천일염 함유 청국장의 항산화효과 (Antioxidative Effects of Chungkukjang Preparation by Adding Solar Salt)

  • 본 연구에서는 천일염의 아직 밝혀지지 않은 기능성과 청국장의 효과를 감안하여 천일염으로 제조된 청국장의 기능적 우수성을 평가하기 위하여 Bacillus subtilis DJI 균주를 접종하여 발효시킨 무염청국장, 3% 천일염 첨가 청국장, 3% 정제염 첨가 청국장과 3% 정제염이 첨가되어진 시판청국장의 에탄올, 물 및 헥산 추출물의 항산화력을 비교 검토하였다. 청국장 에탄올, 헥산 및 물 추출 수율은 각각 $10.9{\sim}11.8%$, $15.7{\sim}16.9%$, $18.6{\sim}19.4%$ 범위로 청국장 용매별 간에는 서로 유사한 경향이었으며, 물 추출 수율이 가장 높게 나타났다. 추출물의 DPPH에 대한 전자소거능, 아질산염 소거능, linoleic acid에 대한 과산화물가 및 항산화지수 측정에 의한 항산화효과는 에탄올 추출 시료군이 물 추출 시료군에 비하여 높았다. 청국장 추출물의 전자소거능과 아질산염 소거능은 무염청국장과 3% 천일염 첨가 청국장 추출물이 가장 우수하였으나, 합성항산화제에 비해서는 낮은 효능을 보였다. 청국장 추출물 중 항산화지수는 3% 천일염 첨가 청국장 추출물이 가장 우수하였다. Bacillus subtilis DJI 균주를 접종하여 발효시킨 청국장인 무염청국장, 3% 천일염 첨가 청국장 및 3% 정제염 첨가 청국장이 시판청국장에 비하여 항산화효과가 우수하였으며, 균주를 접종하여 발효시킨 청국장 중 무염청국장과 3% 천일염 첨가 청국장이 3% 정제염 첨가 청국장에 비하여 항산화효과가 다소 우수하였다.

Enhancing Production of Terpenoids in Metabolically Engineered Transgenic Spearmint (Mentha spicata L.) by Salt and Fungal Elicitors

  • Forest tree species usually takes for long periods to be harvested and cultivated but spearmints are a good model system for woody plant because of reducing and shortening cultivation time. Spearmints are good model plants (Mentha species) for research about terpenoids production and industrial essential oil manufacture. Isopentenyl pyrophosphate isomerase (Iso) and limonene synthase (Limo) are the key enzymes of terpenoid biosynthesis pathway. Transgenic and wild spearmints (Mentha spicata, MS) were cultured in vitro and assessed for the essential oil contents. The content of essential oil of transgenic spearmint also was enhanced slightly depending on the target terpenoid genes. In an attempt to increase productivity of terpenoids further, salt and fungal elicitation strategy was adopted on transgenic Mentha spicata. The salt (800 mM NaCl) as abiotic and two fungi (Botrytis cinerea and Glomerella cingulata) as biotic were used for elicitors. In the absence of salt stress four terpenoids were detected from the spearmint extracts, all of them being monoterpenes. On the other hand, the transgenic (MSIso) extracts contained eleven terpenoids (10 monoterpenes and 1 phenylpropene) while transgenic (MSLimo) extracts contained seven monoterpenes. After 3 days of fungal infection, the resistance indices further increased to 4.38, 3.89 and 2.04 for wild type, MSIso and MSLimo, respectively. The salt and fungal elicitators proved beneficial towards modifying both the terpenoids profile and improvement in the composition of essential oil. These results have important applications for the large-scale production of essential oils and forest biotechnology with respect to spearmint.

Performance Responses, Nutrient Digestibility, Blood Characteristics, and Measures of Gastrointestinal Health in Weanling Pigs Fed Protease Enzyme

  • Although exogenous protease enzymes have been used in poultry diets quite extensively, this has not been the case for pig diets. In general, due to their better gut fermentative capacity and longer transit time, pigs have greater capacity to digest dietary proteins than poultry. However, in early-weaned piglets, the stress brought about by weaning adversely affects the digestion of dietary proteins. Therefore, a study was conducted to determine the effects of a commercial protease enzyme in weanling pigs. Indices of growth, nutrient digestibility, blood profiles, fecal microflora, fecal gas emission and fecal scores were measured during the study. A total of 50 weanling pigs ($6.42{\pm}0.12kg$) at 28 d of age were randomly assigned to receive 1 of 2 dietary treatments: i) control diet (corn-soy based) with no supplemental protease (CON), and ii) control diet+200 g/ton protease (PROT) for 42 d. A completely randomized design consisting of 2 treatments, 5 replicates, and 5 pigs in each replicate was used. Growth performance in terms of body weight ($27.04{\pm}0.38kg$ vs $25.75{\pm}0.39kg$; p<0.05) and average daily gain ($491{\pm}7.40g$ vs $460{\pm}7.46g$; p<0.05) in PROT fed pigs were increased significantly, but gain per feed ($0.700{\pm}0.01$ vs $0.678{\pm}0.01$; p>0.05) was similar between treatments at d 42. Relative to CON pigs, PROT fed pigs had increased (p<0.05) apparent total tract digestibility ($84.66%{\pm}0.65%$ vs $81.21%{\pm}1.13%$ dry matter and $84.02%{\pm}0.52%$ vs $80.47%{\pm}1.22%$ nitrogen) and decreased (p<0.05) $NH_3$ emission ($2.0{\pm}0.16ppm$ vs $1.2{\pm}0.12ppm$) in the feces at d 42. Except for a decreased (p<0.05) in blood creatinine level, no differences were observed in red blood cell, white blood cell, lymphocyte, urea nitrogen, and IgG concentrations between treatments. Fecal score and fecal microflora (Lactobacillus and E. coli) were also similar between CON and PROT groups. Overall, the supplementation of protease enzyme in weanling pigs resulted in improved growth rate and nutrient digestibility. Exogenous protease enzyme reduced fecal $NH_3$ emission, thus, potentially serving as a tool in lowering noxious gas contribution of livestock production in the environment.

($KNO_3$를 첨가한 양액에서 상추의 생육 및 마이크로솜 ATPase 활성 변화 (Growth and Microsomal ATPase Activity of Lettuce(Lactuca sativa. L.) Cultured in the $KNO_3-Added$ Nutrient Solution)

  • 본 시험에서 사용된 상추는 대조구인 양액과 양액에 30 mM $KNO_3$, 혹은 양액에 50 mM $KNO_3$를 첨가한 염류농도가 다른 3가지의 양액에서 재배하였으며 이들 양액의 EC는 각각 1.0, 4.5, 6.5 ds/m이었다. 상추의 생육은 처리간에 차이를 보였으며 $KNO_3$를 첨가하여 염류농도를 높여준 용액에서 재배한 상추는 잎끝이 마르는 생리적 장해를 보이면서 발아율의 감소는 물론 초장, 경태, 엽장 및 엽폭 등 성장이 대조구와 비교하여 현저히 부진하였다. 이들 양액에서 자란 상추의 뿌리로부터 마이크로솜을 분리하여 ATPase의 특성을 조사하였다. 마이크로솜 ATPase의 활성은 대조구에 비하여 양액에 30 mM $KNO_3$와 50 mM $KNO_3$를 첨가한 용액에서 자란 상추에서 더 높았다. 상추뿌리로부터 분리한 마이크로솜 ATPase의 총활성은 재배양액 조건에 관계없이 pH 7.0에서 최대로 나타났다. ATPase의 활성은 반응용액의 $K^+$ 농도를 증가시키면 증가하였고 반응용액에 $Na^+$ 농도를 증가시키면 감소하였다. $K^+$에 의한 활성증가 효과는 양액에서 재배한 대조구보다 $KNO_3$를 첨가하여 EC를 높여준 양액에서 재배한 상추뿌리의 마이크로솜 ATPase에서 더 크게 나타났다. 이러한 결과는 생육환경내 $KNO_3$ 농도의 증가에 따라 뿌리의 생리활성을 조절하는 ATPase의 활성이 증가함을 보여준다.

  • Garlic occupies a special position among the many foods of vegetable origin because it is the sole food for Koreans during the their lives. And vitamin A has been ingested by forms of food or additives. Cadmium has been described as one of the most dangerous trace elements in the food and environment of man and livestocks. Since the de novo synthesis of stress proteins can be detected early after exposure to some agents, analysis of cadmium-induced changes in gene expression , ie. alterations in patterns of protein synthesis, may be useful to develop as biomarkers of exposure and damage for food hygiene. He acute and chronic combine effects of cadmium (Cd, CdCl2 20mg/kg), garlic oil(Dds: diallyl disulfide 50mg/kg, 3 times a week) and vitamin A(Ra: retinol acetate 50,000 IU/kg, 3 times a week) on Wistar male rats were evaluated concerning cadmium contents, tissues enzyme activity, HSP expression histopathological and electron microscopical examinations. The results of the study are as follows ; 1. Less cadmium was absorbed through the digestive tracts, but the ratio of contents in tissue were not changed by the simultaneous adminstration of diallyl disufide or retinol acetate. 2. ALT(alanine aminotransferase) , AST(aspartate aminotransferase), glucose, BUN (blood urea nitrogen), creatinine, the key indices of the clinical changes in hepatic and renal function were significantly hanged by the cadmium treatment after 1 week in liver, after 4 weeks in kidney. 3. Histopathological changes in cadmium treated rats were appeared at 8 weeks age treatment in kidneys. Homogenous eosinophilic material was accumulated in cortical and collecting tubular lumens at 16 weeks. Degenerated or necrotized tubular cells were observed in cortex and medulla. Degenerated seminiferous tubules and homogeneous eosinophilic material was seen in interstitial tissue of rat treated with cadmium for 16 weeks. Calcium deposits were seen in degenerated seminiferous tubules and the tubules showed severe calcification of rat treated with cadmium for 16 weeks. Electron microscope changes in kidney were observed in rats treated with CdCl2 20 mg/kg. Proximal convoluted tubule cells showed selling of cytoplasm and narrow lumen. Capillary endothelial cells showed cytoplasmic vacuoles and swelling. Degenerated epithelial cells were accumulated in tubular lumen of kidney. 4. Enhanced synthesis of 70 KDa relateve molecular mass proteins were detected in 2 hours after cadmium, exposure, with maximum activity occurring at 8~48 hours. Induction of HSP 70 was evident at proximal tubules and glomeruli in kidney. Testicular cells produced enough HSP to be detected normally. From the above results, it could be concluded that HSP70 induction by the cadmium treatment was a rapid reaction to indicated the exposure of xenobiotics, and retinol acetate reduced the cadmium induced nephrotoxicity.

Ganoderma Lucidum Pharmacopuncture for Teating Ethanol-induced Chronic Gastric Ulcers in Rats

  • Objectives: The stomach is a sensitive digestive organ that is susceptible to exogenous pathogens from the diet. In response to such pathogens, the stomach induces oxidative stress, which might be related to the development of both gastric organic disorders such as gastritis, gastric ulcers, and gastric cancer, and functional disorders such as functional dyspepsia. This study was accomplished to investigate the effect of Ganoderma lucidum pharmacopuncture (GLP) on chronic gastric ulcers in rats. Methods: The rats were divided into 4 groups of 8 animals each: the normal, the control, the normal saline (NP) and the GLP groups. In this study, the modified ethanol gastritis model was used. The rats were administrated 56% ethanol orally every other day. The dose of ethanol was 8 g/kg body weight. The normal group received the same amount of normal saline instead of ethanol. The NP and the GLP groups were treated with injection of saline and GLP respectively. The control group received no treatment. Two local acupoints CV12 (中脘) and ST36 (足三里) were used. All laboratory rats underwent treatment for 15 days. On last day, the rats were sacrificed and their stomachs were immediately excised. Results: Ulcers of the gastric mucosa appeared as elongated bands of hemorrhagic lesions parallel to the long axis of the stomach. In the NP and GLP groups, the injuries to the gastric mucosal injuries were not as severe as they were in the control group. Wound healings of the chronic gastric ulcers was promoted by using GLP and significant alterations of the indices in the gastric mucosa were observed. Such protection was demonstrated by gross appearance, histology and immunehistochemistry staining for Bcl-2-associated X (BAX), B-cell lymphoma 2 (Bcl-2) and Transforming growth factor-beta 1 (TGF-${\beta}1$). Conclusion: These results suggest that GLP at CV12 and ST36 can provide significant protection to the gastric mucosa against an ethanol induced chronic gastric ulcer.

Effects of photoperiod and light intensity on milk production and milk composition of dairy cows in automatic milking system

  • The purpose of this study was to determine the effects of photoperiod and light intensity on milk production, milk composition, hormones levels and blood metabolites indices of Korean Holstein dairy cows in automatic milking system (AMS). A total of 24 Holstein dairy cows were selected and used to four subsequent treatments for the experimental periods of 60 days. The light programs consisted of (1) Control: the natural photoperiod with 14.2 h of the light period and 9.4 h of the dark period (below 10 Lux); (2) T1: 16 h of the long day photoperiod (LDPP) with 50 Lux of light; (3) T2: 16 h of LDPP with 100 Lux of light; and (4) T3: 16 h of LDPP with 200 Lux of light, respectively. Importantly, there was a significant difference in the thurl activity of dairy cows between the different light intensity programs (p < 0.05). Milk yield was higher in T1 and T2 (40.80 ± 1.71 and 39.90 ± 2.02 kg/d, respectively) than those of Control and T3 (32.18 ± 1.51 and 35.76 ± 2.80 kg/d, respectively) (p < 0.05), but DMI was lower in T1, T2, and T3 compared to Control (p < 0.05). Also, milk fat percentage, the contents of milk fat and total solids were higher in T2 than those in the others (p < 0.05). The average daily melatonin level in milk was high to T3 (28.20 ± 0.43 pg/mL), T2 (24.62 ± 0.32 pg/mL), T1 (19.78 ± 0.35 pg/mL), and Control (19.36 ± 0.45 pg/mL) in order (p < 0.05). Also, the cortisol levels in milk and blood were lower in treatment groups than in Control (p < 0.05). The results of this study showed that it will be effective to improve the milk yield and milk composition, and to reduce the stress of dairy cows when the light conditions regulate to extend the photoperiod to 16 h at a light emitting diode (LED) intensity of 100 Lux under the AMS in dairy farm.

위성기반 증발산량 및 토양수분량 산정 국내 연구동향 (Research Status of Satellite-based Evapotranspiration and Soil Moisture Estimations in South Korea)

  • 최근 수문 및 수자원 분야에서 위성영상의 활용성이 높아짐에 따라 관련 전용 위성 개발연구와 연계하여 위성을 활용한 증발산량과 토양수분량 산정 연구의 필요성이 강조되고 있다. 본 연구에서는 이러한 위성을 기반으로 증발산량 및 토양수분량의 국내 연구현황과 그 산정 방법론을 조사하여 현재까지의 연구동향을 파악하고자 하였다. 국내 연구현황을 세부 방법론 별로 살펴본 결과 일반적으로 증발산량의 경우는 Surface Energy Balance Algorithm for Land (SEBAL), Mapping Evapotranspiration with Internalized Calibration (METRIC)과 같은 에너지수지 기반 모형과 Penman-Monteith (PM) 및 Priestley-Taylor (PT) 산출식을 기반으로 산정되었으며, 토양수분량의 경우 능동형(AMSR-E, AMSR2, MIRAS, SMAP) 및 수동형(ASCAT, SAR)와 같은 마이크로파 센서를 통한 산정이 주를 이루었다. 통계적 측면에서는 증발산량 및 토양수분량 공통적으로 회귀식 및 인공지능을 이용한 산출사례를 찾을 수 있었다. 또한 위성기반 자료들을 이용한 Evaporative Stress Index (ESI), Temperature-Vegetation Dryness Index (TVDI), Soil Moisture Deficit Index (SMDI) 등의 다양한 지표를 산정하여 가뭄 특성파악에 적용한 연구 사례도 다수 있었으며, 지표모형(Land Surface Model, LSM)을 기반으로 하여 위성 다중센서에서 얻을 수 있는 주요 자료들을 활용해 증발산량과 토양수분량의 수문순환인자를 산출하기도 하였다. 본 논문에서는 이렇게 기존 연구사례 조사 및 내용파악 과정을 통해 위성을 활용한 주요 세부 방법론을 비교·검토 제시함으로써 관련 연구분야 기준 참고자료로의 활용 및 향후 위성기반 관련 수문순환 자료 산출 고도화 연구의 초석을 다지고자 한다.

Inferring Pedestrians' Emotional States through Physiological Responses to Measure Subjective Walkability Indices

  • Walkability is an indicator of how much pedestrians are willing to walk and how well a walking environment is created. As walking can promote pedestrians' mental and physical health, there has been increasing focus on improving walkability in different ways. Thus, plenty of research has been undertaken to measure walkability. When measuring walkability, there are many objective and subjective variables. Subjective variables include a feeling of safety, pleasure, or comfort, which can significantly affect perceived walkability. However, these subjective factors are difficult to measure by making the walkability index more reliant on objective and physical factors. Because many subjective variables are associated with human emotional states, understanding pedestrians' emotional states provides an opportunity to measure the subjective walkability variables more quantitatively. Pedestrians' emotions can be examined through surveys, but there are social and economic difficulties involved when conducting surveys. Recently, an increasing number of studies have employed physiological data to measure pedestrians' stress responses when navigating unpleasant environmental barriers on their walking paths. However, studies investigating the emotional states of pedestrians in the walking environment, including assessing their positive emotions felt, such as pleasure, have rarely been conducted. Using wearable devices, this study examined the various emotional states of pedestrians affected by the walking environment. Specifically, this study aimed to demonstrate the feasibility of monitoring biometric data, such as electrodermal activity (EDA) and heart rate variability (HRV), using wearable devices as an indicator of pedestrians' emotional states-both pleasant-unpleasant and aroused-relaxed states. To this end, various walking environments with different characteristics were set up to collect and analyze the pedestrians' biometric data. Subsequently, the subjects wearing the wearable devices were allowed to walk on the experimental paths as usual. After the experiment, the valence (i.e., pleasant or unpleasant) and arousal (i.e., activated or relaxed) scale of the pedestrians was identified through a bipolar dimension survey. The survey results were compared with many potentially relevant EDA and HRV signal features. The research results revealed the potential for physiological responses to indicate the pedestrians' emotional states, but further investigation is warranted. The research results were expected to provide a method to measure the subjective factors of walkability by measuring emotions and monitoring pedestrians' positive or negative feelings when walking to improve the walking environment. However, due to the lack of samples and other internal and external factors influencing emotions (which need to be studied further), it cannot be comprehensively concluded that the pedestrians' emotional states were affected by the walking environment.

복합재 압력용기의 돔 형상 설계 및 성능 평가 (Dome Shape Design and Performance Evaluation of Composite Pressure Vessel)

  • 시스템 요구 조건에 따른 다양한 돔 형상 설계를 위하여 외부 하중이 고려된 돔 형상 설계 식이 제시되었고, 동일 원통부와 보스 오프닝을 가지면서 다른 돔 형상을 갖는 압력 용기의 성능(파열압력*내부용적/복합재무게) 변화를 유한요소해석을 통하여 평가하였다. 해석 결과는 돔 형상이 낮아질수록 파열 압력이 저하되어 압력 용기 성능이 저하됨을 나타내었다. 특히 원통부 반경과 보스 오프닝간의 비를 나타내는 보스 오프닝 크기 비율(${\rho}\;o$)이 큰 경우는 돔 형상이 낮아짐에 따라 복합재 무게 증가율이 크게 나타남으로서 압력 용기 성능 저하의 원인이 되었고, 파열 압력 상승을 위하여 돔 부위에 추가적인 보강설계가 요구되었다. 예를 들어 ${\rho}\;o=0.35$ 이하의 돔인 경우, 돔 형상 변화가 크지 않음으로서 돔 형상 변화에 따른 압력 용기 성능 변화도 크지 않은 것으로 나타난 반면, ${\rho}\;o=0.54$ 이상의 압력 용기인 경우 낮은 돔으로 변화됨에 따라 파열 압력 감소와 복합재 무게 증가로 압력 용기 성능 저하가 비교적 크게 나타나서 압력 용기 내부 용적 증가나 스커트와 돔간의 공간 확보가 우선인 설계 조건을 제외하고는 낮은 돔 형상 설계가 바람직하지 않은 것으로 나타났다.
