IbMYB1, a transcription factor (TF) for R2R3-type MYB TFs, is a key regulator of anthocyanin biosynthesis during storage of sweet potatoes. Anthocyanins provide important antioxidants of nutritional value to humans, and also protect plants from oxidative stress. This study aimed to increase transgenic potatoes' (Solanum tuberosum cv. LongShu No.3) tolerance to environmental stress and enhance their nutritional value. Transgenic potato plants expressing IbMYB1 genes under the control of an oxidative stress-inducible peroxidase (SWPA2) promoter (referred to as SM plants) were successfully generated through Agrobacterium-mediated transformation. Two representative transgenic SM5 and SM12 lines were evaluated for enhanced tolerance to salinity, UV-B rays, and drought conditions. Following treatment of 100 mM NaCl, seedlings of SM5 and SM12 lines showed less root damage and more shoot growth than control lines expressing only an empty vector. Transgenic potato plants in pots treated with 400 mM NaCl showed high amounts of secondary metabolites, including phenols, anthocyanins, and flavonoids, compared with control plants. After treatment of 400 mM NaCl, transgenic potato plants also showed high DDPH radical scavenging activity and high PS II photochemical efficiency compared with the control line. Furthermore, following treatment of NaCl, UV-B, and drought stress, the expression levels of IbMYB1 and several structural genes in the flavonoid biosynthesis such as CHS, DFR, and ANS in transgenic plants were found to be correlated with plant phenotype. The results suggest that enhanced IbMYB1 expression affects secondary metabolism, which leads to improved tolerance ability in transgenic potatoes.
Hydroxypropylated starches and acetylated starches were prepared by reaction of potato starch with propylene oxide and acetic anhydride, respectively and then degree of retrogradation and swelling power were investigated in different pHs and temperature. The extent of retrogradation determined by glucoamylase method during freeze-thaw treatment and storage in low temperature $(0{\sim}5^{\circ}C)$ showed that modified potato starches were slowly retrograded as the increase of degree of substitution. The order of the retrogradation tendencies in different pHs were pH 4>pH 6>pH 8>pH 10. Retrogradation of hydroxypropylated potato starches were less than that of acetylated potato starches. Swelling power of starches were influenced more by the temperature than by the pH. The order of the swelling power tendencies in different pHs were pH 10>pH 2>pH 8>pH 4>pH 6.

Background: Potato is the main crop of Ardabil Plain (accounting for one-fifth of potato production in Iran). Its health hazard risk to farmers is rising due to the increasing rate of pesticide use. The present study analyzes potato farmers' health hazard risk in the use of chemical pesticides. Methods: The rate of pesticide use by farmers (n = 370) was first compared with the recommended dosage (on pesticide label). Then, a composite index was employed to estimate the health hazard risk of farmers during pesticide use, and the variables accounting for pesticide overuse and nonoveruse were analyzed. Safety behavior was examined in four steps, namely of pesticide purchase and storage, preparation, application, and postapplication. Results: It was found that 74.6 percent of potato farmers used pesticides in higher concentrations than the recommended dosage. The higher average rate of pesticide use versus recommendation (label instruction) was related to Chlorpyrifos and Trifluralin, and the highest average health hazard risk among farmers was related to the use of Chlorpyrifos and Metribuzin. Farmers with a higher risk of health hazard displayed much lower safety behavior than the other farmers at all steps of pesticide use. Conclusion: The most important variables discriminating the health hazard risk of farmers' overuse included health behavior identity, attitude, knowledge and awareness, and cues to action. Therefore, using social media, holding local exhibitions, and engaging local leaders and skilled farmers in the region to improve farmers' attitudes and health behavior identity toward the dangers of chemical pesticides can play a significant role in motivating farmers' display of overuse preventive behaviors.

Potatoes are the world's 4th major food crop after maize, rice, and wheat and also are a staple food for 1.3 billion people. Due to their wide adaptability to various environmental conditions, their yeild capacity, and high commercial value, potatoes have contributed to global food security. Many potato germplasms are commonly preserved as whole plants in fields or in storage to maintain their particular genetic combinations. However, field maintenance is expensive and has the risk of potential losses from diseases, pests, plant ageing and climate change. Over the past four decades, meaningful efforts have been made toward the safe long-term conservation of potatoes through cryopreservation methods such as droplet-vitrification. In this study, we tested 4 Korean potato varieties('Golden Egg', 'Golden Ball', 'Ja-Young' and 'Ha-Ryeong') with the modified potato droplet -vitrification protocol. Potato shoot tips are precultured in a sucrose-enriched medium(0.3 and 0.7M for 7 and 17hrs, respectively) and submitted to a loading step with C4 solution for osmoprotection. The treated explants were dehydrated with Plant Vitrification Solution(PVS)2 which is 80% A3 solution in ice for 30 minutes. Thawing and unloading steps were performed with 0.8M sucrose solution for 30 sec(40℃) followed by 30min(25℃, room temperature). In a potato post-culture medium(MS+0.1 mg·L-1 GA3+0.1 mg·L-1 kinetin), we obtained a survival rates of post-thawed explants ranging 16.1-82.2%. The results suggest that modified and optimized protocols are required dependinig on every cultivar, genetic and ecological types. To achieve higher survival and regeneration rates, each step within the cryoprocedure must be carefully optimized.

Sweet potato (Ipomoea batatas L. Lam) is an economically important root crop and a valuable source of nutrients, processed foods, animal feeds, and pigment materials. However, during post-harvest storage, storage roots of sweet potatoes are susceptible to decay caused by various microorganisms and diseases. Post-harvest curing is the most effective means of healing wounds and preventing spoilage by microorganisms during storage. In this study, we aimed to identify proteins involved in the molecular mechanisms related to curing and study proteomic changes during the post-curing storage period. For this purpose, changes in protein spots were analyzed through 2D-electrophoresis after treatment at 33℃ (curing) and 15℃ (control) for three days, followed by a storage period of eight weeks. As a result, we observed 31 differentially expressed protein spots between curing and control groups, among which 15 were identified. Among the identified proteins, the expression level of 'alpha-amylase (spot 1)' increased only after the curing treatment, whereas the expression levels of 'probable aldo-keto reductase 2-like (spot 3)' and 'hypothetical protein CHGG_01724 (spot 4)' increased in both the curing and control groups. However, the expression level of 'sporamin A (spot 10)' decreased in both the curing and control treatments. In the control treatment, the expression level of 'enolase (spot 14)' increased, but the expression levels of 'chain A of actinidin-E-64 complex+ (spot 19)', 'ascorbate peroxidase (spot 22)', and several 'sporamin proteins (spot 20, 21, 23, 24, 27, 29, 30, and 31)' decreased. These results are expected to help identify proteins related to the curing process in sweet potato storage roots, understand the mechanisms related to disease resistance during post-harvest storage, and derive candidate genes to develop new varieties with improved low-temperature storage capabilities in the future.
In this study, we investigated the physical properties, color values of mixtures with starch from potato in preparing a cartridge for 3D printing using golden threadfin bream (Nemipterus virgatus) surimi as raw material. The results show that the hardness and gumminess of the surimi mixture added potato starch increased but cohesiveness decreased compared to the negative control. This means that the starch makes surimi stronger texture than the negative control. After 7 days of cold storage at 4 ℃, for unheated surimi, firmness is increased but adhesiveness is decreased. And among physical properties of surimi stored after heating, hardness and gumminess were decreased compared to before storage. Lastly, after storage for 7 days, the hardness and gumminess of fried surimi decreased compared to fried surimi before storage. As a result of sensory evaluation, texture, elasticity scent of surimi stored after heating were decreased but overall favorability was similar. Compared to the after storage, result of sensory evaluation of fried surimi, result of the before storage showed no significant differences. From these results, it is suggested that potato starch, which increases overall physical strength and have little effect on preference, can be used as an additive for golden threadfin bream surimi.
The handling and processing of agricultural products in Korea is done manually. Small plastic boxes, nets, and corrugated cardboard boxes are used as containers during harvesting, sorting and other product handling operations. However, these practices are labor-intensive, time-consuming, require various kinds of packing materials, and are expensive because of high operating costs. To overcome these problems, the use of pallet bins with pre-cooling and storage features for handling and processing bulk farm products was investigated. The airflow resistances through bulk potato, onion and mandarin stocks were measured, and the pallet bins and a pressure pre-cooling device were manufactured. The opening ratio, bed depth and airflow rate through bulk potato, onion and mandarin in the pallet bin were defined with regression equations. The cooling rates of bulk potato, onion and mandarin were 0.8C/h ($21.7{\rightarrow}0C$, 14.5 h), 0.4C/h ($15.4{\rightarrow}.0C$, 32.2 h) and 0.7C/h ($13.7{\rightarrow}C$, 18.8 h), respectively, with the pressure pre-cooling system. Temperature deviances for storage of bulk potato, onion and mandarin were 0.12C, 0.12C and 0.17C, respectively.
In order to determine the moisture content level for safe storage of wild vegetables, drying characteristics and water vapor sorption characteristics of four vegetables, i.e., zucchini slice, sweet potato stem, taro stem, and platycodon, were investigated. The drying curves of these vegetables were consisted of three characteristic stages which were the initial settling down period, the constant drying rate period, and the falling drying rate period. And the falling rate period of the vegetables showed 2 or 3 parts of falling rate. All of the falling rate curves of the vegetables showed upwardly convex shape which is known as a characteristic pattern for the drying of fibrous food materials. The critical moisture contents of the vegetables were $8.29{\sim}9.75,\;10.40{\sim}15.08,\;9.51{\sim}14.52\;and\;3.29{\sim}3.56g\;H_2O/g$ dry solids for zucchini slice, sweet potato stem, taro stem, and platycodon, respectively. Activation energy values of drying rate during falling rate period were 2.30, 2.11, 4.97, and 2.80 kcal/mol for zucchini slice, sweet potato stem, taro stem, and platycodon respectively. The BET monolayer moisture contents of the vegetables were $10.05{\sim}13.59,\;9.49{\sim}12.69,\;9.50{\sim}16.48\;and\;5.01{\sim}5.44g\;H_2O/g$ dry solids for zucchini slice, sweet potato stem, taro stem, and platycodon, respectively. And these values were found to be very compatible with the values of the critical moisture content. Consequently, it was found that the moisture of these vegetables should be removed below the BET monolayer moisture content or below the critical moisture content for the long term storage.
This study was attempted to investigate the effects of blanching temperature and cooking methods on the changes in the proportions of vitamin C of fresh potato and potatoes with different storage time. Sensory evaluation of fresh potato by different cooking methods was also conducted. The contents of residual ascorbic acid(AA) and total ascorbic acid(TAA) of fresh potato were maximum at $40^{\circ}C$ followed by rapid decrease at $50-65^{\circ}C$ while leached AA and TAA showed steady increase as the blanching temperature increased. Oxidized AA and dehydroascorbic acid(DHA) hydrolyzed increased at $50-65^{\circ}C$. From these results, it was considered that AA was lost mainly by oxidation up to $65^{\circ}C$ and leaching of AA was the major mode of loss above $65^{\circ}C$. In the case of potatoes stored for 1-4 weeks, they showed similar changes in the proportions of vitamin C as that of fresh potato. However, at $40^{\circ}C$ the content of residual TAA decreased and those of leached TAA and DHA hydrolyzed increased redundant during storage. At $65^{\circ}C$, the content of DHA hydrolyzed decreased The residual TAA of fresh potato by different cooking methods decreased in the order of pressure cooking (PC) > microwave cooking (MC)>boiling. Leached TAA were 49.5% and 36.4%, during boiling and MC, respectively. While DHA hydrolyzed were 22.3% and 4.2%, respectively Leached TAA and DHA hydrolyzed during PC were not determined. From these results, it was considered that AA was lost mainly by leaching during cooking. Residual TAA of stored potatoes by different cooking methods decreased during storage. But leached TAA and DHA hydrolyzed did not show any steady increase or decrease. Overall eating quality of fresh potato by different cooking methods decreased in the order of PC>MC>boiling(p<0.05).

A potato cultivar, Haryeong, was released for table stock in 2005 and has been supplied to farmers since 2010. Black dot has been known as a disease causing tuber blemishes in the cultivar, which is primarily an issue in storage. To investigate the effect of curing periods on disease occurrence, four periods (1, 3, 6 and 9 weeks) of curing were applied on Haryeong tubers harvested from the highlands (800 m a.s.l) in September. Tubers were stored at $2-3^{\circ}C$ and 80-90% RH for 6-7 months and were visually checked for disease occurrence. Tuber infection was characterized by barely visible small black spots on tubers and dark sooty lesions in stolon end of tubers. Another symptoms were observed on the upper surface of tubers which showed irregular, small to large, and depressed areas of brown to blackish skin with necrotic lesions inside. Results showed decrease in black dot depending on the curing period that ranges 18-48, 3-35, 0-14 and 0-3% at 1, 3, 6 and 9 weeks curing, respectively. During the storage, however, percentage weight loss of tubers and percentage of tubers with sprouts increased slightly with increasing the curing period from 1-3 to 6-9 weeks.
