Genetic Studies of Oenothera odorata Populations in Korea Based on Isozyme Analysis

  • The genetic variation in Korean evening primorse (Oeothera odorata L.) populations was examiend to estimate the level of allozyme variation within populatons using starch gel electrophoresis. 7 of 13 loci (Adh, Est-1, Est-2, Mdh-2, Pgd-2, Pgm-1, and Idh) revealed (Ps=43.2%) were polymorphic. The mean number of alleles per locus (A) and polymorphic locus (Ap) for populations were 1.64 and 2.46, respectively. The effective number of alleles (Aep) within populations relatively was low ranging from 1.08 to 1.22 with a mean of 1.14. Within populations, the mean number of allele per polymorphic loci (Ap) was 2.46, the mean number of alleles per locus (A) was 1.64, and the mean genetic diversity was 0.093. About 2.7% of the total allozyme diversity resided among populations (Mean GST=0.0274). FIS, a measure of the deviation from random mating within 13 populations, was relative low (mean FIS=0.03636). The indirect estimate of gene flow, based on the mean GST, was high (Nm=8.88). Estimates of gene flow were consistent with low levels of genetic differentiation among populations.

Isolation, Purification and Characterization of Chitosanase from Bacillus subtilis CH1

  • Bacillus subtilis strain with highly active chitosanase was isolated from the intestine of Sebastiscus marmoratus (scorpion fish). It was named as Bacillus subtilis CH1 by morphological, biochemical and 165 rRNA gene analysis. The optimal conditions for chitosanase production were investigated. The optimum carbon and nitrogen sources for Bacillus stibtilis CH1 were 2% starch and 1% yeast extract respectively. Unlike other chitosanases, the expression of this chitosanase was not induced or slightly induced with chitosan. The chitosanase secreted into the medium were concentrated with ammonium sulfate precipitation and purified by gel permeation chromatography. The molecular weight of purified chitosanase was 30 kDa. The optimum pH and temperature of purified chitosanase were 5.5 and $60^{\circ}C$ respectively. The purified chitosanase was continuously thermostable at $40^{\circ}C$ and showed stable activity between pH 6.0 and 8.0. Chitosanase activity of Bacillus subtilis CH1 under optimum condition was 4.1 units/ml.

Partial Purification and Properties of Polygalacturonase Produced by Botrytis cinerea (잿빛곰팜이병균 Botrytis cinera가 분비하는 Polygalacturonase의 부분정제와 특성)

  • Polygalacturonase (PG) produced by Botrytis cinerea in the culture broth containing citrus pectin as a carbon source was partially purified and characterized. PG was produced on a range of carbon sources such as starch, glycerol, cellobiose, and Na+-PAG with total activities of 34.8, 32.0, 29.2, 27.8 units, respectively. The specific activity was highest with 2316.7 units on Na+-PGA. Proteins of culture filtrate were concentrated with polyethylene glycol and acetone and applied to a hydroxyapatite column. Among three active fractions collected from the column, the reaction containing the highest PG activity was resolved by a Q-sepharose column. The active fraction from the Q-sepharose column was further purified by HPLC Mono Q column. The partially purified enzyme was analyzed by 10% sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Among a few protein bands revealed, the amount of the protein of which molecular weight estimated to be 43 kDa coincided with the PG activity. The partially purified PG had optimal temperatures between 35~55$^{\circ}C$ and pH between 4.5~5.5.

  • Horizontal starch gel electrophoresis, follow by enzyrne-specific staining, separate and visualize several legume esterase isozyme. Using extracts prepared from cotyledon, radicle and plumule of legume seedlings germinated 5 days. The results were as follows. 1. The number and staining intensity of esterase isozyme bands varies depending on the plant species. tissues and developmental stage. 2. Bands in the cotyledon of field bean seedling expressed 4 and 1 in radicle. 3. In soybean cultivars, cotyledon of IIwangkum-kong had 3 bands and 1 band in the examined tissues of Paldal-kong and Jangkyung-kong seedling. 4. The cotyledon and radicle of french bean seedling had 3 bands, respectively. 5. The highest esterase isozyme activity appears to be expressed in the cotyledon and radicle of french bean, as indicated by intensity of stain, with the Paldal-kong particulary active.

A Genetic Analysis of the Common Squid, Todarodes pacificus in the Korean Waters

  • In order to estimate the genetic variability and differentiation in common squid, eleven isozymic loci, coded for nine enzymes detected by starch gel electrophoresis, were scored from nine spawning cohorts in four localities. The expected average heterozygosity ranged from 0.00019 (between II-S$_2$ and Na-W) to 0.00814 (Between Bu-S and Na-W) in nine different spawning cohorts in four localities. A dendrogram, based on genetic distance mentioned, illustrated that nine different spawning cohorts were divided into three groups, similar to the result estimated by their ecological characterizations. From these results, we estimate that the common squid distributed throughout Korean waters will maintain this gene exchange. It is postulated that either the summer or the autumn spawning cohort has developed a local population that is isolated by hydrographic factors.

Genetic Variations of Natural and Hatchery Populations of Korean Ayu (Plecoglossus altivelis) by Isozyme Markers

  • Genetic variability and population structure of 11 natural ayu, Plecoglossus altivelis populations and one hatchery stock were assessed by starch gel electrophoretic analysis with 10 enzyme coding loci. Three loci were polymorphic (lower than 0.95 in major allele frequency) in natural populations,2 loci in hatchery stock. The average number of alleles per locus was 1.38. Observed heterozygosities ranged from 0.0235 to 0.088 (0.055 on the average) in natural population while 0.0925 in hatchery stock. The genetic distance among natural populations measured 0.000047-0.005407 and no significant differentiation was observed among them. On the other hand, a signifcant genetic distance was found between natural populations and the hatchery stock with measuring 0.002032-0.O08605. The results in this study suggest that the hatchery stock has diverged from natural populations, and also that careful to maintain sustainable and effective population size (parents number) should be made.

Electrophoretic Study of Semisulcospira gottschei in Korea (한국산(韓國産) Semisulcospira gottschei의 전기영동적(電氣泳動的) 연구(硏究))

  • Genetic variations of Semisulcospjra gottschei in Korea were investigated by means of starch gel-electrophoresis. The results are as follows; 1) Eight loci of ${\alpha}$-Gpd, Mdh-2, Pept-1, Pgm-2, Gp-1, Gp-2, Gp-3 and Gp-4 were monomorphic, and 8 loci of Mpi, Mdh-1, Pgi, Got, Pept-2, Pept-3, Pgm-1 and Sdh showed genetic variations. 2) Means of allele per locus ($\bar{A}$) and polymorphism (P) were revealed 0.64 and 0.44, respectively. The value of heterozygosity ($H_G$) showed a little more than heterozygosity $H_D$. ($H_D$=0.09, $H_G$=0.13) 3) Genetic variation appears to be due to loci of Got, Pept, Pgi, and Sdh, all of which of the $H_D$ values are high.

  • A horizontal starch gel electrophoresis for enzyme proteins extracted from three Korean species and one Chinese species of Semisulcospira was carried out in order to elucidate their genetic relationships. A total of 10 enzymes were employed in three different of buffer systems. Two loci from each enzyme of GAPDH, GOT, ICDH, IDH and PEP(VL); three loci from each of three enzymes, EST, PEP(LGG) and PGDH; and five loci from GPI were observed. Most of the loci in three pleurocerid species employed showed homozygous monomorphic banding patterns and some of them were specific as genetic markers between two different species. However, EST-2, PEP(LGG-3) and PGDH-1 loci in Korean S. libertina and PEP(LGG-3), PGM-1 and PGM-2 loci in Chinese S. libertina showed polymorphic banding patterns. Three Korean Semisulcospira species including S. libertina were more closely clustered in a dendrogram within the range of genetic identity values of 0.818-0.936, and these clusters were lineated with Chinese S. libertina at the value of 0.621.

Characterization of YS-27, an axenic Korean strain of Entamoeba histolytica

  • Characterization of YS-27, an axenic Entamoeba strain. was performed by three different laboratory methods. Zymodeme analysis using starch gel electrophoresis and PCR with species-specific primers showed that YS-27 is a pathogenic Entamoeba which belongs to the group II zymodeme. Pathogenicity of YS-27 was further confirmed by observing the formation of liver abscess in Mongolian gerbils. These results showed that YS-27 is E. hisolytica.

  • One male in the former group of 2110 Korean males of the Republic of Korea Army stationed Taegu. And two males and one female in the later group of 4590 Korean school children and university students in Seoul and Taejuon were found to have a slow hemoglobin in addition to normal hemoglobin A. In all four subjects the slow component migrated at a rate characteristic of the G hemoglobin. The overall incidence of the variant in the present group of Koreans was low: 4 in 6700 or 0.06 percent. It appears significant that no insistence of hemoglobin E were found among these Korean subjects. Hemoglobin E has been found among numerous ethnic groups of Southeast Asia and the variant most frequently appeared in Chinese subject. By Urea-Starch-Gel Electrophoresis in Alkaline PH and A.I. Chernoff method was demonstrated that another 3 cases of abnormal hemoglobin also were beta-chain variants. This was reconfirmed by Hybridization Experiment with canine hemoglobin. And the results of family test of 3 case of abnormal hemoglobin were heterozygous carrier.
