  • There exist many types of pipe and component fatigue through vibrations, thermal fatigues or shifting. In some cases of thermal stratification/thermal fatigue, pipes & components are receiving thermal stress by means of material expansion and shrinkage by continuous thermal repetitive variation. Small cracks initially occur on the inside surface by thermal stress. These cracks grow in depth the pipe wall and finally come to a rupture. Pipe parts of susceptibility to thermal stratification and thermal fatigue are now being examined by conventional UT(ultrasonic test) as volumetric examination. It is difficult to fully satisfy the code & standards requirements because 3" weldolet weldments of RCS 16" pipe to 3" branch connection lines have complex structural shape. To solve the problems of conventional UT examination, we made a realistic mock-up and UT calibration block. We performed a simulation of phased array UT utilizing CIVA as NDE(Non-Destructive Examination) simulation software. Also we designed phased array UT transducer and wedge, optimal frequency by using simulation data. We performed phased array UT experiment through mock-up including artificial flaws(notch). The phased array UT technique is finally developed to improve the reliability of ultrasonic test at RCS 16" pipe to 3" branch connection weld.

  • Metal fatigue is an important aging mechanism that material characteristics can be deteriorated when even a small load is applied repeatedly. An accurate fatigue evaluation is very important for component structural integrity and reliability. In the design stage of a nuclear power plant, the fatigue evaluations of the Class 1 components have to be performed. However, operating experience shows that the design evaluation can be very conservative due to conservatism in the transient severity and number of occurrence. Therefore, the fatigue monitoring system has been considered as a practical mean to ensure safe operation of the nuclear power plants. The fatigue monitoring system can quantify accumulated fatigue damage up to date for various plant conditions. The purpose of this paper is to describe the fatigue monitoring procedure and to introduce the fatigue monitoring program developed by the authors. The feasibility of the fatigue monitoring program is demonstrated by comparing with the actual operating data and finite element analysis results.

  • The national coordinate system is an essential component for a geographic/land information system, since it provides the spatial reference for expressing position information. The national mapping of Korea has been based on 3-different meridians on the Gauss-Schreiber projection in year 1910s, later this was changed to the Gauss-Kruger projection. Existing map coordinate systems maintaining the national land survey project on 1910s, have some structural shortcomings of unknown computational procedures and projection methods. In this paper, the problems of the map coordinates usage and of longitudes origin shift(10.405") and their solutions are investigated. Also, this study discusses the issues involved in choosing coordinate system for digital mapping and their applications as a basis for spatial data management. The foreign country's coordinate systems are reviewed and the elements to realize a new unified grid coordinate system is proposed. The Transverse Mercator projection with a central meridian of $127^\circ\;30'$, scale factor 0.9996, and GRS80 ellipsoid, is selected in Korean peninsula.sula.

Effect of Cold Stress on Myocardium of Aging Rat (Cold Stress가 노화(老化) 흰쥐 심근(心筋)에 미치는 영향(影響))

  • The present study was performed to determine the effect of cold stress on myocardium of aging rat. Control groups, which aged 6, 12 and 24 months, were compared with age-matched experimental groups that were exposed to moderate cold stress for a hours daily in a week at laboratory cold room $(4{\pm}1^{\circ}C)$. The histological, histochemical and ultrastructural changes of myocardium were observed. The results were summarized as follow: 1. Age-dependent histological change of control groups was observed the formation of contraction band in 24months aged group. The experimental groups submitted to cold stress showed a similar change pattern as seen in control groups. However, the degree of change in the experimental groups was significantly larger than that of control groups. In the 34 months aged group the formation of hypercontraction band was observed. 2. Regarding age-dependent histochemical changes of control groups, we observed the increase activities of PAS and Masson's trichrome. In experimental groups the activities of PAS and Masson's trichrome were also increased with age. Compare with control group, the activities of PAS was increased but the activities of Masson's trichrome was decreased. 3. Age-dependent ultrastructural changes on vacuolization, lysosome were observed. In control groups the structural changes occur at 12 months. The accumulation of lipofuscin, contraction band, hypercontraction band and a component of connective tissue were observed in 24 months. However, the degree of change in the experimental groups was significantly larger than that of control groups. In contract, the myelin body in intercalated discs was observed in 24 months of experimental groups.

  • Soils containing vanishing materials lead changes in the microstructure of particulate media due to water inflow. Thus, dissolution renders some local unstability. As the moisture contents decease, the component of the vanished materials may affects on the cementation of paniculate materials. This cementation phenomenon has a huge influence on the stiffness, strength and stability under lower stress level. The goal of this study is to introduce the cementation effects on a compressional wave velocity, a shear wave velocity, and the resonant frequency of shear waves. The glass bead and salt water with different mole contents are used. Test results show that the changes of shear and compressional wave velocities consist of three stages. In the first region, compressional wave velocities increase and shear wave velocities decrease with a decreases in reducing water contents from 100% to 90~95%. In the second region, shear and compressional wave velocities become stable at 90~95% to 10% of the water contents. In the third region, shear and compressional wave velocities increases dramatically with a decrease in the water content due to the capillary force and cementation of salt. Furthermore, the resonant frequency of the shear waves shows similar phenomenon. Specimens prepared by glass beads and salt water are proved to be able to provide a meaningful insight in under structural behaviors of the cementation.

Purification and refolding of the recombinant subunit B protein of the Aggregatibacter actinomycetemcomitans cytolethal distending toxin

  • Purpose: Aggregatibacter actinomycetemcomitans is associated with localized aggressive periodontitis. It produces cytolethal distending toxin (CDT), which induces cell cycle arrest in the G2/M phase. The CDT holotoxin is composed of CdtA, CdtB, and CdtC. CdtB has structural homology to human DNase I and is an active component of the CDT complex acting as a DNase. In particular, the pattern homology seen in the CdtB subunit has been associated with specific DNase I residues involved in enzyme catalysis, DNA binding, and metal ion binding. So, to study the functions and regulation of recombinant CdtB, we made up a quantity of functional recombinant CdtB and tested it in relation to the metal ion effect. Materials and Methods: We constructed the pET28a-cdtB plasmid from A. actinomycetemcomitans Y4 by genomic DNA PCR and expressed it in the BL21 (DE3) Escherichia coli system. We obtained the functional recombinant CdtB by the refolding system using the dialysis method and then analyzed the DNase activity and investigated the metal ion effect from plasmid digestion. Results: The recombinant CdtB subunit was expressed as the inclusion bodies. We were able to obtain functional recombinant CdtB subunit using refolding system. We confirmed that our refolded recombinant CdtB had DNase activity and was influenced by the metal ions $Mg^{2+}$ and $Ca^{2+}$. Conclusion: We suggest that the factors influencing recombinant CdtB may contribute to CDT associated diseases, such as periodontitis, endocarditic, meningitis, and osteomyelitis.

Immersion grating mount design of IGRINS

  • The IGRINS (Immersion GRating INfrared Spectrometer) is a high resolution wide-band infrared spectrograph developed by Korea Astronomy and Space Science Institute (KASI) and the University of Texas at Austin (UT). Immersion grating is a key component of IGRINS, which disperses the input ray by using a Silicon material with a lithography technology. Opto-mechanical mount for the immersion grating is important to keep the high spectral resolution and the optical alignment in a cold temperature of $130{\pm}0.06K$. The optical performance of immersion grating can maintain within the de-center tolerance of ${\pm}0.05mm$ and the tip-tilt tolerance of ${\pm}1.5arcmin$. The mount mechanism utilizes the flexure and the kinematic support design to satisfy the requirement and the operation condition. When the IGRINS system is cooled down to a cold temperature, three flexures compensate the thermal contraction stress due to the different material between the immersion grating and the mounting part(Aluminum 6061). They also support the immersion grating by an appropriate preload. Thermal stability is controlled by a copper strap with proper dimensions and a heater. Generally structural and thermal analysis was performed to confirm the mount mechanism. This talk presents the opto-mechanical mount design of the immersion grating of IGRINS.

  • This paper shows the development procedure of the low pressure LOX feed system which is used in the liquid rocket with a turbopump. Korean Air has cooperated with KARI in developing the LOX feed system to turbopump. The LOX feed system is characterized with cryogenic temperature and the thin-thickness tube for weight saving. The system in this project is composed with a main feed line and a recirculation line for the LOX temperature conditioning. Each piping system has many components, namely, bellows, filter, orifice, valves, flange and support. In this paper, system design & manufacturing, structural & thermal analyses, and component tests are explained. Finally, the system was assembled to the KARI's PTF test facility and functioned well to meet its required performance.

Direct-Patternable SnO2 Thin Films Incorporated with Conducting Nanostructure Materials (직접패턴형 SnO2 박막의 전도성 나노구조체 첨가연구)

  • There have been many efforts to modify and improve the properties of functional thin films by hybridization with nano-sized materials. For the fabrication of electronic circuits, micro-patterning is a commonly used process. For photochemical metal-organic deposition, photoresist and dry etching are not necessary for microscale patterning. We obtained direct-patternable $SnO_2$ thin films using a photosensitive solution containing Ag nanoparticles and/or multi-wall carbon nanotubes (MWNTs). The optical transmittance of direct-patternable $SnO_2$ thin films decreased with introduction of nanomaterials due to optical absorption and optical scattering by Ag nanoparticles and MWNTs, respectively. The crystallinity of the $SnO_2$ thin films was not much affected by an incorporation of Ag nanoparticles and MWNTs. In the case of mixed incorporation with Ag nanoparticles and MWNTs, the sheet resistance of $SnO_2$ thin films decreased relative to incorporation of either single component. Valence band spectral analyses of the nano-hybridized $SnO_2$ thin films showed a relation between band structural change and electrical resistance. Direct-patterning of $SnO_2$ hybrid films with a line-width of 30 ${\mu}m$ was successfully performed without photoresist or dry etching. These results suggest that a micro-patterned system can be simply fabricated, and the electrical properties of $SnO_2$ films can be improved by incorporating Ag nanoparticles and MWNTs.

Distribution of Lipid and Lipase in Lipid-and Starch-Rich Seeds (지질 및 전분성 종자에서 지질 및 지질가수분해효소의 분포)

  • Formation, cellular distribution and structural changes of storage lipid, and active site and cellular localization of lipase in endosperms and cotyledons of lipid-rich seeds such as Helianthus annuus, Ricinus communis and Pinus koraiensis, and in those of starch-rich seeds such as Pisum sativum and Zea mays were investigated in relation to the seed development by cytochemical methods. In endosperms and storage cotyledons of lipid- and starch-rich seeds after seed-gathering, there were widely distributed storage material which was composed of spherical protein bodies, spherosomes, and starch granules. But cellular organelles were hardly observed in the cytoplasm. Staining pattern of vesicles released from SER, and of low electron dense membraneous granules, which were perhaps at an early stage of spherosomes, were the same as in the spherosome. Electrondense granules released from RER were observed in the vicinity of plasma membrane. As a result of lipid staining, the spherosomes were more electron dense and were uniform as compared with the protein matrix within the protein body and cytoplasmic proteinaceous granules. The major component of the spherosome was determinated to be lipid. Spherosomes and vesicles containing SER-released materials showed the same as in the electron density. Lipase activity was especially strong in the inner region and on the surface of decomposed spherosomes and near the plasma membrane.mbrane.
