Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) from Streptococcus iniae shows potential as a subunit vaccine against various streptococcal species

  • The potential of Streptococcus iniae glyceraldehyde-3-phosphate dehydrogenase (GAPDH) as an antigen for a subunit vaccine was investigated using a zebrafish model. The recombinant S. iniae GAPDH was purified using His-tag column chromatography, and antisera against the recombinant GAPDH (rGAPDH) were produced by intraperitoneal immunization of rats. By immunization with S. iniae rGAPDH, the survival rates of zebrafish against an S. iniae challenge increased, suggesting that GAPDH would be an antigen capable of inducing protective immune responses in fish. Furthermore, we demonstrated using Western blotting, that the antisera against rGAPDH of S. iniae had cross-reactivity with GAPDH from Streptococcus parauberis and Lactococcus garviae, which are also culprits of streptococcosis in cultured fish in Korea. These results suggest that S. iniae GAPDH may be used as an antigen for the development of a subunit vaccine against streptococcosis caused by diverse cocci in cultured fish.

  • In order to develop natural anticariogenic agents, we investigated inhibitory effects of Areca catechu L. extracts on the acid production of Streptococcus mutans JC-2 and its cytotoxicity on NIH 3T3 fibroblasts were also examined. The results are as follows : 1. Major organic acid produced by Streptococcus mutans JC-2 were lactic acid and acetic acid, and their productions were decreased by additions of Areca catechu L. extracts. 2. Areca catechu L. extracts were showed noncytotoxicity on NIH 3T3 fibroblasts.

Development of Streptococcus faecalis Strains Resistant to Rifampicin (리팜피신에 내성인 Streptococcus faecalis 균주의 개발)

  • The preparation of Streptococcus faecalis RSI is used as a medicinal preparation for human intestinal disorders. But the microbe in this preparation is very sensitive to rifampicin. If this preparation is taken with rifampicin, its therapeutic effect can not be expected. To develope rifampicin resistant mutants, the rifampicin sensitive strain S. faecalis RSI was treated with Nmethyl-N'-nitro-N-nitrosoguanidine(MNNG). Twelve strains of the MNNG-induced mutants showed distinct resistance to rifampicin and five mutants were selected for further studies. They also exhibited identical characteristics with the parent S. faecalis RSI when they were tested for lactic acid formation and growth inhibition of E. coli. From in vitro test, it was identified that rifampicin is not inactivated by certain factors of the rifampicin resistant mutants. Conclusively, the rifampicin resistant mutants are efficient strains that have insensitivity against rifampicin and original biochemical characteristics of the parent strain.

  • This report was concerned with the isolation and identification of bacterial flora in the human dental plaque and the dextransucrase activity of isolated species. The results were obtained as follows: 1. The bacterial flora, isolated from the human dental plaque, was identified as 3 species of resembling streptococci, Streptococcus salivarius strain SD-1, Streptococcus bacilli, Lactobacillus brevis strain SD-3, Lactobacillus acidophilus strain SD-2 and SD-7, resembling Staphylococcus sp, and one species of resembling Leuconostoc mesenteroides strain SD-6. 2. The dextransucrase activites of resembling Streptococcus mitis strain SD-9 and Streptococcus salivarius strain SD-1 were exhibited the highest among the isolated species of human dental plaque.

  • Streptococcal nuclease was completely purified by stepwise CM-Sepharose column chromatography from crude extracts isolated from Streptococcus sp. The active enzyme fraction was eluted with the buffer containing 0.2 M NaCl. The purified enzyme showed a homogeneity on SDS PAGE and had a molecular weight of 35,000 daltons. The optimum pH and temperature for the enzyme were 9.0 and $50^{\circ}C$, respectively.

  • Streptococcus pneumoniae (pneumococcus) is the most frequent causative agent of acute bacterial pneumonia. Outstanding characteristic of pneumococcus is an ample polysaccharide capsule that is highly anti-genic agent and is the major factor for classification of pneumococcus into more than 94 serotypes. In this study, production of capsular polysaccharide from Streptococcus pneumoniae type 23 was optimized by supplementation of metal ions or by modulation of culture conditions. When brain heart infusion (BHI) broth was supplemented with 1 mM concentration of $CaCl_2$, $CuSO_4$ and $MgSO_4$, growth of pneumococcus as well as polysaccharide yield were stimulated. Also slight aeration gave rise to better polysaccharide yield.

  • A 5-year-old boy was admitted to the hospital with a high fever and abnormal gait. Magnetic resonance imaging showed extensive swelling of the quadriceps and adductor muscles around the right hip and anterior thigh. A duplex scan demonstrated a thrombus from the external iliac vein to below the popliteal vein. The blood culture revealed Streptococcus pyogenes. The patient was immediately treated with antibiotics and anticoagulants. Follow-up sonography demonstrated complete disappearance of the venous thrombosis. We report a case of streptococcal myositis complicated by deep vein thrombosis.

  • Conditions for efficient formation and regeneration of protoplasts of Streptococcus lactis ATCC 11454 were investigated. Addition of 20mM DL-threonine into growth medium, growth phase and lysozyme concentration had significant effects on protoplast formation. Approximately, 20% regeneration efficiency was obtained by optimizing the medium composition and modifying the plating procedure.

Gliotoxin is Antibacterial to Drug-resistant Piscine Pathogens

  • By activity-guided fractionation, gliotoxin was isolated as an antibacterial metabolite of the fungus Penicillium decumbens which was derived from the jellyfish Nemopilema nomurai. Gliotoxin was further evaluated for antibacterial activity against several piscine and human MDR (multidrug resistance) pathogens. Gliotoxin showed significant antibacterial activity against Gram-positive piscine pathogens such as Streptococcus iniae FP5228, Streptococcus iniae FP3187, Streptococcus parauberis FP3287, Streptococcus parauberis SPOF3K, S. parauberis KSP28, and Lactococcus garvieae FP5245. Gliotoxin showed strong activity especially against S. parauberis SPOF3K and S. iniae FP5228, which are resistant to oxytetracycline. It is noteworthy that gliotoxin effectively suppressed streptococci which are the major pathogens for piscine infection and mortality in aquaculture industry. Gliotoxin also showed strong antibacterial activity against multidrug- resistant human pathogens (MDR) including Enterococcus faecium 5270 and MRSA (methicillin-resistant Staphylococcus aureus) 3089.
