Effect of Chlorine Dioxide and Commercial Chlorine Sanitizer on Inhibiting Foodborne Pathogens and on Preventing the Formation of Chemically Injured Cells on Radish Sprouts

  • This study assessed the efficacy of aqueous chlorine dioxide ($ClO_2$) and commercial chlorine sanitizer in terms of its ability to eliminate Listeria monocytogenes, Salmonella Typhimurium, and Escherichia coli O157:H7 on radish sprouts (Raphanus sativus L.). Radish sprouts were inoculated with a cocktail containing one each of three strains of three different foodborne pathogens, then treated with distilled water (control) or chemical sanitizers (100 ppm commercial chlorine, and 50, 100, 200 ppm $C1O_2$) for 1, 5, and 10 min at room temperature ($22{\pm}2^{\circ}C$). Populations of S. Typhimurium, E. coli O157:H7 and L. monocytogenes were counted at 4.64, 6.05, and 4.29 log CFU/g, respectively, after inoculation. Treatment with water did not significantly reduce the levels of any of the three foodborne pathogens. The levels of all three pathogens were reduced by treatment with chemical sanitizers; however, the observed levels of reduction of E. coli O157:H7 and L. monocytogenes were not significant as compared with the controls. The levels of the three pathogens were reduced most profoundly when treated for 10 min with 200 ppm of $C1O_2$, and the reduction levels of S. Typhimurium, E. coli O157:H7, and L. monocytogenes were 1.17, 1.63, and 0.96 log CFU/g, respectively. When chemically injured cells were investigated using SPRAB for E. coli O157 :H7 and by selective overlay methods for S. Typhimurium and L. monocytogenes, respectively, it was noted that commercial chlorine sanitizer generated more numbers of injured pathogens than did $C1O_2$. These data indicate that $C1O_2$ treatment may prove useful in reducing the numbers of pathogenic bacteria in radish sprouts.

양상추에 오염된 병원성 미생물에 대한 Chlorine Dioxide 및 상업적 Chlorine 살균소독제의 저해효과 평가 (Inhibitory Effects of Chlorine Dioxide and a Commercial Chlorine Sanitizer Against Foodborne Pathogens on Lettuce)

  • This study compared the effects of chlorine dioxide and a commercial chlorine sanitizer for inhibiting foodborne pathogens, including Salmonella enterica serovar Typhimurium, Listeria monocytogenes, and Escherichia coli O157 : H7, on lettuce leaves. The lettuce samples were inoculated with each cocktail of the three strains, and were then treated with chemical sanitizers [distilled water, 100 ppm commercial chlorine and 50 ppm, 100 ppm, 200 ppm chlorine dioxide ($ClO_2$)] for 1 min, 5 min, and 10 min at room temperature($22{\pm}2^{\circ}C$). Following inoculation of the leaves, initial populations of E. coli O157:H7, L. monocytogenes, and S. Typhimurium were approximately 5.54, 4.47, and 5.12 log CFU/g, respectively these levels were not significantly reduced by the treatment with water,whereas the 100 ppm commercial chlorine sanitizer treatment and $ClO_2$ (at all tested concentrations) were effective at reducing levels of all three pathogens. The treatment of 200 ppm $ClO_2$ for 10 min was most effective at inhibiting the three pathogens, and reduction levels of E. coli O157 : H7, L. monocytogenes, and S. Typhimurium were 2.28, 1.95, 1.76 log, respectively. The inhibitory effect of $ClO_2$ increased with increasing treatment concentration of $ClO_2$, but there was no significant difference by the treatment times. When chemically injured cells of E. coli O157 : H7 and L. monocytogenes and S. Typhimurium were examined by SPRAB and selective overlay methods, respectively, it was observed that the commercial chlorine sanitizer generated greater numbers of injured L. monocytogenes than the $ClO_2$ treatment. From the overall results, $ClO_2$ was more effective at inhibiting pathogenic bacteria compared to the commercial chlorine sanitizer therefore, it has potential to be utilized as an alternative sanitizer to increase the microbial safety of fresh produce.

Application of Thermotolerant Yeast at High Temperature in Jar-fermentor Scale.

  • We investigated the possibility of industrial application and economit process of high temperature fermentation by thermotolerant alcohol producing yeasts as previously reported. From the 20% glucose media, the RA-74-2 produced 11.8% (v/v) ethanol at $32^{\circ}C$ (0.5% inoculum) and 10.6% (v/v) ethanol at $40^{\circ}C$ (3% inoculum), respectively. Also, 11.3% (v/v) ethanol was produced for 96 hours in the temperature-gradient fermentation. These results suggest that the RA-74-2 could isuccessfully be applied to save the cooling water and energy in industrial scale without re-investment or modification of established fermentation systems. When potato starch was used as the substrate for the RA-74-2, high temperature fermentation above $40^{\circ}C$ was more appropriate for industrial utilization because organic nitrogen was not necessary to economical fermentation. As the naked barley media just prior to industrial inoculation, taken from the Poongkuk alcohol industry Co., were used, 9.6% (v/v) ethanol was produced at $40^{\circ}C$ for 48 hours in jar-fermentor scale (actually, 9.5-9.8% (v/v) ethanol was produced at 30~$32^{\circ}C$ for 100 hours in industrial scale). The ethanol productivity was increased by the high glucoamylase activity as well as the high metabolic ratio at $40^{\circ}C$ Therefore, if the thermotolerant yeast RA-74-2 would be used in industrial scale, we could obtain a high productivity and saving of the cooling water and energy. Meanwhile, the RA-912 produced 6%(v/v) ethanol in 10% glucose media at $45^{\circ}C$ and showed the less ethanol-tolerance compared with industrial strains. As the produced alcohol was recovered by the vacuum evaporator at $45^{\circ}C$ in 15% glucose media, the final fermentation ratio was enhanced (76% of theoretical yields). This suggest that a hyperproductive process could be achieved by a continuous input of the substrate and continuous recovery of the product under vacuum in high cell-density culture.

A lower cost method of preparing corn stover for Irpex lacteus treatment by ensiling with lactic acid bacteria

  • Objective: This study investigated a method of preparing corn stover for Irpex lacteus (I. lacteus) treatment to improve its in vitro rumen degradability under non-sterile conditions. Methods: Corn stover was inoculated with Lactobacillus plantarum (L. plantarum), Lactobacillus buchneri (L. buchneri), and an equal mixture of these strains, and ensiled for 0, 3, 7, 14, and 28 days. After each period, a portion of the silage was sampled to assess the silage quality, and another portion of the silage was further treated with I. lacteus at 28℃ for 28 d. All the samples were analyzed for fermentation quality, chemical composition, and in vitro gas production (IVGP) as a measure of rumen fermentation capacity. Results: Lactic acid bacteria (LAB) was found to improve the silage quality of the corn stover, and the corn stover silage inoculated with L. plantarum produced more lactic acid and higher IVGP than other silage groups. The I. lacteus colonies flourished in the early stage of corn stover silage, especially on the 3-d corn stover silage inoculated with both L. plantarum and L. buchneri. This led to an 18% decrease in the acid detergent lignin content, and a 49.6% increase in IVGP compared with the raw stover. Conclusion: The combination of ensiling with the mixed LAB inoculation and I. lacteus treatment provided a cost-effective method for the improvement of the IVGP of corn stover from 164.8 mL/g organic matter (OM) to 246.6 mL/g OM.

  • 광교, 동북태, 강림, 육우3호, 은대두등과 같은 대리장려품종이 바이러스에 의하여 심하게 이병되었다. 이 병은 주로 모자익병의 발생이 많은 강원, 경기지방에서 발병이 심하였으나 모자익병이 심하지 않은 전남 등 남부지방에서도 발병되고 있다. 병징으로 보아 tobacco ringspot virus에 의한 대두의 피해와 유사한 것으로 보였으나 지표식물검정과 혈청검정에 의하여 조사한 결과 모두 부정적이었으며 대두품종에 따른 이병정도의 상이, 품종과 접종원에 의한 병징의 변이가 많았다. 이병주에서 분리되는 병징형은 Mottling과 necrosis였으며 지금까지의 연구결과 이 대두병해는 모자익바이러스(SMV)의 계통 내지는 tobacco ringspot virus 이외의 두류바이러스의 복각감염에 의한 것으로 생각할 수 있으나 SMV의 계통에 의한 피해일 가능성이 더욱 유력시되고 있다.

식물 생장촉진 미생물의 외부 유전자 도입과 그 접종효과 (Transfer of Bacillus thuringiensis toxin gene into Bacillus subtilis and its inoculation effects)

  • 토양전염 병원성 사상균인 Fusarium oxyporum, Rhizoctonica solani에 대하여 길항력을 갖는 균주 Bacillus subtilis를 근권 토양에서 분리하여 동정한 후 이들 균주의 사상균에 대한 길항력, 발아율 및 작물의 생육에 미치는 영향력을 검토하였다. 또한 이 균주의 chromosome에 Bacillus thuringiensis(BT) 독소유전자를 삽입하여 균주의 형질변환을 유도하였다. BT 독소 유전자는 southern blotting에 의하여 확인되었으나 이의 최종 생성물인 독소 단백질은 SDS-PAGE에 확인되지 않았다. 이들 형질변환된 균주의 생리 및 생화학적 특성을 조사한 결과 모균주와 차이는 없었으며, BT 독소유전자가 삽입된 균주는 선충의 유충에 대한 생물학적 검정에서는 효과가 인정되지 않았으나 누에에 있어서는 1X 균체 희석액에서 10내지 20% 정도의 치사율이 관찰되었다. 모균주와 BT 독소유전자에 의하여 형질변환된 균주 모두 발아율 및 작물의 생육을 향상시켰다.

인삼 유청음료의 제조 및 관능적 특성 (Ginseng-Whey Beverage Production and Sensory Properties)

  • 렌넷유청, 인삼, 감미료, 꿀, 매실 등에 종류가 다른 유산균주들을 접종하여 인삼 유청음료를 제조한 후, 일부는 발효시키고 일부는 비발효 상태로 4$^{\circ}C$ 및 30$\pm$1$^{\circ}C$에 저장하여 1, 3, 5주에 관능검사를 실시하였다. 관능검사 결과, 감미료와 꿀을 첨가하여 냉장보관 한 유산균 비발효 인삼 유청음료(A)는 단맛이 강하고 신맛, 쓴맛, 떫은맛, 뒷맛이 적어서 전체적인 평가에서 8.64~8.85으로 가장 높은평가를 받았고 Lac. caseisub-sp. casei와 Str. salivarius sbu-sp. thermophilus를 접종하여 냉장보관한 인삼 유청음료 (C)와 감미료, 꿀 및 매실을 첨가하여 실온보관한 인삼 유청음료(D)가 대체로 좋은 평가를 받았으나 실온보관한 유산균 발효 음료(E, F)는 2.92~3.58로 가장 낮은 평가를 받았으므로 이의 개선이 요구된다.

Distribution and Isolation of Soil borne Wheat Mosaic Virus in Korea

  • This study was conducted to investigate the occurrence of Soil borne wheat mosaic virus(SbWMV) in barley fields in Korea and to examine the host pathogenicity of SbWMV. By using the ELISA test, SbWMV was detected in the six regions : Suwon, Milyang, Jinju, Youngkwang, Iksan, and Chonju. SbWMV was isolated from the two strains, Albori strain from Jinju and Eunpamil strain from Milyang. SbWMV was collected from leaves showing mosaic, yellowing and necrosis stripes. SbWMV was inoculated mechanically on 1∼1.5 leaf stages with leaf-rubbing to identify the host pathogenicity of 36 Korean barley cultivars, a wheat cultivar, two rye cultivars, three Japanese barley cultivars and Chenopodium amaranticola. Viral sympoms of inoculated leaves appeared on moulted loaves about 4 to 6 weeks of inoculation. Baegdong and Tapgolbori, infected from Albori strain and Eunpamil strain infected from Samdobori showed much higher susceptibility than C. amaranticola and C. quinoa which showed ring spots and chlorotic spots respectively. Virus particles were observed by the electron microscope. They were rod-shapes, which are bipartite, of 142 nm or 281 nm in length with 20 nm diameter on infected leaves. Specific detection and identification of SbWMV was set up using the RT-PCR. PCR fragments of SbWMV(0.5kb) were obtained by using the designed primers for SbWMV RNA 2.

와송 에틸아세테이트 분획물의 항균효능 평가에 관한 연구 (A Study on the Evaluation of Antimicrobial Effect of Orostachys Japonicus A. Berger Ethyl Acetate Fraction)

  • 본 연구에서 와송 에틸아세테이트 분획물을 항균활성 및 화장품 소재로 활용하기 위해 연구를 진행하였다. 와송 에틸아세테이트 분획물의 항균활성을 측정한 결과 Paper disc method에서 S. aureus, S. epidermidis, P. aeruginosa 균의 생육을 저해하였으며, 특히 S. aureus에 대한 생육저해환은 0.5 g/mL농도에서 18.35 ± 1.5 mm로 대조군인 methyl paraben(16.83 ± 1.0 mm)보다 우수한 항균활성을 나타내었다. MIC 측정을 통한 최소저해농도를 평가에서는 Candida. A를 제외한 나머지 S. aureus, S. epidermidis, E. coli P. aeruginosa 균주에서 17.5 mg/mL의 최소저해농도를 보였으며 화장품 에멀션 제형에 적용하여 Challenge test를 통해 방부 효능을 평가한 결과, S. aureus에 대하여 와송 에틸아세테이트 분획물을 0.3% 첨가한 에멀션에서 7일 후 100.0%균의 생장을 사멸시켜 화장품에 적용 시 우수한 방부효능을 나타내었다.

Isolation and identification of mammalian orthoreovirus type 3 from a Korean roe deer (Capreolus pygargus)

  • Mammalian reovirus (MRV) causes respiratory and intestinal disease in mammals. Although MRV isolates have been reported to circulate in several animals, there are no reports on Korean MRV isolates from wildlife. We investigated the biological and molecular characteristics of Korean MRV isolates based on the nucleotide sequence of the segment 1 gene. In total, 144 swabs from wild animals were prepared for virus isolation. Based on virus isolation with specific cytopathic effects, indirect fluorescence assays, electron microscopy, and reverse transcription-polymerase chain reaction, only one isolate was confirmed to be MRV from a Korean roe deer (Capreolus pygargus). The isolate exhibited a hemagglutination activity level of 16 units with pig erythrocytes and had a maximum viral titer of 105.7 50% tissue culture infectious dose (TCID50)/mL in Vero cells at 5 days after inoculation. The nucleotide and amino-acid sequences of the partial segment S1 of the MReo2045 isolate were determined and compared with those of other MRV strains. The MReo2045 isolate had nucleotide sequences similar to MRV-3 and was most similar (96.1%) to the T3/Bat/Germany/342/08 strain, which was isolated in Germany in 2008. The MReo2045 isolate will be useful as an antigen for sero-epidemiological studies and developing diagnostic tools.
