Enhancement of Osteogenic Differentiation by Combination Treatment with 5-azacytidine and Thyroid-Stimulating Hormone in Human Osteoblast Cells

  • Background and Objectives: The role of thyroid-stimulating hormone (TSH) signaling on osteoblastic differentiation is still undetermined. The aim of this study was to investigate the effects of 5-aza-2'-deoxycytidine (5-azacytidine) on TSH-mediated regulations of osteoblasts. Materials and Methods: MG63, a human osteoblastic cell-line, was treated with 5-azacytidine before inducing osteogenic differentiation using osteogenic medium (OM) containing L-ascorbic acid and ${\beta}$-glyceophosphate. Bovine TSH or monoclonal TSH receptor stimulating antibody (TSAb) was treated. Quantitative real-time PCR analyses or measurement of alkaline phosphatase activities were performed for evaluating osteoblastic differentiation. Results: Studies for osteogenic-related genes or alkaline phosphatase activity demonstrated that treatment of TSH or TSAb alone had no effects on osteoblastic differentiation in MG63 cells. However, treatment of 5-azacytidine, per se, significantly increased osteoblastic differentiation and combination treatment of 5-azacytidine and TSH or TSAb in the condition of OM showed further significant increase of osteoblastic differentiation. Conclusion: Stimulating TSH signaling has little effects on osteoblastic differentiation in vitro. However, in the condition of epigenetic modification using inhibitor of DNA methylation, TSH signaling positively affects osteoblastic differentiation in human osteoblasts.

Immune Evasion of G-CSF and GM-CSF in Lung Cancer

  • Tumor immune evasion is a complex process that involves various mechanisms, such as antigen recognition restriction, immune system suppression, and T cell exhaustion. The tumor microenvironment contains various immune cells involved in immune evasion. Recent studies have demonstrated that granulocyte colony-stimulating factor (G-CSF) and granulocyte-macrophage colony-stimulating factor (GM-CSF) induce immune evasion in lung cancer by modulating neutrophils and myeloid-derived suppressor cells. Here we describe the origin and function of G-CSF and GM-CSF, particularly their role in immune evasion in lung cancer. In addition, their effects on programmed death-ligand 1 expression and clinical implications are discussed.

Effects of Extracellular Signaling on the Endogenous Expression of Self-Renewal-Stimulating Factor Genes in Mouse Embryonic Stem Cells

  • In order to provide the basis for developing practical mouse embryonic stem cells (mESCs) culture method, how the endogenous level of self-renewal-stimulating factor genes was altered in the mESCs by different extracellular signaling was investigated in this study. For different extracellular signaling, mESCs were cultured in 2 dimension (D), 3D and integrin-stimulating 3D culture system in the presence or absence of leukemia inhibitory factor (LIF) and transcriptional level of $Lif$, $Bmp4$ and $Wnt3a$ was evaluated in the mESCs cultured in each system. The expression of three genes was significantly increased in 3D system relative to 2D system under LIF-containing condition, while only $Wnt3a$ expression was increased by 3D culture under LIF-free condition. Stimulation of integrin signaling in mESCs within 3D system with exogenous LIF significantly up-regulated transcriptional level of $Bmp4$, but did not induce transcriptional regulation of $Lif$ and $Wnt3a$. In the absence of LIF inside 3D system, the expression of $Lif$ and $Bmp4$ was significantly increased by integrin signaling, while it significantly decreased $Wnt3a$ expression. Finally, the signal from exogenous LIF significantly caused increased expression of $Lif$ in 2D system, decreased expression of $Bmp4$ in both 2D and 3D system, and decreased expression of $Wnt3a$ in integrin-stimulating 3D system. From these results, we identified that endogenous expression level of self-renewal-stimulating factor genes in mESCs could be effectively regulated through artificial and proper manipulation of extracellular signaling. Moreover, synthetic 3D niche stimulating endogenous secretion of self-renewal-stimulating factors will be able to help develop growth factor-free maintenance system of mESCs.

  • To enhance our understanding of immunological stimulating effects through the pathway by which nitric oxide (NO) activity and alkaline phosphatase(ALP) enzyme activity from the marine algae, Ulva lactuca, we have investigated NO activity by using mouse RAW264.7 cell line. And ALP enzyme activity performed by spleen of ICR mouse. The results showed that NO activity of the $H_2O$ fraction is the most effective than activities of other solvent fractions.

A Growth-Stimulating Protein in Cow's Milk

  • 이 실험에서는 소의 유즙에 있는 성장인자를 세포배양을 통해 확인하고 크로마토그래피를 사용하여 분리 정제하였다. 5%의 우유를 포함하는 배지를 FBS(fetal bovine serum)를 포함한 배지와 비교했을때 Africal green monkey kidney cell의 성장 촉진 효과가 비슷하게 나타났으며, 우유의 성장인자를 분리 정제한 결과 FBS배지에서보다 2,000배 이상의 성장 촉진 효과가 나타났다. 이 성장인자는 hydrophobic column(phenyl-sepharose)과 gel-filtration column(sephadex G-100)을 사용하여 분리했으며 그 결과 milk-deriven growth factor는 phenyl-sepharose에서 50% ethylene glycol step에서 용출되므로 hydrophobic protein임이 증명되었고 size exclusion column chromatography로 부터 분자량이 100,000-15,0000 범위의 고분자 물질임이 밝혀졌다. 또한 우유의 성장인자는 매우 다양하며 그중 본 실험으로부터 정제된 물질이 주요 성장인자로 밝혀졌다.

  • Melanin determines phenotypic appearance and its election-opaque property protects cells from physical, including ultraviolet (UV) radiation, and chemical stimuli such as free radicals. However hyper-pigmentation is associated with various skin diseases such as keloid scar. The aim of present study was to investigate the effects of aqueous extracts from Angelica dahurica Benth. (AEAD) on ${\alpha}-Melanocyte$ stimulating hormone $({\alpha}-MSH)-induced$ melanogenesis in B16 mouse melanoma cell. Relative high doses ($5\;mg/m{\ell}$) of AEAD could inhibit melanin formation without apoptotic death in cells treated with ${\alpha}-MSH$. And also, ${\alpha}-MSH-induced$ activation of tyrosinase was inhibited in cells treated with AEAD. These results suggest that AEAD inhibit melanogenesis through inhibiting tyrosinase activity, and also, AEAD may apply to develop whitening drugs and cosmetics.

  • Purpose: The purpose of this study was to identify the effects of hand acupuncture therapy on sleep quality by means of a sleep questionnaire, polysomnography and a cerebral blood flow test in $30{\sim}59$ year old adults. Methods: The study was a sham controlled design. Twenty-two adults were assigned to the pellet stimulating group (11) or sham group (11). The pellet stimulating group received hand acupuncture therapy using New Seoam Press Pellets number 1 for 4 weeks. On the other hand, the sham group used the same Adhesive tape in terms of shape, size and quality as New Seoam Press Pellets number 1 for 4 weeks. A Transcranial Doppler Ultrasonography and Carotid Duplex Ultrasonography examination were used for evaluating cerebral blood flow. Data was analyzed using the SPSS 12.0 version program with $X^2-test$, Fisher's exact test and Mann Whitney U-test. Results: In the pellet stimulating group, subjective sleep quality significantly improved more than that of the sham group. Among the sleep indices of the polysomnography, total sleep time and sleep latency of the sham group significantly improved. The cerebral blood flow test didn't show any differences. Conclusion: These results suggest that hand acupuncture therapy is effective for subjective sleep improvement only, not polysomnographical sleep indices and cerebral blood flow.

Immune-stimulating Effect of Lactobacillus plantarum Ln1 Isolated from the Traditional Korean Fermented Food, Kimchi

  • This study aimed to determine the immune-stimulating effects of heat-killed Lactobacillus plantarum Ln1 (HK-Ln1) through the production of nitric oxide (NO) and pro-inflammatory cytokine achieved by inducing NF-κB and mitogen-activated protein kinase (MAPK)-signaling pathways in macrophages. HK-Ln1 showed higher NO and cytokine production compared to control (nonstimulated lipopolysaccharide); in addition, the expression of inducible nitric oxide synthase (iNOS) was induced through HK-Ln1treatment. The phosphorylation of IκB-α and p65 increased following treatment by HK-Ln1, which implicates IκB-α degradation and the translocation of p65 to nucleus. In addition, the phosphorylation of MAPKs, ERK 1/2, JNK, and p38 was induced following HK-Ln1 treatment.

TERATOGENICITY STUDY OF RECOMBINANT GRANULOCYTE-MACROPHAGE COLONY STIMULATING FACTOR (LBD-005) IN RABBITS

  • LBD-005, a newly developed recombinant granulocyte-macrophage colony-stimulating factor, was at dose levels of 0, 20, 80 and 320ng/kg/day administered subcutaneously to pregnat New Zealand White rabbits during the organogenetic period. The dams were subjected to caesarean section on day 28 of pregnancy. Effects of test substance on dams and embryonal development of fetuses were examined. No treatment-related changes in clinical signs and necropsy findings of dams were observed in all groups. At 80 and 320 ng/kg, a significant decrease in food consumption followed by a loss in body weight was found.

  • Administration of 3 type KGCs [recombinant human granulocyte colony-stimulating factor (rhG-CSF)] to mice with cyclophosphamide (CPA)-induced neutropenia for 4 consecutive days from the day after the CPA dosing (100 mg/kg) resulted in a dose dependent increase in the peripheral blood neutrophil count 6 hours after the final KGC injection. Within the KGC dose range of 0.1 to 40$\mu$g Per mouse Per day, there was a sigmoidal relationship between the logarithm of the dose and the peripheral blood neutrophil count (relative value for neutrophil count of the basal dose) in the treated mice. The sigmoidal relationship of test KGC preparations shows that there is a saturation point in terms of efficacy. Compared with e(fact of KGC-Orange, Green, and Blue, KGC-Orange recovers neutrophils more effectively than the others do.
