Ultrastructure of Acinar Secretory Granules of Submandibular and Parotid Salivary Gland in the Korean Striped Field Mouse, Apodemus agrarius (Rodentia, Murinae)

  • The ultrastructures of the secretory acinar granules of submandibular and parotid salivary gland were examined in the Korean striped field mouse, Apodemus agraius. The acini of the submandibular salivary gland had serous and mucous acinar cells filled with numerous secretory granules. The serous acinar granules had uniformly fine dense contents and were round typed with a definite boundary between the granules. The mucous acinar granules were relatively coarse, with moderate density, and clustered together as a result of the indistinct boundaries between the granules. The acini of the parotid salivary glands contained only serous cells filled with numerous round-typed serous acinar granules. Serous acinar granules had uniformed dense matrix and definite boundaries. The ultrastructures without substructure in a matrix of serous and mucous acinar granules in the submandibular and parotid salivary glands of A. agraius were similar to those of species of Rodentia but different from those of Soricidae in Korea with a characteristic substructure in a matrix. This ultrastructure and charateristics in secretory acinar granules provide fundamental data for molecular comparisions of genetic relationships and are one of the key methods for classifying A. agraius.

  • Seasonal variation of the population structure and the reproductive pattern of the Korean striped field mouse, Apodemus agrarius, were investigated. High capture ratios in juveniles, young adult, and old adult mice were found during the period from October to November, from November to March, and from May to September, respectively, and extremely low capture ratios of old adults during the period from November to February were characteristic. It seemed that the young adults that survived during the winter might become older by summer and have been counted as the old adults. The breeding in the mice began earlier in males (from mid February or early March to late October) than in females (from mid March to late October), having a peak in August and September, and both the male and female mice weighing more than 20 g generally reached sexual maturation in general. In the breeding season, both young and old adult males had large testes with enlarged seminiferous tubules filled with numerous germ and Sertoli cells, and expanded caudal epididymides with a vast number of spermatozoa; the females had many Graafian follicles and corpora lutea in large ovaries, and developed uterine glands in the thick endometria. The lower ratios of the testis weight to the body weight in July and August in 1994 compared to 1995 seemed due to the extreme drought and considerably higher temperature in 1994, but the decrease in the ratio in mid-summer, only in the old mice, in both years might be explained partially by aging.

  • Four external and 27 cranial characters of three subspecies of striped field mice (Apodemus agrarius) from 15 regions in far eastern Asia(South Korea, North Korea, northeastern China, and far eastern Russia) were analyzed by multivariate methods in order to determine taxonomic status of striped field mice from North Korea. Three subgroups were recognized: a large-size form from two localities in South Korea(Cheju Island, Wando Island): a medium-size form from eight localities in South Korea and North Korea(Jindo Island, Kunsan, Mt. Chirisan, Cheongju, Mt. Weoleksan, Mt. taebaksan; Haeju, Sineuiju); a small-size form from five localities in North Korea, northeastern China, and far eastern Russia (Mt. Kumkangsan, Mt. Myohyangsan; Jirin, Yichun; Vladivostok). It is confirmed that the large-size form, the medium-size form, and the small-size form are subspecies chejuensis, coreae, and manchuricus, respectively, as noted by Koh et al.(1997). It is revealed that striped field mice(the medium-size form) from western region of North Korea (Haeju, Sineuiju) is classified into A. agrarius coreae, as noted by Thomas(1908), but striped field mice(the small-size form) from eastern region of North Korea(Mt. Kumkangsan, Mt. Myohyangsan) is A. agrarius manchuricus.

Intestinal Helminthic Infections in Striped Field Mice, Apodemus agrarius, from Two Southern Regions of Korea

  • The present study was performed to know the infection status of intestinal helminths in a most common species of field mice, Apodemus agrarius, from 2 southern regions of Korea. Total 133 and 103 mice were collected by the mouse trap in Hapcheon-gun, Gyeongsangnam-do and Gurye-gun, Jeollanam-do, respectively, from July 2005 to June 2006. The small intestine of each mouse was resected and longitudinally opened with a pair of scissors. The intestinal contents were washed with 0.85% saline until the supernatant became clear. Helminths were collected with naked eyes or under a stereomicroscope from the sediment of the intestinal content. More than 11 species of helminths (4 nematode spp., 5 trematode spp., and 2 cestode spp.) were recovered. Among these, heligmosomoid nematodes (97.5%) was the most highly and heavily infected species. As the members of trematodes, Plagiorchis muris, Brachylaima sp., Echinostoma hortense, Echinostoma cinetorchis, and unidentified echinostome larvae were found in the small intestines of 35 (14.8%), 12 (5.1%), 6 (2.5%), 1 (0.4%), and 1 (0.4%) mice respectively. Two species of tapeworms, Hymenolepis nana and Hymenolepis diminuta were also detected in 79 (33.5%) and 21 (8.9%) mice, respectively. Conclusively, heligmosomoid nematodes were the most prevalent (dominant) species among more than 11 helminth species detected, and Brachylaima sp. fluke is newly added in the list of intestinal trematodes in Korea.

Ecological Studies of the Field Mouse (들쥐의 生態學的 硏究)

  • The present investigation has been done to observe the ecological habits of field mice to protect the rice from damages during the growing season in paddy-field and during the storge period. The results obtained are summarized as follows: 1. Of 155 mice captured in the period of April-November 1970, which belong to four genera (Apodemus, Cricetulus, Rattus, and Micromys), 148 mice(95%) were found as striped field mice (Apodemus agrarius coreae). The population density of striped field mouse was revealed by the present study as 55/ha, which is quite a low level compared with that in Japan of 900/ha. 2. The age distribution of the mice as judged by their body weight was found mainly composed of adult and the sex ratio was found to be 1.8 as determined with 147 individuals. The nest was found to be occupied by an adult and was composed of at least three openings and more than one food storage tunnels. The mice usually keep hulled rice rather than unhulled one in storage tunnel. The weight of food found in a nest was about 50 grams on an average. 3. The mice show a most active behaviour 1-2 hours after the sunset, around midnight, and an hour before the sunrise, but they are active even in daytime in order for searching for food and for breeding. 4. The ratio (%) of damage appeared in high stem of sweet corn in August was 30 ~ 40 percent, whereas that in low stem was 80 ~ 90 percent. The weight of spoiled grains in paddy-field was 11, 400gm/0.4ha and this gives an estimate of 349, 695 for whole country. 5. The female striped field mouse weighs average of about 30 grams and gives birth to average of 4.8 younglings which wean away from female mouse three weeks after delivery. 6. The natural enemies to the mice are found to be carnivores (weasel, cat, mountain cat, fox, raccoon, and otter), raptatores(eagle, owl, kete, buzzard), and snakes. Two kinds of field rats(Rattus norvegicus, Cricetulus tritor) are also the predator to the mice. 7. The feeding preference of striped field mice follows in decreasing order of sweet corn, soybean, sweet potatoes, chestnut, and wheat. The mice do not have a preference for barley, millet, rough millet, red bean, and green bean. 8. The starvation experiment, in which water alone was supplied, revealed that the mice in good physical and nutritional conditions survived for 71 ~ 79 hours, whereas those in worse conditions survived for only 32 ~ 39 hours.

Diversity of Mitochondrial DNA cytochrome b Gene in Two Subspecies of Striped Field Mouse, Apodemus asrarius coreae Thomas and A. asrarius manchuricus Thomas (Mammalia, Rodentia) from Korea and Northeast China (한국과 북동 중국에 서식하는 등줄쥐 2아종, Apodemus agrarius coreae Thomas and A. agirarius manchuricus Thomas (포유강, 설치목)의 미토콘드리아 DNA cytochrome b 유전자의 다양성)

  • The partial sequences of mtDNA cytochrome b gene in two subspecies of striped field mouse(Apodemus agrarius coreae and A. agrarius manchuricus) from Korea and northeast China were analyzed to determine the degree of genetic diversity in ech subspecies and to confirm their subspecific difference. In 18 specimens of A. agrarius coreae, ten haplotypes were resulted, and in two specimens of A. agrarius manchuricus. one haplotype was revealed. Tamura-Nei nucleotide distance among ten haplotypes in subspecies coreae ranged 0.36 to 1.86%. and nucleotide distance between two subspecies (coreae and manchuricus) was 0.37 to 1.47%: maximum infrasubspecific divergence in coreae was greater than maximum intersubspecific difference between two subspecies. Moreover, no major subgroup was resulted when 11 haplotypes in two subspecies were compared. Our sequence result was not cancordant with the morphological data studied so far, and it is concluded that cytochrome b gene sequence is not a good genetic marker to distinguish two subspecies of A. agrarius. In futurem, mtDNA control region analyses seemded to be necessary to reveal genetic relationship between these two subspecies of A. agrarius.

Environmental Factors on the Use of Wildlife Bridge by Striped Field Mouse (Apodemus agraius) (등줄쥐의 육교형 생태통로 이용에 미치는 환경 특성)

  • Although wildlife bridge are built as a way to reduce habitat fragmentation caused by road construction, there is still a lot of debate about their effectiveness. Monitoring methods such as footprint traps and camera traps are used evaluate the effectiveness of wildlife bridge, but there is a limit to evaluate of effectiveness. In this study, the degree of use the wildlfe bridge was surveyed by striped field mouse that is likely use the wildlife bridge and surrounding as a habitat with capture-mark-recapture method.(Apodemus agraius). The distance and route of movement were identified by connecting the capture points, and the environmental factors on the use of the wildlife bridge implemented a generalized linear model(GLM) with the capture number of captured as a dependent variable. Consequently of capture, no individuals crossing the wildlife bridge, striped field mouse use the wildlife bridge as a habitat.The environmental factors affecting the use of mice were vegetation cover(1~2m, 2~8m, over 8m), vegetation construction, maximum diameter at breast height were positively correlated and slope was nagatively correlated. In conclusion, it is expected that the effectiveness of the wildlife bridge will be further improved by planting shrubs and trees and preventing high slope and cut slope increasing the utilization of the rat, such as being used as a food source in the ecosystem.

Morphological Changes of Epithelial Cells of the Epididymides by Sperm Entrance in the Korean Striped Field Mouse, Apodemus agrarius coreae (등줄쥐(Apodemus agrarius coreae)의 정자유입에 따른 정소상체 상피세포의 형태적 변화)

  • To investigate the morphological changes of epithelial cells of ductus epididymides by sperm entrance, the Korean striped field mouse, Apodemus agrarius coreae was studied with light and transmission electron microscopy. 1. The diameters of the caput epididymis (Cp) and its lumen were $135\sim145{\mu}(140{\pm}0.5{\mu})$ and $115\sim120{\mu}m(117{\pm}0.1{\mu}m)$. The length and width of the epithelium were $27.0\sim28.5{\mu}m(28.0{\pm}0.1{\mu}m)$ and $4.8\sim5.4{\mu}m(5.1{\pm}0.2{\mu}m)$. 2. The diameters of the corpus epididymis (Cr) and its lumen were $160\sim170{\mu}m(166{\pm}0.2{\mu}m)$ and $140\sim150{\mu}m(145{\pm}0.3{\mu}m)$. The length and width of the epithelium were $17.4\sim18.0{\mu}m(17.6{\pm}0.5{\mu}m)$ and $8.8\sim10.4{\mu}m(9.5{\pm}0.2{\mu}m)$. 3. The diameters of the cauda epididymis (Cu) and its lumen were $270\sim280{\mu}m(275{\pm}0.2{\mu}m)$ and $265\sim275{\mu}m(268{\pm}0.3{\mu}m)$. The length and width of the epithelium were $11.2\sim13.4{\mu}m(12.3{\pm}0.3{\mu}m)$ and $9.2\sim11.2{\mu}m(10.0{\pm}0.2{\mu}m)$. Therefore, the size of diameter and lumen of the ductus epididymides and the width of the epithelium increased from Cp and Cr to Cu, but the length of epithelial cells decreased from Ca and Cr to Cu. These data suggest that the morphological changes of epithelial cells of the ductus epididymides may be the results of the sperm entrance.
