In an attempt to identify the neuroprotective effect of Cudrania tricuspidata (CT) leaves against ROS (reactive oxygen species)-induced oxidative stress in neuronal cells, the extracts from CT leaves were investigated using PC12 cells and N18-RE-105 cells. The methanolic and ethanolic extracts from CT were denoted as CTM (Cudrania tricuspidata Leaves methanolic extracts) and CTE (Cudrania tricuspidata Leaves ethanolic extracts), respectively. The neuroprotective effects of the extracts were measured by DCF-DA assay, MTT reduction assay, and LDH release assay. The PC12 cells exposed to $H_2O_2$-induced oxidative stress and the N18-RE-105 cells exposed to glutamate-induced oxidative stress were treated with various concentrations of CTM and CTE. The results, CTM treatments resulted in the induction of a dose-dependent protective effect in PC12 cells and N18-RE-105 cells. Interestingly, CTE also showed neuroprotective effect in PC12 cells and N18-RE-105 cells. Therefore, these results suggest that CTM and CTE could be a new potential candidate as neuroprotective agents against ROS-induced oxidative stress in neuronal cells.
The present study was conducted to investigate the effects of flowers ethanol extract of Elsholtzia splendens (ESE) on the antioxidant defence system in mice with 7,12-dimethylbenz(a)anthracene (DMBA)-induced oxidative stress. The ESE was pre-administered orally to 2 groups of mice at 10 and 50mg/kg body weight (BW) for 5 weeks. Subsequently, mice with pretreatment of ESE received DMBA intragastrically at a dose of 34 mg/kg BW twice a week for 2 weeks. In DMBA alone group, biomarkers of oxidative stress (TBARS value, carbonyl content, and serum 8-OH-dG) were significantly increased. Also, the antioxidant enzymes were down-regulated. ESE significantly restored the TBARS value and carbonyl content at both doses, while a decrease in the elevated serum 8-OH-dG content was observed only at the higher dose. The DMBA-induced decreases in catalase and superoxide dismutase (SOD) activities were restored to nearly control levels by ESE. Glutathione peroxidase (GSH-px) and glutathione reductase (GR) activities, as well as the reduced glutathione (GSH)/oxidized GSH (GSSG) ratio, were significantly affected by ESE at the higher dose. These results suggest that ESE possesses antioxidant activity, which plays a protective role against DMBA-induced oxidative stress.
[Purpose] The purpose of this study was to investigate the effects of treadmill exercise on oxidative stress in the hippocampal tissue and mitochondrial dynamic-related proteins in rats fed a long-term high-fat diet (HFD). [Methods] Obesity was induced in experimental animals using high fat feed, and the experimental groups were divided into a normal diet-control (ND-CON; n=12), a high fat diet-control (HFD-CON; n=12) and a high fat diet-treadmill exercise (HFD-TE; n=12) group. The rats were subsequently subjected to treadmill exercise (progressively increasing load intensity) for 8 weeks (5 min at 8 m/min, then 5 min at 11 m/min, and finally 20 min at 14 m/min). We assessed weight, triglyceride (TG) concentration, total cholesterol (TC), area under the curve, homeostatic model assessment of insulin resistance, and AVF/body weight. Western blotting was used to examine expression of proteins related to oxidative stress and mitochondrial dynamics, and immunohistochemistry was performed to examine the immunoreactivity of gp91phox. [Results] Treadmill exercise effectively improved the oxidative stress in the hippocampal tissue, expression of mitochondrial dynamic-related proteins, and activation of NADPH oxidase (gp91phox) and induced weight, blood profile, and abdominal fat loss. [Conclusion] Twenty weeks of high fat diet induced obesity, which was shown to inhibit normal mitochondria fusion and fission functions in hippocampal tissues. However, treadmill exercise was shown to have positive effects on these pathophysiological phenomena. Therefore, treadmill exercise should be considered during prevention and treatment of obesity-induced metabolic diseases.

The present study has numerically predicted both the flow -induced and thermally-induced residual stresses and birefringence in injection o. injection/compression molded center -gated disks. Analysis system for entire molding process was developed based on an ap propriate physical modeling including a nonlinear viscoelastic fluid model, stress-optical law, a linear viscoelastic solid model, free volume theory for density relaxation phenomena and a photoviscoelasticity and so on. Part I presents physical modeling a nd typical numerical analysis results of residual stresses and birefringence in the injection molded center-gated disk. Thermal residual stress was found to be extensional near the center, compressive near the surface and tend to become toward tensional at the surface. A double-hump profile was obtained across the thickness in birefringence distribution: nonzero birefringence is found to be thermally induced, the outer peak is due to the shear flow and subsequent stress relaxation during the filling stage a nd the inner peak is due to the additional shear flow and stress relaxation during the packing stage. Predicted birefringence including both the flow -induced and thermally-induced one becomes quite similar to the experimental one.

The accompanying paper, Part 1, has presented the physical modeling and basic numerical analysis results of both the flow-induced and thermally-induced residual stress and birefringence in injection molded center gated disks. The present paper, Part II, has attempted to investigate the effects of various processing conditions of injection/compression molding process on the residual stress and birefringence. The birefringence is significantly affected by injection melt temperature, packing pressure and packing time. Birefringence in the shell layer increases as melt temperature gets lower. The inner peak of birefringence increases with packing time and packing pressure. On the other hand, packing pressure, packing time and mold wall temperature affect the thermally-induced residual stress rather significantly in the shell layer, but insignificantly in the core region. Injection/compression molding has been found to reduce the birefringence in comparison with the conventional injection molding process. In particular, mold closing velocity and initial opening thickness in the compression stage of injection/compression molding process have significant effect on the flow-induced birefringence, but not on tile thermal residual stress and the thermally induced birefringence.
This study was done to know the effects of the water extracts of Gyogamdan(GGD) on the function of macrophages, the most important cells of the innate immune system, and the rosette forming ability of splenocytes in the mouse under stress. The effects of GGD on the immunosuppression induced by noise stress are as follows. 1. Administration of GGD water extracts normalized the bo요 weight which might be decreased by noise stress. 2. Administration of GGD water extracts increased the production of the such reactive oxygen intermediates as superoxide and hydrogen peroxide from macrophsges in vivo & in vitro which were decreased by noise stress. 3. Administration of GGS water extracts did not affect the production of reactive nitrogen intermediates. 4. Administration of GGD water extracts increased the rosette forming ability of splenocytes which was decreased by noise stress. The above effects of GGD might be useful for the treatment of stress-induced infections diseases which could be caused by the suppression of immune responeses which are initiated by the functions of macrophages of the innate immune system.
The fatigue crack growth behavior of the SA516/70 steel which is used for pressure vessels was examined experimentally at room temperature, $-60^{\circ}C$,$-80^{\circ}C$ and $-100^{\circ}C$ with stress ratio of R=0.05, 0.1 and 0.3. Fatigue crack propagation rate da/dN related with stress intensity factor range ${\Delta}K$ was influenced by stress ratio in stable of fatigue crack growth (Region II) with an increase in ${\Delta}K$. The resistance of fatigue crack growth at low temperature is higher compared with that at room temperature, which is attributed to the extent of plasticity-induced by compressive residual stress according to the cyclic loads. Fractographic examinations reveal that the differences of the fatigue crack growth characteristics between room and low temperatures are mainly explained by the crack closure and the strengthening due to the plasticity induced and roughness induced.

The fatigue crack growth behavior of the SA516/60 steel which is used for pressure vessels was examined experimentally at room temperature $25^{\circ}C, -30^{\circ}C, -60^{\circ}C, -80^{\circ}C, -100^{\circ}C$ and -l2$0^{\circ}C$ with stress ratio of R=0.05, 0.1 and 0.3. Fatigue crack propagation rate da/dN related with stress intensity factor range ΔK was influenced by stress ratio in stable of fatigue crack growth (Region II) with an increase in ΔK. The resistance of fatigue crack growth at low temperature is higher compared with that at room temperature, which is attributed to the extent of plasticity-induced by compressive residual stress according to the cyclic loads. Previous studies reported that exposure to noise during pregnancy adversely influenced the development of the fetus and neonate. In Oriental medicine, medications based on Angelicae gigantis radix have been known to be of efficacy in the treatment of various diseases. c-Fos, an immediate early gene whose expression is sometimes used as a marker for stimulus-induced changes in the metabolic activity of neurons. In the present study, the influence of postnatal Angelicae gigantis radix administration on c-Fos expression in the each region of hippocampus of offspring rats with prenatal noise stress during pregnancy was investigated. From the present results, exposure to the prenatal stress during pregnancy enhanced c-Fox expression, whereas exposure to postnatal Angelice gigantis radix suppressed c-Fos expression in the offsprings with prenatal noise stress during pregnancy. Based on the present study, Angelicae gigantis radix may provide new therapeutic opportunities as an agent to counteract the effects of prenatal noise stress- induced hippocampal dysfunction, and may be useful in the treatment of psychiatric problems in children of mothers who have experienced noise stress during pregnancy.
Although reactive oxygen species (ROS) are inevitable by-products of many redox reactions in eukaryotic cells, they play a crucial role as signaling molecules in many cellular processes for development and defense response to abiotic stresses. The biphasic ROS production which was peaked twice in a first transient phase and a second massive phase was occurred after treatment of abiotic stress such as oxidative stress, high salinity. This biphasic generation of ROS was followed by the biphasic production of stress hormone, ethylene. The mechanism of interactions between ROS and ethylene biosynthesis is studied in tobacco (Nicotiana tabaccum L.) plants under the abiotic stresses. The stress-induced ethylene production was significantly inhibited in RbohD-AS and RbohF-AS, in which antisense expression of NADPH oxidase genes was performed. The accumulation of ROS, which was determined by DAB and DCFH-DA staining, was significantly decreased after abiotic stresses in transgenic plants. The suppression of signaling with ethylene and ROS induced more tolerance in response to abiotic stress. The transgenic plants were more tolerant in MS medium supplemented with salinity stress in contrast with wild-type. Stress-induced cell damage determined by DNA fragmentation was decreased at phase II in those transgenic plants. Therefore, the first burst of ROS is more responsible for making a role as a signaling molecule during stress-induced response. These results suggested that ethylene and ROS act in a positive feedback cycle that results in mutual enhancement of ethylene and ROS production during stress-induced cell death.
