Arabidopsis thaliana의 Ethylene Triple Response Mutant에서 에틸렌 생합성 과정의 생리 생화학적 특성

  • The physiological and biochemical characterizations of the ethylene-related mutants in Arabidopsis thaliana - ethylene overproducing mutant (eto1-l) and ethylene insensitive mutants (etrl-3, ein2-l) - were detailed in this studies. Two or three week.old mature rosette leaves (before bolting) were used as the plant materials. Ethylene productions of eto1-l, etrl-3, and ein2-l mutants were about 200%, 400%, and 450% compared to that of wild type, respectively. ACC synthase and ACC oxidase activities of eto1-l mutant were similar to those of wild type. ACC content and ACC N-malonyltransferase activity, however, were 4.5 times and 3 times higher than those of wild type, respectively. SAM synthetase activity increased by 50% in eto1-l mutant plant. These results indicated that the alteration in the eto1-l mutant occured before the step of the conversion of SAM to ACe. In etrl-3 and ein2-l mutants, ACC synthase activities increased, but ACC oxidase activities decreased. ACC content and ACC N-malonyltransfcrase activity were 2 times higher than those of wild type. SAM synthetase activity in etrl-3 is similar to those of wild type, while it increased by 73% in ein2-l. These results showed that the block in ethylene action affected the autoregulation of ethylene biosynthesis, so that ACC synthase activity was not autoinhibited and ACC oxidase activity was not auto stimulated by ethylene. When the leaf tissues were used for in vitro kinase assay, a cytosolic protein (approximately 36 kDa) was phosphorylated only in eto1-l and ein2-l mutants.utants.

A Novel Pyruvate Kinase M2 Activator Compound that Suppresses Lung Cancer Cell Viability under Hypoxia

  • Pyruvate kinase M2 isoform (PKM2), a rate-limiting enzyme in the final step of glycolysis, is known to be associated with the metabolic rewiring of cancer cells, and considered an important cancer therapeutic target. Herein, we report a novel PKM2 activator, PA-12, which was identified via the molecular docking-based virtual screening. We demonstrate that PA-12 stimulates the pyruvate kinase activity of recombinant PKM2 in vitro, with a half-maximal activity concentration of $4.92{\mu}M$, and effectively suppresses both anchorage-dependent and -independent growth of lung cancer cells in non-essential amino acid-depleted medium. In addition, PA-12 blocked the nuclear translocalization of PKM2 in lung cancer cells, resulting in the inhibition of hypoxia response element (HRE)-mediated reporter activity as well as hypoxia-inducible factor 1 (HIF-1) target gene expression, eventually leading to the suppression of cell viability under hypoxia. We also verified that the effects of PA-12 were dependent on PKM2 expression in cancer cells, demonstrating the specificity of PA-12 for PKM2 protein. Taken together, our data suggest that PA-12 is a novel and potent PKM2 activator that has therapeutic implications for lung cancer.

Middle School Students' Evaluation of Scientific Information: From the Perspective of Hypothetico-deductive Reasoning (가설-연역적 추론 관점에서 본 중학생의 과학적 정보 평가 양상)

  • The purpose of this study is to find out how middle school students evaluate scientific information in terms of hypothetico-deductive reasoning. A total of 66 middle school students completed a paper-and-pencil test on scientific information evaluation and 14 of them were individually interviewed for triangulation. The test includes six topics related to scientific or pseudoscientific information, and questions about each topic were sequenced based on a hypothetico-deductive reasoning. The hypothetico-deductive process consists of three steps: identifying predictions made by explanations in the information, identifying data actually obtained, and determining the fit between predictions and data to judge the validity of the explanations. Data analyses have focused on students' response types at each step, whether students used hypoethetico-deductive reasoning, and students' preference to evidence types in making decisions. The middle school students in this study answered the questions in various ways based on how they used the information given or personal knowledge and beliefs. A small portion of students evaluated information based on hypothetico-deductive reasoning. These students tended to give priority to scientific data in determining the validity of the information. On the other hand, students who did not use hypoethetico-deductive reasoning tended to prefer first-hand experience in the decision. The results provide implications for science lessons and the curriculum for scientific literacy. Further research should include student evaluation of the validity of data and other types of reasoning.

Anti-CD3 Antibody Induces IL-10-producing $CD4^+CD25^+$ Regulatory T Cells, Which Suppress T Cell Response in Rheumatoid Arthritis Patients

  • Background: Regulatory T cells (Tregs) have been investigated intensively for some decades. These cells regulate the immune system, prevent overactivated immune responses and can be used therapeutically. For rheumatoid arthritis (RA), understanding the functions and status of Tregs is an important step for understanding immune regulation in this autoimmune disease. Methods: We investigated the percentages, phenotypes and suppressive functions of $CD4^+CD25^+$ Tregs in peripheral blood (PB) of patients with RA. Results: The percentages were higher in the patients (n=12) than in healthy controls (n=10), and the cells expressed the $CD45RB^{low}$, CTLA-4 and CCR7 phenotypes. We also investigated the expression of Foxp3 and secretion of interleukin (IL)-10 induced $CD4^+CD25^+$ Tcells by anti-CD3 antibody treatment. A suppressive function of the patients' cells was shown through coculture with $CD4^+CD25^-$ T cells in vitro. Conclusion: We suggest that, despite their increased numbers and suppressive function, they manage the ongoing inflammation ineffectively. It might be possible to apply IL-10 to induce the proliferation of IL-10-producing Tregs as therapy for RA.

  • Differently from an existing analog control, because the digital control includes microprocessor basically, the digital control is enable to monitor internal parameters of DC-DC converter and to control output voltage remotely by communicating with a Windows based PC. These things are impossible in an analog control and there are more advantages in a digital control than an analog control. In this paper, with the advantages mentioned above, the feasibility of digital controlled DC-DC converter in low price is proposed. In order to implement these functions, it is used the inexpensive H8/3672 made by Renesas that has built in AD converters and PWM logic generators. The proposed digital controller is applied to a flyback converter that is designed to output DC 5[V] from DC 20∼30[V] and is remotely controlled to make variable outputs from DC 0[V] to 5[V] above in PC. The PWM controller adopts the PD controller in PID. In the last, the response characteristics of a step reference voltage and in a steady state are experimented to verify the feasibility and the usefulness of the proposed flyback converter that is implemented inexpensively.

The Constitutional Ideas in the 『Huangdi Neijing』 (『황제내경(黃帝內經)』의 체질론(體質論))

  • One's constitution is formed congenitally and is also acquired, and is relatively stable in its individually manifested function, structure and temperament. It has characteristics of universality, complexity, generality and continuity. It is also manifested in physiological responses and expresses pathologic tendencies including susceptibility. Attempts at understanding constitution has had a long history throughout the world. In Oriental Medicine, the constitution had been acknowledged from the "Huangdi Neijing", which has been a bible in Oriental Medicine for about two thousand years. In many aspects, the "Huangdi Neijing" provides the basis for both basic and practical fields. In order to derive the ideal structure from the "Huangdi Neijing" to Lee Je-ma's(Lee Jayma's) Sasang Constitutional ideas, which can strengthen the understanding of ideal core of Oriental Medicine's constitutional characteristics, the author launched his analysis of the constitutional understandings in the "Huangdi Neijing" as a first step and came to the following: 1. The constitutional understanding in the "Huangdi Neijing" was done both via the physiological responses to heat, pain and acupuncture treatment and the pathologic tendencies via body structure, strength, body heat, courage and obesity. 2. It was recognized that the constitution was formed congenitally and also acquired factors like living conditions including food customs, residents and geographical conditions were explained in detailed. 3. The constitutional typology was suggested both by the general and systematic criterion according to the Yin-yang and the Five phase theories and by single criterion such as obesity, courage and the response to acupuncture treatment. 4. As diagnosis is related to the constitution, courage was adopted for the disease caused by emotions, obesity was for its manifestations of qi and blood, the quantities of qi-blood and Yin-yang in the typology by the Yin-yang and Five phase theories were suggested. 5. In the case of obesity, Yin-yang and Five phase theory, treatment according to the constitutional ideas was mentioned, and their goals were concluded as "being the balancing between Yin and Yang" which means the Yin-yang theory is more practical than the Five phase theory in Oriental Constitutional Medical field. According to the above understandings, the author would like to suggest that the constitutional ideas in the "Huangdi Neijing" based on the Yin-yang theory blossomed via the practical spirit of Zhang Zhong-jing's "Shanghanlun" and at last from Lee Je-ma's(Lee Jayma's) Sasang Constitutional Medicine which realized "the balancing between Yin and yang" through the practical applications of herbal treatments.

A Study on the Correlation between Categorization of the Individual Exposure Levels to Agent Orange and Serum Dioxin Levels Among the Korean Vietnam Veterans (베트남 참전 제대 군인의 범주화된 에이전트 오렌지 개인 폭로량과 혈청 다이옥신 측정치와의 상관성에 관한 연구)

  • Objectives : In an epidemiologic study on the health impact of Agent Orange exposure, the valid estimation of exposure level is the most important step. Based on recent studies, we examined the correlation between exposure levels categorized by personal exposure estimates and serum 2,3,7,8-tetrachlorodibenzo-p-dioxin (2,3,7,8-TCDD, Dioxin), exploring the possibility of utilizing the exposure level as a surrogate for the estimate of exposure to agent orange. Methods : During the study period (Jan 1996-Feb 1996), blood specimens of 745 subjects taken randomly among 1,329 persons and kept frozen, were analyzed for 2,3,7,8-TCDO and six other dioxin congeners. The serum dioxin and congeners were measured in 1998 by CDC, adjusted for serum lipids. We categorized the total exposure scores into five groups based on Agent Orange exposure data collected by interview and military records. Pearson and Spearman's correlation coefficients & multiple regression analysis were used to identify the relationship of the exposure level categorized with serum concentration of 2,3,7,8-TCDD, and six other dioxin congeners. Results : Dioxin and the other congeners, except 1,2,4,6,7,8-HpCDD, showed significant correlations to exposure categories (p<0.005): 2,3,7,8-TCDD and OCDD showed positive correlations, whereas the other congeners did negative. The values of 2,3,7,8-TCDD differed according to exposure category and proportionally increased from the low exposure group to the high, a dose-response relationship, even after other possible confounding variables were adjusted for. In multiple regression analysis, age$(\beta=0.033)$, dioxin$(\beta=0.433)$, 1,2,3,7,8-PeCDD$(\beta=-0.998)$, 1,2,3,4,7,8-HxCDD$(\beta=-0.773)$, 1,2,3,6,7,8-HxCDD$(\beta=0.255)$, 1,2,3,7,8,9-HxCDD$(\beta=-3.468)$, 1,2,3,4,6,7,8-HpCDD$(\beta=0.109)$ we re found to be significantly related to the total exposure score(p<0.005). Conclusion : This study demonstrated that the use of such categorizations as a surrogate measure of agent orange exposure in identifying exposure degrees in a health impact study is valid.

Synthesis of (4-$[^{18}F]$Fluorophenyl)triphenylphosphonium as a Mitochondrial Voltage Sensor for PET (PET영상용 미토콘드리아 막전위 감지기 (4-$[^{18}F]$Fluorophenyl)triphenylphosphonium의 합성)

  • Purpose: Lipophilic cations including tetraphenylphosphonium (TPP) salts penetrate the hydrophobic barriers of the plasma and mitochondrial membranes, resulting in accumulation in mitochondria in response to the negative inner transmembrane potentials. The development of radiolabeled phosphonium cations as a noninvasive imaging agent may serve as a new molecular "voltage sensor" probe to investigate the role of mitochondria in the pathophysiology and diagnosis of cancer. Materials and Methods: We have synthesized a reference compound (4-fluorophenyl)triphenylphosphonium (TPP) and a labeled compound $[^{18}F]$TPP via two step nucleophilic substitution of no-carrier-added $[^{18}F]$fluoride with the precursor, 4-iodophenyltrimethylammonium iodide, in the presence of Kryptofix-2.2.2 and $K_2CO_3$. Result: The reference compound (4-fluorophenyl)triphenylphosphonium (TPP) was synthesized in 60% yield. The radiolabeled compound $[^{18}F]$TPP was synthesized in $10\sim15%$ yield. The radiochemical purity of the $[^{18}F]$TPP was $95.57{\pm}0.51%$ (n=11). Conclusion: $[^{18}F]$TPP was successfully synthesized that might have a potential to be utilized as a novel myocardial or cancer imaging agent for PET. However, it is required to improve the radiochemical yield to apply $[^{18}F]$TPP in preclinical or clinical researches.

  • This paper developed the algorithm for improving the performance the auto pilot in the autonomous vehicle system consisting of the Track keeping control, the Automatic steering, and the Automatic mooring control. The automatic steering is the control device that could save the voyage distance and cost of fuel by reducing the unnecessary burden of driving due to the continuous artificial navigation, and avoiding the route deviation. During the step of the ship autonomic navigation control, since the wind power or the tidal force could make the ship deviate from the fixed course, the automatic steering calculates the difference between actual sailing line and the set course to keep the ship sailing in the vicinity of intended course. first, we could get the transfer function for the modeling of ship according to the Nomoto model. Considering the maneuverability, we propose it as linear model with only 4 degree of freedoms to present the heading angle response to the input of rudder angle. In this paper, the model of ship is derived from the simplified Nomoto model. Since the proposed model considers the maximum angle and rudder rate of the ship auto pilot and also designs the Fuzzy controller based on existing PID controller, the performance of the steering machine is well improved.

  • This is an explicit-Implicit, finite element analysis for linear as well as nonlinear hygrothermal stress problems. Additional features, such as moisture diffusion equation, crack element and virtual crack extension(VCE ) method for evaluating J-integral are implemented in this program. The Linear Elastic Fracture Mechanics(LEFM) Theory is employed to estimate the crack driving force under the transient condition for and existing crack. Pores in materials are assumed to be saturated with moisture in the liquid form at the room temperature, which may vaporize as the temperature increases. The vaporization effects on the crack driving force are also studied. The Ideal gas equation is employed to estimate the thermodynamic pressure due to vaporization at each time step after solving basic nodal values. A set of field equations governing the time dependent response of porous media are derived from balance laws based on the mixture theory Darcy's law Is assumed for the fluid flow through the porous media. Perzyna's viscoplastic model incorporating the Von-Mises yield criterion are implemented. The Green-Naghdi stress rate is used for the invariant of stress tensor under superposed rigid body motion. Isotropic elements are used for the spatial discretization and an iterative scheme based on the full newton-Raphson method is used for solving the nonlinear governing equations.
