Alternanthera mosaic virus - an alternative 'model' potexvirus of broad relevance

  • Alternanthera mosaic virus (AltMV) is a member of the genus Potexvirus which has been known for less than twenty years, and has been detected in Australasia, Europe, North and South America, and Asia. The natural host range to date includes species in at least twenty-four taxonomically diverse plant families, with species in at least four other families known to be infected experimentally. AltMV has been shown to differ from Potato virus X (PVX), the type member of the genus Potexvirus, in a number of ways, including the subcellular localization of the Triple Gene Block 3 (TGB3) protein and apparent absence of interactions between TGB3 and TGB2. Differences between AltMV variants have allowed identification of viral determinants of pathogenicity, and identification of residues involved in interactions with host proteins. Infectious clones of AltMV differing significantly in symptom severity and efficiency of RNA silencing suppression have been produced, suitable either for high level protein expression (with efficient RNA silencing suppression) or for Virus-Induced Gene Silencing (VIGS; with weaker RNA silencing suppression), demonstrating a range of utility not available with most other plant viral vectors. The difference in silencing suppression efficiency was shown to be due to a single amino acid residue substitution in TGB1, and to differences in subcellular localization of TGB1 to the nucleus and nucleolus. The current state of knowledge of AltMV biology, including host range, strain differentiation, host interactions, and utility as a plant viral vector for both protein expression and VIGS are summarized.

Evaluation and Application of T-Ray Nondestructive Characterization of FRP Composite Materials (FRP 복합재료의 T-Ray 비파괴특성 평가 및 적용)

  • Recently, (terahertz ray) applications have emerged as one of the most promising new powerful nondestructive evaluation (NDE) techniques. In this study, a new T-ray time-domain spectroscopy system was utilized for detecting and evaluating layup effect and flaw in FRP composite laminates. Extensive experimental measurements in reflection and thru-transmission modes were made to map out the T-ray images. Especially this was demonstrated in thick GFRP laminates containing double saw slots. In carbon composites the penetration of terahertz waves is limited to some degree and the detection of flaws is strongly affected by the angle between the electric field(E-field) vector of the terahertz waves and the intervening fiber directions. The artificial defects investigated by terahertz waves were bonded foreign material, simulated disbond and delamination and mechanical impact damage. The effectiveness and limitations of terahertz radiation for the NDE of composites are discussed.

Beamforming and User Scheduling for Device-to-Device Underlaying Cellular Networks (셀룰러 네트워크에서 D2D 통신을 고려한 빔 형성 및 스케줄링 기법)

  • In this paper, the beamforming and user scheduling scheme for device-to-device (D2D) underlaying celullar networks with partial channel state information is investigated. In the proposed scheme, cellular users whose channels are nearly orthogonal to the channel between base station and D2D receiver are selected for unitary beamforming so as to minimize the interference to the D2D receiver. The interference caused by underlaying D2D communication can be minimized by reducing the D2D transmitter's transmission power. To enhance the cellular users' throughput further, we also investigate codebook generation method in which each code vector belongs to multiple unitary code matrices. From simulation results, we show that the proposed beamforming and user scheduling scheme mitigates D2D interference to the cellular networks and the codebook generation method improves cellular users' throughput.

  • In this paper we analyze the CA formed by combining the uniform 102 CA $\mathbb{C}_u$ and the m-cell 90/150 hybrid CA $\mathbb{C}_h$ whose characteristic polynomial is $(x+1)^m$. We analyze cycle structures of complemented group CA derived from $\mathbb{C}_u$ and propose a condition of complemented CA dividing the entire state space into smaller cycles of equal lengths. And we analyze the cycle structure of complemented group CA $\mathbb{C}^{\prime}$ derived from the CA $\mathbb{C}$ formed by combining $\mathbb{C}_u$ and $\mathbb{C}_h$ with complement vector F such that $(T+I)^{q-1}F{\neq}0$ where $(x+1)^q$ is the minimal polynomial of $\mathbb{C}$.

Characterization of porcine cytokine inducible SH2-containing protein gene and its association with piglet diarrhea traits

  • Objective: The cytokine inducible SH2-containing protein (CISH), which might play a role in porcine intestine immune responses, was one of the promising candidate genes for piglet anti-disease traits. An experiment was conducted to characterize the porcine CISH (pCISH) gene and to evaluate its genetic effects on pig anti-disease breeding. Methods: Both reverse transcription polymerase chain reaction (RT-PCR) and PCR were performed to obtain the sequence of pCISH gene. A pEGFP-C1-CISH vector was constructed and transfected into PK-15 cells to analysis the distribution of pCISH. The sequences of individuals were compared with each other to find the polymorphisms in pCISH gene. The association analysis was performed in Min pigs and Landrace pigs to evaluate the genetic effects on piglet diarrhea traits. Results: In the present research, the coding sequence and genomic sequence of pCISH gene was obtained. Porcine CISH was mainly localized in cytoplasm. TaqI and HaeIII PCR restriction fragment length polymorphism (RFLP) assays were established to detect single nucleotide polymorphisms (SNPs); A-1575G in promoter region and A2497C in Intron1, respectively. Association studies indicated that SNP A-1575G was significantly associated with diarrhea index of Min piglets (p<0.05) and SNP A2497C was significantly associated with the diarrhea trait of both Min pig and Landrace piglets (p<0.05). Conclusion: This study suggested that the pCISH gene might be a novel candidate gene for pig anti-disease traits, and further studies are needed to confirm the results of this preliminary research.

  • 최근 기상변동성 증가와 극치수문사상의 발생빈도 증가로 인한 기상재해가 빈번하게 일어나고 있다. 이러한 기상현상으로 인한 재해의 예방을 위해서 사전에 위험을 인지하고 그 규모를 예측할 수 있는 여러 기법들이 기상레이더 또는 수치예보자료 등을 이용하여 개발 및 적용되고 있다. 이 과정에서 해결해야 할 여러 문제점들이 있는데, 우선 수치예보자료 또는 기상레이더자료를 종관기상관측소 및 자동기상관측지점의 지상관측 강수량과 연계하여 평가하는 과정이 필요하고, 현재시점에 형성되어 있는 강우장의 공간 이동 예측 기법이 확보되어야 할 것이다. 전북지역은 게릴라성 집중호우가 빈번한 산악형 강수와 산지유역의 급한 하천경사가 맞물려 인명 및 재산피해가 매년 발생하고 있으며, 과거 돌발홍수가 발생한 사례가 있어 이상기후 및 기후변화로 인한 홍수 위험도가 커질 것으로 전망되고 있다. 본 연구는 전라북도의 기상재해 예측모형 개발을 위한 사전 분석과정으로 전라북도지역에서 관측된 기존의 대규모 강수사상을 이용한 강수사상의 특성 분류 및 관측소간 공간상관성을 분석하는데 목적을 두고 있다. 강수사상의 특성분류를 통해 강수 발생형태에 따른 기상학적 영향인자, 강수의 발생량 및 이동특성 예측의 정도를 향상시킬 수 있으며, 분류 기법으로 SVM(support vector machine)을 이용한 자동분류를 적용한다. 또한 관측소간 공간상관성 분석을 위하여 각 관측소 강수량간의 조건부 확률을 이용한다. 예로써 부안관측소에 강수가 발 생했을 때, 부안관측소의 강수량 조건에 의한 전주관측소 강수량 확률을 다음과 같이 구성할 수 있다. �揚滑斂�수량�咀刮활�수량��. 공간상관성 분석과정에서 관측소간 강수 이동시간에 따른 강수 발생 시간의 차이 또한 고려하며, 과거 기상관측 자료의 분석을 통해 전라북도지역의 관측소간 강수발생의 공간적 상관성을 규명하고, 단기예측 모델 개발을 위한 기초자료로 활용할 수 있을 것이다. 또한, 기후변화시나리오에 의한 미래 강수량의 지역적 상세화 과정에도 본 연구를 통한 결과를 이용할 수 있을 것이라 판단된다.

Ground-based Observations of the Polar Region Space Environment at the Jang Bogo Station, Antarctica

  • Jang Bogo Station (JBS), the second Korean Antarctic research station, was established in Terra Nova Bay, Antarctica ($74.62^{\circ}S$ $164.22^{\circ}E$) in February 2014 in order to expand the Korea Polar Research Institute (KOPRI) research capabilities. One of the main research areas at JBS is space environmental research. The goal of the research is to better understand the general characteristics of the polar region ionosphere and thermosphere and their responses to solar wind and the magnetosphere. Ground-based observations at JBS for upper atmospheric wind and temperature measurements using the Fabry-Perot Interferometer (FPI) began in March 2014. Ionospheric radar (VIPIR) measurements have been collected since 2015 to monitor the state of the polar ionosphere for electron density height profiles, horizontal density gradients, and ion drifts. To investigate the magnetosphere and geomagnetic field variations, a search-coil magnetometer and vector magnetometer were installed in 2017 and 2018, respectively. Since JBS is positioned in an ideal location for auroral observations, we installed an auroral all-sky imager with a color sensor in January 2018 to study substorms as well as auroras. In addition to these observations, we are also operating a proton auroral imager, airglow imager, global positioning system total electron content (GPS TEC)/scintillation monitor, and neutron monitor in collaboration with other institutes. In this article, we briefly introduce the observational activities performed at JBS and the preliminary results of these observations.

Finite Element Modal Analysis of a Spinning Flexible Disk-Spindle System Supported by Hydro Dynamic Bearings and Flexible Supporting Structures In a HDD (유연한 지지 구조와 유체 동압 베어링으로 지지되는 HDD의 회전 유연 디스크-스핀들 시스템에 대한 유한 요소 고유 진동 해석)

  • The free vibration of a spinning flexible disk-spindle system supported by hydro dynamic bearings in a HDD is analyzed by FEM. The spinning flexible disk is described using Kirchhoff plate theory and von Karman non-linear strain, and its rigid body motion is also considered. It is discretized by annular sector element. The rotating spindle which includes the clamp, hub, permanent magnet and yoke, is modeled by Timoshenko beam including the gyroscopic effect. The flexible supporting structure with a complex shape which includes stator core, housing, base plate, sleeve and thrust pad is modeled by using a 4-node tetrahedron element with rotational degrees of freedom to satisfy the geometric compatibility. The dynamic coefficients of HDB are calculated from the HDB analysis program, which solves the perturbed Raynolds equation using FEM. Introducing the virtual nodes and the rigid link constraints defined in the center of HDB, beam elements of the shaft are connected to the solid elements of the sleeve and thrust pad through the spring and damper element. The global matrix equation obtained by assembling the finite element equations of each substructure is transformed to the state-space matrix-vector equation, and the associated eigenvalue problem is solved by using the restarted Arnoldi iteration method. The validity of this research is verified by comparing the numerical results of the natural frequencies with the experimental ones. Also the effect of supporting structures to the natural modes of the total HDD system is rigorously analyzed.

Sensitivity Analysis of Vortical Hull Girder Vibration (선체 저차 상하고유진동 감도해석)

  • This paper presents a prediction method of natural frequencies of vertical hull girder vibration based on design sensitivity analysis in case of design modification and the variation of loading condition. The resented method premises the vibration analysis by the transfer matrix method. Governing sensitivity equation is derived from the direct differentiation of state vector and transfer matrix to parameters and its transfer over all the hull girder elements. Derivatives of natural frequencies and mode shapes are determined by two trial calculation of the governing equation. Using the derivatives, the changes of natural frequencies and mode shapes can be predicted when mass and stiffness parameter's are changed. As results, it is possible to optimize ship structure as well as to avoid troublesome calculation in hull girder vibration analysis rationally and efficiently. To verify the accuracy and efficiency of the resented method numerical results obtained by both the sensitivity analysis and the ordinary reanalysis far a real ore/bulk carrier in case of the change of mass and stiffness parameters are compared.

Free and Forced Vibration Analysis of a Hard Disk Drive Considering the Flexibility of Spinning Disk-Spindle, Actuator and Supporting Structure (회전 디스크-스핀들, 액츄에이터와 지지구조의 유연성을 고려한 하드 디스크 드라이브의 고유 및 강제 진동 해석)

  • This paper presents a finite element method to analyze the free and forced vibration of a hard disk drive (HDD) considering the flexibility of a spinning disk-spindle with fluid dynamic bearings (FDBs), an actuator with pivot bearings, an air bearing between head-disk interface and the base with complicated geometry. Finite element equation of each component is consistently derived with the satisfaction of the geometric compatibility of the internal boundary between each component. The spinning disk, hub and FDBs are modeled by annular sector elements, beam elements and stiffness and damping elements, respectively. The actuator am, E-block, suspension and base plate are modeled by tetrahedral elements. The pivot bearing in the actuator and the air bearing between head-disk interfaces are modeled by the stiffness element with five degrees of freedom and the axial stiffness, respectively. A global matrix equation obtained by assembling the finite element equations of each substructure is transformed to a state-space matrix-vector equation, and both damped natural frequencies and modal damping ratios are calculated by solving the associated eigenvalue problem with the restarted Arnoldi iteration method. Modal and shock testing are performed to show that the proposed method well predicts the vibration characteristics of a HDD.
