS A ISO Guide 35, KS Q ISO 13528 and EURL-PT protocol. The values of the within-bottle standard variation(swb) and the between-bottle standard variation(sbb) were 0.9~6.5% of assigned value and the uncertainty(u*bb) due to inhomogeneity was also calculated as 0.6~1.9% for all pesticides. This indicated that it was satisfactory to be used as a analytical reference material. The storage stabilities of twenty-pesticides at room temperature and freezing conditions were assessed according to the requirement of the KS Q ISO Guide 35. All pesticides were stable at room temperature (20~30℃) for 8 days and freezing (-20℃) for 23 days.CONCLUSION: The feasibility of analytical reference material for pesticide multi-residue analysis in a tomato matrix was investigated. Homogeneity of within/between-bottle, uncertainty and stabilities at room temperature and freezing condition were satisfactory for a use of proficiency test and quality control. From these results, a analytical reference material would be applicable to monitor the proficiency test of pesticide analysis organizations to improve the reliability and consistency.
Rancidity changes were examined for 6 herbal medicines, namely Persicae Semen, Armeniacae Semen, Lini Semen, Trichosanthis Semen, Arecae Semen, Myristicae Semen known to have relatively high fat content. In order to reduce rancidity of herbal medicines, samples were stored at 3 different conditions of room, refrigerating and freezing temperatures, and the rancidity was measured for 10 months with every 2 month interval. Fat content was extracted by using ethyl ether, and acid values and peroxide values, which are generally accepted indicators of fat rancidity, were measured. When storing Persicae Semen, Lini Semen and Arecae Semen at room temperature, the acid values increased as the storage period increased, and it was higher than when stored in refrigeration or freezing. The measurement of peroxide value showed more significantly higher initial degree of rancidity when Persicae Semen, Trichosanthis Semen, Arecae Semen and Myristicae Semen were stored at room temperature. It was observed that storing herbal medicines in refrigeration or freezing inhibited their rancidity compared to storing them at room temperature. To investigate the quality changes according to rancidity, the analysis of aflatoxins and indicator components showed that aflatoxins B1 and B2 were detected in Armeniacae Semen, Arecae Semen and Myristicae Semen, and the amount of amygdalin was well maintained within the specification standard.

Microalgae are of significant importance for future biotechnological applications. Many microalgae banks or laboratories attempt to maintain various microalgae for further research purposes. Cryopreservation has been preferred to reduce a labor-intensive and costly routine sub-culturing. Cryopreservation can also diminish the genetic drift risk. However, cryopreservation as a long term storage of microalgae method are still in developing progress because it cannot be generalized for all microalgae. Microalgae types, cryoprotectant agents (CPAs) types, freezing and thawing methods are the most important factors that should be considered for cryopreservation. In this short review the basic principles and the current advanced of microalgae cryopreservation methods are discussed with a suggested starting parameters for microalgae cryopreservation.

BACKGROUND: Rabbiteye blueberry(Vaccinium ashei) is one of the most widely grown blueberry types in the world, together with Northern and Southern highbush blueberry(Vaccinium corymbosum). Rabbiteye blueberry have higher soil adaptability and fruit productivity but less cold tolerance to low temperature than highbush blueberry. The objective of this study is to investigate freezing tolerance of floral buds and establish a cultivation zone for rabbiteye blueberry cultivars. METHODS AND RESULTS: Bearing branches which have similar thickness and same number of floral buds were collected in the early January at the blueberry germplasm preservation plot located in Namhae Sub-station, National Institute of Horticultural and Herbal Science in Gyeongsangnamdo. Cold response of bearing branches were investigated by electrolyte leakage and freezing tolerance of floral buds were determined by ovary browning ratio of 50%($LT_{50}$). Cultivation zone was established based on mean annual extreme minimum temperature for 30 years, from 1981 to 2010. The electrolyte leakage of bearing branches in rabbiteye blueberry increased as temperature decreased and was lowest in 'Brightwell' but highest in 'Bluegem' when they were kept in $-5^{\circ}C$. Besides, the electrolyte leakage increased in 'Brightblue', 'Brightwell', 'Climax', 'Delite', 'Gardenblue', 'Southland' and 'Woodard' in $-20^{\circ}C$. Freezing tolerance($LT_{50}$) was lowest in 'Bluegem' and 'Homebell'($-13.3^{\circ}C$), and highest in 'Tifblue'($-25^{\circ}C$) among different rabbiteye blueberry cultivars. $LT_{50}$ of 'Southland' was from -15.0 to $-16.7^{\circ}C$, that of 'Delite', 'Brightwell',' Austin' and 'Climax' was $-18.3^{\circ}C$, and that of 'Bluebelle', 'Woodard' and 'Powderblue' was $-20^{\circ}C$. CONCLUSION: This study indicate that The hardiness zones of rabbiteye blueberry were classified into Six cultivation zones and cultivation zones of most cultivars were the south of Jeollanam-do and Gyeongdangnam-do, except for 'Tifblue.'

When producing recycled asphalt mix, it is important that the old binder of reclaimed asphalt pavement(RAP) should be well melted during blending in the mixer. The recycled asphalt mix is produced by instant mixing(IM) of all materials(RAP, virgin asphalt and new aggregates) all together in the mixer. However, in the same recycled mix, the binder around RAP aggregate was found to show higher oxidation level than the binder coated around the virgin aggregate because the old binder of RAP was not rejuvenated properly while instant mixing. The partially-rejuvenated RAP binder is assumed to be a high stiffness point in IM recycled mix. In this study, the stage mixing(SM) method was introduced; blending RAP and virgin asphalt for the first stage, and then mixing all together with hot new aggregates for the second stage. To compare the effect of the two mixing methods on moisture resistance of recycled mixes, a statistical t-test was performed between SM and IM using indirect tensile strength(ITS) and tensile strength ratio(TSR). Three conditioning methods were used; a 16-h freezing and then 24-h submerging, 48-h submerging, and 72-h submerging in 60℃ water. It was found that the TSR(=ITSwet/ITSdry) values of the mixes prepared by SM was clearly higher than the IM mixes, and coefficients of variation of SM mixes were lower than the IM mixes. It was also observed that the ITSWET of SM was significantly different from the IM at α=0.05 level by statistical t-test. The ITSWET of SM mix was reduced less than the IM mix in severer conditioned mixes. Therefore, it was concluded that the stage mixing method was an important blending technique for producing better-quality of recycled asphalt mixes, which would show higher moisture resistance than the recycled mixes produced by conventional instant mixing.
